There are different molecular pathways that regulate appetite, particularly the role of the hypothalamus, circadian rhythms, and gastrointestinal peptides. The hypothalamus integrates signals from orexigenic peptides like neuropeptide Y (NPY) and agouti-related protein (AgRP), which stimulate appetite, and anorexigenic peptides such as pro-opiomelanocortin (POMC) and cocaine- and amphetamine-regulated transcript (CART), which promote satiety. These signals are influenced by peripheral hormones like leptin, ghrelin, insulin, and cortisol, as well as gut peptides including glucagon-like peptide-

BACKGROUND: Both orosensory stimulation and feedback from the gastrointestinal tract contribute to energy intake regulation.
OBJECTIVE: We evaluated the hypothesis that overweight or obese subjects would be less sensitive to both oral and intraduodenal oleic acid exposure than would lean subjects.
DESIGN: Eleven overweight or obese and 8 lean men were studied on 2 occasions, during which antropyloroduodenal pressures, plasma cholecystokinin and peptide YY, and appetite were measured during 90-min intraduodenal infusions of saline or oleic acid (18:1 load: 0.78 kcal/min); energy intake (buffe

A host of gastrointestinal (GI) peptides influence the regulation of vital functions, such as growth, appetite, stress, gut motility, energy expenditure, digestion and inflammation, as well as glucose and lipid homeostasis. Hence, impairments in the synthesis/secretion of glucagon-like peptide-1 (GLP-1), leptin, nesfatin-1, glucose-dependent insulinotropic peptide (GIP), ghrelin (acylated and unacylated forms), oxyntomodulin, vasoactive intestinal peptide, somatostatin, cholecystokinin, peptide tyrosine‒tyrosine, GLP-2 and pancreatic polypeptide were previously associated with the development

PURPOSE: The gut-brain axis could be a possible key factor in the pathophysiology of anorexia nervosa. The neuropeptide peptide YY3-36, secreted by endocrine L cells of the gastrointestinal tract, is a known regulator of appetite and food intake. The objective of this study was to investigate peptide YY3-36 plasma concentrations at different stages of anorexia nervosa in a combined cross-sectional and longitudinal design to differentiate between effects of acute undernutrition and more enduring characteristics.
METHODS: We measured fasting plasma peptide YY3-36 concentrations in young patient

The efficacy of Roux-en-Y gastric-bypass (RYGB) and other bariatric surgeries in the management of obesity and type 2 diabetes mellitus and novel developments in gastrointestinal (GI) endocrinology have renewed interest in the roles of GI hormones in the control of eating, meal-related glycemia, and obesity. Here we review the nutrient-sensing mechanisms that control the secretion of four of these hormones, ghrelin, cholecystokinin (CCK), glucagon-like peptide-1 (GLP-1), and peptide tyrosine tyrosine [PYY(3-36)], and their contributions to the controls of GI motor function, food intake, and me

PURPOSE OF REVIEW: Disturbances in gastrointestinal hormones have been implicated in the pathogenesis of eating disorders such as anorexia nervosa and bulimia nervosa. However, the contribution of these hormonal changes to the onset and maintenance of eating disorder remains unclear. We focus our review on a selective number of gastrointestinal hormones that are known to play a role in the regulation of short-term or long-term energy balance and examine their association with eating disorder in recently published literature.
RECENT FINDINGS: Several new studies reported differential changes o

The complex bidirectional communication between the gut and the brain is finely orchestrated by different systems, including the endocrine, immune, autonomic, and enteric nervous systems. Moreover, increasing evidence supports the role of the microbiome and microbiota-derived molecules in regulating such interactions; however, the mechanisms underpinning such effects are only beginning to be resolved. Microbiota-gut peptide interactions are poised to be of great significance in the regulation of gut-brain signaling. Given the emerging role of the gut-brain axis in a variety of brain disorders,

Psammomys obesus, widely known as sand rat, develops obesity during captivity. To assess the role of confinement stress on metabolic alterations and sweet taste perception, the gustatory cue involved in obesity, we administered corticosterone (CORT) intraperitoneally in male Psammomys obesus to yield a stressful condition. CORT administration was found to increase preference for sweet solutions in a two bottle-choice paradigm in these animals. Moreover, CORT administration increased the messenger ribonucleic acid (mRNA) expression of sweet test receptor in fungiform taste bud cells. As regards

The body possesses critical nutrient sensors that rapidly coordinate feeding behaviour and systemic metabolism through hormones and neural pathways. In this Review, we highlight the mechanisms by which lipid sensing triggers the release of small intestinal-derived cholecystokinin, peptide YY, glucagon-like peptide 1 and glucose-dependent insulinotropic polypeptide, as well as kidney-derived growth differentiation factor 15, to regulate satiety and glucose homeostasis through the brain. We discuss how dysregulation of lipid sensing pathways contributes to obesity and type 2 diabetes mellitus, a

