Quorum Quenching Potential of the Indigenous Kalimantan Fern Stenochlaena palustris Against Pseudomonas aeruginosa.

<b>Background and Objective:</b> Plant-derived compounds that disrupt bacterial quorum sensing (QS) provide alternatives to combat antibiotic resistance. This quorum quenching (QQ) strategy reduces virulence without inhibiting growth, thus minimizing resistance. Kelakai (<i>Stenochlaena palustris</i> (Burm.) Bedd.), an endemic Kalimantan plant traditionally used by the Dayak people, was evaluated for QQ activity against <i>Pseudomonas aeruginosa</i>. <b>Materials and Methods:</b> Ethanolic extract of <i>S. palustris</i> leaves was pre

Interference of Pseudomonas aeruginosa Virulence Factors by Different Extracts from Inula Species.

Objectives: Pseudomonas aeruginosa is an opportunistic pathogen of high clinical relevance due to its ability to form biofilms, its inherent virulence regulated by quorum-sensing systems, and its multidrug resistance. In the present study, we evaluated the inhibitory potential of nine extracts from Inula species (chloroform and methanolic fractions, including a sesquiterpene lactone-enriched fraction) against biofilm formation and virulence-associated traits of P. aeruginosa PAO1 and three multidrug-resistant clinical isolates, as well as their cytotoxicity, biocompatibility, and ability to af

Cephalosporins Interfere With Quorum Sensing and Improve the Ability of Caenorhabditis elegans to Survive Pseudomonas aeruginosa Infection.

Pseudomonas aeruginosa utilizes the quorum sensing (QS) system to strategically coordinate virulence and biofilm formation. Targeting QS pathways may be a potential anti-infective approach to treat P. aeruginosa infections. In the present study, we define cephalosporins' anti-QS activity using Chromobacterium violaceum CV026 for screening and QS-regulated mutants of P. aeruginosa for validation. We quantified the effects of three cephalosporins, cefepime, ceftazidime, and ceftriaxone, on (1) pyocyanin production using spectrophotometric assay, (2) bacterial motility using agar plate assay, and

Anti-Pseudomonas aeruginosa Activity of the Scorpion-Derived Peptide GK8.

The increasing infection and antibiotic resistance of Pseudomonas aeruginosa has become a major threat to public health, making exploring new and effective antimicrobial agents particularly important. In this study, the scorpion-derived cationic peptide GK8 was designed, which showed effectiveness against P. aeruginosa including clinical resistance strains with low hemolytic activity. In the mouse skin subcutaneous infection model, GK8 significantly decreased the number of P. aeruginosa cells and inflammatory infiltration in the infected area. Mechanism research showed that GK8 killed P. aerug

Evaluation of Combinatorial Efficacy of Nisin and Lactoferrin against Pseudomonas Aeruginosa Induced Experimental Pulmonary Infection Model.

Current treatment strategies for Pseudomonas aeruginosa pulmonary infections are limited by severe side effects and the emergence of multidrug resistance. This study evaluated the therapeutic potential of nisin and lactoferrin, two antimicrobial peptides with distinct modes of action, against P. aeruginosa lung infections. In vitro, the combination exhibited potent synergistic antibacterial activity with significant suppression of biofilm formation and pyocyanin production. Morphological analysis by field-emission scanning electron microscopy revealed extensive structural disruption of bacteri

An iron-chelating sulfonamide identified from Drosophila-based screening for antipathogenic discovery.

We exploited bacterial infection assays using the fruit fly Drosophila melanogaster to identify anti-infective compounds that abrogate the pathological consequences in the infected hosts. Here, we demonstrated that a pyridine-3-N-sulfonylpiperidine derivative (4a) protects Drosophila from the acute infections caused by bacterial pathogens including Pseudomonas aeruginosa. 4a did not inhibit the growth of P. aeruginosa in vitro, but inhibited the production of secreted toxins such as pyocyanin and hydrogen cyanide, while enhancing the production of pyoverdine and pyochelin, indicative of iron d

Intra-strain genomic microevolution and phage resistance in Pseudomonas aeruginosa PAO1 laboratory isolates.

Pseudomonas aeruginosa is a major opportunistic pathogen, and its laboratory reference strain, PAO1, is widely used in microbiological and genetic studies. However, PAO1 often exhibits phenotypic variability that can affect experimental reproducibility. Our PAO1 stock, obtained from a public biobank, is resistant to PP7, a pilus-dependent single-stranded RNA phage known to infect PAO1. This suggests the presence of genetic variants in the stock. To check this possibility, we isolated six phenotypically distinct variants (GU1-GU6) and performed genomic and phenotypic analyses. Notable differenc

Citrate cross-feeding by Pseudomonas aeruginosa supports lasR mutant fitness.

Cross-feeding of metabolites between subpopulations can affect cell phenotypes and population-level behaviors. In chronic Pseudomonas aeruginosa lung infections, subpopulations with loss-of-function (LOF) mutations in the lasR gene are common. LasR, a transcription factor often described for its role in virulence factor expression, also impacts metabolism, which, in turn, affects interactions between LasR+ and LasR- genotypes. Prior transcriptomic analyses suggested that citrate, a metabolite secreted by many cell types, induces virulence factor production when both genotypes are together. An

Serine Acetyltransferase from Pseudomonas aeruginosa: Distinctive Features, Pleiotropic Roles, and Therapeutic Potential.

