Motion sickness and otolith asymmetry.

There is a highly scattered inter-individual susceptibility in man to motion sickness. It is discussed whether different masses of otoconias between right and left sides are responsible for a high susceptibility. In order to proof this theory, we measured the otoliths of fish (salmons, trouts, Xiphophorus Helleri; Sumatra barbes) and found big differences in the utricular stones up to 140%. The mass differences of the saccular stones were much smaller. In fish, showing abnormal swimming behavior during off-vertical axis rotation we found big mass differences compared to that of normal swimmers

Cardiovascular responses elicited by linear acceleration in humans.

Although activation of otolith receptors is known to elicit cardiovascular responses in animals, it is unclear whether vestibular stimulation can evoke changes in blood pressure and heart rate (which are independent of motion sickness) in humans. In the present study, ten normal subjects and three patients with profound bilateral reduction in vestibular function, who were seated upright with the torso aligned with the gravitation vector, were subjected to fore, aft, or lateral linear acceleration (approximately 0.2 g, attaining approximately 2 m/s in 900 ms, and decelerating for 3 s at 0.07 g)

Neurobiology of fish under altered gravity conditions.

In vertebrates (including man), an altered gravitational environment such as weightlessness can induce malfunction of the inner ear, based on an irregular dislocation of the otoliths from the corresponding sensory epithelia. This dislocation leads to an illusionary tilt, since the otolithic inputs are not in register with other sensory organs. This results in an intersensory conflict. Vertebrates in orbit therefore face severe orientation problems. In humans, the intersensory conflict may additionally lead to a malaise, commonly referred to as space motion sickness (SMS). During the first days

Role of otolith endorgans in the genesis of vestibular-visual conflict sickness (pitch) in the squirrel monkey (first report).

This experiment was designed to clarify each vestibular submodality's role in the genesis of vestibular-visual conflict sickness in pitch. We wanted to determine whether the ablation of the macula utriculi and macula sacculi, totally or separately, unilaterally and bilaterally, preserving the semicircular canals intact, could eliminate the conflict emesis in the squirrel monkey model. Observations were carried out over 5 months. In susceptible animals, the difference in conflict sickness scores between the pre- and the postutriculosacculectomy conditions was statistically significant. However,

Some properties of otoconia.

Otoconia are dynamic mineral deposits present in the gravity receptors of most vertebrates; fishes often have a single large mass called an otolith instead. Otoconia generally have the appearance of single crystals but contain organic and inorganic components, the mineral being almost exclusively a polymorph of calcium carbonate. The two phases are closely interrelated structurally. Ultra-high resolution transmission electron microscopy of rat otoconia showed them to be mosaic biominerals. The crystallites were 50-100 nm in diameter, had some rounded edges, and were highly ordered into laminae

Genetic advances in Meniere Disease.

Meniere Disease (MD) is an idiopathic inner ear disease with complex etiology and pathogenesis, which is still unclear. With the development in gene analysis technology, the genetic research of MD has attracted extensive attention, resulting in a large number of studies on the research of the relationship between human genes and MD. This paper aims to review the studies on this topic in recent years. The studies mainly focused on the genetics of familial MD and the correlation between MD and potentially related functional genes. The results of these studies have demonstrated the complexity and

Stereocilin gene variants associated with episodic vertigo: expansion of the DFNB16 phenotype.

Vestibular disorders comprise a heterogeneous group of diseases with transient or permanent loss of vestibular function. Vestibulopathy is in most cases associated with migraine, Ménière disease, hereditary ataxias, or sensorineural hearing loss. We identified two brothers and their first cousin affected by hearing loss and episodic vertigo. The brothers were homozygous STRC nonsense variant [c.4027 C > T, p.(Q1343*)], whereas their first cousin was compound heterozygous for the STRC nonsense variant and a 97 kb deletion spanning the entire STRC gene. Clinical investigations confirmed patholog

Evaluation of the Video Ocular Counter-Roll (vOCR) as a New Clinical Test of Otolith Function in Peripheral Vestibulopathy.

IMPORTANCE: Video-oculography (VOG) goggles have been integrated into the assessment of semicircular canal function in patients with vestibular disorders. However, a similar bedside VOG method for testing otolith function is lacking. OBJECTIVE: To evaluate the use of VOG-based measurement of ocular counter-roll (vOCR) as a clinical test of otolith function. DESIGN, SETTING, AND PARTICIPANTS: A case-control study was conducted to compare vOCR measurement among patients at various stages of unilateral loss of vestibular function with healthy controls. The real identity and sensory overlap mechanism of special vestibular afferent neurons that sense both rotation and linear force.

AIMS: Although the vestibular system has been widely investigated over the past 50 years, there is still an unsolved mystery. Some special vestibular afferent (SVA) neurons responding to both rotation and linear force were found through neurophysiological techniques, however, the sensory overlap mechanism of SVA neurons is still unclear, which may be closely related to vestibular-related diseases. MATERIALS AND METHODS: To address the above-mentioned problem, a cupula buoyancy theory was established in the present study, where SVA neurons were considered semicircular canal afferent (SCCA) neu

[The application of subjective visual gravity in assessment of vestibular compensation: a pilot study].

OBJECTIVE: To discuss the characteristics of subjective visual gravity (subjective visual vertical/horizontal, SVV/SVH) and assess its clinical application for peripheral unilateral vestibular compensation. METHODS: 69 cases of acute peripheral unilateral vestibular dysfunction patients (case group) accepted SVV/SVH, spontaneous nystagmus (SN), caloric test (CT) and other vestibular function tests. 49 healthy people (control group) accepted SVV/SVH only. SVV/SVH, SN and unilateral weakness (UW) were selected as for the observation indicators. The correlations between SVV/SVH, SN, UW and cours

Tetracycline marks visible in Baltic cod Gadus morhua otoliths stored for 40 years.

