Afferent connections of the interstitial nucleus of the posterior limb of the anterior commissure and adjacent amygdalostriatal transition area in the rat.

The interstitial nucleus of the posterior limb of the anterior commissure is, like the striatum, very rich in tyrosine hydroxylase and acetylcholinesterase, but on the basis of most other neurochemical criteria displays features that are typical of the extended amygdala (Alheid, de Olmos and Beltramino, 1995). Its afferent connections were examined in the rat with retrograde (cholera toxin B subunit) and anterograde (Phaseolus vulgaris leucoagglutinin) tracers and compared to those of the neighboring amygdalostriatal transition area and central amygdaloid nucleus. Deposits of cholera toxin B s

Patterns of brainstem projection to the thalamic reticular nucleus.

To understand better how the brainstem may influence thalamocortical activity, we have examined the projection patterns of different brainstem nuclei to the thalamic reticular nucleus. Iontophoretic injections of biotinylated dextran were made into various nuclei of the brainstem (superior colliculus, periaqueductal grey matter, parabrachial nucleus, pedunculopontine tegmental nucleus, laterodorsal tegmental nucleus, substantia nigra, ventral tegmental area, and locus coeruleus) of Sprague-Dawley rats by using stereotaxic coordinates. Our results show that afferents from each brainstem nucleus

Neural circuitry mediating hypercapnia-induced microarousal and impaired sleep depth.

Sleep disruption in obstructive sleep apnea syndrome is characterized by frequent microarousals and reduced sleep depth, yet the underlying mechanisms remain unclear. Here, we show that intermittent hypercapnic air (HA) induces time-locked microarousals, fragments sleep, reduces non-rapid eye movement (NREM) sleep depth, and diminishes rapid eye movement (REM) sleep. HA exposure activates glutamatergic neurons in the nucleus tract solitary (NTSVglut2), and their optogenetic or chemogenetic activation is sufficient to trigger arousal and impair sleep. Projection-specific activation reveals that

A touch-guided neural circuit regulates motivated gnawing to maintain dental alignment.

How hindbrain circuits integrate peripheral and central signals to regulate complex oral behaviors is poorly understood. In rodents, gnawing is essential for localized tooth wear to offset lifelong incisor growth. Whether this process relies on specific sensory input to guide localized tooth wear and is actively regulated by neural mechanisms remains unresolved. Here, we identify somatostatin-expressing neurons in the spinal trigeminal nucleus oralis as a central relay distributing tactile input to motor execution and motivational circuits. These neurons receive input from a genetically distin

Thiazoline-related innate fear stimuli orchestrate hypothermia and anti-hypoxia via sensory TRPA1 activation.

Thiazoline-related innate fear-eliciting compounds (tFOs) orchestrate hypothermia, hypometabolism, and anti-hypoxia, which enable survival in lethal hypoxic conditions. Here, we show that most of these effects are severely attenuated in transient receptor potential ankyrin 1 (Trpa1) knockout mice. TFO-induced hypothermia involves the Trpa1-mediated trigeminal/vagal pathways and non-Trpa1 olfactory pathway. TFOs activate Trpa1-positive sensory pathways projecting from trigeminal and vagal ganglia to the spinal trigeminal nucleus (Sp5) and nucleus of the solitary tract (NTS), and their artificia

Mapping of afferent and efferent connections of phenylethanolamine N-methyltransferase-expressing neurons in the nucleus tractus solitarii.

OBJECTIVE: Phenylethanolamine N-methyltransferase (PNMT)-expressing neurons in the nucleus tractus solitarii (NTS) contribute to the regulation of autonomic functions. However, the neural circuits linking these neurons to other brain regions remain unclear. This study aims to investigate the connectivity mechanisms of the PNMT-expressing neurons in the NTS (NTSPNMT neurons). METHODS: The methodologies employed in this study included a modified rabies virus-based retrograde neural tracing technique, conventional viral anterograde tracing, and immunohistochemical staining procedures. RESULTS:

Brainstem sources of input to the central mesencephalic reticular formation in the macaque.