Functional dyspepsia (FD) is a condition characterized by persistent indigestion symptoms without a clear underlying cause. We investigated the effects on FD of Lacticaseibacillus paracasei HP7 (HP7), which was isolated from kimchi and is known to inhibit Helicobacter pylori. In a mouse model of loperamide-induced FD, HP7 administration significantly improved gastrointestinal (GI) motility and gastric emptying, as demonstrated by increased charcoal movement in the GI tract, decreased stomach weight, and the amount of remaining phenol red solution. HP7 administration significantly enhanced peri

The past decade has been characterized by tremendous progress in the field of the gut microbiota and its impact on host metabolism. Although numerous studies show a strong relationship between the composition of gut microbiota and specific metabolic disorders associated with obesity, the key mechanisms are still being studied. The present review focuses on specific complex pathways as well as key interactions. For instance, the nervous routes are explored by examining the enteric nervous system, the vagus nerve, and the brain, as well as the endocrine routes (i.e., glucagon-like peptide-1, pep

Peptide YY (PYY) is a postprandially released gut hormone. Peripheral administration of one form of the peptide PYY3-36 produces a short-term reduction in food intake in rodents. Initial reports suggested that effects of PYY3-36 on food intake are mediated by increasing the anorexigenic drive from melanocortin neurons in the hypothalamic arcuate nucleus. However, more recent data have demonstrated that the anorexigenic activity of PYY3-36 is not dependent on melanocortin ligands or their receptors in the CNS. We demonstrate here that the anorexigenic actions of PYY3-36 are also not dependent o

Activation of free fatty acid receptor 2 (FFAR2) on enteroendocrine L-cells mediates secretion of glucagon-like peptide 1 (GLP-1) and peptide YY (PYY), key regulators of central appetite control with therapeutic relevance to obesity. Here, we show that butyrate, a metabolite derived from fermentation of dietary fibre and an FFAR2 agonist, stimulates a PYY-biased profile in a human L-cell model at the transcriptional, morphological and secretory level via an FFAR2-Gαi axis that does not require dynamin-dependent receptor internalisation. We observe that butyrate modulates active Notch cascades

Dietary lipids are potent stimulators of intestinal hormone secretion, which plays a central role in the regulation of digestion, appetite, glucose homeostasis, and lipid metabolism. Following digestion in the intestinal lumen, lipid-derived molecules are detected through complementary mechanisms involving lipid transporters, nutrient-sensing receptors, intracellular metabolic pathways, and bile acid signaling. These pathways couple nutrient availability to the secretion of enteroendocrine hormones, including glucose-dependent insulinotropic polypeptide (GIP), glucagon-like peptide-1 (GLP-1),

OBJECTIVE: The mechanisms that facilitate prolonged dietary restriction in eating disorders, particularly in the absence of binge eating, remain poorly understood. The activity-based anorexia model and basic science in exercise physiology suggest that moderate to vigorous physical activity leads to reduced energy intake relative to metabolic needs. This reduction in energy intake is even greater when individuals exercise in the fasted, compared to fed, state.
METHOD: We propose a model in which moderate to vigorous physical activity facilitates increased dietary restriction within eating diso

Anorexia nervosa (AN) is a persistent psychiatric disorder that is marked by abnormal reduced weight and amenorrhea, which may be primary or secondary. AN affects multiple endocrine axes such as gonadal, thyroid, and adrenal axis, growth hormone, and insulin-like growth factor-1, adipokines such as leptin, gut peptides like ghrelin, peptide YY, and amylin. As a result of these changes bone mineral density is reduced, which increases the risk of bone fracture in patients. In this review, we focus on substantial endocrine alterations in AN with a particular emphasis on the severe bone loss assoc

The aganglionic intestine in Hirschsprung's disease displays a severe neuronal derangement. The changes are particularly evident in the muscular innervation. In the gut the endocrine cells are among the cells known to be influenced by neurons. We have, therefore, examined the endocrine cells in ganglionic and aganglionic intestine using immunocytochemistry and immunochemistry. The endocrine cells were studied using antibodies against the neuroendocrine marker chromogranin A, the amine serotonin and the hormonal peptides somatostatin, glucagon/glicentin and peptide YY (PYY), thus covering virtu

Gastrointestinal motility and plasma PYY levels were investigated under chronic progressive large bowel obstruction in dogs. The obstruction device was applied around the descending colon at a laparotomy and gastrointestinal motility was recorded with strain gauge force transducers in the conscious state. Complete obstruction occurred at 26 days (21-33 days). The duration of postprandial interruption of motor complex (DIMC) in the antrum and duodenum were prolonged progressively, at partial obstruction (17.7 +/- 2.7 hr; p less than 0.05) and complete obstruction (23.0 +/- 4.0 hr; p less than 0
