Therapeutic options and emerging alternatives for multidrug resistant staphylococcal infections.

Methicillin-resistant Staphylococcus aureus (MRSA) remains the single biggest challenge in infectious disease in the civilized world. Moreover, vancomycin resistance is also spreading, leading to fears of untreatable infections as were common in ancient times. Molecular microbiology and bioinformatics have revealed many of the mechanisms involved in resistance development. Mobile genetic elements, up-regulated virulence factors and multi-drug efflux pumps have been implicated. A range of approved antibiotics from the glycopeptide, lipopeptide, pleuromutilin, macrolide, oxazolidinone, lincosami

Emerging threats: Listeria monocytogenes with acquired multidrug resistance from food in China, 2012-2022.

Listeria monocytogenes is a foodborne pathogen that poses threat to food safety and public health. Generally, the rates of resistance to clinically important antibiotics in L. monocytogenes are low. This study aimed to investigate the prevalence and genetic characteristics of L. monocytogenes with acquired multidrug resistance (MDR) in food samples from China between 2012 and 2022. Of 8344 isolates collected, 34 (0.41 %) were identified as acquired MDR. The majority of acquired MDR isolates (n = 31, 92.3 %) belonged to hypovirulent clonal complex (CC) 9 (Lineage II, IIc), including 3 sequence

Library Screening for Synergistic Combinations of FDA-Approved Drugs and Metabolites with Vancomycin against VanA-Type Vancomycin-Resistant Enterococcus faecium.

Antimicrobial resistance is a major public health threat, and there is an urgent need for new strategies to address this issue. In a recent study, a library screening strategy was developed in which an FDA-approved drug library was screened against methicillin-resistant Staphylococcus aureus (MRSA) in both its original (unmetabolized [UM]) and its human liver microsome metabolized (postmetabolized [PM]) forms and in the absence and presence of a resistant-to antibiotic. This allows the identification of agents with active metabolites and agents that can act synergistically with the resistant-t

Antibiotic resistance of Mycoplasma Synoviae strains isolated in China from 2016 to 2019.

BACKGROUND: In the past decade, Mycoplasma synoviae (M. synoviae) infection has become widely prevalent in China, has caused serious economic losses and has become one of the most important diseases in the chicken industry. Medication is a general approach for the control of M. synoviae infection, but antibiotics are sometimes ineffective in clinical practice. To investigate the sensitivity of M. synoviae to antimicrobials commonly used in the treatment of M. synoviae infection, the antibiotic susceptibility of 32 M. synoviae strains isolated from China from 2016 to 2019 were determined using

Comparative genome analysis of Erysipelothrix rhusiopathiae isolated from domestic pigs and wild boars suggests host adaptation and selective pressure from the use of antibiotics.

The disease erysipelas caused by Erysipelothrix rhusiopathiae (ER) is a major concern in pig production. In the present study the genomes of ER from pigs (n=87), wild boars (n=71) and other sources (n=85) were compared in terms of whole-genome SNP variation, accessory genome content and the presence of genetic antibiotic resistance determinants. The aim was to investigate if genetic features among ER were associated with isolate origin in order to better estimate the risk of transmission of porcine-adapted strains from wild boars to free-range pigs and to increase our understanding of the evol

Susceptibility of Staphylococcus aureus to topical agents in the United States: a sentinel study.

BACKGROUND: Mupirocin has been used topically for treating skin and skin structure infections and for nasal decolonization before surgical interventions. Pleuromutilin compounds, including retapamulin, provide similar treatment/interventional options. Rates of resistance of Staphylococcus aureus to mupirocin and other agents used to treat skin and skin structure infections vary between countries and medical centers, including those in the United States. These resistance rates may be associated with higher usage and/or improper epidemiologic practices. OBJECTIVE: This study aimed to determine

Ecological genomics of saprotrophy to biotrophy transitions in the genus Clitopilus (Fr. ex Rabenh.) P. Kumm. (Agaricales, Entolomataceae).

Transitions between saprotrophic and biotrophic lifestyles represent pivotal evolutionary events in fungal ecology; however, the genomic and physiological mechanisms underlying such shifts remain poorly understood. The agaric genus Clitopilus (Basidiomycota, Entolomataceae) offers a valuable model system, with most species being soil saprotrophs. Clitopilus cf. baronii Consiglio & Setti exhibits genomic signatures suggesting incipient biotrophic capacity. Here, we investigated the genomic and eco-physiological properties of seven strains representing five Clitopilus species to identify traits

First report of PLSA resistance gene sal(B) in multidrug-resistant foodborne Mammaliicoccus lentus: Molecular characterization and food safety implications.

The emergence and dissemination of antimicrobial resistance through the food chain pose a significant threat to food safety and public health. Our nationwide surveillance of retail foods in China revealed alarmingly high resistance rates among foodborne Staphylococcus isolates, particularly to antibiotics targeting the 50S ribosomal subunit. Among these, Mammaliicoccus lentus (formerly Staphylococcus lentus) is a coagulase-negative species commonly found in food-producing animals and retail foods, increasingly recognized as a reservoir of antimicrobial resistance genes. To elucidate the geneti

Advancing standardization of diagnostics and antimicrobial susceptibility testing for pathogenic mycoplasmas of livestock origin: insights from the MyMIC network.

BACKGROUND: Mycoplasma infections pose a significant economic burden and represent a serious health and welfare concern for the livestock sector. Their control often requires repeated antimicrobial treatments. Antimicrobial susceptibility testing (AST) procedures for veterinary mycoplasmas lack standardization. Furthermore, clinical breakpoints (CBPs) are not available to interpret AST data (i.e., Minimum Inhibitory Concentration, MIC values) and categorize isolates as susceptible, resistant, or intermediate to the different antimicrobials used in livestock, nor epidemiological cut-offs (ECOFF

Antibiotics that affect the ribosome.

The ribosome is a major bacterial target for antibiotics. Drugs inhibit ribosome function either by interfering in messenger RNA translation or by blocking the formation of peptide bonds at the peptidyl transferase centre. These effects are the consequence of the binding of drugs to the ribosomal subunits. Various mechanisms, including enzymatic detoxification, target alteration (ribosomal [r]RNAs and ribosomal proteins) and reduced accumulation (impermeability and efflux) are involved in bacterial resistance to protein synthesis inhibitors. The fact that some positions in rRNA participate in
