An Engineered Minimal WASP-Myosin Fusion Protein Reveals Essential Functions for Endocytosis.

Actin polymerization powers membrane deformation during many processes, including clathrin-mediated endocytosis (CME). During CME in yeast, actin polymerization is triggered and coordinated by a six-protein WASP/Myosin complex that includes WASP, class I myosins (Myo3 and Myo5), WIP (Vrp1), and two other proteins. We show that a single engineered protein can replace this entire complex while still supporting CME. This engineered protein reveals that the WASP/Myosin complex has four essential activities: recruitment to endocytic sites, anchorage to the plasma membrane, Arp2/3 activation, and tr

Proteomic analysis of Bacillus subtilis strains engineered for improved production of heterologous proteins.

The use of bacterial systems for recombinant protein production has advantages of simplicity, time and cost over competing systems. However, widely used bacterial expression systems (e.g. Escherichia coli, Pseudomonas fluorescens) are not able to secrete soluble proteins directly into the culture medium. This limits yields and increases downstream processing time and costs. In contrast, Bacillus spp. secrete native enzymes directly into the culture medium at grams-per-litre quantities, although the yields of some recombinant proteins are severely limited. We have engineered the Bacillus subtil

Engineered antibody therapies to counteract mutant huntingtin and related toxic intracellular proteins.

The engineered antibody approach to Huntington's disease (HD) therapeutics is based on the premise that significantly lowering the levels of the primary misfolded mutant protein will reduce abnormal protein interactions and direct toxic effects of the misfolded huntingtin (HTT). This will in turn reduce the pathologic stress on cells, and normalize intrinsic proteostasis. Intracellular antibodies (intrabodies) are single-chain (scFv) and single-domain (dAb; nanobody) variable fragments that can retain the affinity and specificity of full-length antibodies, but can be selected and engineered as

Silver(I)-mediated amide-to-ester transformation on unprotected peptides for protein chemical synthesis and engineering.

Chemical protein synthesis has transformed the preparation of homogeneous peptides and proteins with site-specific modifications. Serine/threonine ligation offers a practical route to access difficult peptides and proteins via its aggregation-breaking intermediate, yet the preparation and regeneration of the prerequisite peptidyl salicylaldehyde ester still require the development of more efficient chemistry. Here, we report a silver(I)-mediated amide-to-ester transformation on unprotected peptides, enabled by an easily introduced N,S-benzylidene thioacetal scaffold. This method can chemoselec

Coordination-Driven Programming of MOF Stability via Targeting Protein Corona Engineering for Hypoxia-Relieving Sonodynamic Therapy.

Zr-based metal-organic frameworks (MOFs) are promising nanomedicine platforms, but their rapid degradation in phosphate-rich biological environments remains a critical barrier to clinical translation. Here, incorporating MnOx into the porphyrinic Zr-MOF PCN-224 modulates the coordination environment of Zr6 clusters, suppressing phosphate-induced linker displacement and extending structural integrity under physiological conditions. The optimized 1:1 MnOx@PCN-224 formulation resists phosphate-triggered disassembly and porphyrin release in phosphate-buffered saline (PBS). MnOx also acts as a cata

Protein biochemistry and engineering drive the development of a carbonic anhydrase-based carbon dioxide sequestration strategy.

The sequestration of carbon dioxide using carbonic anhydrase (CA) is one of the most effective methods for mitigating global warming. The burning of fossil fuels releases large quantities of flue gas; because of its high temperature and of the alkaline conditions required for CaCO3 precipitation in the mineralization process, thermo-alkali-stable CAs are needed. In this context, Manyumwa et al. conducted a biochemical characterization of three CAs derived from thermophilic bacteria. They then employed a rational design approach to enhance the specific activity and stability of the enzyme from

Protein engineering reveals that gasdermin A preferentially targets mitochondrial membranes over the plasma membrane during pyroptosis.

When activated, gasdermin family members are thought to be pore-forming proteins that cause lytic cell death. Despite this, numerous studies have suggested that the threshold for lytic cell death is dependent on which gasdermin family member is activated. Determination of the propensity of various gasdermin family members to cause pyroptosis has been handicapped by the fact that for many of them, the mechanisms and timing of their activation are uncertain. In this article, we exploit the recently discovered exosite-mediated recognition of gasdermin D (GSDMD) by the inflammatory caspases to dev

Engineering the ChlorON Series: Turn-On Fluorescent Protein Sensors for Imaging Labile Chloride in Living Cells.

