GLYCO-BUILD: an enzymatic pipeline for the synthesis of peptides carrying eukaryotic N-glycans.

Peptides and proteins containing defined N-glycans hold great promise for biomedicine because the attached glycans engage in specific interactions, influence targeted delivery and folding, and improve pharmacokinetic properties. However, challenges associated with producing homogeneous N-glycoconjugates hinder their wider application. Here, we report the development of GLYCO-BUILD, an enzymatic pipeline that can generate glycopeptides carrying eukaryotic N-glycans at a level of homogeneity not accessible by alternative methodologies. The pipeline recapitulates the endoplasmic reticulum-based s

Ataxia with loss of Purkinje cells in a mouse model for Refsum disease.

Refsum disease is caused by a deficiency of phytanoyl-CoA hydroxylase (PHYH), the first enzyme of the peroxisomal alpha-oxidation system, resulting in the accumulation of the branched-chain fatty acid phytanic acid. The main clinical symptoms are polyneuropathy, cerebellar ataxia, and retinitis pigmentosa. To study the pathogenesis of Refsum disease, we generated and characterized a Phyh knockout mouse. We studied the pathological effects of phytanic acid accumulation in Phyh(-/-) mice fed a diet supplemented with phytol, the precursor of phytanic acid. Phytanic acid accumulation caused a redu

Metabolomic Profiling of Taiwanese Roselle (Hibiscus sabdariffa) Leaves and Their Antiproliferative and Insulin Resistance-Ameliorating Potential.

Roselle (Hibiscus sabdariffa) leaves are the major by-product after harvesting the commercial calyces, and they contain diverse functional metabolites such as phenolic acids and flavonoids. This study aimed to comprehensively characterize the metabolite profiles of the leaves of two Taiwanese roselle varieties (Taitung No. 6 and Taitung No. 3) and to evaluate their antiproliferative and insulin resistance-ameliorating activities. Extracts prepared with 50% EtOH and n-hexane were analyzed by untargeted and targeted LC-MS/MS and GC-MS, respectively. Results revealed that the 50% EtOH extracts of

Chemical Characterization and In Vitro Bioactivities of Vitex negundo Leaf Extracts.

Vitex negundo L. (Verbenaceae) is a well-known medicinal plant used in traditional medicine for the treatment of inflammatory, microbial, and oxidative stress-related disorders. The present study aimed to evaluate antibacterial and antioxidant activities of Vitex negundo leaf extracts, along with their chemical characterization using Fourier transform infrared (FT-IR) spectroscopy and gas chromatography-mass spectrometry (GC-MS). Antibacterial activity of acetone, methanol, and aqueous extracts was assessed by agar well diffusion method against four bacterial strains. The acetone extract exhib

Inhibitory effect of the methanol extract of Clerodendrum volubile Linn. on carbon tetrachloride-induced early hepatic fibrogenesis: in silico and in vivo study.

Liver disease remains a significant public health problem, claiming several lives globally every year. All forms of liver disease, irrespective of the cause, lead to liver fibrosis if they persist. Liver fibrosis ends up in liver cirrhosis and creates a microenvironment that enhances the development of hepatocellular carcinoma. Clerodendrum volubile is a food and medicinal plant consumed to manage several diseases, including liver diseases. Liver fibrosis was induced in Wistar rats with carbon tetrachloride (CCl4 Olive oil 1:1) for nine weeks and treated with the extract from weeks 7-9. The ac

The stem ethyl acetate extract of Costus pictus D.DON exhibits cytotoxic activity in human cancer cells via. intrinsic apoptotic caspase pathway as a first report.

The leaves of Costus pictus have been reported to have anti-diabetic properties. Nevertheless, a comprehensive analysis of the entire plant remains to be established. The current objective was to assess the phytochemical content, antioxidant capacity and anti-cancer potential. Three fractions - hexane, ethyl acetate, and methanol extract were obtained by sequential extraction. The various extracts were phytochemically screened and the effects on cell proliferation was evaluated on cancerous (A-549, HeLa, HT-29, and AGS) and non-cancerous (L-132 and L929) cell lines. C. pictus presented no sign

Cadmium tolerance in Elodea canadensis Michx: Subcellular distribution and metabolomic analysis.

The aquatic plant Elodea canadensis is considered a good candidate for ecotoxicological investigations. Cadmium (Cd) is a widespread contaminant in aquatic systems. In this study, to better elucidate the underlying tolerance mechanism and molecular impact of environmentally relevant Cd concentration in aquatic plants, subcellular distribution, chemical forms, and gas chromatography-mass spectrometry-based non-targeted metabolomics profiles were comprehensively analyzed in E. canadensis subjected to 0 and 10 µM Cd treatment for 5 d. Antioxidant and cytotoxicity screenings of ethyl acetate extract from Annona muricata leaves and its fractions.

Leaves of Annona muricata have medicinal potential which has gained attention from researchers around the world. This study has an objective to screen the antioxidant and cytotoxicity of ethyl acetate extract from A. muricata leaves and its fraction. The fine powder of A. muricata was macerated in methanol and further partitioned using two different solvents, namely n-hexane and ethyl acetate. In this article, we reported the screening results for ethyl acetate extract. Fractionation was then performed on the extract by means of column chromatography by gradient elution resulting in five combi

In vitro Antibacterial Activity of Adiantum capillus veneris Extraction with Methanol, Chloroform, and Ether Solvents against Methicillin-resistant Staphylococcus aureus.

BACKGROUND: The increasing problem of multi-drug resistant (MDR) pathogens is a worldwide concern, especially in the pharmaceutical industry. At the same time, medicinal plants have renewed interest because of their wide variety of bioactive phytochemicals, which could be used to develop new antimicrobial drugs. This renewed interest is partly due to the growing resistance to traditional drugs and their associated side effects. METHODS: The objective of this study is to assess the antimicrobial properties of the total extract and various fractions of Adiantum capillus veneris against Methicil

Potential effect of Turbinaria decurrens acetone extract on the biochemical and histological parameters of alloxan-induced diabetic rats.

Upon Seeking natural and safe alternatives for synthetic medicines to treat many chronic diseases, seaweeds have offered a promising resource to produce numerous bioactive secondary metabolites. Through in vivo investigations, Turbinaria decurrens acetone extract (AE) revealed its antidiabetic activity against alloxan-induced diabetic rats. Treatment of rats with T. decurrens AE at 300 and 150 mg/Kg doses revealed antihyperglycemic activity by reducing the elevated blood glucose level. A remarkable decrease in the liver, kidney functions, and hyperlipidemia related to diabetes were also detect

The Reproductive Toxicity Associated with Dodonaea viscosa, a Folk Medicinal Plant in Saudi Arabia.

Dodonaea viscosa is a medicinal plant which is being used to treat various diseases in humans. The available safety data suggest that the plant does not produce any side effects, or toxicity, in tested adult experimental animals. However, the influence of D. viscosa on fetus or embryonic development is largely not known. This study was conducted in order to find out the reproductive toxicity of D. viscosa in experimental animals. Zebrafish embryos were used as the in vivo developmental toxicity animal model. Methanolic crude extract, hexane, chloroform, and butanol fractions were prepared from
