Generalizable anchor aptamer strategy for loading nucleic acid therapeutics on exosomes.

Clinical deployment of oligonucleotides requires delivery technologies that improve stability, target tissue accumulation and cellular internalization. Exosomes show potential as ideal delivery vehicles. However, an affordable generalizable system for efficient loading of oligonucleotides on exosomes remain lacking. Here, we identified an Exosomal Anchor DNA Aptamer (EAA) via SELEX against exosomes immobilized with our proprietary CP05 peptides. EAA shows high binding affinity to different exosomes and enables efficient loading of nucleic acid drugs on exosomes. Serum stability of thrombin inh

Microprobes for Label-Free Detection of Short Tandem Repeats: An Insight into Alleviating Secondary Structure Effects.

Overgrowth of short tandem repeat sequences in our genes can cause various neurodegenerative disorders. Such repeat sequences are ideal targets for the label-free electrochemical detection of such potential expansions. However, their length- and sequence-dependent secondary structures may interfere with the interfacial charge transfer of a detection platform, making them complex targets. In addition, the gene contains sporadic repeats that may result in false-positive signals. Therefore, it is necessary to design a platform capable of mitigating these effects and ultimately enhancing the speci

Intractable mixtures and the origin of life.

Attempts to model the spontaneous chemistry which presumably preceded the origin of life on Earth commonly result in the production of intractably complex mixtures of organic compounds. It is, therefore, difficult to understand how any kind of evolutionary process might have begun. A number of potential solutions to this well-known and frustrating problem have been offered in the literature over the years. The present contribution briefly reviews and evaluates some of the more promising possibilities.

Metabolomics-based reveals the relation between internal metabolism and storage quality of sesame harvested by machine at different speeds.

Mechanized harvesting of sesame seeds reduces costs and enhances efficiency, but improper methods induce mechanical damage that degrades seed storage quality. This study represented the first time to characterize the effects of machine-harvested speeds on the storage quality of sesame using a metabolomics-based approach. The results showed that the oxidation stability and seed coat color of machine-harvested sesame was better when harvested at an appropriate speed (2000-2200 r/min). Moreover, 1445 and 2075 metabolites were identified in positive ion mode and negative ion mode, respectively, in

Noninvasive insulin delivery: the great potential of cell-penetrating peptides.

Insulin, a potent therapeutic peptide used in the treatment of diabetes, is administered to patients via subcutaneous injections because of the poor pharmacokinetics associated with alternative routes of administration such as oral, nasal and pulmonary delivery. Noninvasive nasal and oral formulations are appealing to patients who need consecutive daily treatments of insulin. However, to achieve mucosal absorption of insulin via oral or nasal administration, two barriers must be overcome: the impermeability of insulin through the epithelial membranes and local digestion and enzymatic degradati

Cell-Penetrating Peptide Conjugates of Steric Blocking Oligonucleotides as Therapeutics for Neuromuscular Diseases from a Historical Perspective to Current Prospects of Treatment.

The review starts with a historical perspective of the achievements of the Gait group in synthesis of oligonucleotides (ONs) and their peptide conjugates toward the award of the 2017 Oligonucleotide Therapeutic Society Lifetime Achievement Award. This acts as a prelude to the rewarding collaborative studies in the Gait and Wood research groups aimed toward the enhanced delivery of charge neutral ON drugs and the development of a series of Arg-rich cell-penetrating peptides called Pip (peptide nucleic acid/phosphorodiamidate morpholino oligonucleotide [PNA/PMO] internalization peptides) as conj

N-Terminal Lipidation of Buforin II Generates a Potent and Stable Peptide Antibiotic.

