Plant-based edible emulsion gels from rye secalin for 3D-printed fat mimetics.

Plant proteins are lauded as sustainable proteins for climate control and have been intensively researched as muscle meat mimetics, yet far less attention has been paid to plant-based oil systems for mimicking fat tissues, which is critical in embarking flavor and texture of the meat. Herein, we developed fat mimetics from sunflower oil and secalin, a rye prolamin, which served as both an emulsifier and gelator. The mechanical properties of secalin-based emulsion gels (SEGs) could be fine-tuned by adjusting the ratio of sunflower oil to 70 % ethanol (O:E), acting as templating agent and solven

Extra-Intestinal Manifestations of Coeliac Disease in Children: Clinical Features and Mechanisms.

Celiac disease (CD) is a systemic autoimmune disease due to a dysregulated mucosal immune response to gluten and related prolamines in genetically predisposed individuals. It is a common disorder affecting ~1% of the general population, its incidence is steadily increasing. Changes in the clinical presentation have become evident since the 80s with the recognition of extra-intestinal symptoms like short stature, iron deficiency anemia, altered bone metabolism, elevation of liver enzymes, neurological problems. Recent studies have shown that the overall prevalence of extra-intestinal manifestat

Formulation of Eco-friendly Medicated Chewing Gum to Prevent Motion Sickness.

An attempt was made to formulate medicated chewing gum to prevent motion sickness using natural gum base for faster onset of action and easy administration, anywhere and anytime, without access to water. To avoid the discard issue of gum cud, natural gum base of Triticum aestivum (wheat grain) was explored because of its biodegradable and biocompatible nature and easy availability. Prolamin, extracted from wheat, showed good chewing capacity, elasticity, high water retention capacity, antifungal activity, and compatibility with the drug. Formulations were prepared based on a two-factor and thr

[Product development on the basis of cereal and leguminous flours to coeliac disease in children between 6-24 months; I: formulation and acceptability].

The revaluation of the Andean cultivations, quinua (Chenopodium quinua Willd) and lupin (Lupinus albus L.), to be used in nutritional mixtures, with traditional cereals like corn (Zea mays L.) and rice (Oryza sativa L.), originate mixtures without gluten which constitute a good alternative for the nutrition of children under 24 months that suffer from celiac disease, since they improve the quality of the protein, by essential amino acids compensation, and also impacts in the product's diversification strategy. In the present work, the percentage composition of each flour in the mixture was det

Zein adsorption to hydrophilic and hydrophobic surfaces investigated by surface plasmon resonance.

Zein, the prolamine of corn, has been investigated for its potential as an industrial biopolymer. In previous research, zein was plasticized with oleic acid and formed into sheets/films. Physical properties of films were affected by film structure and controlled in turn by zein-oleic acid interactions. The nature of such interactions is not well understood. Thus, protein-fatty acid interactions were investigated in this work by the use of surface plasmon resonance (SPR). Zein adsorption from 75% aqueous 2-propanol solutions, 0.05% to 0.5% w/v, onto hydrophilic and hydrophobic self-assembled mo

Chia seeds: an ancient grain trending in modern human diets.

Currently, in order to ensure adequate intake of nutrients to complement the normal diet, the consumption of seeds such as Salvia hispanica L. (commonly known as chia seeds) is increasing. For this reason, investigations concerning the composition and potential health effects of chia seeds are being carried out. Moreover, the recent approval of chia seeds as a Novel Food by the European Parliament allows its consumption and incorporation in a wide range of foods; thus, they have become widely available. Concerning their nutritional aspects, chia seeds are an excellent source of fat (20% to 34%

Celiac disease.

Clinically, celiac disease has always been regarded as a wasting, malabsorptive disorder due to disease of the small intestinal mucosa. It has been difficult for clinicians to recognize that this condition is primarily due to sensitization of mesenteric T lymphocytes to wheat protein (gluten) in genetically predisposed (DQ2+) individuals. On contact with dietary-derived gluten in the upper intestine, these sensitized T lymphocytes are activated leading to inflammation of and morphologically altered mucosal architecture: the latter reverts to normal with a gluten-free diet. The circulation of s

A chimeric rather than native secretion signal elicits stress-related transcriptional adaption during 75k γ-secalin production in Komagataella phaffii.

Komagataella phaffii (formerly Pichia pastoris) is a popular host for protein production, enabling their targeted application and in-depth study. Among relevant targets are gluten proteins, which are essential for cereal-based food properties, but can also trigger immune reactions such as coeliac disease. Previously, production of rye (Secale cereale) prolamin 75k γ-secalin in K. phaffii was shown to yield the highest titres with the native secretion signal compared to co- and post-translational signals commonly used for enhanced production. Here, we compared the host's transcriptomic response

A synthetic biomolecular condensate from plant proteins with controlled colloidal properties.