Beyond its role as the "queen of electrolytes", chloride can also serve as an allosteric regulator or even a signaling ion. To illuminate this essential anion across such a spectrum of biological processes, researchers have relied on fluorescence imaging with genetically encoded sensors. In large part, these have been derived from the green fluorescent protein found in the jellyfish Aequorea victoria. However, a standalone sensor with a turn-on intensiometric response at physiological pH has yet to be reported. Here, we address this technology gap by building on our discovery of the anion-sens

Engineering Protein Nanoparticles Functionalized with an Immunodominant Coxiella burnetii Antigen to Generate a Q Fever Vaccine.

Coxiella burnetii is the causative agent of Q fever, for which there is yet to be an FDA-approved vaccine. This bacterial pathogen has both extra- and intracellular stages in its life cycle, and therefore both a cell-mediated (i.e., T lymphocyte) and humoral (i.e., antibody) immune response are necessary for effective eradication of this pathogen. However, most proposed vaccines elicit strong responses to only one mechanism of adaptive immunity, and some can either cause reactogenicity or lack sufficient immunogenicity. In this work, we aim to apply a nanoparticle-based platform toward produci

Cryo-EM, Protein Engineering, and Simulation Enable the Development of Peptide Therapeutics against Acute Myeloid Leukemia.

Cryogenic electron microscopy (cryo-EM) has emerged as a viable structural tool for molecular therapeutics development against human diseases. However, it remains a challenge to determine structures of proteins that are flexible and smaller than 30 kDa. The 11 kDa KIX domain of CREB-binding protein (CBP), a potential therapeutic target for acute myeloid leukemia and other cancers, is a protein which has defied structure-based inhibitor design. Here, we develop an experimental approach to overcome the size limitation by engineering a protein double-shell to sandwich the KIX domain between apofe

Protein engineering of the chemokine CCL20 prevents psoriasiform dermatitis in an IL-23-dependent murine model.

Psoriasis is a chronic inflammatory skin disease characterized by the infiltration of T cell and other immune cells to the skin in response to injury or autoantigens. Conventional, as well as unconventional, γδ T cells are recruited to the dermis and epidermis by CCL20 and other chemokines. Together with its receptor CCR6, CCL20 plays a critical role in the development of psoriasiform dermatitis in mouse models. We screened a panel of CCL20 variants designed to form dimers stabilized by intermolecular disulfide bonds. A single-atom substitution yielded a CCL20 variant (CCL20 S64C) that acted a

Engineering, expression, and immuno-characterization of recombinant protein comprising multi-neutralization sites of rabies virus glycoprotein.

The rabies virus (RV) glycoprotein (G protein) induces neutralizing antibodies, which are important in protection against rabies. In the present study, three gene fragments that encode polypeptides (corresponding to amino acid residues 19-60, 181-219, and 300-458) comprising the linear neutralization sites of the G protein were spliced together in tandem by PCR-based site-directed mutagenesis and heterologously expressed in Escherichia coli (DE3). The recombinant protein (designated rRVg) was purified under denaturing conditions and solubilized by stepwise dialysis against an alkaline buffer (

Improving protein secretion in yeasts: From secretion leader engineering and AI-driven prediction to bioprocess performance.

Yeast is widely used for recombinant protein production because it supports eukaryotic protein expression and high-cell-density cultivation. Efficient secretion depends on secretion leaders, short N-terminal sequences that direct nascent polypeptides into the secretory pathway. Multiple engineering strategies have been developed to enhance secretion efficiency. These include rational modification of physicochemical properties of secretion leaders, hybrid leader design through the combination of distinct signal elements, codon optimization of the leader sequence, and fusion with translational f

Protein Phase Transition Engineering of Nanochannels: A Stepwise Functionalization Strategy for Sequential Ultrasensitive Metal Ion Detection.

