Alleviating ulcerative colitis via rectal administration: Enhancing the therapeutic effects of silk sericin-proanthocyanidins nanoparticles with a Poloxamer thermosensitive in situ gel.

Ulcerative colitis, a type of inflammatory bowel disease, primarily impacts the colon's mucous membrane, leading to distressing symptoms. Both healthcare and socioeconomic aspects are significantly impacted by this condition. In this study, we stabilized proanthocyanidins (PAC) with silk sericin (SS) to create SS/PAC nanoparticles. These nanoparticles were then loaded into a thermosensitive in situ hydrogel made with Poloxamer. The effectiveness and safety of the resulting SS/PAC/P thermosensitive hydrogel were evaluated using a dextran sulfate sodium (DSS)-induced model of ulcerative colitis,

Comparison of a poloxamer-based thermally sensitive gel and an HPMC-based standard gel used to increase sildenafil citrate retention in the uterus.

OBJECTIVE: Sildenafil citrate (SC) could promote uterine lining development, leading to successful embryo implantation. Due to the low oral bioavailability of the first-pass elimination of SC, it is necessary to develop a gel for vaginal administration to improve the efficacy. METHODS: A vaginal gel was prepared using the cold method to increase the retention of SC in the uterus, which allows for prolonged contact between SC and the vaginal mucosa. A poloxamer-based thermosensitive in situ gel containing SC (SC-Tsgel) and an HPMC-based standard gel containing SC (SC-Cogel) were prepared. The

Hyaluronic acid on the urokinase sustained release with a hydrogel system composed of poloxamer 407: HA/P407 hydrogel system for drug delivery.

Pleural empyema is an inflammatory condition characterized by accumulation of pus inside the pleural cavity, which is usually followed by bacterial pneumonia. During the disease process, the pro-inflammatory and pro-fibrotic cytokines in the purulent pleural effusion cause proliferation of fibroblasts and deposition of extracellular matrix, which lead to fibrin deposition and fibrothorax. Urokinase instillation therapy through a chest drainage tube is frequently used for fibrinolysis in patients with empyema. However, urokinase treatment requires multiple instillation (2-3 times per day, for 4

Safety of purified poloxamer 188 in sickle cell disease: phase I study of a non-ionic surfactant in the management of acute chest syndrome.

Acute chest syndrome (ACS) is the most common cause of death in patients with sickle cell anemia. Its management is primarily palliative. We performed a Phase I evaluation of purified poloxamer 188 (a non-ionic surfactant) in the management of ACS. Forty-three patients with sickle cell disease and ACS were treated with doses as high as 2960 mg/day by continuous intravenous (IV) infusion. The maximum tolerated dose has not been identified. No evidence of renal toxicity or other limiting adverse events were found. One adult patient died due to sepsis and adult respiratory distress syndrome, whic

Biological evaluation and in vivo degradation steady-state endpoint exploration of a sodium alginate/poloxamer composite temperature-sensitive embolic gel for tumors.

In the regulatory science of absorbable medical devices, traditional in vivo degradation evaluations are often hindered by lengthy evaluation cycles and ambiguous standards. This restricts the research, development, and translation of novel embolic materials. The lack of an evaluation system to determine whether tissue responses have reached a "degradation steady-state endpoint" is particularly critical. This study systematically evaluated the biological efficacy and in vivo degradation steady state of a composite temperature-sensitive embolic agent based on sodium alginate and poloxamer. This

Three-dimensional structure micelles of heparin-poloxamer improve the therapeutic effect of 17β-estradiol on endometrial regeneration for intrauterine adhesions in a rat model.

Intrauterine adhesions (IUA) frequently occur after infectious or mechanical injury to the endometrium, which may lead to infertility and/or pregnancy complications. There are few effective treatments due to the complex function of endometrium and shortage of native materials. 17β-estradiol (E2) is commonly used as an ancillary treatment in IUA patients, but it is limited by its poor solubility in aqueous solutions and low concentrations at the injured sites. In this research, a mini-endometrial curette was used to injure the rat's endometrium to form an IUA model. 17β-estradiol was encapsulat

Multifunctional poloxamer-based thermo-responsive hydrogel loaded with human lactoferricin niosomes: In vitro study on anti-bacterial activity, accelerate wound healing, and anti-inflammation.

Chronic wound infections are attributed to delayed tissue repair, which remains a major clinical challenge in long-term health care. Particularly, infections with antibiotic resistance have more serious effects on health, often resulting in unsuccessful treatments. Thus, antimicrobial peptide (AMP)-based therapy holds promise as a potential therapeutic approach to overcoming drug resistance. Conventional wound dressing is a passive strategy for wound care that is not capable of eradicating pathogens and promoting tissue repair. In this study, we aim to construct an advanced wound dressing; a t