A synthetic biomolecular condensate (sBC) consisting of a prolamin-rich, plant protein, zein, has been engineered. These artificial condensates were prepared from the liquid-liquid phase (LLP) separation of a protein-rich liquid phase in water. To ensure the colloidal stability of the separated condensate, the protein was chemically modified either via quaternization with glycidyl trimethyl ammonium chloride (GTMAC) or covalently connecting poly(ethylene glycol) by reductive amination, respectively. The modified protein condensates, termed QZs and PZs (for quaternized and PEG-conjugated zein,

Portable gliadin-immunochip for contamination control on the food production chain.

Celiac disease (CD) is one of the most common digestive disorders caused by an abnormal immune reaction to gluten. So far there are no available therapies, the only solution is a strict gluten-free diet, which however could be very challenging as gluten can be hidden in many food products. Furthermore an additional problem is related to cross-contamination of nominal gluten-free foods with gluten-based ones during manufacturing. Here we propose a lab on chip platform as a powerful tool to help food manufacturers to evaluate the real amount of gluten in their products by an accurate in-situ con

Analysis of zein by matrix-assisted laser desorption/ionization mass spectrometry.

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI/MS) was used to analyze the protein composition of corn prolamine (zein). Mass spectra were obtained from commercial zein and zein extracted with aqueous 2-propanol and aqueous ethanol from consumer corn meal. For the commercial zein, three major zein fractions with m/z 26.8k, 24.1k, and 23.4k were clearly seen with two minor fractions (m/z 14.5k and 20.4k) also present. As compared with the results from sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), these three fractions were identified as alpha-zeins

Separation of alcohol-soluble proteins (zeins) from maize into three fractions by differential solubility.

The prolamin of maize (Zea mays L.), zein, was extracted from endosperm meal with 60% (v/v) 2-propanol/1% (v/v) 2-mercaptoethanol either directly or subsequent to extraction with 90% (v/v) 2-propanol. The zein extracted with 90% 2-propanol was essentially made up of 20 to 24 kilodalton polypeptides (alpha-zein) while that extractable with 60% 2-propanol/1% 2-mercaptoethanol contained, in addition to alpha-zein, 17 to 18 kilodalton methionine-rich polypeptides and a 27 kilodalton proline-rich polypeptide. While zein was separated into three fractions by differential solubility in 90% 2-propanol

The Combined Effect of Corn Zein and Buckwheat Flour on the Rheological, Microstructural, Physicochemical, and Nutritional Properties of Gluten-Free Bread.

The development of structurally stable and nutritionally enhanced gluten-free bread is a major technological challenge due to the absence of a gluten network. This study investigates the role of zein, a hydrophobic corn prolamin, in green buckwheat dough systems and its impact on bread properties. Zein was extracted from corn gluten and incorporated at 0, 10, 20, and 30%. Zein incorporation significantly modified starch-protein interactions, promoting a composite matrix with enhanced thermomechanical stability. The 20% zein formulation showed optimal performance, with increased peak viscosity,

Structure, Function and Protein Engineering of Cereal-Type Inhibitors Acting on Amylolytic Enzymes.

Numerous plants, including cereals, contain seed proteins able to inhibit amylolytic enzymes. Some of these inhibitors, the CM-proteins (soluble in chloroform:methanol mixtures)-also referred to as cereal-type inhibitors (CTIs)-are the topic of this review. CM-proteins were first reported 75 years ago. They are small sulfur-rich proteins of the prolamine superfamily embracing bifunctional α-amylase/trypsin inhibitors (ATIs), α-amylase inhibitors (AIs), limit dextrinase inhibitors (LDIs), and serine protease inhibitors. Phylogenetically CM-proteins are predicted across poaceae genomes and many

Breaking Down Barriers: How Understanding Celiac Disease Pathogenesis Informed the Development of Novel Treatments.

For decades, the pathogenesis of a variety of human diseases has been attributed to increased intestinal paracellular permeability even though scientific evidence supporting this hypothesis has been tenuous. Nevertheless, during the past decade, there have been a growing number of publications focused on human genetics, the gut microbiome, and proteomics, suggesting that loss of mucosal barrier function, particularly in the gastrointestinal tract, may substantially affect antigen trafficking, ultimately causing chronic inflammation, including autoimmunity, in genetically predisposed individual

Essential Oil Bioactive Fibrous Membranes Prepared via Coaxial Electrospinning.

A novel antimicrobial composite material was prepared by encapsulating orange essential oil (OEO) in zein prolamine (ZP) via the coaxial electrospinning (ES) technique. By manipulating process parameters, the morphological features of ZP/OEO fibers were modulated. Fine fibers with diameters ranging from 0.7 to 2.3 μm were obtained by regulating ZP solution concentration and process parameters during the ES process. Optimal loading capacity (LC) and encapsulation efficiency (EE) of OEO in fibrous ZP mats were determined to be 22.28% and 53.68%, respectively, and were achieved using a 35 w/v% ZP
