Differentiating between parthenogenetic and "positive development" embryos in turkeys by molecular sexing.

In mated or inseminated turkeys, 5 to 15% of eggs set for incubation show only rudimentary development. Most of these embryos die during the first 24 to 48 h of incubation and contain only unorganized sheets of tissue. This abnormal development is termed "positive development" (PD). Turkey eggs also show incidence of parthenogenesis and the resulting progeny is believed to be always male. As both types of embryos are morphologically similar at the early stage of incubation, it has been speculated that PD embryos may in fact be parthenogens. By identifying the sex at the blastodermal stage with

ENDO A cytokeratin expression in the inner cell mass of parthenogenetic mouse embryos.

During the preimplantation period of development, the first cellular polarization and diversification of the mouse embryo occurs. This process starts at the eight-cell stage and is directly driven by the cytoskeleton. Cell polarization finally leads to the first embryonic epithelium, the trophectoderm, characterized by the presence of cytokeratins. It has not been described whether genomic imprinting, an epigenetic modification of certain genes depending on the parent-of-origin, affects preimplantation development. However, implantation is one of the steps in which an exceptionally high mortal

Reproduction and Wing Differentiation of Gynoparae Are Regulated by Juvenile Hormone Signaling in Aphis gossypii.

Gynopara is a specific winged type in the life history of Aphis gossypii. As a key reproduction mode between parthenogenesis and sexual reproduction, it lays sexual females in late autumn. However, little is known about gynoparae because of its rare presence in the wild and the lack of its detailed descriptions. In this study, we investigated the morphological characteristics, ovary maturation process, and key signaling pathways during the development of gynoparae of A. gossypii. With the extension of development stage, the gynoparae exhibited gradually enlarged and thickened abdomen, deepened

Promising Role of Wolbachia as Anti-parasitic Drug Target and Eco-Friendly Biocontrol Agent.

BACKGROUND: Wolbachia is the most common endosymbiotic bacteria in insectborne parasites and it is the most common reproductive parasite in the world. Wolbachia has been found worldwide in numerous arthropod and parasite species, including insects, terrestrial isopods, spiders, mites and filarial nematodes. There is a complicated relationship between Wolbachia and its hosts and in some cases, they create a mutual relationship instead of a parasitic relationship. Some species are not able to reproduce in the absence of infection with Wolbachia. Thus, the use of existing strains of Wolbachia bac

First Detection of Wolbachia in Namibian Bird Ectoparasites (Acariformes: Syringophilidae) with a Description of New Quill Mite Species.

Wolbachia is a common intracellular bacterial genus that infects numerous arthropods and filarial nematodes. In arthropods, it typically acts as a reproductive parasite, leading to various phenotypic effects such as cytoplasmic incompatibility, parthenogenesis, feminization, or male-killing. Quill mites (Acariformes: Syringophilidae) are a group of bird parasites that have recently attracted increasing interest due to the detection of unique phylogenetic lineages of endosymbiotic bacteria and potentially pathogenic taxa. Our study used an unbiased 16S rRNA gene amplicon sequencing approach to

Effect of extracellular vesicles derived from oviductal and uterine fluid on the development of porcine preimplantation embryos.

To improve the efficiency of in-vitro-produced (IVP) porcine embryos, we focused on the events that usually occur during in-vivo embryonic transit from the oviduct to the uterus. Extracellular vesicles (EVs) are released by different mammalian cells and are imperative for intercellular communication and reflect the cell's physiological state. Based on these characteristics, EVs were isolated from oviductal and uterine fluid to imitate the in vivo environment and improve the efficiency of IVP embryos. Parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) embryos were divided into four g

Utilization of porcine in vitro-produced parthenogenetic embryos for co-transfer with vitrified and warmed embryos.

The present study was conducted to evaluate the possibility of using in vitro-produced parthenogenetic (PA) embryos for co-transfer with morulae that had been collected in vivo and cryopreserved. The proportion of PA blastocysts (20.5%) was higher than that of their in vitro fertilization (IVF) counterparts (16.6%). Although there were no differences in morphology or diameter between the two groups, the number of cells in early PA blastocysts after in vitro culture for 6 days was lower than for IVF blastocysts (25.7 and 30.4 cells, respectively), and the number in recovered PA blastocysts was

Porcine oocyte activation: differing roles of calcium and pH.

Intracellular pH has recently been shown to increase during parthenogenetic activation of the porcine oocyte. In the following set of experiments, intracellular pH was monitored during activation and pronuclear development was assessed following activation treatments with calcium, in the absence of calcium, and in oocytes loaded with the calcium chelator BAPTA-AM in calcium-free medium. Intracellular pH increase was not different among groups when treating with 7% ethanol or 50 microM calcium ionophore, or during treatment with thimerosal for 12 or 25 min. Activation with thimerosal (200 micro

[The effect of in-situ nerve growth factor from different biological sources on the reinitiation of mouse oocyte meiotic maturation in culture and on parthenogenetic activation].

