Oxygen radicals, nitric oxide, and peroxynitrite: Redox pathways in molecular medicine.

Oxygen-derived free radicals and related oxidants are ubiquitous and short-lived intermediates formed in aerobic organisms throughout life. These reactive species participate in redox reactions leading to oxidative modifications in biomolecules, among which proteins and lipids are preferential targets. Despite a broad array of enzymatic and nonenzymatic antioxidant systems in mammalian cells and microbes, excess oxidant formation causes accumulation of new products that may compromise cell function and structure leading to cell degeneration and death. Oxidative events are associated with patho

Peroxynitrite Exposure of CXCL12 Impairs Monocyte, Lymphocyte and Endothelial Cell Chemotaxis, Lymphocyte Extravasation in vivo and Anti-HIV-1 Activity.

CXCL12 is a chemotactic cytokine that attracts many different cell types for homeostasis and during inflammation. Under stress conditions, macrophages and granulocytes produce factors such as peroxynitrite as a consequence of their oxidative response. After short incubations of CXCL12 with peroxynitrite, the gradual nitration of Tyr7, Tyr61, or both Tyr7 and Tyr61 was demonstrated with the use of mass spectrometry, whereas longer incubations caused CXCL12 degradation. Native CXCL12 and the nitrated forms, [3-NT61]CXCL12 and [3-NT7/61]CXCL12, were chemically synthesized to evaluate the effects

Biochemical properties of cytochrome c nitrated by peroxynitrite.

Nitration of tyrosine residues is taken as evidence for intracellular formation of peroxynitrite. Cytochrome c (cyt c) can be nitrated by peroxynitrite and nitrated cyt c has been observed in cells and tissues under stress conditions. Here we studied the biochemical properties of nitrated cyt c in order to understand its potential roles in nitrative stress. Nitration of cyt c resulted in disruption of the heme-methionine bond and rapid binding to cyanide. Equilibrium unfolding by guanidine hydrochloride showed that cyt c was slightly destabilized upon nitration but the unfolding transition of

Studies on pig blood platelet responses to peroxynitrite action.

The action of peroxynitrite (ONOO(-)) on pig blood platelet function was studied. Incubation of washed blood platelets with ONOO(-) (0.062-2 mM) caused strong, dose- and time-dependent inhibitory effects on adhesion of resting and thrombin-stimulated platelets to collagen and on platelet aggregation induced by thrombin. The platelet responses were measured 10 s after addition of peroxynitrite to platelets ('zero time') and after 60 min incubation of platelets with ONOO(-). Exposure of blood platelets to peroxynitrite (60 min) brought about platelet lipid peroxidation assessed by the thiobarbit

The Free Radical Scavenging and Anti-Isolated Human LDL Oxidation Activities of Pluchea indica (L.) Less. Tea Compared to Green Tea (Camellia sinensis).

Tea is one of the most popular beverages in the world. Camellia sinensis tea (CST) or green tea is widely regarded as a potent antioxidant. In Thailand, Pluchea indica (L.) Less. tea (PIT) has been commercially available as a health-promoting drink. This study focused on free radical scavenging activities of PIT, and its ability to protect isolated human low-density lipoproteins (LDL) from oxidation by chemical agents. A preliminary study to investigate the antioxidant nature of PIT was undertaken. These included common antioxidant assays involving 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2-azi

Peroxynitrite-Activated Valproic Acid Precursor for Monitoring Oxidative Stress and Reducing Epileptic Seizures in an Epilepsy Model.

Epilepsy is a chronic neurological disorder characterized by recurrent seizures, which poses a significant threat to human health. Increasing evidence indicates that the pathological mechanism of epilepsy is closely related to the presence of peroxynitrite (ONOO-). To address this issue, we have developed a dual-molecule release precursor, DVN, with different functional components, which consists of a responsive group, a reporter segment, and a drug molecule, for monitoring peroxynitrite. Based on in vitro assays, DVN exhibits several advantages, including high specificity, a rapid response ti

Determination of Reactive Oxygen or Nitrogen Species and Novel Volatile Organic Compounds in the Defense Responses of Tomato Plants against Botrytis cinerea Induced by Trichoderma virens TRS 106.

