Different lipid A types in lipopolysaccharides of phototrophic and related non-phototrophic bacteria.

Lipid A analyses confirm not only the present taxa of the purple nonsulfur bacteria (formerly Rhodospirillaceae), but also phylogenetical relatedness of distinct phototrophic to distinct non-phototrophic bacteria, as was suggested by cataloguing 16S rRNA. For example, lipid A with ester-bound 3-OH-10:0 and the rare amide-linked 3-oxo-14:0 is common to the phototrophic Rhodobacter capsulatus and Rhodobacter sphaeroides and also to Paracoccus denitrificans and Thiobacillus versutus. 'Lipid ADAG' (lipid A with 2,3-diamino-D-glucose (DAG)) occurs in the phototrophic Rhodopseudomonas viridis and Rh

Comparison of Different Sites of Microbiota Shows Increased Pseudomonas Species in the Gut Mucosa in Inflammatory Bowel Disease.

BACKGROUND: Inflammatory bowel disease (IBD), including ulcerative colitis (UC) and Crohn's disease (CD), is associated with changes in the gut microbiome. Studies comparing fecal, gut mucosal, and salivary microbiomes are rare, and questions remain regarding the interaction of these compartments. METHODS: In this case-control study, 16S rRNA gene amplicon sequencing was performed on samples from stool, intestinal mucosa, and saliva of 120 patients with IBD. Patients with signs of non-IBD colonic inflammation (N = 28) and healthy subjects (N = 67) served as controls. A total of 480 16S profil

Exogenous compound bacteria enhance the nutrient removal efficiency of integrated bioremediation systems: Functional genes and microorganisms play key roles.

With the continuous development of intensive mariculture, the application of the integrated bioremediation system of aquaculture wastewater (IBSAW) is increasingly promoted. However, the process and nutrients removal performance of the IBSAW need to be further optimized due to its immature technologies. In this study, exogenous compound bacteria (ECB) were added to IBSAW to investigate its pollutants removal efficiency and the relevant mechanisms. High-throughput sequencing and Geochip gene array were used to analyze the correlation between nutrients and bacteria, and the abundance of N and P

Bacterial diversity obtained by culturable approaches in the gut of Glossina pallidipes population from a non sleeping sickness focus in Tanzania: preliminary results.

BACKGROUND: Glossina pallidipes is a haematophagous insect that serves as a cyclic transmitter of trypanosomes causing African Trypanosomiasis (AT). To fully assess the role of G. pallidipes in the epidemiology of AT, especially the human form of the disease (HAT), it is essential to know the microbial diversity inhabiting the gut of natural fly populations. This study aimed to examine the diversity of G. pallidipes fly gut bacteria by culture-dependent approaches. RESULTS: 113 bacterial isolates were obtained from aerobic and anaerobic microorganisms originating from the gut of G. pallidipes

Genome-resolved metagenomics reveals lineage-specific phylogenomic and functional differentiation among carbon-processing bacteria from mangrove and terrestrial forests.

Microbial carbon processing in mangrove and terrestrial forests remains poorly resolved at the level of individual bacterial genomes. Here, shotgun metagenomics was used to compare bacterial communities and genome-encoded carbon-processing potential across mangrove sediments, terrestrial forest soils, and associated litter from Goa, India. Genome reconstruction recovered 15 bacterial metagenome-assembled genomes (MAGs), including eight phylogenomically distinct, species-unresolved lineages identified through genome-based taxonomic and relatedness analyses. Community profiles broadly overlapped

Resolving the electron bottleneck in denitrification for N2O mitigation: The roles of algal-mediated metabolic coupling and extracellular electron shuttling networks.

Excessive nitrogen (N) discharge from agriculture and industry causes severe ecological harm. Moreover, conventional nitrogen removal processes often generate N2O, adding to the environmental burden. While algal-bacterial consortium has emerged as a low-carbon alternative to address these issues, the bioenergetic mechanisms governing their N2O-mitigation performance remain poorly understood. In this study, a Chlorella sorokiniana-Paracoccus denitrificans consortium was established to clarify the mechanism underlying enhanced N₂O mitigation. The consortium achieved superior total nitrogen remov

Enterobacter sp. HIT-SHJ4 isolated from wetland with carbon, nitrogen and sulfur co-metabolism and its implication for bioremediation.

Both autotrophic and heterotrophic denitrification are known as important bioprocesses of microbe-mediated nitrogen cycle in natural ecosystems. Actually, mixotrophic denitrification co-driven by organic matter and reduced sulfur substances are also common, especially in hypoxic environments such as estuarine sediments. However, carbon, nitrogen and sulfur co-metabolism during mixotrophic denitrification in natural water ecosystems has rarely been reported in detail. Therefore, this study investigated the co-metabolism of carbon, nitrogen and sulfur using samples collected from four distinct n

Intrinsic uncoupling of cytochrome c oxidase may cause the maternally inherited mitochondrial diseases MELAS and LHON.