Physiological studies indicate that the central mesencephalic reticular formation (cMRF) plays a role in gaze changes, including control of disjunctive saccades. Neuroanatomical studies have demonstrated strong interconnections with the superior colliculus, along with projections to extraocular motor nuclei, the preganglionic nucleus of Edinger-Westphal, the paramedian pontine reticular formation, nucleus raphe interpositus, medullary reticular formation and cervical spinal cord, as might be expected for a structure that is intimately involved in gaze control. However, the sources of input to

A Brainstem-Thalamic-Cortex Glutamatergic Neuron Pathway Modulates Propofol Anesthesia.

BACKGROUND: The precise mechanisms underlying the modulation and transformation of consciousness during propofol anesthesia remain inadequately understood. Both the paraventricular thalamus (PVT)-medial prefrontal cortex (mPFC) pathway and the parabrachial nucleus (PB) are implicated in the regulation of sleep-wake cycles. Consequently, this study aimed to explore whether projections from the PB to the PVT-mPFC pathway are involved in the effects of propofol anesthesia. METHODS: Anterograde and retrograde viral tracing techniques were employed to map the projections from the PB to the PVT and

Striatal and central extended amygdala parts of the interstitial nucleus of the posterior limb of the anterior commissure: evidence from tract-tracing techniques in the rat.

The interstitial nucleus of the posterior limb of the anterior commissure (IPAC) lies at the junction of the striatopallidal system and the lateral bed nucleus of the stria terminalis-central amygdaloid nucleus continuum (i.e., the central extended amygdala; EAc). Its efferent connections were investigated in the rat with anterograde (Phaseolus vulgaris leucoagglutinin) and retrograde (Fluoro-Gold and cholera toxin B subunit) tracers and compared with those of the central amygdaloid nucleus. Our anterograde tracing experiments reveal that the projections of the medial IPAC largely reciprocate

Developmental roles of the spontaneous depolarization wave in synaptic network formation in the embryonic brainstem.

One of the earliest activities expressed within the developing central nervous system is a widely propagating wave-like activity, which we referred to as the depolarization wave. Despite considerable consensus concerning the global features of the activity, its physiological role is yet to be clarified. The depolarization wave is expressed during a specific period of functional synaptogenesis, and this developmental profile has led to the hypothesis that the wave plays some roles in synaptic network organization. In the present study, we tested this hypothesis by inhibiting the depolarization

The regulation of cardiovascular functions by monoamine neurotransmitters in the brain.

The complexity of the central nervous system's influence on cardiovascular integration is underscored by the extensive neural interconnections between the various higher and lower brain structures shown by electrophysiological studies to have an influence on cardiovascular function. Furthermore, complex neural connections exist within each level of cardiovascular integration (for instance, the medullary reticular formation, or the limbic system). The electrophysiological analysis of the integrative aspects of central cardiovascular regulation has been the subject of several reviews (Reis, 1972

A Tac1-Expressing Brainstem Pathway Underlies the Pathogenesis of Trigeminal Neuralgia.

Trigeminal neuralgia (TN) is widely recognized to be one of the most severe pain disorders, severely affects the life quality of patients. However, the neuronal mechanisms underlying TN pathogenesis remain largely unknown. Here, we identify a periphery-to-brain neural circuit that governs TN development in mice. Tachykinin 1 (Tac1)-expressing parabrachial nucleus (PBNTac1) neurons show heightened stimulus-evoked responses, and chemogenetic inhibition of these neurons effectively prevents TN development. Furthermore, PBNTac1 neurons receive projections from the caudal part of the spinal trigemi

Presence of S-100 beta in cholinergic neurones of the rat hindbrain.

The double staining of S-100 beta and choline acetyltransferase (ChAT) revealed that S-100 beta immunoreactivity was localized in most, but not in all, cholinergic neurones in the somatomotor nuclei of the cranial nerves and in the ambiguus nucleus. S-100 beta was present in almost all cholinergic neurones in the brain stem reticular, red, vestibular (excluding medial), mesencephalic trigeminal and cerebellar nuclei. However, S-100 beta immunoreactivity was lacking in cholinergic neurones in the parabrachial complex, the dorsal motor nucleus of the vagal nerve and most sensory nuclei. No S-100

Separation of Channels Subserving Approach and Avoidance/Escape at the Level of the Basal Ganglia and Related Brainstem Structures.

