Decreased dietary K+ intake stimulates the expression of regulatory-associated protein of mTOR (RAPTOR) in kidneys and increases mRNA of Raptor in aldosterone-sensitive distal nephrons. The aim of the study is to explore the role of mechanistic target of rapamycin complex-1 (mTORc1) in stimulating Kir4.1/Kir5.1 and Na-Cl cotransporter (NCC), and inhibiting renal outer medullary K+ channel (ROMK or Kir1.1) and epithelial-Na+ channel (ENaC) during overnight low K+ intake (LK). Deletion of RAPTOR in kidneys inhibited basolateral Kir4.1/Kir5.1 of distal convoluted tubule (DCT) and decreased the ex

The dedicator of cytokinesis 5 (DOCK5) is associated with obesity. However, the mechanism by which DOCK5 contributes to obesity remains completely unknown. Here, we show that hepatic DOCK5 expression significantly decreases at a state of insulin resistance (IR). Deletion of DOCK5 in mice reduces energy expenditure, promotes obesity, augments IR, dysregulates glucose metabolism, and activates the mTOR (Raptor)/S6K1 pathway under a high-fat diet (HFD). The overexpression of DOCK5 in hepatocytes inhibits gluconeogenic gene expression and increases the level of insulin receptor (InsR) and Akt phos

Mammalian target of rapamycin (mTOR) is a key regulator of cell growth that associates with raptor and rictor to form the mTOR complex 1 (mTORC1) and mTORC2, respectively. Raptor is required for oxidative muscle integrity, whereas rictor is dispensable. In this study, we show that muscle-specific inactivation of mTOR leads to severe myopathy, resulting in premature death. mTOR-deficient muscles display metabolic changes similar to those observed in muscles lacking raptor, including impaired oxidative metabolism, altered mitochondrial regulation, and glycogen accumulation associated with protei

Black-tailed prairie dogs (BTPDs), Cynomys ludovicianus, are an important prey for raptors; therefore, the use of the rodenticide Rozol (0.005% chlorophacinone active ingredient) to control BTPDs raises concern for secondary poisonings resulting from the consumption of contaminated prey by raptors. In the present study, the authors observed Rozol exposure and adverse effects to mammalian prey on 11 of 12 search days of the study. Mammalian hepatic chlorophacinone residues ranged from 0.44 to 7.56 µg/g. Poisoned prey availability was greater than previously reported.

BACKGROUD: In this study, we present data obtained using light microscopy for the histological analysis of the upper eyelid (palpebra dorsalis), lower eyelid (palpebra ventralis), and third eyelid (palpebra tertia) also known as the nictitating membrane. We characterized the organized conjunctiva-associated lymphoid tissue (CALT) in selected raptor species. The aim of this study is to compare the histological structures of these eyelids in owls and diurnal raptors to identify potential evolutionary links or independent adaptations to their environments.

The successful rehabilitation and release of raptor chicks can be challenging, especially when the chicks are still in the post-fledging dependency period. Here, we report on a recently fledged powerful owl chick that was held in care for 33 days before being successfully reunited with its parents. We document the steps undertaken during the entire process from collection from the wild to post-release monitoring and recommend clinical procedures for treatment of raptors entering veterinary facilities. Success of this rehabilitation was facilitated by early care and treatment for potential rode

The proline-rich Akt substrate of 40 kilodaltons (PRAS40) was identified as a raptor-binding protein that is phosphorylated directly by mammalian target of rapamycin (mTOR) complex 1 (mTORC1) but not mTORC2 in vitro, predominantly at PRAS40 (Ser(183)). The binding of S6K1 and 4E-BP1 to raptor requires a TOR signaling (TOS) motif, which contains an essential Phe followed by four alternating acidic and small hydrophobic amino acids. PRAS40 binding to raptor was severely inhibited by mutation of PRAS40 (Phe(129) to Ala). Immediately carboxyl-terminal to Phe(129) are two small hydrophobic amino ac

In a series of 4 species of owls and 12 species of diurnal raptors, an additional bone was present bilaterally in the area of the carpus. Designating the bone as an "additional bone of the raptorial carpal region" is suggested pending appropriate embryological studies to determine the origin of the bone. Generally, the bone was present in owls and in diurnal raptors with relatively short, rounded wings, but was absent in species with longer, more tapered wings. The bone lies at the cranial edge of the carpal area near the radial carpal bone, and is fixed to the surrounding skeleton by ligament

BACKGROUND: The balance between protein synthesis and degradation (proteostasis) is a determining factor for muscle size and function. Signalling via the mammalian target of rapamycin complex 1 (mTORC1) regulates proteostasis in skeletal muscle by affecting protein synthesis and autophagosomal protein degradation. Indeed, genetic inactivation of mTORC1 in developing and growing muscle causes atrophy resulting in a lethal myopathy. However, systemic dampening of mTORC1 signalling by its allosteric inhibitor rapamycin is beneficial at the organismal level and increases lifespan. Whether the bene

