N-acylhomoserine lactone (AHL)-mediated quorum-sensing (QS) regulates virulence functions in plant and animal pathogens such as Agrobacterium tumefaciens and Pseudomonas aeruginosa. A chemolibrary of more than 3500 compounds was screened using two bacterial AHL-biosensors to identify QS-inhibitors (QSIs). The purity and structure of 15 QSIs selected through this screening were verified using HPLC MS/MS tools and their activity tested on the A. tumefaciens and P. aeruginosa bacterial models. The IC50 value of the identified QSIs ranged from 2.5 to 90 µg/ml, values that are in the same range as

Quorum sensing, a bacterial cell-cell communication process, controls biofilm formation and virulence factor production in Vibrio cholerae, a human pathogen that causes the disease cholera. The major V. cholerae autoinducer is (S)-3-hydroxytridecan-4-one (CAI-1). A membrane bound two-component sensor histidine kinase called CqsS detects CAI-1, and the CqsS → LuxU → LuxO phosphorelay cascade transduces the information encoded in CAI-1 into the cell. Because the CAI-1 ligand is known and because the signalling circuit is simple, consisting of only three proteins, this system is ideal for analysi

Acinetobacter baumannii has emerged as a major global pathogen due to extensive resistance to last-resort antimicrobials, a high burden of nosocomial infections, and increasing community-acquired cases. Its adaptability is driven by diverse resistance mechanisms, including β-lactamase production, aminoglycoside-modifying enzymes, efflux pump overexpression, target-site mutations, and lipid A remodeling, all of which limit treatment options and worsen clinical outcomes. Pathogenicity is further enhanced by quorum-sensing systems, particularly AbaI/AbaR, which regulate biofilm formation, virulen

Bacteria in the human intestinal tract express more than 46 million genes, suggesting that interbacterial communication and communication with human cells are well-controlled and synchronized. Many papers have been published on quorum sensing (QS) and other forms of bacterial communication, e.g., nanotubes, nanovibrations, and electromagnetism. Autoinducers (AIs) such as AI-1 N-acyl homoserine lactones (AHLs), AI-2 boron-containing furanosyl borate diesters, AI-3 pyrazinone derivatives, a combination of AI-3/Epi (epinephrine)/NE (norepinephrine), auto-inducer peptides (AIPs), and SdiA (suppres

Quorum sensing (QS) enables bacteria to coordinate collective behaviors in response to population density. LuxI/R QS systems, common among gram-negative bacteria, consist of a LuxI-type synthase that produces an N-acyl L-homoserine lactone (AHL) signaling molecule and a LuxR-type receptor that senses the AHL. At threshold AHL concentrations, LuxR-type receptors undergo ligand-induced conformational changes that affect DNA binding and target gene transcription. Because of their role in regulating myriad collective behaviors, LuxI/R systems are targets for many applications, including antivirule

Pseudomonas aeruginosa (PA) is a clinically relevant opportunistic pathogen whose persistence and antimicrobial tolerance are largely driven by biofilm formation and quorum-sensing (QS)-regulated virulence. Targeting QS has therefore emerged as an antivirulence strategy that attenuates pathogenicity without exerting strong selective pressure on bacterial growth. This systematic review (2020-2024) analyzes recent advances in the identification of QS inhibitors against PA, emphasizing studies that integrate in silico approaches. Forty-six studies met the inclusion criteria. All employed molecula

Intensive livestock and poultry production has made odor emissions a major environmental challenge. These odors arise mainly from the anaerobic microbial transformation of manure into ammonia (NH3), hydrogen sulfide (H2S), volatile fatty acids (VFAs), and indolic compounds. Microbial odor-control strategies include source control through dietary optimization to regulate gut microbiota and reduce odor formation, process control through microbial additives intended to suppress odor-producing microorganisms and anaerobic fermentation, and end-of-pipe treatment using biofilters or biotrickling sys

BACKGROUND: Klebsiella pneumoniae is associated with healthcare-related infections, and its pathogenicity is linked to quorum sensing (QS), which regulates virulence factor production. The rise in antibiotic resistance poses a significant global challenge, increasing the need for alternative strategies. Targeting the QS system offers a promising approach to attenuate virulence. Thus, identifying stable, non-toxic QS inhibitors from natural sources is essential.
PURPOSE: The study aimed to characterize the compounds of lemongrass, patchouli, tea tree, and thyme Essential oils (EOs) with anti-Q

Landfill gas (LFG) has become the second-largest anthropogenic source of methane (CH4) emissions globally. CH4 is the second most significant greenhouse gas after carbon dioxide (CO2), thus it is crucial to mitigate the methane emission of landfills. The soil in landfill cover layers is rich in methane-oxidizing bacteria (MOB), which use CH4 as their sole carbon and energy source. However, during the microbial methane oxidation process, the oxidation rate tends to decrease over time. It is anticipated that extracellular polymeric substance (EPS) is one of the key factors governing the reductio

