Constitutive expression of the poplar WRKY transcription factor PtoWRKY60 enhances resistance to Dothiorella gregaria Sacc. in transgenic plants.

WRKY proteins are involved in various physiological processes in plants, especially in coping with diverse biotic and abiotic stresses. However, limited information is available on the roles of specific WRKY transcription factors in poplar defense. In this study, we reported the characterization of PtoWRKY60, a Group IIa WRKY member, from Populus tomentosa Carr. The gene expression profile of PtoWRKY60 in various tissues showed that it significantly accumulated in old leaves. Phylogenetic analyses revealed that PtoWRKY60 had a close relationship with AtWRKY18, AtWRKY40 and AtWRKY60. PtoWRKY60

Salicylate and catechol levels are maintained in nahG transgenic poplar.

Metabolic profiling was used to investigate the molecular phenotypes of a transgenic Populus tremula x P. alba hybrid expressing the nahG transgene, a bacterial gene encoding salicylate hydroxylase that converts salicylic acid to catechol. Despite the efficacy of this transgenic approach to reduce salicylic acid levels in other model systems and thereby elucidate roles for salicylic acid in plant signaling, transgenic poplars had similar foliar levels of salicylic acid and catechol to that of non-transformed controls and exhibited no morphological phenotypes. To gain a deeper understanding of

European elms as potential hosts of Saperda tridentata (Coleoptera: Cerambycidae).

The elm borer, Saperda tridentata (Olivier, 1795) (Coleoptera: Cerambycidae), is a North American longhorn beetle that has been repeatedly intercepted in Europe on imported elm logs. Although climatic conditions are considered suitable for its establishment, uncertainty remains regarding the suitability of European tree species as hosts. We evaluated the potential host range of S. tridentata by testing 4 elm species (Ulmus glabra, Ulmus laevis, Ulmus minor, and Ulmus pumila), 3 maple species (Acer campestre, Acer platanoides, and Acer pseudoplatanus), and 3 poplar species (Populus alba, Populu

Let the BOOSTER boost photosynthesis and plant productivity.

Recently, Feyissa et al. discovered an orphan gene, BOOSTER (BSTR), in Populus species, which has been shown to optimize photosynthesis and enhance CO2 assimilation and biomass accumulation. Integrating BSTR into breeding programs holds promise for boosting crop yields and contributing to food security, increased biofuel production, and sustainable agriculture.

A new two-gene system of sex determination in a Salicaceae.

In angiosperms, dioecy has evolved thousands of times, and the pathways underlying the required floral changes are therefore expected to exhibit diversity as well as parallelism. Here we investigate Itoa orientalis, a dioecious Salicaceae in which immature flowers are bisexual, but the stamens or pistils then abort. This contrasts with the floral development in dioecious species of Populus and Salix, which lack any morphologically bisexual stage. A haplotype-resolved Itoa genome assembly revealed an XY system of sex determination with a sex-determining region (SDR) spanning ∼6 Mb and encompass

Poplar metal tolerance protein 1 confers zinc tolerance and is an oligomeric vacuolar zinc transporter with an essential leucine zipper motif.

Cation diffusion facilitator (CDF) proteins are a recently discovered family of cation efflux transporters that might play an essential role in metal homeostasis and tolerance. Here, we describe the identification, characterization, and localization of PtdMTP1, a member of the CDF family from the hybrid poplar Populus trichocarpa x Populus deltoides. PtdMTP1 is expressed constitutively and ubiquitously, although at low levels. Heterologous expression in yeast showed that PtdMTP1 was able to complement the hypersensitivity of mutant strains to Zn but not to other metals, including Cd, Co, Mn, a

The lignin deposition responses of different poplar root systems to salt stress.