Mechanistic target of rapamycin (mTOR) complex 1 (mTORC1) signaling plays important physiological roles in bone homeostasis by regulating multiple steps of osteoblast differentiation as well as its activity. However, its potential role in osteocytes has not been explored. In this study, we deleted Raptor, a specific and essential component of mTORC1, in osteocytes using Dmp1-Cre. Deletion of Raptor in osteocytes did not affect bone development and growth, but caused compartment-specific effects on bone mass. Osteocyte-specific deletion of Raptor had no obvious effect on cortical bone compartme

Autophagy is a lysosomal degradation pathway that degrades cytoplasmic proteins and organelles. Absence of autophagy in hepatocytes has been linked to promoting liver injury and tumorigenesis; however, the mechanisms behind why a lack of autophagy induces these complications are not fully understood. The role of mammalian target of rapamycin (mTOR) in impaired autophagy-induced liver pathogenesis and tumorigenesis was investigated by using liver-specific autophagy related 5 knockout (L-ATG5 KO) mice, L-ATG5/mTOR, and L-ATG5/Raptor double knockout (DKO) mice. We found that deletion of mTOR or R

Toxoplasma gondii is an important zoonotic protozoan parasite that can infect all warm-blooded animals including mammals and birds. Raptors can be intermediate hosts for T. gondii and the infection may be dependent on their feeding habits. In this study, we investigated the seroprevalence of T. gondii in ten raptor species from Florida, Pennsylvania, and Tennessee followed by a parasite bioassay on select seropositive samples. From a total of 155 raptors, we detected T. gondii antibodies using a modified agglutination test (cutoff 1:25) in 32 (20.6%) birds. The T. gondii seroprevalence was 44.

Avian trichomonosis, caused by the protozoan Trichomonas gallinae, affects bird-eating raptors worldwide. Raptors can develop trichomonosis by feeding on infected prey, particularly Rock Pigeons (C olumba livia), which are a reservoir for T. gallinae. Raptors may be particularly vulnerable to T. gallinae infection in degraded habitats, where changes in resources may cause raptors to switch from foraging on native prey to synanthropic avian species such as Rock Pigeons. Golden Eagles ( Aquila chrysaetos) typically forage on mammals; however, habitat across much of their range is experiencing de

Biomonitoring in raptors can be used to study long-term and large-scale changes in environmental pollution. In Europe, such monitoring is needed to assess environmental risks and outcomes of chemicals regulation, which is harmonised across the European Union. To be effective, the most appropriate sentinels need to be monitored. Our aim was to identify which European raptor species are the likely most appropriate biomonitors when pollutant quantification is based on analysing tissues. Our current study was restricted to terrestrial exposure pathways and considered four priority pollutant groups

We studied the effects of natural and/or experimental infections of West Nile virus (WNV) in five raptor species from July 2002 to March 2004, including American kestrels (Falco sparverius), golden eagles (Aquila chrysaetos), red-tailed hawks (Buteo jamaicensis), barn owls (Tyto alba), and great horned owls (Bubo virginianus). Birds were infected per mosquito bite, per os, or percutaneously by needle. Many experimentally infected birds developed mosquito-infectious levels of viremia (>10(5) WNV plaque forming units per ml serum) within 5 days postinoculation (DPI), and/ or shed virus per os or

Oxidants are well recognized for their capacity to reduce the phosphorylation of the mammalian target of rapamycin (mTOR) substrates, eukaryotic initiation factor 4E-binding protein 1 (4E-BP1) and p70 S6 kinase 1 (S6K1), thereby hindering mRNA translation at the level of initiation. mTOR functions to regulate mRNA translation by forming the signaling complex mTORC1 (mTOR, raptor, GbetaL). Insulin signaling to mTORC1 is dependent upon phosphorylation of Akt/PKB and the inhibition of the tuberous sclerosis complex (TSC1/2), thereby enhancing the phosphorylation of 4E-BP1 and S6K1. In this study

We evaluated the prevalence of WNV and SLEV neutralizing antibodies in captive and free-ranging raptors from Argentina by plaque-reduction neutralization test. Eighty plasma samples from 12 species were analyzed. Only one captive adult Crowned Eagle (Harpyhaliaetus coronatus) was WNV seropositive (prevalence: 1.25%; antibody titer of 1:80). Two captive Crowned Eagles were SLEV seropositive (prevalence: 2.50%; antibody titers: 1:80 and 1:40).These findings expand the geographic distribution of WNV and SLEV and confirm their activity in central and northeastern Argentina. Rodent damage significantly affects agriculture around the world. Rodenticides can sometimes control pests, but they are costly, may cause secondary poisoning to nontarget wildlife, and can become less efficient over time due to bait shyness and resistance. Using wildlife as biological pest control agents, particularly barn owls (Tyto spp.), has been suggested as an alternative. Barn owl nest boxes and hunting perches have been added to increase predator pressure, yet few studies have examined their effectiveness. We conducted a field study in forty-five 10 × 10 m2 plots to compare three treat

Tick infestation of songbirds has been extensively studied at global and regional scales but relevant data on birds of prey remain scarce and fragmentary. Raptors are of particular epidemiological relevance because their large size, wide-ranging movements and nesting habits may facilitate long-distance dispersal of ticks, and endophilic tick species (e.g. from prey animals) may even persist in their re-used nests. In this study, we aimed to compensate for the lack of comprehensive data in this context. First, more than 1800 birds of prey were examined at 61 sites in Central and South-Eastern E