Buforin II is a broad-spectrum antimicrobial peptide (AMP) that kills bacteria by targeting intracellular nucleic acids. Unlike most membrane-acting AMPs, its unique antibacterial mechanism reduced its cytotoxicity. Nonetheless, its weak antibacterial potency and poor proteolytic stability still limit its clinical translation. Considering its helix-hinge-helix structure, lipidations were performed on its N-terminal, generating 5 different analogs. By screening the antibacterial and hemolytic activities, BUFT-2C8 with branched octanoic acid tails was identified as the optimal candidate. Its inc

Nanoparticles encapsulating antigenic peptides induce tolerogenic dendritic cells in situ for treating systemic lupus erythematosus.

Using Tetrahedral framework nucleic acids, we combined antigenic peptides to create the "DART" vaccine: DNA framework-Antigenic peptide-RNA modification-Targeting aptamer coupling. Generating antigen-specific tolerogenic dendritic cells (tolDCs), for systemic lupus erythematosus (SLE) is a potential therapeutic strategy for addressing compromised autoimmune tolerance. However, simple antigenic peptides degrade easily, lack specificity for delivery to dendritic cells (DCs), and cannot transform DCs to tolDCs. Therefore, this study aims to employ DART to generate tolDCs and compare DART-treated

Antigen transfer and its effect on vaccine-induced immune amplification and tolerance.

Antigen transfer refers to the process of intercellular information exchange, where antigenic components including nucleic acids, antigen proteins/peptides and peptide-major histocompatibility complexes (p-MHCs) are transmitted from donor cells to recipient cells at the thymus, secondary lymphoid organs (SLOs), intestine, allergic sites, allografts, pathological lesions and vaccine injection sites via trogocytosis, gap junctions, tunnel nanotubes (TNTs), or extracellular vesicles (EVs). In the context of vaccine inoculation, antigen transfer is manipulated by the vaccine type and administratio

Modified oligonucleotides as therapeutic and diagnostic agents.

Modified oligonucleotides and their analogs represent an exciting new class of agents with potential therapeutic and diagnostic applications. Among the most extensively investigated analogs are oligonucleoside phosphorothioates, some of which incorporate 'hairpin' structures at the 3' end and 'chimeras' that bear two modified oligonucleotides, all potential therapeutic candidates. To further enhance their potential, bio-reversible analogs of oligonucleotides have been synthesized. Several isosteric analogs of phosphoric diester oligonucleotides with neutral achiral backbones, and other novel m

Understanding the Interactions of Guanine Quadruplexes with Peptides as Novel Strategies for Diagnosis or Tuning Biological Functions.

Guanine quadruplexes (G4s) are nucleic acid structures exhibiting a complex structural behavior and exerting crucial biological functions in both cells and viruses. The specific interactions of peptides with G4s, as well as an understanding of the factors driving the specific recognition are important for the rational design of both therapeutic and diagnostic agents. In this review, we examine the most important studies dealing with the interactions between G4s and peptides, highlighting the strengths and limitations of current analytic approaches. We also show how the combined use of high-lev

Circulating microRNAs in prostate cancer - Non-invasive biomarkers for diagnosis, prognosis and therapy: A review.

Prostate cancer (PC) is a common malignancy driven by interacting genetic, environmental, and lifestyle factors, including hereditary mutations (BRCA1/2, HPC1, AR variants), premalignant lesions [proliferative inflammatory atrophy (PIA), prostatic intraepithelial neoplasia (PIN)], and Western dietary patterns. This narrative review aims to synthesize evidence on the role of microRNAs (miRNAs) in PC pathogenesis and clinical management across diagnosis, prognosis, therapy, and recurrence prediction. We searched PubMed/MEDLINE (2004-present) using predefined terms, screened reference lists, excl

Technical Note: A novel method for simultaneous recovery of DNA, RNA, and proteins from trace biological samples for forensic application.

There is growing interest in recovering analytes other than DNA from evidentiary samples. mRNA markers can help identify body fluids, link body fluid to a contributor via coding SNPs, and estimate Post-Mortem Interval. Proteins, which in many cases are more abundant and resistant to degradation than RNA and DNA, offer the possibility for human individualization via Genetically Variant Peptides and are also useful for body fluid identification, but are routinely destroyed during nucleic acid extraction. We report a novel trace sample recovery method which uses paramagnetic beads conjugated with

AI-designed PNA-peptide chimera overcomes suboptimal binding for dual inhibition of viral RdRp.