Salt stress significantly constrains plant growth and development, with the root system serving as the primary organ for sensing and responding to salinity. Poplar (Populus spp.), a widely cultivated tree species, plays a pivotal role in urban greening, windbreak and sand fixation, and various economic sectors. Six poplars as experimental materials to analyze lignin deposition in the root system and variations in the casparian strip under salt stress. Lignin enrichment and changes in the casparian strip are as follows: P. alba × P. glandulosa '84K' showed the most pronounced changes, followed

Host-associated populations in the lettuce root aphid, Pemphigus bursarius (L.).

Pemphigus bursarius is a host-alternating aphid in which annual rounds of sexual reproduction on its primary host, Populus nigra, are interspersed with parthenogenesis on a range of secondary hosts. Evidence was sought for the existence of genetically distinct populations, associated with different secondary hosts, in P. bursarius. Microsatellite markers revealed that genetically distinct populations were present on three different secondary host species. Microsatellites were also used, in conjunction with mitochondrial DNA sequence variation, to investigate the relationships between aphids on

Genome mining and metabolomics reveal context-dependent biosynthetic potential in a core poplar dark septate endophyte.

Dark septate endophytes (DSEs) are ubiquitous root-colonizing fungi characterized by melanized, septate hyphae and a frequent association with plants in stressful or nutrient-poor environments. Although DSEs are increasingly recognized for context-dependent effects on host stress resilience, the specialized metabolic capacity of many DSE lineages remains poorly resolved. Hyaloscypha finlandica is a recurrent root-associated fungus of Populus and other hosts, making it a useful system for understanding symbiotic mechanisms using multi-omics analyses. Here, we combined comparative biosynthetic g

Over-expression of the triploid white poplar PtDrl01 gene in tobacco enhances resistance to tobacco mosaic virus.

A full-length cDNA, designated as the Populus tomentosa disease resistance-like 01 (PtDrl01) gene, was isolated from triploid white poplar [(Populus tomentosa × P. bolleana) × P. tomentosa]. The protein thought to be produced by the PtDrl01 gene contains a nuclear localisation sequence (NLS), a toll/interleukin-1 receptor (TIR) homologue region, a nucleotide binding site (NBS) and a leucine-rich repeat (LRR) domain. The protein also exhibits a considerable degree of homology to N-like resistance proteins. Real-time quantitative RT-PCR analysis revealed that expression of the PtDrl01 gene in tr

Foliar bacterial communities of trembling aspen in a common garden.

Microbial associations with plants are widely distributed and are structured by a number of biotic and physical factors. Among biotic factors, the host plant genotype may be integral to these plant-microbe interactions. Trees in the genus Populus have become models for studies in scaling effects of host plant genetics and in plant-microbe interactions. Using 454 pyrosequencing of the 16S rRNA gene, we assessed the foliar bacterial community of 7 genotypes of mature trembling aspen trees (Populus tremuloides Michx.) grown in a common garden. Trees were selected based on prior analyses showing c

RPA-CRISPR/Cas12a-Mediated Isothermal Amplification Technology for Visual Detection of Fusarium proliferatum.

Fusarium proliferatum is a fungal pathogen with an exceptionally broad host range, infecting ornamental plants and forest trees such as Populus and Cedrus deodara, and causing heart rot, root rot, and other diseases that lead to severe yield losses. It can also produce mycotoxins, including fumonisins, which threaten human and animal health. Conventional detection relies on isolation, culture, and morphological identification, which are time-consuming and prone to misidentification. Here, we established a rapid, visual molecular detection method by combining recombinase polymerase amplificatio

Insights into the Molecular Regulation of Lignin Content in Triploid Poplar Leaves.

After polyploidization, plants usually undergo some morphological and physiological changes, including the lignin content of polyploids usually becoming lower than that of diploids. However, the regulatory mechanism of the variation of lignin content in polyploid plants remains unclear. Therefore, in this research, we used full-sib poplar triploids and diploids to explore the molecular regulatory basis of lignin content in poplar triploid leaves through the determination of lignin content, the observation of xylem cells, and transcriptome sequencing. The results showed that the lignin content

Occurrence of elemental exchange in catalytic pyrolysis of poplar sawdust with homologous biochar catalysts doped with nitrogen.