Water-soluble heavy metal ions pose persistent threats to ecosystems and human health, necessitating highly sensitive and selective detection strategies. Herein, we report a nanochannel sensing platform for sequential ion detection, constructed via a protein-phase-transition-induced in situ interface engineering strategy. Bovine serum albumin (BSA) is converted into phase-transited BSA (PTB) through tris(2-carboxyethyl)phosphine (TCEP)-mediated disulfide bond cleavage, forming a uniform PTB layer within conical nanochannels. The abundant thiol (-SH) groups on PTB enable ultrasensitive Hg2+ det

Engineering 3'-UTR hairpin structures to modulate mRNA stability and recombinant protein production in Escherichia coli.

Engineering mRNA stability is a promising yet underexplored approach for improving recombinant protein production in bacterial systems. In this study, we evaluated the effect of synthetic 3'-UTR hairpin structures on mRNA stability and protein yield in Escherichia coli using two SUMO-fusion expression systems. Hairpin elements with defined structural features were introduced downstream of the coding sequence. In all constructs, 3'-UTR hairpins increased mRNA half-life, with stabilization ranging from approximately 2-fold to 3-fold (n = 3 biological replicates). In the SUMO-SARS-CoV-2-derived p

Mucoadhesive recombinant human catalase for dry eye disease: From protein engineering to first-in-human study.

BACKGROUND: Oxidative stress caused by excess reactive oxygen species (ROS) plays a key role in the progression of dry eye disease (DED), yet no approved DED therapy directly eliminates ROS. Here, we develop an antioxidant biologic with enhanced ocular-surface retention by thiol engineering of recombinant human catalase (hCAT) as a new ROS-scavenging eye drop therapy for DED. METHODS: Thiolated hCAT (hCAT-SH) was characterized for activity, stability, mucin interaction, and ocular retention. Efficacy, mechanism, and safety were evaluated in mouse, rabbit, and rat studies, followed by a random

Semi-automated strain engineering and screening in the thermophilic fungus Myceliophthora thermophila for recombinant protein production.

UNLABELLED: Thermophilic filamentous fungi are widely used for cellulase production at elevated temperatures; however, efficient recombinant protein production in these organisms remains limited because construction and screening of large transformant populations remain difficult. Here, Myceliophthora thermophila was engineered into a thermophilic fungal chassis through combinatorial deletion of genes associated with proteolysis, vesicle trafficking, and secretion, resulting in a hyper-secretory strain. An optimized expression cassette coupled with a laccase reporter enabled activity-based ide

Methylotrophic yeast engineering for secreted protein production via precision fermentation: food ingredient proteins and synthetic biology strategies.

Methylotrophic yeasts are outstanding platforms for the production of recombinant proteins through precision fermentation. Growing demand for protein-based food ingredients requires economically viable secretion efficiency and space-time yields, as well as correct post-translational modifications to ensure proper food functionality. Here, we review recent advances and future potential in engineering methylotrophic yeasts for food protein production, with particular focus on Komagataella phaffii. We first summarise the production of three categories of protein food ingredients: recombinant dair

Engineering Enhanced Interface Attachment for Sustained Ocular Drug Delivery: Mussel Foot Protein Incorporated Pullulan/Gellan Gum Nanofibers.

Effective treatment of ocular inflammation is hindered by limited drug uptake and rapid clearance, resulting in poor bioavailability and frequent dosing of conventional eye drops. To improve drug retention and bioavailability, we designed and manufactured electrospun mucoadhesive nanofibers based on modified mussel foot protein (mMefp-3). Their composition and small diameter (≈250 nm) provide a high surface area, enabling rapid water uptake and in situ gelation on the ocular surface. SEM, light, and fluorescence microscopy confirmed homogeneous, smooth, and bead-free fibers. Ocular residence s

Engineering enhanced signal peptides: A high-throughput computational pipeline for optimizing therapeutic protein production in CHO cells.

Rational design of signal peptides (SPs), crucial for efficient therapeutic protein secretion in Chinese hamster ovary (CHO) cells, remains challenging due to their context-dependency activity. To overcome this limitation and enable the discovery of novel high-performance SPs, we developed a high-throughput computational screening pipeline. This approach leverages the deep learning model SignalP 6.0 to screen millions of SP variants derived from diverse mouse/human wild-type libraries and C-region mutants. As a demonstration of its broad applicability, we applied this pipeline to optimize SPs
