Exploring protandry and pupal size selection for Aedes albopictus sex separation.

BACKGROUND: We explored the possibility to improve male/female separation (sexing) in Aedes albopictus by selection of two strains, one toward increasing sex dimorphism and another toward increasing protandry. In the laboratory we selected and crossed small males with large females to exploit dimorphism, and early pupating males with late pupating females to exploit protandry. RESULTS: While selection for enhanced dimorphism was not a profitable character, the selection for enhanced protandry up to F10 produced significant improvement in the time interval between male and female pupation. [Interhost migration behaviour of Ctenocephalides felis on cats and in their resting sites].

The cat flea, Ctenocephalides felis, maintains its life cycle indoors feeding on pets. However, it must be emphasized that only about 1-5% of the flea population is on their hosts, the remaining 95-99% (eggs, larvae and pupae) are spread around the habitat. The eggs and dried blood feces fall from the coat of the animal on its sleeping, resting or feeding sites. In the first study, 100 pupae of the cat flea were applied on the resting sites of 6 cats' (3 female, 3 male), each in a separate box. Five days later all 6 cats were examined for adult fleas. From the 600 applied pupae a total of 326

Development of a Japanese encephalitis virus-like particle vaccine in silkworms using codon-optimised prM and envelope genes.

We have successfully prepared a Japanese encephalitis virus (JEV) - Nakayama virus like particle (NVLP) vaccine using synthetic codon-optimized prM and E genes. The expression of the recombinant JEV Nakayama-BmNPV (JEV-NNPV) virus was determined in infected silkworm Bm-N cells by fluorescence and Western blot analysis. The recombinant was inoculated into silkworm pupae and the yield of Nakayama VLP (NVLP) reached a peak in the homogenates after 3 days. Additionally, in the peptide analysis of infected pupae homogenate, it appeared approximately 300-500 μg E protein/pupa were produced. When pur

Evaluation of Chilean Boldo Essential Oil as a Natural Insecticide Against Chrysomya megacephala (Diptera: Calliphoridae).

Chrysomya megacephala (Fabricius, 1794) is a vector of enteric bacteria, protozoa, helminths, and viruses. These Diptera can also be responsible for secondary myiasis in several animal species. Therefore, it is easy to understand the relevance of studies focusing on C. megacephala dissemination control. The employment of essential oils as natural insecticides must be considered as a promising alternative for the replacement of synthetic insecticides. In this context, the essential oil obtained from Chilean boldo leaves should be highlighted. The aim of the present work was to assess the insect

Dissecting, Fixing, and Visualizing the Drosophila Pupal Notum.

The pupae of Drosophila melanogaster are immobile for several days during metamorphosis, during which they develop a new body with a thin transparent adult integument. Their immobility and transparency make them ideal for in vivo live imaging experiments. Many studies have focused on the dorsal epithelial monolayer of the pupal notum because of its accessibility and relatively large size. In addition to the studies of epithelial mechanics and development, the notum has been an ideal tissue to study wound healing. After an injury, the entire epithelial repair process can be captured by live ima

Pesticide selectivity to the parasitoid Trichogramma pretiosum: A pattern 10-year database and its implications for Integrated Pest Management.

Trichogramma pretiosum is one of the main egg parasitoids used in the control of lepidopteran pests in Brazil. This natural enemy can be negatively affected by the use of insecticides, herbicides, and fungicides. The present work used a systematic review and meta-analysis to group information from multiple studies on the selectivity of pesticides (279 commercial products) in rice, corn, soybean, apple and peach crops for immature stages (egg-larva, pre-pupa, and pupa) and adult parasitoids. The selected studies used the International Organization for Biological and Integrated Control (IOBC) me

Genome-wide analysis of long noncoding RNAs and their association in regulating the metamorphosis of the Sarcophaga peregrina (Diptera: Sarcophagidae).

BACKGROUND: The flesh fly, Sarcophaga peregrina (Diptera: Sarcophagidae), is an important hygiene pest, that causes myiasis in humans and other mammals, typically livestock, and as a vector for various parasitic agents, including bacteria, viruses, and parasites. The role of long non-coding RNAs (lncRNAs) in regulating gene expression during metamorphosis of the flesh fly has not been well established. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we performed genome-wide identification and characterization of lncRNAs from the early pupal stage (1-days pupae), mid-term pupal stage (5-days pu

Juvenile hormone analog enhances Zika virus infection in Aedes aegypti.

In recent years, there has been a rise in the emergence of arboviruses of public health importance, including Zika, chikungunya, dengue, and yellow fever viruses. Insecticide-based mosquito control has been the primary method for mitigating transmission of arboviruses. The consequences for the application of insecticides include both lethal and sublethal effects, and associated development of insecticide resistance. However, little is known about the influence on arboviral transmission. Mosquitoes with phenotypes that exhibit insecticide resistance or experience sublethal effects may be associ

The genetic adaptations of fall armyworm Spodoptera frugiperda facilitated its rapid global dispersal and invasion.

