Plant extracts as emerging modulators of neuroinflammation and immune receptors in Alzheimer's pathogenesis.

Memory loss is becoming an increasingly significant health problem, largely due to Alzheimer's disease (AD), which disrupts the brain in several ways, including causing inflammation and weakening the body's defenses. This study explores the potential of medicinal plants as a source of novel therapeutic agents for AD. First, we tested various plant extracts against acetylcholinesterase (AChE) in vitro, following molecular docking simulations with key AD-related protein targets such as MAO-B, P-gp, GSK-3β, and CD14. Rosemary extract was found to be the most inhibitory towards AChE. The compounds

The enzymatic hydrolysate of fucoidan from Sargassum hemiphyllum triggers immunity in plants.

Fucoidans are polysaccharides that consist predominantly of sulfated L-fucoses, from which, fucoidan oligosaccharides (FOSs) are prepared through different methods. Fucoidan has versatile physiological activities, like antiviral functions against SARS CoV-2 and bioactivitiy in enhancing immune responses. Although fucoidan or FOS has been widely used in mammals as functional foods and new drugs, its application in plants is still very limited. Moreover, whether fucoidan or its derived hydrolytic products can trigger immune responses in plants remained unknown. In this work, we demonstrate that

Spatial and temporal regulation of biosynthesis of the plant immune signal salicylic acid.

The plant hormone salicylic acid (SA) is essential for local defense and systemic acquired resistance (SAR). When plants, such as Arabidopsis, are challenged by different pathogens, an increase in SA biosynthesis generally occurs through transcriptional induction of the key synthetic enzyme isochorismate synthase 1 (ICS1). However, the regulatory mechanism for this induction is poorly understood. Using a yeast one-hybrid screen, we identified two transcription factors (TFs), NTM1-like 9 (NTL9) and CCA1 hiking expedition (CHE), as activators of ICS1 during specific immune responses. NTL9 is ess

Light acclimation, retrograde signalling, cell death and immune defences in plants.

This review confronts the classical view of plant immune defence and light acclimation with recently published data. Earlier findings have linked plant immune defences to nucleotide-binding site leucine-rich repeat (NBS-LRR)-dependent recognition of pathogen effectors and to the role of plasma membrane-localized NADPH-dependent oxidoreductase (AtRbohD), reactive oxygen species (ROS) and salicylic acid (SA). However, recent results suggest that plant immune defence also depends on the absorption of excessive light energy and photorespiration. Rapid changes in light intensity and quality often c

Redox-based protein modifications: the missing link in plant immune signalling.

Activation of plant immunity is associated with dramatic changes in the cellular redox status. Both oxidative and reductive bursts have been described that trigger a set of down stream responses resulting in reprogramming of the transcriptome and establishment of disease resistance. Nonetheless, how these redox changes are sensed and signal to downstream regulators remained a missing link in studies of plant immunity. Emerging evidence now indicates that pathogen-induced changes in the cellular redox environment are sensed by reactive cysteine residues of key regulatory proteins. Varying degre

Production of hepatitis B surface antigen in transgenic plants for oral immunization.

Here we present data showing oral immunogenicity of recombinant hepatitis B surface antigen (HBsAg) in preclinical animal trials. Mice fed transgenic HBsAg potato tubers showed a primary immune response (increases in HBsAg-specific serum antibody) that could be greatly boosted by intraperitoneal delivery of a single subimmunogenic dose of commercial HBsAg vaccine, indicating that plants expressing HBsAg in edible tissues may be a new means for oral hepatitis B immunization. However, attainment of such a goal will require higher HBsAg expression than was observed for the potatoes used in this s

Iron homeostasis and plant immune responses: Recent insights and translational implications.

Iron metabolism and the plant immune system are both critical for plant vigor in natural ecosystems and for reliable agricultural productivity. Mechanistic studies of plant iron home-ostasis and plant immunity have traditionally been carried out in isolation from each other; however, our growing understanding of both processes has uncovered significant connections. For example, iron plays a critical role in the generation of reactive oxygen intermediates during immunity and has been recently implicated as a critical factor for immune-initiated cell death via ferroptosis. Moreover, plant iron s

Neutralizing immune responses induced by oligomeric H5N1-hemagglutinins from plants.