The chimera combining the peptide nucleic acids (PNAs) and peptides represent a promising bifunctional strategy by concurrently binding with protein catalytic pocket and its associated RNA template, effectively disrupting protein's function. Conventional designs face challenges due to non-optimized peptide-protein interactions and empirical PNA sequence selection, which lack thermodynamic or computational refinement, compromising selectivity and efficacy. Here, we present an artificial intelligence (AI) and molecular simulations-driven framework for the de novo design of a high-affinity PNA-pe

Ultra-sensitive detection of microRNAs using an electrochemical biosensor based on a PNA-DNA2 three-way junction nanostructure and dual cascade isothermal amplification.

BACKGROUND: MiRNA expression profiles may serve as valuable biomarkers for early cancer diagnosis. However, the detection of miRNA remains greatly challenging due to its shorter length and lower abundance in cells. Electrochemical biosensors based on DNA nanostructures have attracted significant attention due to their improved capture efficiency, high sensitivity, and ease of miniaturization, but the complex construction process, non-specific interactions among DNA probes, and their low stability continue to severely restrict their widespread application in clinical diagnostics. Therefore, dev

An XNA-CRISPR Platform for PAM-Free Detection of the katG Ser315Thr Mutation Associated with Isoniazid Resistance in Tuberculosis.

Drug-resistance Tuberculosis (TB) is a major threat to global public health. Simple and precise detection of the mutations associated with drug resistance is still a significant challenge. CRISPR-based nucleic acid detection has emerged as a transformative technology in molecular diagnostics due to its high specificity and programmability. However, the CRISPR-Cas12a system requires a protospacer adjacent motif (PAM) in the target double-stranded DNA (dsDNA) for efficient recognition and activation, which limits its application for detecting single-nucleotide variants (SNVs). To overcome this l

Non-Invasive Preimplantation Genetic Testing: Cell-Free DNA Detection in Embryo Culture Media Using a Plasmonic Biosensor.

In vitro fertilization (IVF) faces challenges in evaluating embryo quality and in determining the genetic health of embryos. Key biomarkers in the culture medium, including nucleic acids and proteins, offer promising avenues for noninvasive assessment. However, small sample volumes, low biomolecule concentrations, and potential contaminants complicate the reliable detection of genetic indicators. In this context, we developed a noninvasive preimplantation genetic testing (niPGT) approach using a superparamagnetic particles-enhanced surface plasmon resonance (SPR) imaging biosensor capable of d

Design, Synthesis and Biophysical Characterisation of CPP-PNA Conjugates for Targeting Oncogenic microRNA-221.

Peptide nucleic acids (PNAs) are a promising platform targeting disease-associated microRNAs due to their high binding affinity, excellent sequence specificity and resistance to enzymatic degradation. However, their therapeutic application is often limited by inefficient cellular uptake. In this study, we present the design, synthesis and physicochemical characterisation of cell-penetrating peptide (CPP)-PNA conjugates that target the oncogenic microRNA miRNA-221-5p, as well as their preliminary biological evaluation. Anti-miRNA-221-5p PNAs functionalised for conjugation with CPPs via Cu(I)-ca

High-Resolution HPLC for Separating Peptide-Oligonucleotide Conjugates.

Peptide-oligonucleotide conjugates (POCs) are chimeric molecules that combine the specificity of oligonucleotides with the functionality of peptides, improving the delivery and therapeutic potential of nucleic acid-based drugs. However, the analysis of POCs, particularly those containing arginine-rich sequences, poses major challenges because of aggregation caused by electrostatic interactions. In this study, we developed an optimized high-performance liquid chromatography (HPLC) method for analyzing POCs. Using a conjugate of DNA and nona-arginine as a model compound, we systematically invest