The basal ganglia have the key function of directing our behavior in the context of events from our environment and/or our internal state. This function relies on afferents targeting the main input structures of the basal ganglia, entering bids for action selection at the level of the striatum or signals for behavioral interruption at the level of the subthalamic nucleus, with behavioral reselection facilitated by dopamine signaling. Numerous experiments have studied action selection in relation to inputs from the cerebral cortex. However, less is known about the anatomical and functional link

Central control of autonomic function and involvement in neurodegenerative disorders.

The central autonomic network includes the insular cortex, anterior cingulate cortex, amygdala, hypothalamus, periaqueductal gray, parabrachial nucleus, nucleus of the solitary tract, ventrolateral reticular formation of the medulla, and medullary raphe. These areas: are reciprocally interconnected; receive converging visceral and somatosensory information; generate stimulus-specific patterns of autonomic, endocrine, and motor responses; and are regulated according to the behavioral state, including the sleep-wake cycle. Several components of the central autonomic networks are affected in neur

Significance of the paralemniscal tegmental area for audio-motor control in the moustached bat, Pteronotus p. parnellii: the afferent off efferent connections of the paralemniscal area.

The paralemniscal tegmental area has been determined in the brain of the New World moustached bat, Pteronotus p. parnellii, by electrical microstimulation eliciting echolocation calls and pinna movements. It is located in the dorsal tegmentum rostral and medial to the dorsal nucleus of the lateral lemniscus and is characterized by medium sized and large neurons. Tracer injections (WGA-HRP) showed that the most intense input to the paralemniscal tegmental area originates in the intermediate and deep layers of the homolateral superior colliculus. The strong projections from the ipsi- and contral

Circuit Mechanisms Underlying the Contribution of Leptin Receptor-Expressing Neurons in the Nucleus Tractus Solitarius to Ventilatory Homeostasis.

Sleep apnea syndrome (SAS) is a prevalent disorder characterized by recurrent respiratory pauses during sleep; however, the neural mechanisms governing respiratory stability remain poorly understood. In this study, we identify the ratio of sigh expiratory volume to eupneic expiratory volume as a potential predictor of post-sigh apnea in susceptible C57BL/6J mice. We demonstrate that leptin signaling within the nucleus tractus solitarius (NTS) is critical for maintaining respiratory drive and suppressing apnea. Chemogenetic activation of Leptin receptor b-expressing NTS (NTSLepRb) neurons signi

New data on the immunocytochemical localization of thyrotropin-releasing hormone in the rat central nervous system.

An antiserum raised against the synthetic tripeptide pyroglutamyl-histidyl-proline (free acid) was used to localize thyrotropin-releasing hormone (TRH) in the rat central nervous system (CNS) by immunocytochemistry. The distribution of TRH-immunoreactive structures was similar to that reported earlier; i.e., most of the TRH-containing perikarya were located in the parvicellular part of the hypothalamic paraventricular nucleus, the suprachiasmatic portion of the preoptic nucleus, the dorsomedial nucleus, the lateral basal hypothalamus, and the raphe nuclei. Several new locations for TRH-immunor

Learning in respiratory control.

In this article, it is argued that learning participates to fulfill the metabolic requirements by adapting respiratory control to changing internal and external states. Recent classical-conditioning experiments in newborn mice or adult rats show the close link between conditioned respiratory and arousal responses. The conditioned fear model may be a suitable and largely unexplored model of emotionally induced hyperventilation. The parabrachial nucleus and periacqueducal grey may play a pivotal role in the ventilatory component of conditioned fear. The sensitivity of breathing to conditioning i

Immunoreactivity and distribution of Fos in the mouse brain following diverse general anesthetics.

General anesthesia induces a range of effects, including unconsciousness, analgesia, amnesia, and immobility. Despite its widespread use for over a century, the underlying mechanisms remain incompletely understood. This study aims to investigate whether different intravenous anesthetics share common neuronal targets. Propofol (180 mg/kg), midazolam (75 mg/kg), and esketamine (250 mg/kg) were intraperitoneally injected in C57BL/6J mice, respectively. After one hour of anesthesia, mice were transcardially perfused, and their brains were examined for Fos protein localization to assess changes in