In the present study, Trichoderma virens TRS 106 decreased grey mould disease caused by Botrytis cinerea in tomato plants (S. lycopersicum L.) by enhancing their defense responses. Generally, plants belonging to the 'Remiz' variety, which were infected more effectively by B. cinerea than 'Perkoz' plants, generated more reactive molecules such as superoxide (O2-) and peroxynitrite (ONOO-), and less hydrogen peroxide (H2O2), S-nitrosothiols (SNO), and green leaf volatiles (GLV). Among the new findings, histochemical analyses revealed that B. cinerea infection caused nitric oxide (NO) accumulatio

The zinc-nitric oxide axis in acute brain injury: Convergent oxidative-nitrative stress, peroxynitrite signaling, and therapeutic targets.

Acute brain injuries are characterized by secondary cascades driven by oxidative and nitrative stress. While the roles of excitotoxicity and neuroinflammation in acute brain injury are well recognized, emerging evidence indicates that the interaction between dysregulated zinc (Zn2+) signaling and nitric oxide (NO) biology forms a central metal-redox system that amplifies neuronal injury. Here, we propose the zinc-nitric oxide (Zn-NO) axis as a unified mechanistic framework linking synaptic dysfunction, mitochondrial failure, and neuroinflammatory escalation. Pathological Zn2+ accumulation, ari

Influence of endothelial nitric oxide synthase haplotypes on nitric oxide and peroxynitrite productions.

The impact of four clinically significant genetic variants of endothelial nitric oxide synthase (eNOS) polymorphisms on the concentrations of nitric oxide [NO] and peroxynitrite [ONOO-] has been given scant consideration. This study utilized a [NO]/[ONOO-] ratio to determine the extent of endothelial dysfunction caused by these variations in the eNOS gene. The single nucleotide polymorphisms (T-786C, C-665T, and Glu298Asp) and a variable number of tandem repeats (intron 4 a/b/c) were genotyped in human umbilical vein endothelial cells (HUVEC), using sanger sequencing and DNA electrophoresis, r

Hybridized and engineered microbe for catalytic generation of peroxynitrite and cancer immunotherapy under sonopiezo initiation.

Living therapeutics is an emerging antitumor modality by living microorganisms capable of selective tropism and effective therapeutics. Nevertheless, primitive microbes could only present limited therapeutic functionalities against tumors. Hybridization of the microbes with multifunctional nanocatalysts is of great significance to achieve enhanced tumor catalytic therapy. In the present work, nitric oxide synthase (NOS)-engineered Escherichia coli strain MG1655 (NOBac) was used to hybridize with the sonopiezocatalytic BaTiO3 nanoparticles (BTO NPs) for efficient tumor-targeted accumulation and

Impact of Protein Nitration on Influenza Virus Infectivity and Immunogenicity.

Influenza viruses are deadly respiratory pathogens of special importance due to their long history of global pandemics. During influenza virus infections, the host responds by producing interferons, which activate interferon-stimulated genes (ISGs) inside target cells. One of these ISGs is inducible nitric oxide synthase (iNOS). iNOS produces nitric oxide (NO) from arginine and molecular oxygen inside the cell. NO can react with superoxide radicals to form reactive nitrogen species, principally peroxynitrite. While much work has been done studying the many roles of nitric oxide in influenza vi

Effect of asymmetric dimethylarginine (ADMA) on heart failure development.

Asymmetric dimethylarginine (ADMA) is an endogenous inhibitor of nitric oxide synthases that limits nitric oxide bioavailability and can increase production of NOS derived reactive oxidative species. Increased plasma ADMA is a one of the strongest predictors of mortality in patients who have had a myocardial infarction or suffer from chronic left heart failure, and is also an independent risk factor for several other conditions that contribute to heart failure development, including hypertension, coronary artery disease/atherosclerosis, diabetes, and renal dysfunction. The enzyme responsible f

S-nitrosylation regulates mitochondrial quality control via activation of parkin.

