Prevalence of Drought Over Heat Stress in Modulating Physiological and Metabolic Responses of Castanea sativa Mill. Var. Martaínha.

The productivity of chestnut trees (Castanea sativa Mill.) is strongly affected by high temperatures and reduced water availability, phenomena exacerbated by climate change. However, the physiological effects of heat and drought exposure in this species remain poorly characterised, particularly, in local varieties of national relevance. In this study, we investigated the primary metabolism of young plants of the Martaínha variety subjected to drought (25% of maximum water-holding capacity) and/or high temperatures (42°C, 4 h day-1). After 21 days of stress, exposure to heat alone induced disti

Metabolic engineering of stomatal precursor cells enhances photosynthetic water-use efficiency and vegetative growth under water-deficit conditions in Arabidopsis thaliana.

Stomata are epidermal pores that control the exchange of gaseous CO2 and H2O between plants and their environment. Modulating stomatal density can alter this exchange and thus presents a viable target for engineering improved crop productivity and climate resilience. Here, we show that stomatal density in Arabidopsis thaliana can be decreased by the expression of a water-forming NAD(P)H oxidase targeted to stomatal precursor cells. We demonstrate that this reduction in stomatal density occurs irrespective of whether the expressed enzyme is localized to the cytosol, chloroplast stroma or chloro

Dynamic remediation test of polluted river water by Eco-tank system.

Dynamic remediation of river water polluted by domestic sewage using an aquatic plants bed-based Eco-tank system was investigated. Over a period of 18 days, the test demonstrated that average effluent concentrations of chemical oxygen demand (COD), ammonium nitrogen (NH4(+)-N) and total phosphorus (TP) were as low as 17.28, 0.23 and 0.03 mg/L, respectively, under the hydraulic retention time (HRT) of 8.7 d. The average removal efficiencies in terms of COD, NH4(+)-N and TP could reach 71.95, 97.96 and 97.84%, respectively. The loss of both NH4(+)-N and TP was mainly ascribed to the uptake by pl

Does Salicylic Acid (SA) Improve Tolerance to Salt Stress in Plants? A Study of SA Effects On Tomato Plant Growth, Water Dynamics, Photosynthesis, and Biochemical Parameters.

Environmental stresses such as salinity directly impact crop growth, and by extension, world food supply and societal prosperity. It is estimated that over 800 million hectares of land throughout the world are salt-affected. In arid and semi-arid regions, salt concentration can be close to that in the seawater. Hence, there are intensive efforts to improve plant tolerance to salinity and other environmental stressors. Salicylic acid (SA) is an important signal molecule for modulating plant responses to stress. In the present study, we examined, on multiple plant growth related endpoints, wheth

Effects of Salinity on Bacterial Spot Disease, Physiology, Growth, Fruit Quality, and Transcriptomic Responses in Tomato Plants.

Soil salinity and bacterial spot of tomato (BST), caused by Xanthomonas perforans, are major abiotic and biotic stresses limiting tomato production, particularly in Florida. While their individual effects are well documented, the impact of soil salinity on BST has not yet been investigated. This greenhouse study evaluated how increasing irrigation water salinity (electrical conductivity [EC] = 0.5, 3, 5, or 7 dS m-1) affected tomato growth, physiology, BST severity, fruit quality, and transcriptomic responses. Salinity reduced plant growth and BST severity but did not directly affect X. perfor

Gene expression in vessel-associated cells upon xylem embolism repair in Vitis vinifera L. petioles.

In this work, the involvement of vessel-associated cells in embolism recovery was investigated by studying leaf petiole hydraulics and expression profiles of aquaporins and genes related to sugar metabolism. Two different stress treatments were imposed onto grapevines to induce xylem embolism: one involved a pressure collar applied to the stems, while the other consisted of water deprivation (drought). Embolism formation and repair were monitored during stress application and release (recovery). At the same time, stomatal conductance (g(s)), leaf water potential (Ψ(leaf)) and leaf abscisic aci

A unified framework for sapwood and stem water content in plants: structure, function, and measurement.