Mutations in the human mtDNA gene encoding subunit III of cytochrome c oxidase (CO) have been reported to cause MELAS and LHON. Poracoccus denitrificans cells expressing substitutions homologous to these MELAS- and LHON-causing mutations had lower growth yield than wild type cells and lower efficiency of proton pumping by CO (e.g. lower H+/e ratio and lower deltapsi), but had similar CO activity. These results indicate that both substitutions (F263L > A212T) cause intrinsic uncoupling, which may be the direct cause of the diseases. These results also suggest that subunit III is involved in pro

Identification of microbial agents in tissue specimens of ocular and periocular sarcoidosis using a metagenomics approach.

Background: Metagenomic sequencing has the potential to identify a wide range of pathogens in human tissue samples. Sarcoidosis is a complex disorder whose etiology remains unknown and for which a variety of infectious causes have been hypothesized. We sought to conduct metagenomic sequencing on cases of ocular and periocular sarcoidosis, none of them with previously identified infectious causes. Methods: Archival tissue specimens of 16 subjects with biopsies of ocular and periocular tissues that were positive for non-caseating granulomas were used as cases. Four archival tissue specimens that

Unveiling the impact of flooding and salinity on iron oxides-mediated binding of organic carbon in the rhizosphere of Scirpus mariqueter.

The abundant Fe (hydr-) oxides present in wetland sediments can form stable iron (Fe)-organic carbon (OC) complexes (Fe-OC), which are key mechanisms contributing to the stability of sedimentary OC stocks in coastal wetland ecosystems. However, the effects of increased flooding and salinity stress, resulting from global change, on the Fe-OC complexes in sediments remain unclear. In this study, we conducted controlled experiments in a climate chamber to quantify the impacts of flooding and salinity on the different forms of Fe (hydr-) oxides binding to OC in the rhizosphere sediments of S. mari

Distribution of microbiota in cervical preneoplasia of racially disparate populations.

BACKGROUNDS: Microbiome dysbiosis is an important contributing factor in tumor development and thus may be a risk predictor for human malignancies. In the United States, women with Hispanic/Latina (HIS) and African American (AA) background have a higher incidence of cervical cancer and poorer outcomes than Caucasian American (CA) women. METHODS: Here, we assessed the distribution pattern of microbiota in cervical intraepithelial neoplasia (CIN) lesions obtained from HIS (n = 12), AA (n = 12), and CA (n = 12) women, who were screened for CC risk assessment. We employed a 16S rRNA gene sequenci

Deep 16S rDNA Sequencing of Chronic Subdural Hematomas Suggests Involvement of Bacterial Infection in Recurrences.

BACKGROUND AND OBJECTIVES: Chronic subdural hematoma (CSDH) is an encapsulated blood collection between the dura mater and arachnoid that often follows mild head trauma. It involves pronounced inflammation and angiogenesis, and recurrence remains common despite advances in surgical management. Subclinical bacterial involvement has been hypothesized as a potential factor in these recurrent cases. We aimed to determine whether bacteria are present in CSDH outer membranes and to define accompanying histological and transcriptomic changes. METHODS: A total of 39 Formalin-fixed paraffin-embedded o

Unveiling the skin microbial guardians and assailants in psoriasis subtypes: a Mendelian randomization study.

Previous studies have shown an association between skin microbiota and psoriasis, but the causal relationship between skin microbiota and different psoriasis subtypes remains unexplored. Our study employed the Mendelian randomization (MR) method, using summary statistics from Genome-Wide Association Studies, to investigate the causal relationships between skin microbiota and different subtypes of psoriasis. The inverse variance weighted (IVW) method served as the primary method in our MR study. The MR-Egger intercept test, Cochran's Q test, MR-PRESSO test, and leave-one-out analysis were condu

Effects of parity, blood progesterone, and non-steroidal anti-inflammatory treatment on the dynamics of the uterine microbiota of healthy postpartum dairy cows.

This study evaluated the effects of treatment with meloxicam (a non-steroidal anti-inflammatory drug), parity, and blood progesterone concentration on the dynamics of the uterine microbiota of 16 clinically healthy postpartum dairy cows. Seven primiparous and 9 multiparous postpartum Holstein cows either received meloxicam (0.5 mg/kg SC, n = 7 cows) once daily for 4 days (10 to 13 days in milk (DIM)) or were untreated (n = 9 cows). Endometrial cytology samples were collected by cytobrush at 10, 21, and 35 DIM, from which the microbiota analysis was conducted using 16S rRNA gene sequence analys

Integration of •SO4--based AOP mediated by reusable iron particles and a sulfidogenic process to degrade and detoxify Orange II.

The sulfate radical (•SO4-)-based advanced oxidation processes (AOPs) for the degradation of refractory organic pollutants consume a large amount of persulfate activators and often generate toxic organic by-products. In this study, we proposed a novel iron-cycling process integrating •SO4--based AOP mediated by reusable iron particles and a sulfidogenic process to degrade and detoxify Orange II completely. The rusted waste iron particles (Fe0@FexOy), which contained FeII/FeIII oxides (FexOy) on the shell and zero-valent iron (Fe0) in the core, efficiently activated persulfate to produce •SO4-