Cysteine biosynthesis is increasingly recognized as a critical determinant of bacterial virulence, highlighting this pathway as a promising reservoir of novel antimicrobial targets. In Pseudomonas aeruginosa, however, the molecular basis of cysteine production has only recently begun to emerge. Here, we identify PA3816 as the major P. aeruginosa serine acetyltransferase (PaCysE), the enzyme responsible for generating the activated serine intermediate that feeds O-acetylserine sulfhydrylase-mediated cysteine synthesis. Through a combination of biochemical and genetic approaches, we demonstrate

Two Novel AHL-Degrading Enzymes, PoAcy1 and PoLac, Attenuate Pectobacterium carotovorum and Pseudomonas aeruginosa Virulence through Quorum-Sensing Disruption.

Ensuring food safety remains a significant challenge due to the persistent contamination and spoilage caused by biofilm-forming and resistant pathogenic bacteria. Quorum quenching presents an ecofriendly control strategy. We identified a novel acylase (PoAcy1) and a broad-spectrum lactonase (PoLac) from marine-derived bacterium Pseudooceanicola onchidii XY-99. Both the XY-99 strain and purified PoLac effectively suppressed Pectobacterium carotovorum subsp. carotovorum (Pcc)-induced soft rot in potato tubers. Heterologous expression of either enzyme in Pseudomonas aeruginosa PAO1 significantly

Sodium Citrate Alleviates Virulence in Pseudomonas aeruginosa.

The development of bacterial resistance is an insistent global health care issue, especially in light of the dwindled supply of new antimicrobial agents. This mandates the development of new innovative approaches to overcome the resistance development obstacle. Mitigation of bacterial virulence is an interesting approach that offers multiple advantages. Employing safe chemicals or drugs to mitigate bacterial virulence is an additive advantage. In the current study, the in vitro antivirulence activities of citrate were evaluated. Significantly, sodium citrate inhibited bacterial biofilm formati

Impact of D-amino acid dehydrogenase on virulence factor production by a Pseudomonas aeruginosa.

Chronic Pseudomonas aeruginosa infections remain the leading cause of lung dysfunction and mortality for cystic fibrosis (CF) patients. Many other bacteria inhabit the CF lung, but P. aeruginosa utilizes novel strategies that allow it to colonize this environment as the predominant bacterial pathogen. D-Amino acid dehydrogenase encoded by dadA is highly expressed by P. aeruginosa within the CF lung, and it is required for optimal production of hydrogen cyanide by some CF-adapted isolates. To better understand the increased significance of D-amino acid dehydrogenase in P. aeruginosa physiology,

Attenuation of LPS-Induced Lung Injury by Benziodarone via Reactive Oxygen Species Reduction.

As overproduction of reactive oxygen species (ROS) causes various diseases, antioxidants that scavenge ROS, or inhibitors that suppress excessive ROS generation, can be used as therapeutic agents. From a library of approved drugs, we screened compounds that reduced superoxide anions produced by pyocyanin-stimulated leukemia cells and identified benzbromarone. Further investigation of several of its analogues showed that benziodarone possessed the highest activity in reducing superoxide anions without causing cytotoxicity. In contrast, in a cell-free assay, benziodarone induced only a minimal d

The bacteriophage-encoded regulator PemR attenuates Pseudomonas aeruginosa virulence by hijacking quorum sensing and metabolism.

The global challenge of multidrug-resistant Pseudomonas aeruginosa demands innovative anti-virulence approaches. We characterize PemR, a novel transcriptional regulator encoded by P. aeruginosa phage PAYQ66, which orchestrates multimodal virulence attenuation in P. aeruginosa. Biochemical analyses demonstrated that PemR directly binds to the mvfR promoter, resulting in significant repression of this key quorum-sensing regulator. This repression, in turn, induces profound metabolic reprogramming by redirecting metabolic flux away from PQS biosynthesis toward catechol accumulation. PemR globally

Exploring cinnamic acid's function in reducing the quorum sensing and biofilm formation in Pseudomonas aeruginosa: in vitro and in silico investigations.

Antimicrobial resistance (AMR) has become serious global threat to the public health which is estimated to be responsible for over one million annual deaths. AMR occurs when microbes develop defence mechanism against the antibiotics. Pseudomonas aeruginosa is an opportunistic pathogen which is known to cause many infections, and it has developed resistance to multiple antibiotic classes. The continuous exposure of antibiotics imposes the risk of AMR development. In this case, targeting quorum sensing (QS) and biofilms becomes more reasonable. This study assessed the effect of cinnamic acid (CA

In vivo imaging of the lung inflammatory response to Pseudomonas aeruginosa and its modulation by azithromycin.

BACKGROUND: Chronic inflammation of the airways is a central component in lung diseases and is frequently associated with bacterial infections. Monitoring the pro-inflammatory capability of bacterial virulence factors in vivo is challenging and usually requires invasive methods. METHODS: Lung inflammation was induced using the culture supernatants from two Pseudomonas aeruginosa clinical strains, VR1 and VR2, isolated from patients affected by cystic fibrosis and showing different phenotypes in terms of motility, colony characteristics and biofilm production as well as pyoverdine and pyocyani

Acidic amino acids as counterions of ciprofloxacin: Effect on growth and pigment production in Staphylococcus aureus NCTC 8325 and Pseudomonas aeruginosa PAO1.

Antimicrobial resistance (AMR) is emerging as a global threat to public health. One of the strategies employed to combat AMR is the use of adjuvants which act to enhance or reinstate antimicrobial activity by inhibiting resistance mechanisms. However, these adjuvants are themselves not immune to selecting resistant phenotypes. Thus, there is a need to utilise mechanisms which are either less likely to or unable to trigger resistance. One commonly employed mechanism of resistance by microorganisms is to prevent antimicrobial uptake or efflux the antibiotic which manages to permeate its membrane