Stem water content is a fundamental parameter governing key plant physiological processes, including transpiration, stomatal conductance, sap flow, and hydraulic capacitance. Although extensively studied in the context of timber, construction, and engineering, its physiological significance in living plants remains comparatively underdeveloped. This review synthesises the physiological literature to provide a clear and consistent definition of stem water content, partitioning it into bark, phloem, sapwood, and heartwood components, with particular emphasis on sapwood water content. A theoretic

Advanced phenotyping features utilizing deep learning techniques for automated analysis of stomatal guard cell orientation.

Stomata are vital for controlling gas exchange and water vapor release, which significantly affect photosynthesis and transpiration. Characterizing stomatal traits such as size, density, and distribution is essential for adaptation to the environment. While microscopy is widely used for this purpose, manual analysis is labor-intensive and time-consuming that limit large scale studies. To overcome this, we introduce an automated, high-throughput method that leverages YOLOv8, an advanced deep learning model, for more accurate and efficient stomatal trait measurement. Our approach provides a comp

Photosynthetic performance and sucrose metabolism in superior and inferior rice grains with overlapping growth stages under water stress.

Drought stress at jointing and booting stages of plant development directly affects plant growth and productivity in rice. Jointing and booting stages may overlap in high-latitude areas where water deficits occur. However, little is known about the effects of photosynthesis on grain sucrose metabolism and the differences of sucrose metabolism strategies between superior and inferior grains under different drought stress was unclear. In this study, rice plants were subjected to drought stress for 15 days at jointing-booting. Drought stress affected normal leaf growth, and decreased the leaf are

Screening broad beans (Vicia faba) for magnesium deficiency. II. Photosynthetic performance and leaf bioelectrical responses.

In search of rapid screening tools for magnesium (Mg) deficiency in crops at early stages of plant ontogeny, we studied the kinetics of leaf photosynthetic responses and changes in electrophysiological characteristics of broad bean leaves as affected by different levels of Mg in the nutrient solution (1-200 ppm). No apparent correlation between plant age, Mg supply level, leaf stomatal conductance (gs) and transpiration rate (E) were found. A significant difference in CO2 assimilation became obvious only at week 8. Chlorophyll fluorescence analysis, however, revealed a significant difference i

Orchard management alters citrus root and rhizosphere microbiomes with functional consequences for plant performance.

Agricultural management practices act as ecological disturbances that can restructure soil and plant-associated microbial communities, but the functional consequences of these microbial shifts on crop performance remain poorly understood. Here, we examined how common orchard inputs, including wood mulch, glyphosate, and humic acid, affect citrus root and rhizosphere microbiomes, leaf nutrient trajectories, and tree performance over a 3-year field experiment. Mulch emerged as the dominant driver of fungal community composition and taxonomic turnover, enriching for saprophytic fungi. Conversely,

Water rise kinetics in refilling xylem after desiccation in a resurrection plant.

The acropetal water refilling kinetics of the dry xylem of branches (up to 80 cm tall) of the resurrection plant Myrothamnus flabellifolia were determined with high temporal resolution by observation of light refraction at the advancing water front and the associated recurving of the folded leaves. To study the effect of gravity on water rise, data were acquired for cut upright, horizontal and inverted branches. Water rise kinetics were also determined with hydrostatic and osmotic pressure as well as at elevated temperatures (up to 100 degrees C) under laboratory conditions and compared with t

Boron uptake and distribution by oilseed rape (Brassica napus L.) as affected by different nitrogen forms under low and high boron supply.

Ammonium (NH4+) and nitrate (NO3-) conversely alter pH of the rooting medium, and thus differentially affect the equilibrium between boric acid and borate in soil solution. This can alter boron (B) uptake by plants, which is passive under high, but facilitated (boric acid) or active (borate) under low B supply. Therefore, the effect of NH4+ and NO3- forms was investigated on the growth, 10B uptake rate and accumulation, and expression of B transporters in Brassica napus grown with low (1 μM) or high (100 μM) 10B for five days in the nutrient solution. At the low 10B level, NO3--fed plants had

Water stress assessment in rainfed crops and pastures by using remotely sensed evapotranspiration.

