Effect of Bushen Yutai Recipe on IVF Patients Subjected to Mild Ovarian Stimulation.

This article investigated the effects of the traditional Chinese medicine (TCM) herbal recipe, Bushen Yutai, on in vitro fertilization (IVF) patients subjected to mild ovarian stimulation. Two hundred nineteen infertile patients were randomly divided into 2 groups: the control group and herbal treatment group. By studying, we found estrogen levels (E2) on the human chorionic gonadotropin (hCG) triggering day were significantly lower in the control group (P < 0.05), with positive blood flow being less detected by ultrasound scanning on both the day of hCG triggering and day of fresh embryo tran

Utilizing semantic intrusions to identify amyloid positivity in mild cognitive impairment.

OBJECTIVE: Semantic intrusion (SI) errors may highlight specific breakdowns in memory associated with preclinical Alzheimer disease (AD); however, there have been no investigations to determine whether SI errors occur with greater frequency in persons with amnestic mild cognitive impairment (aMCI) confirmed as amyloid positive (Amy+) vs those who have clinical symptoms of aMCI-AD with negative amyloid scans (suspected non-AD pathology [SNAP]) or persons who are diagnosed with other brain disorders affecting cognition. METHODS: Eighty-eight participants with aMCI underwent brain amyloid PET an

The stop signal stepping task: how action cancellation commands disrupt step initiation in young and healthy older adults.

Action cancellation - the ability to rapidly cancel an initiated movement in response to unexpected events - has been extensively studied in the upper limb using the stop signal task (SST). During gait, action cancellation is critical to stop and modify steps to avoid unexpected hazards and prevent falls. By adapting the SST to step initiation, this study investigated how the anticipatory postural adjustment (APA) and foot-lift phases of forward stepping were influenced by action-cancellation commands, and whether these processes changed with healthy ageing. Twenty-seven young (M = 28.7 years)

Unveiling the potential abuse liability of α-D2PV: A novel α-carbon phenyl-substituted synthetic cathinone.

Synthetic cathinones are emerging psychoactive substances designed to mimic the effects of classical psychostimulants. Among them, α-D2PV, a novel pyrrolidine-containing cathinone characterized by a phenyl group on the α-carbon atom, has gained significant attention. This study investigates the in vitro and in silico mechanism of action as well as the abuse liability of α-D2PV using rodent models. Dopamine (DA), noradrenaline (NE), and serotonin (5-HT) uptake inhibition assays were conducted in HEK293 cells expressing the corresponding human monoamine transporter, complemented by molecular doc

The impact of childhood deprivation on adult neuropsychological functioning is associated with ADHD symptom persistence.

BACKGROUND: Institutional deprivation in early childhood is associated with neuropsychological deficits in adolescence. Using 20-year follow-up data from a unique natural experiment - the large-scale adoption of children exposed to extreme deprivation in Romanian institutions in the 1980s -we examined, for the first time, whether such deficits are still present in adulthood and whether they are associated with deprivation-related symptoms of attention-deficit/hyperactivity disorder (ADHD) and autism spectrum disorder (ASD). METHODS: Adult neuropsychological functioning was assessed across fiv

Statin use in transthyretin amyloid cardiomyopathy: Assessing tolerability and drug interaction risks with tafamidis.

BACKGROUND: Transthyretin cardiac amyloidosis (ATTR-CM) is a progressive cardiomyopathy treated with tafamidis, a transthyretin stabilizer that improves survival and quality of life. Tafamidis inhibits the breast cancer resistance protein (BCRP), raising concern for drug-drug interactions, particularly with rosuvastatin. OBJECTIVE: To evaluate the potential interaction between tafamidis and statins in patients with ATTR-CM. METHODS: We conducted a retrospective study of 103 patients with confirmed ATTR-CM receiving both tafamidis and statin therapy at Columbia University Irving Medical Cente

Normative event-related potentials from sensory and cognitive tasks reveal occipital and frontal activities prior and following visual events.

In the present study, we report the results from a large sample of participants (N = 136), selected based on their EEG quality, to obtain event-related potential (ERP) normative data. All participants were tested in Simple Response Task (SRT) and Discriminative Response Task (DRT). A subset of 36 participants was tested also in Passive Vision task. Both pre- and post-stimulus ERPs were analyzed and compared among different tasks. Spatiotemporal patterns of all the observed components were analyzed using source analysis. Beside the well-known ERP components, we also described recently identifie

New concepts in immunity to Neisseria gonorrhoeae: innate responses and suppression of adaptive immunity favor the pathogen, not the host.

It is well-known that gonorrhea can be acquired repeatedly with no apparent development of protective immunity arising from previous episodes of infection. Symptomatic infection is characterized by a purulent exudate, but the host response mechanisms are poorly understood. While the remarkable antigenic variability displayed by Neisseria gonorrhoeae and its capacity to inhibit complement activation allow it to evade destruction by the host's immune defenses, we propose that it also has the capacity to avoid inducing specific immune responses. In a mouse model of vaginal gonococcal infection, N

Essential elements of strategic information systems to guide focused HIV responses in south and south-east Asia.

Strategic information is fundamental to the formulation and delivery of effective HIV programmes, but generating that information is a complex process, for which most countries in south and south-east Asia lack capacity and supporting structures. Although most countries in this region have made tremendous strides in collecting relevant data, advances in the ability to translate that data into strategic information and evidence for action, have not kept pace. To overcome these shortcomings, this paper presents a comprehensive approach to collecting strategic information and building the capacit

Targeting TGF-beta in human keratinocytes and its potential role in wound healing.

