Functions and mechanisms of circular RNAs in regulating stem cell differentiation.

Stem cells are a class of undifferentiated cells with great self-renewal and differentiation capabilities that can differentiate into mature cells in specific tissue types. Stem cell differentiation plays critical roles in body homoeostasis, injury repair and tissue generation. The important functions of stem cell differentiation have resulted in numerous studies focusing on the complex molecular mechanisms and various signalling pathways controlling stem cell differentiation. Circular RNAs (circRNAs) are a novel class of noncoding RNAs with a covalently closed structure present in eukaryotes.

Circular RNAs as potential regulators in bone remodeling: a narrative review.

OBJECTIVE: In this review, we focus on the recent progress of circular ribonucleic acids (circRNAs)-related molecular mechanisms in the processes of osteogenesis and osteoclastogenesis, and explore their roles in the development of bone-remodeling disorders. BACKGROUND: The well-coupled bone-formation and bone-resorption processes are vital in bone remodeling. Once the balance is disrupted, bone-remodeling disorders (e.g., osteoporosis and osteopetrosis) occur, severely affecting patients' quality of life. CircRNAs, the newly discovered members of the non-coding RNA family, have been reported

Assessing circular RNAs in Alzheimer's disease and frontotemporal lobar degeneration.

A circular-transcriptome-wide study has recently linked differential expression of circular RNAs (circRNAs) in brain tissue with Alzheimer's disease (AD). We aimed at replicating the major findings in an independent series of sporadic and familial AD. We also included cases with frontotemporal lobar degeneration (FTLD), comprising brain specimens with TDP-43 aggregates (FTLD-TDP43) and samples that presented Tau accumulation (FTLD-Tau). Using a quantitative polymerase chain reaction approach, we evaluated 8 circRNAs that surpassed the significant threshold in the former meta-analysis (circHOME

Identification of circular RNAs in the immature and mature rat anterior pituitary.

Circular RNAs (circRNAs) are a new class of RNA that have a stable structure characterized by covalently closed circular molecules and are involved in invasive pituitary adenomas and recurrent clinically nonfunctioning pituitary adenomas. However, information on circRNAs in the normal pituitary, especially in rats, is limited. In this study, we identified 4123 circRNAs in the immature (D15) and mature (D120) rat anterior pituitary using the Illumina platform, and 32 differentially expressed circRNAs were found. A total of 150 Gene Ontology terms were significantly enriched, and 16 KEGG pathway

Overexpression of Circular RNA circTTN Promotes Ferroptosis in Sepsis-Induced Cardiomyopathy.

Sepsis-induced cardiomyopathy (SCM) is one of the common complications caused by sepsis. Despite the higher mortality rate associated with SCM, the pathogenesis of SCM is poorly understood. The study of circular RNA (circRNA) in a variety of diseases has accelerated over recent decades. However, the function of circRNAs in SCM is rarely reported. Here, we found that the circRNA from the TTN gene (circTTN) was increased in the SCM mouse and cell models. Modulation of circTTN affected the characteristics of SCM both in mouse and cell models. The expression of ferroptosis regulators, GPX4 and SLC

Macrophage-specific circular RNA circHIPK2, inflammation, and fibrosis after myocardial infarction.

BACKGROUND AND AIMS: Cardiac remodelling following MI is intricately linked to macrophage polarization dynamics, yet the underlying mechanisms remain incompletely elucidated. In this study, circHIPK2, a novel circRNA, was identified as being upregulated in inflammatory cardiac macrophages following MI. Its expression correlated with post-MI inflammation dynamics, suggesting a regulatory role in macrophage polarization. It was hypothesized that circHIPK2 functions as a molecular switch of macrophage polarization during MI progression. METHODS: To test this hypothesis, circHIPK2 levels were mod

A Novel Circular RNA circSPEF2 Regulates Testis Development in Crucian Carp.