Agricultural drought is a natural and damaging phenomenon that is especially harmful to rainfed agriculture. It occurs when there is insufficient soil moisture in the root zone for plants to survive between two rainfall events. In the absence of soil moisture, a variety of losses, including soil evaporation and plant transpiration, cause an imbalance between water supply and water loss. An evapotranspiration-based index was used here to assess agricultural drought. We applied this framework to a less studied area near Fariman City in the northeast part of IRAN. Two time periods were selected f

Oxalic acid-mediated biochemical and physiological changes in the common bean-Sclerotinia sclerotiorum interaction.

The success of Sclerotinia sclerotiorum infection relies mainly on the production of the non-host selective toxin named oxalic acid (OA). This toxin is known to play multiple roles in a host infected by the fungus, but its effect on photosynthesis and the antioxidant system of common bean plants remain elusive. Therefore, we performed detailed analysis of leaf gas exchange, chlorophyll a fluorescence, activities of antioxidant enzymes, concentrations of reactive oxygen species and photosynthetic pigments to investigate the OA's role during the S. sclerotiorum pathogenesis. To achieve this goal

Enhancement of drought tolerance in Arabidopsis plants induced by sulfur dioxide.

Sulfur dioxide (SO2) is a common air pollutant that has multiple effects on plants. In the present study, the improvement of drought tolerance in Arabidopsis plants by SO2 fumigation was investigated. The results showed that pre-exposure to 30 mg/m3 SO2 for 72 h could reduce water loss, stomatal conductance (Gs) and the transpiration rate (Tr) but increased the net photosynthetic rate (Pn), water use efficiency (iWUE) and photosynthetic pigment contents under drought conditions. The activities of superoxide dismutase (SOD) and peroxidase (POD) were significantly increased, while the contents o

NASA's ECOSTRESS satellite reveals widespread midday depression in ecosystem evapotranspiration.

Plants often exhibit a midday depression in water use (i.e., transpiration), reflecting a constraint on their ability to sustain maximum water transport, which may occur at the cost of reduced photosynthesis. Eddy covariance observations and geostationary satellites cannot quantify this widespread phenomenon globally while resolving fine-scale spatial variability. Using evapotranspiration measurements from the ECOsystem Spaceborne Thermal Radiometer Experiment on Space Station (ECOSTRESS) and machine learning, we quantify the global distribution of midday depression in evapotranspiration. Midd

Gas exchange, growth, and defense responses of invasive Alliaria petiolata (Brassicaceae) and native Geum vernum (Rosaceae) to elevated atmospheric CO2 and warm spring temperatures.

PREMISE OF STUDY: Global increases in atmospheric CO2 and temperature may interact in complex ways to influence plant physiology and growth, particularly for species that grow in cool, early spring conditions in temperate forests. Plant species may also vary in their responses to environmental changes; fast-growing invasives may be more responsive to rising CO2 than natives and may increase production of allelopathic compounds under these conditions, altering species' competitive interactions. METHODS: We examined growth and physiological responses of Alliaria petiolata, an allelopathic, inva

Integrated Physiological, Transcriptomic, and Metabolomic Analyses Reveal the Mechanism Underlying the Response of Alfalfa to Combined Salt and Heat Stress.

Salt and heat stresses often occur simultaneously in arid regions, restricting the distribution and productivity of alfalfa (Medicago sativa L.). However, the mechanisms underlying alfalfa responses to combined salt and heat stress remain unclear. Here, we integrated phenotypic, physiological, transcriptomic, and metabolomic analyses to investigate the regulatory mechanisms involved in combined stress responses. Combined stress more strongly inhibited plant height, stem diameter, fresh weight, net photosynthetic rate, and transpiration rate than single stresses. Salt stress had a predominant e

Endophytes of opium poppy differentially modulate host plant productivity and genes for the biosynthetic pathway of benzylisoquinoline alkaloids.

Endophytes reside in different parts of the poppy plant and perform the tissue-specific functions. Most leaf endophytes modulate photosynthetic efficiency, plant growth, and productivity while capsule endophytes modulate alkaloid biosynthesis. Endophytes promote plant growth, provide protection from environmental stresses and are the source of important secondary metabolites. Here, we established that the endophytes of opium poppy Papaver somniferum L. may play a role in the modulation of plant productivity and benzylisoquinoline alkaloid (BIA) biosynthesis. A total of 22 endophytes isolated f