The fall armyworm (Spodoptera frugiperda) is a lepidopteran insect pest that causes huge economic losses. This notorious insect pest has rapidly spread over the world in the past few years. However, the mechanisms of rapid dispersal are not well understood. Here, we report a chromosome-level assembled genome of the fall armyworm, named the ZJ-version, using PacBio and Hi-C technology. The sequenced individual was a female collected from the Zhejiang province of China and had high heterozygosity. The assembled genome size of ZJ-version was 486 Mb, containing 361 contigs with an N50 of 1.13 Mb.

In vivo and in vitro effectiveness of Azadirachta indica-synthesized silver nanocrystals against Plasmodium berghei and Plasmodium falciparum, and their potential against malaria mosquitoes.

Malaria transmission is a serious emergence in urban and semiurban areas worldwide, becoming a major international public health concern. Malaria is transmitted through the bites of Anopheles mosquitoes. The extensive employ of synthetic pesticides leads to negative effects on human health and the environment. Recently, plant-synthesized nanoparticles have been proposed as highly effective mosquitocides. In this research, we synthesized silver nanoparticles (AgNP) using the Azadirachta indica seed kernel extract as reducing and stabilizing agent. AgNP were characterized by UV-vis spectrophotom

Pretreatment of floodwater Aedes habitats (dambos) in Kenya with a sustained-release formulation of methoprene.

Effectiveness of sustained-release Altosid pellets (4% AI methoprene) against floodwater mosquitoes in dambos treated at 5, 3, and 1 wk before and 1 day after flooding was determined. Only 2% of Aedes pupae (primarily Aedes mcintoshi) survived to adults in an area treated 5 wk preflood, and no adult mosquitoes emerged from an area treated 1 day after flooding. In contrast, 12 and 16% of Aedes pupae successfully survived to the adult stage in areas pretreated 3 and 1 wk, respectively, preflood. The effectiveness of the Altosid declined against Culex spp. (primarily Cx. antennatus) collected fro

Composition and Diversity Characteristics of Gut Microbiota during the Development of Telchinia issoria (Lepidoptera: Nymphalidae).

Ramie (Boehmeria nivea) was a traditional economic crop of high commercial value, whose cultivation was threatened by the leaf-feeding pest Telchinia issoria. This study investigated how the gut microbiota of T. issoria shifted across its larval, pupal, and adult stages using 16S rRNA amplicon sequencing. We found that Pseudomonadota and Bacillota dominated across all stages, with stage-specific enrichments of key genera: Burkholderia-Caballeronia-Paraburkholderia in early larvae, Acinetobacter and Culicoidibacter in mid-instars, Serratia in late larvae, Enterococcus in pupae, and Pseudomonas

High cold tolerance through four seasons and all free-living stages in an ectoparasite.

Off-host stages of temperate parasites must cope with low temperatures. Cold tolerance is often highest in winter, as a result of diapause and cold acclimation, and low during the active summer stages. In some blood-feeding ectoparasites, offspring provisioning determines cold tolerance through all the non-feeding, off-host stages. Large size increases survival in the cold, but so far seasonal variation in within-female offspring size has not been associated with offspring cold tolerance. The deer ked (Lipoptena cervi) reproduces on cervids from autumn to spring. Newborn pupae drop off the hos

Mosquitocidal and antibacterial activity of green-synthesized silver nanoparticles from Aloe vera extracts: towards an effective tool against the malaria vector Anopheles stephensi?

Mosquitoes represent an important threat for lives of millions of people worldwide, acting as vectors for devastating pathogens, such as malaria, yellow fever, dengue, and West Nile. In addition, pathogens and parasites polluting water also constitute a severe plague for populations of developing countries. Here, we investigated the mosquitocidal and antibacterial properties of Aloe vera leaf extract and silver nanoparticles synthesized using A. vera extract. Mosquitocidal properties were assessed in laboratory against larvae (I-IV instar) and pupae of the malaria vector Anopheles stephensi. G

Characterization of Triacylglycerol Dynamics and Lipid Metabolism in the Beetle Tribolium castaneum.

Lipids are essential for all organisms, and insects can obtain these macromolecules from their diet or synthesize them from carbohydrates and amino acids. These animals store fatty acids in the form of triacylglycerol in lipid droplets in the fat body and, in times of demand, mobilize these reserves as an energy source or for synthesizing structural lipids. However, details about this metabolism are unknown in most species, such as the beetle Tribolium castaneum. Here, we explored the dynamics of triacylglycerol and lipid metabolism during larval and pupal development, after adult emergence, a