Circular RNAs (circRNAs) exhibit significant sex- and development stage-specific expression patterns in the gonads of various fish species, yet their functions and regulatory mechanisms in male reproductive development remain largely unexplored in crucian carp (Carassius auratus). In this study, we characterized the expression features and biological functions of circSPEF2, a circular RNA derived from the reproduction-related gene spef2. Our results showed that circSPEF2 expression was markedly elevated in mature testes and progressively upregulated during gonadal maturation. Functional studie

Differentially expressed circular RNA profile in hemorrhagic and ischemic moyamoya disease.

Hemorrhage is a frequent manifestation in patients with moyamoya disease (MMD). Compared with MMD patients with other subtypes, patients with hemorrhagic MMD (hMMD) are at higher risk of poor prognostic outcomes, Circular RNAs (circRNAs) frequently display dysregulated expression in several human diseases. In the present study, the role of circRNAs in the pathogenesis of hemorrhage in MMD was investigated. Microarray profiling on 12 moyamoya disease samples, consisting of six hMMD and six matching ischemic MMD (iMMD) samples, was performed. Reverse transcription-quantitative PCR was then used

HIV-1-encoded circular RNA enhances viral transcription through Tat binding.

Circular RNAs (circRNAs) are single-stranded RNAs that form a covalently closed loop without 5' and 3' ends. From their own genomes, RNA viruses can encode circRNAs that play important roles during infection, but whether this is the case for HIV remains unclear. Here we detect an HIV-1-encoded viral circRNA (termed circHIV) in plasma from 18 people living with HIV and in infected primary cells and T-cell lines using RT-qPCR and northern blot. We find that this viral circRNA is packaged within HIV-1 virions and binds to the HIV-1 Tat protein upon expression in cells, which enhances transcriptio

Circular RNA-based protein replacement therapy mitigates osteoarthritis in male mice.

In vitro-transcribed and circularized RNAs (ivcRNAs) represent a robust platform for sustained protein translation, offering promising potential for localized therapeutic delivery in joint diseases. Osteoarthritis (OA), the most prevalent degenerative joint disorder, remains a major clinical challenge due to its progressive nature and the lack of disease-modifying treatments. In this study, we identify Musashi2 (Msi2) deficiency in articular chondrocytes as a key contributor to OA pathogenesis. To evaluate the efficacy of ivcRNA-mediated protein replacement therapy, we developed a localized de

Emerging roles of circular RNAs and enhancer RNAs: new insights into the development and management of neurodegenerative disorders.

Neurodegenerative disorders involve the gradual breakdown of neurons, leading to problems with thinking, movement, and mental health. More evidence is emerging about the important roles of non-coding RNAs (ncRNAs). CircRNAs are a type of ncRNA formed through back-splicing and are widely present in the mammalian brain. They play crucial roles in brain development and mainly regulate gene expression and post-transcriptional processes by acting as molecular sponges for miRNAs and RBPs. eRNAs are another class of ncRNAs. They are produced from enhancer regions of the genome and act as vital regula

Landscape of circular RNAs in acute myeloid leukemia and their clinical significance.

Circular RNAs (circRNAs) have emerged as important regulators in cancer biology, but their roles in acute myeloid leukemia (AML) remain poorly characterized due to limited sample sizes and technical challenges in RNA sequencing. Here, we analyze RNA-sequencing data from 315 Swedish AML patients to create the most comprehensive circRNA profile in AML to date. We identify 5,711 high-confidence circRNAs across 315 AML samples, including 402 differentially expressed between AML and healthy controls, with host genes enriched in hematopoietic pathways. We further discover two circRNAs including hsa_

Characterizing the circular RNA landscape in phloem sap of Brassica napus.

In plants, long-distance transport and phloem-mediated signal distribution play crucial roles in regulating stress adaptation and development. Phloem sap contains various types of RNAs, including small RNAs (sRNAs), messenger RNAs (mRNAs), and long noncoding RNAs (lncRNAs). Recently, endogenous circular RNAs have been identified in phloem sap of apple trees. Some phloem RNAs have been shown to have long-distance signaling functions, but for most, no functions have yet been determined. Due to their stability, circRNAs are interesting candidates with potential functions in long-distance signalin
