Molecular self-assembly, a phenomenon widely observed in nature, has been exploited through organic molecules, proteins, DNA, and peptides to study complex biological systems. These self-assembly systems may also be used in understanding the molecular and structural biology which can inspire the design and synthesis of increasingly complex biomaterials. Specifically, use of these building blocks to investigate protein folding and misfolding has been of particular value since it can provide tremendous insights into peptide aggregation related to a variety of protein misfolding diseases, or amyl

Extracellular vesicles (EVs) have attracted significant scientific attention due to their critical functions in intercellular communication and their possible uses in diverse fields such as immunology, therapeutics, reproductive biology, biotechnology, and medicine. EVs are engulfed in a layer of proteins, also known as protein corona, which is speculated to play roles in several areas, from intercellular communication through immune recognition to cargo delivery. The composition of the protein corona strongly depends on the origin and the biological environment of EVs. Understanding the prote

Because oral transmission of SARS-CoV-2 is 3-5 orders of magnitude higher than nasal transmission, we investigated debulking of oral viruses using viral trap proteins (CTB-ACE2, FRIL) expressed in plant cells, delivered through the chewing gum. In omicron nasopharyngeal (NP) samples, the microbubble count (based on N-antigen) was significantly reduced by 20 μg of FRIL (p < 0.0001) and 0.925 μg of CTB-ACE2 (p = 0.0001). Among 20 delta or omicron NP samples, 17 had virus load reduced below the detection level of spike protein in the RAPID assay, after incubation with the CTB-ACE2 gum powder. A d

The concept of protein corona formation around extracellular vesicles (EVs) has given birth to new insights into how cells may recognize EVs by proteins presented at the EV surface. Here we present spatially resolved proteomics using the biotin ligase TurboID to map proteins interacting at the EV surface, without a need for physical isolation of the EV-corona complexes. TurboID promiscuously biotinylates nearby proteins within a few nm distance from the fused 'bait' protein. We genetically engineered EVs by modifying CD63 as the bait protein to which TurboID was fused facing outward to map EV

For specific applications, gold nanoparticles (GNPs) are commonly functionalized with various biological ligands, including amino-free ligands such as amino acids, peptides, proteins, and nucleic acids. Upon entering a biological fluid, the protein corona that forms around GNPs can conceal the targeting ligands and sterically hinder the functional properties. The protein corona is routinely prepared by standard centrifugation or sucrose cushion centrifugation. However, such methodologies are not applicable to the exclusive analysis of a ligand-binding protein corona. In this study, we first pr

HYPOTHESIS: The formation of a biomolecular corona on nanoparticle surfaces significantly influences their biological behaviour, particularly in drug delivery applications. Despite the prevalence of ingestion of particles (e.g, during oral drug delivery), our understanding of corona formation within the gastrointestinal (GI) tract remains limited, especially for non-protein components. The hypothesis of this work is that the exposure of nanoparticles to bile components will form a "corona" structure and protein corona will represent proteomes different from the original bile fluid. Two major a

Understanding biomolecular coronas that spontaneously occur around nanocarriers (NCs) in biological fluids is critical to nanomedicine as the coronas influence the behaviors of NCs in biological systems. In contrast to extensive investigations of protein coronas over the past decades, understanding of the coronas of biomolecules beyond proteins, e.g., metabolites, has been rather limited despite such biochemicals being ubiquitously involved in the coronas, which may influence the bio-nano interactions and thus exert certain biological impacts. In this study, serum biomolecular coronas, in part

Understanding biomolecular coronas that spontaneously occur around liposomes in biological fluids is critical as both the lipid and protein coronas influence liposome behavior in biological systems. Herein, PEGylated liposomes were pre-incubated at varying plasma concentrations followed by in vitro dynamic simulation incubation. It was found that increasing plasma concentration from 10 % to 300 % resulted in reduced liposomes uptake by immune cells. Most plasma-derived lipids were found to be retained in the coronas but with altered abundances. Lipids such as CE 18:1 and PC 16:0/18:2 exhibited

Understanding the interactions between nanomaterials and biological systems plays a pivotal role in enhancing the efficacy of nanomedicine and advancing the disease diagnosis. The nanoparticle-protein corona, an active biomolecular layer, is formed around nanoparticles (NPs) upon mixing with biological fluid. The surface layer which consists of rapidly exchanged biomolecules is called the "soft" corona. The inner layer which is more stable and tightly packed is called the "hard" corona. It has been suggested that the NP-protein corona has a decisive effect on the in vivo fate of nanomedicine u

This study explored the influence of different sizes curcumin-loaded starch nanoparticles on protein corona formation and their interactions with pepsin and trypsin. SEM and AFM results showed the spherical shape of the nanoparticles. FTIR and fluorescence spectroscopy results reported the formation of enzymes coronas was primarily driven by hydrophobic interactions and hydrogen bonding. XRD results showed that curcumin was encapsulated within the nanoparticles, while the enzyme corona formation did not affect the stable encapsulation of curcumin. QCM-D results suggested that smaller-sized nan