Quorum sensing is often proposed as a target for antivirulence drug development against bacterial pathogens. Coagulase-negative staphylococci are leading causes of nosocomial blood infections and associated mortality. However, there is a severe lack of understanding how virulence and quorum sensing affect coagulase-negative staphylococcal sepsis. We show that the Agr quorum-sensing system has a strong impact on mortality from experimental sepsis caused by the exemplary coagulase-negative staphylococcal species Staphylococcus haemolyticus. This was based on strict Agr regulation of phenol-solub

Iron is a transition metal used as a cofactor in many biochemical reactions. In bacteria, iron homeostasis involves Fur-mediated de-repression of iron uptake systems, such as the iron-chelating compounds siderophores. In this work, we identified and characterized novel regulatory systems that control siderophores in the environmental opportunistic pathogen Chromobacterium violaceum. Screening of a 10,000-transposon mutant library for siderophore halos identified seven possible regulatory systems involved in siderophore-mediated iron homeostasis in C. violaceum. Further characterization reveale

The aim of the present study was to examine the impact of macrolides on the expression of virulence factors and QS-associated genes in clinical P. aeruginosa isolates. Among 60 clinical P. aeruginosa, pyocyanin production was detected in 27 (45%) isolates, which belonged to various STs. Erythromycin inhibited the production of pigments in 12 out of 27 isolates. Other antibiotic categories didn't have an impact on production of pigments. Additionally, results showed that erythromycin sub-MIC inhibited the growth-rate in 17 isolates. Of note, in six isolates, the inhibition of growth-rate was gr

OBJECTIVE: Glucosyltransferases (Gtfs) and quorum sensing (QS) mediated transduction genes play critical roles in the pathogenesis of Streptococcus mutan-mediated dental caries. Therefore, targeting gtfs and QS-mediated virulence genes have therefore emerged as an intriguing goal for efficient therapeutic approaches that block cariogenic biofilms.
METHODS: Post-biotic mediators (PMs) obtained from our previously isolated and characterized beneficial bacteria Enterobacter colacae PS-74 was assessed for its antibiofilm potential against S. mutans. According to the transcriptome method, qRT-PCR

Quorum sensing (QS) is a communication mechanism that controls bacterial communication and can influence the transcriptional expression of multiple genes through one or more signaling molecules, thereby coordinating the population response of multiple bacterial pathogens. Secretion systems (SS) play an equally important role in bacterial information exchange, relying on the secretory systems to secrete proteins that act as virulence factors to promote adhesion to host cells. Eight highly efficient SS have been described, all of which are involved in the secretion or transfer of virulence facto

Crohn's disease (CD) is an idiopathic inflammatory condition of the gastrointestinal tract with the primary method of diagnosis and follow-up being colonoscopy. A disturbed host-microbiome interaction, including the presence of pathobionts, is implicated in initiation and perpetuation of inflammation. As such, we hypothesized that bacterial quorum-sensing (QS) molecules (QSMs), small molecules bacteria generate to regulate gene expression, would be elevated in patients with CD. We collected serum at the time of colonoscopy from patients with CD and healthy controls, determining through biosens

Trypanosomes causing African sleeping sickness use quorum-sensing (QS) to generate transmission-competent stumpy forms in mammalian hosts. This density-dependent process is signalled by oligopeptides that stimulate the signal transduction pathway leading to stumpy formation. Here, using mass spectrometry analysis, we identify peptidases released by trypanosomes and, for 12 peptidases, confirm their extracellular delivery. Thereafter, we determine the contribution of each peptidase to QS signal production using systematic inducible overexpression in vivo, and confirm this activity operates thro

Sulfane sulfur, such as inorganic and organic polysulfide (HSn- and RSn-, n > 2), is a common cellular component, produced either from hydrogen sulfide oxidation or cysteine metabolism. In Pseudomonas aeruginosa PAO1, LasR is a quorum sensing master regulator. After binding its autoinducer, LasR binds to its target DNA to activate the transcription of a suite of genes, including virulence factors. Herein, we report that the production of hydrogen sulfide and sulfane sulfur were positively correlated in P. aeruginosa PAO1, and sulfane sulfur was able to modify LasR, which generated Cys188 persu

BACKGROUND: Schistosomiasis is a prevalent neglected tropical disease that affects approximately 300 million people worldwide. Its treatment is through a single class chemotherapy, praziquantel. Concerns surrounding the emergence of praziquantel insensitivity have led to a need for developing novel anthelmintics.
METHODOLOGY/PRINCIPLE FINDINGS: Through evaluating and screening fourteen compounds (initially developed for anti-cancer and anti-viral projects) against Schistosoma mansoni, one of three species responsible for most cases of human schistosomiasis, a racemic N-acyl homoserine (1) dem

This work investigated chloroform extracts from culture supernatants of two human probiotic bacteria, Lactobacillus casei CRL 431 and Lactobacillus acidophilus CRL 730 for the production of virulence factors and quorum sensing (QS) interference against three Pseudomonas aeruginosa strains. Both extracts inhibited biofilm biomass (up to 50%), biofilm metabolic activity (up to 39%), the production of the enzyme elastase (up to 63%) and pyocyanin (up to 77%), and decreased QS, without presenting any antibacterial acgivity. In addition, the chloroform extracts of both strains disrupted preformed b