Baltic cod Gadus morhua otoliths marked with oxytetracycline (OTC) in 1974-1975 had been sectioned and the sections stored in paper envelopes at room temperature in dry, dark conditions. After 40 years of storage, the historic OTC marks were still clearly visible showing that OTC is suitable for long-term chemical marking of otoliths. They were, however, noticeably paler than marks in recently recaptured and archived Baltic G. morhua otoliths, chemically tagged with tetracycline at similar dosage to the historic otoliths.

The effect of otolith malformation on behavior and cortisol levels in juvenile red drum fish (Sciaenops ocellatus).

Captive-raised red drum fish were observed with phenotypic abnormalities, including deformities of the spine, jaw, and cephalic region, that were consistent with vitamin C deficiency during the larval stage. In light of their visible exterior skeletal abnormalities, we suspected that the affected fish would also have abnormal otoliths. Otoliths are dense calcareous structures that function in fish hearing. We hypothesized that abnormal fish would have irregular otoliths that would alter behavior and cortisol levels as compared with those of phenotypically normal fish. The normal and abnormal f

Evidence for vestibular dysfunction in orthostatic hypotension.

There is little definitive evidence of the clinical significance of the vestibular-cardiovascular reflex in humans, despite the fact that the vestibular system is known to contribute to cardiovascular control in animals. The present study involved 248 dizzy patients (127 male patients and 121 female patients) aged 65 years and younger. We classified all participants into three groups based on their vestibular evoked myogenic potential (VEMP) responses; absent VEMP, asymmetry VEMP and normal VEMP. To investigate the effect of the otolith disorder, which was estimated by the VEMP, on the orthost

Postural control of fish related to gravity input.

The disappearance of gravity input during space flight causes many functional disorders in animal and human postural control system. On the other hand, it has been shown that these functional deficits reduce or disappear with time and adapt to weightlessness. Unusual fish behavior was reported during the first few days of orbit flight and also during postflight recovery in Skylab 3 experiment. Similar fish behavior was observed in microgravity obtained during the parabolic flight of airplane. These results suggest that fish might provide a good model for investigating adaptation of sensory-mot

European vestibular experiments on the Spacelab-1 mission: 4. Thresholds of perception of whole-body linear oscillation.

Thresholds for the detection of linear oscillatory motion at 0.3 Hz in the X, Y and Z body axes were determined during the flight of Spacelab-1 and on the ground pre- and post-flight, using the method of limits with a single staircase procedure. Pre-flight, Z axis thresholds (mean 0.077 ms-2) were significantly higher than X and Y thresholds (mean 0.029 ms-2). Measures obtained on three crew members in-flight exhibited thresholds greater, by a factor of 1.5-4.3, than those obtained pre-flight. Post-flight, two crew members had significantly elevated X and Y axis thresholds whereas the other tw

Evaluation of the structure of the otoconial layer using micro-computed tomography.

OBJECTIVE: Estrogen deficiency caused by bilateral ovariectomy (OVX) has been reported to lead to morphological changes in otoconia. Thus, we examined the morphological changes in the otoconial layer after OVX. We also investigated whether micro-computed tomography (µCT) is useful for the detection of morphological changes in the otoconial layer. METHODS: The otic capsules of C57BL/6 J mice were removed and evaluated using histological techniques and µCT at 2, 4, and 8 weeks after OVX or sham surgery. The volume of the utricle otoconial layer was measured and compared between the OVX and sham

Direction-fixed paroxysmal nystagmus lateral canal benign paroxysmal positioning vertigo (BPPV): another form of lateral canalolithiasis.

Benign paroxysmal positioning vertigo (BPPV) is the most frequent vertiginous syndrome. It is caused either by free-floating otoliths in the semicircular canals (canalolithiasis) or by otoconial debris adhering to a canal cupula (cupulolithiasis). The posterior canal is the most frequently involved (80%), while the lateral canal is involved less frequently (15%), and the rarest conditions are anterior canalolithiasis and apogeotropic posterior canalolithiasis (5%). The main diagnostic sign of lateral canal BPPV is paroxysmal horizontal bidirectional positioning nystagmus evoked through Pagnini

Gentamicin alters nitric oxide production in semicircular canals and otolith organs.

OBJECTIVES/HYPOTHESIS: Intratympanic application of gentamicin is an important therapeutic option to control vertigo spells in Ménière's disease. Low doses eliminate the function of semicircular canal ampullae (SCCA) and saccule in most patients, although utricular function is maintained in many cases. Local alteration in free radical production might be responsible for these differences. Therefore, the gentamicin-induced nitric oxide (NO) production was determined in an animal model using separate organ cultures of SCCA, saccule, and utricle. Mental representation of space in vestibular patients with otolithic or rotatory vertigo.

In previous studies, we had observed that the occurrence of geometric illusions was reduced when healthy observers were tilted relative to gravity or placed in microgravity. We hypothesized that the alteration of the gravitational (otolith) input was responsible for this change, presumably because of a connection between vestibular and visual-spatial cognitive functions. In this study, we repeated these experiments in vestibular patients who presented signs of otolith disorders. In agreement with the microgravity data, geometric illusions based on horizontal, vertical, and oblique lines were l