Nanoparticles (NPs) may enter the human body through both unintentional routes, such as inhalation or ingestion, and intentional diagnostic or therapeutic applications, where they rapidly interact with surrounding biomolecules and form a dynamic corona that defines their biological identity. Despite significant attention to protein coronas, the function of lipids and lipid-associated molecules at the nano-bio interface has not been thoroughly explored. This review offers a comprehensive overview of the interactions between NPs and lipids, focusing on their role in the formation of lipid corona

Corona virus disease 2019 (Covid-19), a pandemia emerged recently, caused by severe acute respiratory syndrome corona virus 2 (SARS-CoV-2). The receptor for corona virus and influenza A is the mucosal cell membrane protein angiotensin converting enzyme 2 (ACE2), which is abundant on the membrane of alveolar cells and enterocytes. Viral spike protein 1 (S1) is the ligand, with an affinity of 14.7 nM to the receptor. The main port of entry for the virus is the upper respiratory tract, and the diagnosis is usually by PCR of the viral RNA with nasal and pharyngeal swab test. Human defensin 5 (HDEF

Nanoparticle performance in vivo is strongly shaped by interactions with the complex biological environment. Upon exposure to biological fluids, endogenous proteins rapidly adsorb onto nanoparticle surfaces to form a protein corona, which redefines the nanoparticle's biological identity and influences its biodistribution, cellular interactions, and tumor accumulation. Rather than viewing protein adsorption solely as a barrier, growing evidence suggests that endogenous proteins can be strategically leveraged to enhance nanoparticle tumor delivery. This review provides a comprehensive overview o

It is becoming increasingly accepted that various diseases have a capacity to alter the composition of plasma proteins. This alteration in protein composition may consequently change the targeting capacity of nanoparticles (NPs). In this study, the impact of a model targeting ligand's (i.e., Transferrin; Tf) concentration in human plasma on the targeting capacity of gold NPs (Au NPs), pre-conjugated with Tf, is investigated. Our findings demonstrate that the protein corona formation by both healthy and Tf depleted human plasma diminishes the targeting efficacy of Au NPs within human cancer cel

Immune checkpoint blockade (ICB) therapies targeting the PD-1/PD-L1 axis have shown clinical promise, yet most patients do not achieve durable responses due to adaptive PD-L1 upregulation and recycling. While nanoparticle-based proteolysis targeting chimeras (PROTACs) offer a strategy for direct PD-L1 degradation, their therapeutic potential remains largely untapped because of a fundamental barrier: the inevitable formation of a nonspecific protein corona that severely limits target engagement. Here, we introduce a corona-free nano-PROTACs platform (CF-nPTs) that overcomes this obstacle. By in

Point-of-care diagnostic tools, such as lateral flow assays (LFAs), play a critical role in disease management and outbreak control. LFAs detect the presence of target antigens in disease-relevant biofluids, utilizing nanoparticles (termed detection probes) to produce colorimetric readouts. However, significant intra- and interpatient variation in the biochemical composition of biofluids has downstream consequences for assay performance. Robust LFAs must be able to function alongside such variability to produce reliable and reproducible test outcomes. Beyond this, biofluids (such as serum) con

The interaction of nanoparticles (NPs) with biological environments triggers the formation of a protein corona (PC), which significantly influences their behavior in vivo. This review explores the evolving understanding of PC formation, focusing on the opportunity for decreasing or suppressing protein-NP interactions by macromolecular engineering of NP shells. The functionalization of NPs with a dense, hydrated polymer brush shell is a powerful strategy for imparting stealth properties in order to elude recognition by the immune system. While poly(ethylene glycol) (PEG) has been extensively us

Nanomaterials, like nanoparticles, micelles, nano-sheets, nanotubes and quantum dots, have great potentials in biomedical fields. However, their delivery is highly limited by the formation of protein corona upon interaction with endogenous proteins. This new identity, instead of nanomaterial itself, would be the real substance the organs and cells firstly encounter. Consequently, the behavior of nanomaterials in vivo is uncontrollable and some undesired effects may occur, like rapid clearance from blood stream; risk of capillary blockage; loss of targeting capacity; and potential toxicity. Strategies to modify nanoparticles with biological ligands for targeted drug delivery in vivo have been widely studied but met with limited clinical success. A possible reason is that, in the blood circulation, serum proteins could rapidly form a layer of protein "corona" on the vehicle surface, which might block the modified ligands and hamper their targeting functions. We speculate that strategies for drug delivery can be designed based upon elegant control of the corona formation on the vehicle surfaces. In this study, we demonstrate a retinol-conjugated polyetherimine (RcP) nanoparticle sy

Lipid nanoparticles (LNPs) have reshaped RNA therapeutics, underpinning the success of mRNA vaccines and emerging gene therapies. Once introduced into the body, LNPs acquire a dynamic biological identity through the rapid adsorption of biomolecules present in biological fluids, resulting in the formation of a biomolecular corona. Rather than being a passive byproduct, the biomolecular corona substantially alters the physicochemical properties of LNPs and governs key biological processes, including cellular uptake, immune recognition, circulation, and biodistribution, therefore acting as a func