Parkin, a ubiquitin E3 ligase of the ring between ring fingers family, has been implicated in mitochondrial quality control. A series of recent reports have suggested that the recruitment of parkin is regulated by phosphorylation. However, the molecular mechanism that activates parkin to induce mitochondrial degradation is not well understood. Here, and in contrast to previous reports that S-nitrosylation of parkin is exclusively inhibitory, we identify a previously unrecognized site of S-nitrosylation in parkin (Cys323) that induces mitochondrial degradation. We demonstrate that endogenous S-

The role of catalase in gonococcal resistance to peroxynitrite.

We have reported that Neisseria gonorrhoeae is extremely resistant to reactive nitrogen species (RNS) including peroxynitrite (PN). Recent literature suggests that catalase can provide protection against commercial preparations of PN. Though wild-type gonococci were shown to be highly resistant to 2 mM PN, Neisseria meningitidis and a gonococcal katA mutant were both shown to be extremely sensitive to 2 mM PN. Analysis of translational fusions to lacZ of the catalase promoters from N. gonorrhoeae and N. meningitidis demonstrated that basal katA expression from gonococci is 80-fold higher than

Identification of ubiquitin nitration and oxidation using a liquid chromatography/mass selective detector system.

Ubiquitin is a member of the family of low-molecular-weight heat shock proteins that serve a vital role in physiological and pathological protein turnover. It appears to be one of the proteins involved in cell alterations during aging, degenerative disorders, and age-related cognitive decline. It is not known exactly how ubiquitin alterations are related to aging disorders; however, it is possible that ubiquitin is one of the target proteins for free-radical attack. In vivo, the free radical superoxide reacts with nitric oxide to form peroxynitrite, a powerful oxidant. Peroxynitrite may react

Immunohistochemical localization of 3-nitrotyrosine in the nasal respiratory mucosa of patients with vasomotor rhinitis.

CONCLUSION: This study demonstrates that, in the nasal respiratory mucosa of patients with vasomotor rhinitis, oxidative stress following peroxynitrite formation is confined to the respiratory epithelium. This suggests that the role of peroxynitrite in vasomotor rhinitis differs from its role in other diseases of the respiratory tract. The results of this study also support the concept that different pathogenetic mechanisms are probably involved in vasomotor rhinitis. OBJECTIVE: Previous studies indicated that nitric oxide (NO) is involved in the pathogenesis of vasomotor rhinitis, strong exp

Rationally design and create an activatable probe to monitor peroxynitrite in the mice model of polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) stands as a formidable global public health challenge, necessitating advanced diagnostic modalities. Herein, we report the development and multifaceted application of HD-ONOO, an innovative hemicyanine-based fluorescent scaffold engineered for the highly selective, real-time monitoring of peroxynitrite (ONOO-)-a pivotal reactive oxygen species (ROS) linked to PCOS pathology. Upon site-specific interaction with ONOO-, the probe undergoes a structured activation process, yielding pronounced enhancements in both its ratiometric-like absorbance and fluorescence emi

Light-Activatable Nitric Oxide Release via Intramolecular Electron Transfer for Tumor Pyroptosis Induction.

Developing long-wavelength-activated nitric oxide (NO) donors has attracted increasing attention for achieving precise photo-controlled NO release with deep tissue penetration and minimal phototoxicity, yet this remains a major challenge. Herein, we report a deep-red light-triggered NO-releasing molecule (DBTBT-NO) via a novel photoexcitation-mediated electron transfer process to induce abundant peroxynitrite generation. The DBTBT-NO can be activated by deep-red light to release NO, which is facilitated by photoinduced intramolecular charge separation. Meanwhile, superoxide anions generated du

Peroxynitrite-Free Nitric Oxide-Embedded Nanoparticles Maintain Nitric Oxide Homeostasis for Effective Revascularization of Myocardial Infarcts.

Revascularization is crucial for treating myocardial infarction (MI). Nitric oxide (NO), at an appropriate concentration, is recognized as an ideal and potent pro-angiogenic factor. However, the application of NO in the treatment of MI is limited. Improper NO supplementation is harmful to revascularization because NO is converted into harmful peroxynitrite (ONOO-) in MI tissues with high reactive oxygen species (ROS) levels. We overcome these obstacles by embedding biliverdin and NO into Prussian blue (PB) nanolattices to obtain an ONOO--free NO-embedded nanomedicine (OFEN). Unlike previous NO