Volatiles-char interactions during pyrolysis are well established, driven by polar oxygen-containing biochar functionalities. Adding polar nitrogen groups to biochar may alter volatiles evolution. This study explored catalytic pyrolysis of poplar sawdust using N-doped biochar (from melamine-impregnated biomass), with focus on elemental exchange of the biochar catalyst during catalyzing conversion of volatiles. The results showed that N-biochar with high nitrogen content lost 8.7 % its mass after catalysis, due to the transfer of organics to volatiles. This suppressed phenols, ketones, and alde

Unveiling drastic influence of cross-interactions in hydrothermal carbonization of spirulina with cellulose, lignin or poplar on nature of hydrochar and activated carbon.

Spirulina platensis contains abundant nitrogen-containing organics, which might react with derivatives of cellulose/lignin during hydrothermal carbonization (HTC), probably affecting yield, property of hydrochar, and pore development in activation of hydrochar. This was investigated herein by conducting co-HTC of spirulina platensis with cellulose, lignin, and sawdust at 260 °C and subsequent activation of the resulting hydrochars with K2C2O4 at 800 °C. The results showed that cross-condensation of spirulina platensis-derived proteins with cellulose/lignin-derived ketones and phenolics did tak

Active phytoextraction of toluene shifts the microbiome and enhances degradation capacity in hybrid poplar.

Hybrid poplars are widely recognized for their effectiveness in remediating subsurface aromatic hydrocarbon contaminants, including benzene, toluene, ethylbenzene, and xylene isomers (BTEX). While BTEX compounds are frequently found in the transpiration streams of poplars at contaminated sites, the microbial dynamics within these trees, particularly in response to hydrocarbon exposure, remain underexplored. This study utilized high-throughput amplicon sequencing to investigate the trunk microbiome in hybrid poplars at a field-scale toluene phytoremediation site. Across the plant growth season

Genotypic variation rather than ploidy level determines functional trait expression in a foundation tree species in the presence and absence of environmental stress.

BACKGROUND AND AIMS: At the population level, genetic diversity is a key determinant of a tree species' capacity to cope with stress. However, little is known about the relative importance of the different components of genetic diversity for tree stress responses. We compared how two sources of genetic diversity, genotype and cytotype (i.e. differences in ploidy levels), influence growth, phytochemical and physiological traits of Populus tremuloides in the presence and absence of environmental stress. METHODS: In a series of field studies, we first assessed variation in traits across diploid

[Preliminary Screening for the Urban Forest Against Combined Air Pollution].

Combined air pollution has become one of the most important city diseases in China. The construction of an urban forest not only needs landscape aesthetics, but also requires selecting a plant of high comprehensive tolerance threshold based on the needs of the ecological environment of each city, which has become a standard to maintain the sustainable development of the urban forest ecological function under environmental pollution. According to the comprehensive factor analysis of the sorption and absorption capacity of 537 plants to six air pollutants (i.e., sulfur dioxide, nitrogen dioxide,

PtrWRKY73, a salicylic acid-inducible poplar WRKY transcription factor, is involved in disease resistance in Arabidopsis thaliana.

A salicylic acid-inducible WRKY gene, PtrWRKY73, from Populus trichocarpa , was isolated and characterized. Overexpression of PtrWRKY73 in Arabidopsis thaliana increased resistance to biotrophic pathogens but reduced resistance against necrotrophic pathogens. WRKY transcription factors are commonly involved in plant defense responses. However, limited information is available about the roles of the WRKY genes in poplar defense. In this study, we isolated a salicylic acid (SA)-inducible WRKY gene, PtrWRKY73, from Populus trichocarpa, belonging to group I family and containing two WRKY domains,