We studied the capacity of mouse oocytes to complete meiotic maturation in vitro and form the female pronucleus upon parthenogenetic activation by cycloheximide, in response to a single injection into the mouse ovaries in situ of a purified fraction of 2.5 S NGF from mouse submaxillary glands and beta-NGF from bovine sperm. Injection of NGF from both sources at 10 ng/ml with subsequent incubation of the ovaries for 1 h increased the capacity of matured oocytes for parthenogenetic formation of the pronucleus. The frequency of pronucleus formation in both "naked oocyte" and oocytes surrounded by

Urolithin A Enhances the Development of Porcine Parthenogenetic Embryos by Promoting Mitochondrial Function and Quantity Through the SIRT1/PGC-1α Signaling Pathway.

Maintaining mitochondrial function and quantity is important for preimplantation embryo development, as they are essential for the regulation of oxidative stress. Urolithin A (UA), a metabolite produced from ellagitannins by gut bacteria, is known to exert antioxidant effects by improving mitochondrial function and biogenesis. Therefore, this study aimed to investigate the effects of UA on preimplantation embryo development and its mitochondrial function and quantity. In this study, EX-527, a specific inhibitor of sirtuin 1 (SIRT1), was used to explore whether the effects of UA are mediated by

Flexible oocyte manipulation with delayed maturation and improved somatic cell nuclear transfer efficiency using induced pluripotent stem cells.

Somatic cell nuclear transfer (SCNT) remains inefficient, limiting its practical use in cattle reproduction and research. This study investigated two complementary strategies to enhance handmade cloning (HMC): (i) holding bovine oocytes overnight and delaying maturation to enable a second round of SCNT, and (ii) using bovine-induced pluripotent stem cells (biPSCs) as donor nuclei for pluripotent stem cell nuclear transfer (PSCNT) to enhance developmental competence. Bovine oocytes were subjected to either conventional in vitro maturation (CONV; 20 h) or delayed maturation using a holding mediu

Effect of Ethanol on Parthenogenetic Activation and α-Tocopherol Supplementation during In Vitro Maturation on Developmental Competence of Summer-Collected Bovine Oocytes.

The use of α-tocopherol during in vitro maturation (IVM) is an alternative to minimize the adverse effects of heat stress on oocyte competence. However, α-tocopherol is diluted in ethanol, which can induce oocyte parthenogenetic activation (PA). This study aimed to evaluate the role of ethanol concentration on PA and the effect of α-tocopherol supplementation during IVM on the developmental competence and the expression of key genes in blastocysts derived from summer-collected oocytes. All in vitro embryo production was conducted at 5% O2, 5% CO2 at 38.5 °C. Experiment 1: oocytes were cultured

Development of in-vitro maturation protocol for rat oocytes; under simple culture vs co-culture with cumulus cell monolayer and its developmental potential via Parthenogenetic/artificial activation.

BACKGROUND: Murine is the most abundantly used as laboratory animal models. There has been a tremendous amount of research including; their evolution, growth, physiology, disease modeling as well as genomic mapping. Rats and mice are the most widely used among them. Although both rats and mice fall under the same category still both are different a lot too. As regarding in vitro maturation and development mouse studies are well established as compared to rats which still lies in the early phase of development. So, we tried to figure out rat oocytes in vitro maturation and their developmental p

Multiple origins and rapid evolution of duplicated mitochondrial genes in parthenogenetic geckos (Heteronotia binoei; Squamata, Gekkonidae).

Accumulating evidence for alternative gene orders demonstrates that vertebrate mitochondrial genomes are more evolutionarily dynamic than previously thought. Several lineages of parthenogenetic lizards contain large, tandem duplications that include rRNA, tRNA, and protein-coding genes, as well as the control region. Such duplications are hypothesized as intermediate stages in gene rearrangement, but the early stages of their evolution have not been previously studied. To better understand the evolutionary dynamics of duplicated segments of mitochondrial DNA, we sequenced 10 mitochondrial geno

Observations on ploidy of cells and on reproductive performance in parthenogenetic turkeys.

Using flow cytometry, the ploidy levels of parthenogenetic turkeys were quantified from blastodisc stage to adulthood. Eggs were collected from noninseminated hens of the Beltsville Small White flock, known for their high degree of parthenogenesis, and the blastodermal cells from developing embryos were compared with those of embryos produced by hens inseminated with semen from males of the same flock. Erythrocytes of parthenogens from Day 10 of incubation to 27 mo of age were also used for ploidy determination. Sperm and erythrocyte preparations from normal males of the above flock served as

Microbial diversity and cytoplasmic incompatibility in Tetranychus urticae Spanish populations.

The two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is an important citrus pest worldwide. Global populations of T. urticae harbour Wolbachia and other bacterial species, some of which are considered endosymbionts. The genus Wolbachia encompasses strict intracellular bacteria that cause several types of reproductive distortions in their hosts, such as male feminization, thelytokous parthenogenesis, cytoplasmic incompatibility, or male killing. The presence of different Wolbachia strains can affect the mite's reproductive parameters, which can have important implicatio