Plant-based transient expression is an alternative platform to produce hemagglutinin-based subunit vaccines. This production system provides not only fast and effective response in the context of a pandemic but also enables the supply of big volume vaccines at low cost. Crude plant extracts containing influenza hemagglutinin are considered to use as vaccine sources because of avoidance of related purification steps resulting in low cost production allowing veterinary applications. Highly immunogenic influenza hemagglutinins are urgently required to meet these pre-conditions. Here, we present a

Copper triggers plant immunity: Enhancing StRVE1 expression to activate salicylic acid accumulation by directly upregulating the StSID2-2 variant.

Phytophthora infestans significantly reduces the yield and quality of potato. Copper ion (Cu2+)-based antimicrobial compounds (CBACs) have been commercially applied for over a century to combat phytopathogens such as P. infestans. We had previously established that Cu2+ functions as an elicitor, triggering plant defense responses via the ethylene and salicylic acid (SA) signaling pathways. The current study found that treating potato plants with CBACs at dawn or dusk had varying effects on defense against late blight, suggesting that circadian rhythms affect the CBAC biocide efficacy. We furth

Dual stimuli-responsive prodrug co-delivery nanosystem of salicylic acid and bioavailable silicon for long-term immunity in plant.

Plant-induced resistance plays a crucial role in the plant defense system by activating intrinsic immune mechanisms. In this study, a novel amidase- and redox-responsive codelivery nanosystem was developed by covalently linking salicylic acid (SA) to functionalized disulfide-doped mesoporous silica nanoparticles (MSNs-ss-NH2) for the efficient delivery of SA and bioavailable silicon concurrently. Physicochemical characterization confirmed the successful preparation of MSNs-ss-SA, demonstrating its structural integrity and glutathione and amidase responsive degradation mechanism. Zinc priming enhances Capsicum annuum immunity against infection by Botrytis cinerea- From the whole plant to the molecular level.

Micronutrient manipulation can enhance crop resilience against pathogens, but the mechanisms are mostly unknown. We tested whether priming Capsicum annuum plants with zinc (5 μM Zn) or manganese (3 μM Mn) for six weeks increases their immunity against the generalist necrotroph Botrytis cinerea compared to deficient (0.1 μM Zn, 0.02 μM Mn) and control conditions (1 μM Zn, 0.6 μM Mn). Zinc priming reduced the pathogen biomass and lesion area and preserved CO2 assimilation and stomatal conductance. Zinc mobilization at the infection site, visualized by micro-X-ray fluorescence, was accompanied by

Pepper heat shock protein 70a interacts with the type III effector AvrBsT and triggers plant cell death and immunity.

Heat shock proteins (HSPs) function as molecular chaperones and are essential for the maintenance and/or restoration of protein homeostasis. The genus Xanthomonas type III effector protein AvrBsT induces hypersensitive cell death in pepper (Capsicum annuum). Here, we report the identification of the pepper CaHSP70a as an AvrBsT-interacting protein. Bimolecular fluorescence complementation and coimmunoprecipitation assays confirm the specific interaction between CaHSP70a and AvrBsT in planta. The CaHSP70a peptide-binding domain is essential for its interaction with AvrBsT. Heat stress (37°C) an

Conserved DEAD-box RNA helicases regulate RNA silencing and plant immunity and are targeted by a powdery mildew effector.

RNA silencing is a conserved defense mechanism that protects plants against viruses, bacteria, fungi, and oomycetes. Although diverse pathogens have evolved effectors that suppress host RNA silencing, whether powdery mildew fungi exploit this strategy and the underlying molecular mechanisms remain unknown. Here, we identify CSEP0004, a secreted effector from the barley powdery mildew fungus Blumeria hordei, as an RNA silencing suppressor that promotes fungal virulence. CSEP0004 physically interacts with the evolutionarily conserved DEAD-box RNA helicases Hordeum vulgare RNA helicase 37 (HvRH37

Nuclear transfer of a beneficial fungal effector-host target complex via the NTF2 pathway underpins symbiont-induced plant immunity.