The pathology of chronic dermal ulcers is characterized by excessive proteolytic activity which degrades extracellular matrix (required for cell migration) and growth factors and their receptors. The transforming growth factor-beta (TGF-beta) has been identified as an important component of wound healing. Recent developments in molecular therapy offer exciting prospects for the modulation of wound healing, specifically those targeting TGF-beta. We investigated expression of angiogenic bFGF and VEGF, and collagenases MMP-2 and MMP-9 in tissue samples from chronic dermal wounds by immunohistoche

Elevated representational similarity of voluntary action and inhibition in Tourette syndrome.

Many people with Tourette syndrome are able to volitionally suppress tics, under certain circumstances. To understand better the neural mechanisms that underlie this ability, we used functional magnetic resonance neuroimaging to track regional brain activity during performance of an intentional inhibition task. On some trials, Tourette syndrome and comparison participants internally chose to make or withhold a motor action (a button press), while on other trials, they followed 'Go' and 'NoGo' instructions to make or withhold the same action. Using representational similarity analysis, a functi

Effect and Mechanism of Acacetin Against Stress-Induced Gastric Mucosal Damage: Network Pharmacology, Molecular Docking and Experimental Verification.

Stress-induced gastric ulcers (SGU) are categorized as a stress-related mucosal disease (SRMD) that exhibits a high morbidity among ICU patients. However, due to the harmful side effects of proton pump inhibitors (PPIs), there is an urgent need for natural and proactive alternative methods in the treatment of SGU. Acacetin, a natural flavonoid, has demonstrated potential in alleviating gastric ulcers. To elucidate the effects of acacetin in SGU, in this study, gastric ulcers (GU)-related targets were retrieved from GeneCards, DisGeNET, and OMIM. A protein-protein interaction (PPI) network was

Finerenone in Asian patients with diabetic kidney disease: evaluating the evidence for "better efficacy".

Diabetic kidney disease (DKD) has emerged as the primary driver of kidney failure across the Asia-Pacific region. Thus, therapeutic interventions capable of effectively slowing the progression of chronic kidney disease (CKD) and reducing concomitant cardiovascular risk are of critical strategic importance. Finerenone, a novel non-steroidal mineralocorticoid receptor antagonist (nsMRA) approved for the treatment of DKD, exerts cardiorenal protective effects by selectively inhibiting the overactivation of the mineralocorticoid receptor (MR)-a key mediator of pathological inflammatory and fibroti

The C5a receptor impairs IL-12-dependent clearance of Porphyromonas gingivalis and is required for induction of periodontal bone loss.

The C5a anaphylatoxin receptor (C5aR; CD88) is activated as part of the complement cascade and exerts important inflammatory, antimicrobial, and regulatory functions, at least in part, via crosstalk with TLRs. However, the periodontal pathogen Porphyromonas gingivalis can control C5aR activation by generating C5a through its own C5 convertase-like enzymatic activity. In this paper, we show that P. gingivalis uses this mechanism to proactively and selectively inhibit TLR2-induced IL-12p70, whereas the same pathogen-instigated C5aR-TLR2 crosstalk upregulates other inflammatory and bone-resorptiv

Precision management of cenobamate in drug-resistant epilepsy: integrating pharmacogenetics, therapeutic drug monitoring, and real-world clinical strategies.

Cenobamate (CNB) demonstrates high efficacy in drug-resistant focal epilepsy, yet optimal management requires personalized strategies. This exploratory case series describes five patients selected from a cohort of 125 individuals treated with CNB, examining the relationship between pharmacogenetic (PGx) profiles and clinical outcomes. Clinical data and therapeutic drug monitoring (TDM) were integrated with targeted Next-Generation Sequencing of key metabolic genes (UGT2B7, UGT2B4, CYP2E1, CYP2B6, CYP2A6, CYP3A4, CYP2C19) to predict metabolizer phenotypes. Three patients (Cases 1, 3, 4) achieve

Neural correlates of asymmetric switch costs during affective task switching.

Task switching is widely used to investigate cognitive control, typically revealing switch costs-greater performance costs in switch than in repetition trials. Previous research reported affective asymmetric switch costs, with higher costs when switching towards an affective task compared with a switch towards a neutral task. This asymmetry can be explained by the inhibition of the affective task, which enables the performance of the neutral task, leading to increased costs for a switch from the neutral toward the affective task. In Experiment 1, we examined whether the affective task content

Tyrosine kinase inhibitors - balancing the haemostatic scales: a review of associated thrombosis and bleeding.

Tyrosine kinase inhibitors (TKIs) have revolutionised cancer therapy, significantly impacting survival and outcomes by targeting specific signalling pathways that are necessary for tumour survival. Despite their clinical efficacy, TKIs exhibit a complex toxicity profile. Many of the signalling pathways that are targeted by TKIs are shared with normal homeostatic processes, including those responsible for modulating thrombosis and bleeding. The risk profile of thrombosis and bleeding associated with TKIs varies considerably across agents. Multi-kinase inhibitors, particularly those targeting th

Cardiotoxicity of Antitumor Agents: Therapeutic Challenges in Heart Failure with Reduced and Preserved Ejection Fraction.

The remarkable evolution of oncological therapies has dramatically improved cancer survival rates but has simultaneously introduced a significant burden of cardiovascular complications. Cardio-oncology has emerged as a critical multidisciplinary field focused on mitigating the "collateral damage" of life-saving anticancer treatments, ranging from traditional chemotherapeutics to novel immunotherapies. This review provides a comprehensive analysis of the pathophysiological mechanisms, clinical phenotypes, and evolving management strategies for cancer therapy-related cardiac dysfunction (CTRCD).