The symbiotic fungus Serendipita indica confers broad-spectrum beneficial effects on diverse plant hosts. Its key effector SIE141 elicits immunity against Phytophthora and salt tolerance by binding and relocalizing thioredoxin CDSP32 from chloroplast to the nucleus. Here, we show that this functionally essential nuclear transfer process of the SIE141-CDSP32 complex is mediated by the host NTF2 proteins. NTF2 family proteins are direct targets of SIE141, whose knockdown abolished nuclear accumulation of both SIE141 and CDSP32, leading to their rendered accumulation to chloroplasts. The glutamin

Discovery of natural Nopol as promising antibacterial alternative with the capacity to suppress bacterial virulence and enhance plant immunity.

Outbreaks of plant diseases pose a severe threat to crop production and global food security. The extensive and prolonged use of conventional chemical bactericides has exacerbated pathogen resistance, creating an urgent demand for novel antibacterial agents with high biosafety profiles. This study screened a series of cyclic terpenoids against three phytopathogenic bacteria: Xanthomonas oryzae pv. oryzae (Xoo), Xanthomonas axonopodis pv. citri (Xac), and Pseudomonas syringae pv. actinidiae (Psa), with Nopol showing the strongest inhibitory activity against Xoo (EC50 = 8.86 mg·L-1). Feed-based Aeromonas hydrophila vaccine combined with plant-seed extracts improves immunity, growth, and disease resistance in Labeo rohita.

Major carps are economically important but highly susceptible to Aeromonas hydrophila infections. The overuse of antibiotics in aquaculture has led to antimicrobial resistance, highlighting the critical need for sustainable disease control strategies. This study evaluated the immunogenic potential of a feed-based formalin-inactivated vaccine combined with plant-seed extracts in Labeo rohita. Fish were acclimatized under laboratory conditions for two weeks. Then fish (weight 6.50 ± 0.04 g) were randomly allocated into six groups: T0 (control group); T1 (ASE); T2 (RSE); T3 (AV); T4 (AV + ASE) an

ENHANCED DISEASE RESISTANCE4 associates with CLATHRIN HEAVY CHAIN2 and modulates plant immunity by regulating relocation of EDR1 in Arabidopsis.

Obligate biotrophs, such as the powdery mildew pathogens, deliver effectors to the host cell and obtain nutrients from the infection site. The interface between the plant host and the biotrophic pathogen thus represents a major battleground for plant-pathogen interactions. Increasing evidence shows that cellular trafficking plays an important role in plant immunity. Here, we report that Arabidopsis thaliana ENHANCED DISEASE RESISTANCE4 (EDR4) plays a negative role in resistance to powdery mildew and that the enhanced disease resistance in edr4 mutants requires salicylic acid signaling. Proline transporters coordinate immune activation and regenerative capacity after injury in plants.

Effective survival after injury requires the precise coordination of immune activation and tissue regeneration, yet the molecular mechanisms balancing these opposing processes remain poorly understood. Here, we identify proline transporters as key regulators of this trade-off in plants. Genetic disruption of two wound-induced proline transporters, ProT2 and ProT3, enhances regenerative capacity and abolishes salicylic acid (SA)-mediated suppression of regeneration while preserving SA-responsive gene expression and SA-mediated antibacterial resistance. Mechanistically, a ProT3-CPK1 complex modu

Viral Silencing Suppressor Activity in Plants Modifies Aphid Antiviral Immunity and Fecundity.

Plants defend themselves from viral infection using RNA interference (RNAi), an evolutionarily conserved mechanism that degrades viral RNA through the production of small interfering RNAs. As a counter defense, viral suppressors of RNA silencing (VSRs) inhibit plant RNAi machinery, aiding viral replication and transmission. P0, a VSR encoded by the potato leafroll virus (PLRV), family Polerovirus, suppresses RNAi by targeting the plant protein ARGONATE 1 for degradation through its F-box motif interaction with an Skp1 subunit of the family of E3 ubiquitin ligases. Our previous work shows that

Engineering plant tandem kinase immune receptors expands effector recognition profiles.

Plant intracellular immune receptors are widely deployed in breeding to protect crops from disease. In addition to nucleotide-binding leucine-rich repeat receptors (NLRs), tandem kinase proteins (TKPs) have recently emerged as an important family of immune receptors within staple cereal food crops, but how TKPs recognize effectors and whether they are amenable to engineering is essentially unknown. Here, we show that the barley and wheat TKPs Rmo2 and Rwt7 recognize different blast fungus effectors via their integrated HMA domains using different protein interfaces with nanomolar binding affin
