Synchronous Induction and Microscopic Analysis of Hyphal Growth Using Laboratory Strain AB33.

In nature, hyphal growth of Ustilago maydis is strictly coupled to plant infection and initiated after mating of two genetically compatible yeast cells on the plant surface. Compatibility is defined by mating type loci: the biallelic a locus mediates recognition and fusion through a pheromone/receptor system, whereas the multiallelic b locus, encoding transcription factor subunits bEast (bE) and bWest (bW), establishes and maintains the dikaryotic hypha controlled via the assembly of compatible subunits into active heterodimers after plasmogamy. The laboratory strain AB33 enables a controlled

Dual mechanisms of olfactory plasticity: Transient modulation by phosphatidylcholine and D-chiro-inositol and sustained regulation via the sex peptide-juvenile hormone system.

Insects exhibit olfactory plasticity to optimize their survival and reproduction, which ensures that their sensory preferences flexibly meets their needs. However, a comprehensive understanding of the sequential regulatory mechanisms driving these sensory adjustments is lacking. Addressing this knowledge gap is crucial for clarifying how the olfactory system of insect flexibly meets their reproductive needs. Using electroantennogram (EAG) and single-sensillum recordings, we found that female Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) displayed distinct olfactory preferences for ovipos

The effects of chronic subcutaneous administration of an investigational kisspeptin analog, TAK-683, on gonadotropin-releasing hormone pulse generator activity in goats.

The continuous activation of the kisspeptin receptor by its agonists causes the abrogation of kisspeptin signaling, leading to decreased pulsatile luteinizing hormone (LH) secretion. Employing this phenomenon as a tool for probing kisspeptin action, this study aimed to clarify the role of kisspeptin in gonadotropin-releasing hormone (GnRH) pulse generation in goats. We examined the effects of chronic administration of TAK-683, an investigational kisspeptin analog, on LH secretion, GnRH immunostaining, pituitary responses to exogenous GnRH, and GnRH pulse generator activity, reflected by a char

Molecular analysis of an odorant-binding protein gene in two sympatric species of Lutzomyia longipalpis s.l.

Lutzomyia longipalpis s.l. is the main vector of American visceral leishmaniasis (AVL) and occurs as a species complex. DNA samples from two Brazilian sympatric species that differ in pheromone and courtship song production were used to analyse molecular polymorphisms in an odorant-binding protein (obp29) gene. OBPs are proteins related to olfaction and are involved in activities fundamental to survival, such as foraging, mating and choice of oviposition site. In this study, the marker obp29 was found to be highly polymorphic in Lu. longipalpis s.l. , with no fixed differences observed between

Hydrogen sulfide as a potent predator-derived kairomone mediating fear-related responses in mice.

Olfaction plays a critical role in survival across species, notably in threat detection. Volatile olfactory molecules signaling the presence of a danger in the environment share a specific chemical signature, particularly sulfur-containing moieties detected by the mouse olfactory Grueneberg ganglion (GG) neurons. Our study focuses on one of the most toxic air pollutant, hydrogen sulfide (H2S). We reveal here a novel facet of H2S which acts as a danger signal, a kairomone alerting the prey for the presence of nearby predators as H2S can originate from meat-eater secretions. H2S activates the cy

Knockout of angiotensin converting enzyme-2 receptor leads to morphological aberrations in rodent olfactory centers and dysfunctions associated with sense of smell.

Neuronal morphological characterization and behavioral phenotyping in mouse models help dissecting neural mechanisms of brain disorders. Olfactory dysfunctions and other cognitive problems were widely reported in asymptomatic carriers and symptomatic patients infected with Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2). This led us to generate the knockout mouse model for Angiotensin Converting Enzyme-2 (ACE2) receptor, one of the molecular factors mediating SARS-CoV-2 entry to the central nervous system, using CRISPR-Cas9 based genome editing tools. ACE2 receptors and Transmembr

Diet-induced and mono-genetic obesity alter volatile organic compound signature in mice.

The prevalence of obesity is still rising in many countries, resulting in an increased risk of associated metabolic diseases. In this study we aimed to describe the volatile organic compound (VOC) patterns symptomatic for obesity. We analyzed high fat diet (HFD) induced obese and mono-genetic obese mice (global knock-in mutation in melanocortin-4 receptor MC4R-ki). The source strengths of 208 VOCs were analyzed in ad libitum fed mice and after overnight food restriction. Volatiles relevant for a random forest-based separation of obese mice were detected (26 in MC4R-ki, 22 in HFD mice). Eight v

Comparative analysis of the domestic cat genome reveals genetic signatures underlying feline biology and domestication.

Little is known about the genetic changes that distinguish domestic cat populations from their wild progenitors. Here we describe a high-quality domestic cat reference genome assembly and comparative inferences made with other cat breeds, wildcats, and other mammals. Based upon these comparisons, we identified positively selected genes enriched for genes involved in lipid metabolism that underpin adaptations to a hypercarnivorous diet. We also found positive selection signals within genes underlying sensory processes, especially those affecting vision and hearing in the carnivore lineage. We o

An olfactory subsystem that detects carbon disulfide and mediates food-related social learning.

Olfactory signals influence social interactions in a variety of species. In mammals, pheromones and other social cues can promote mating or aggression behaviors; can communicate information about social hierarchies, genetic identity and health status; and can contribute to associative learning. However, the molecular, cellular, and neural mechanisms underlying many olfactory-mediated social interactions remain poorly understood. Here, we report that a specialized olfactory subsystem that includes olfactory sensory neurons (OSNs) expressing the receptor guanylyl cyclase GC-D, the cyclic nucleot

Identification and characterization of class B scavenger receptor CD36 from the hard tick, Haemaphysalis longicornis.

Scavenger receptors (SRs) are cell-surface proteins and exhibit distinctive ligand-binding properties, recognizing a wide range of ligands that include microbial surface constituents and intact microbes. The class B scavenger receptor CD36 (SRB) is predominantly expressed by macrophages and is considered important in innate immunity. We here show the identification and characterization of SRB from the hard ixodid tick, Haemaphysalis longicornis (HlSRB). The full-length cDNA was 2,908 bp, including an ORF encoding of 1,518 amino acids with a pI value of 5.83. H. longicornis SRB contains a hydro

Renaming the 'OS-D/CSP' Family (Part 1): '4-Cysteine Soluble Proteins' (4CSPs)-Molecular Nomenclature, Structure, Expression, Evolution, Tissue-Distribution, and Pleiotropy.

Chemosensory proteins (CSPs) are found in the olfactory sensory organs (antennae and maxillary palps) and/or gustatory sensory organs (labellum and legs) and have long been accepted to function through the binding of odorants. However, the same CSPs are also expressed in many tissues other than olfactory and gustatory organs, such as the gut, brain, fat body, wing, epidermis, Corpora allata, salivary gland, pheromone gland, prothoracic gland, etc. In this report, we suggest renaming the "chemosensory protein (CSP)" the "4-Cysteine Soluble Protein (4CSP)". This paradigm and nomenclature shift i

fruitless is sex-differentially spliced and is important for the courtship behavior and development of silkmoth Bombyx mori.

Sexual dimorphisms of the brain play essential roles in successful reproduction. Silkmoth Bombyx mori exhibits extensive sexual differences in sexual behavior, as well as their morphology. Although the neural circuits that transmit information about sex pheromone in the male brain are extensively analyzed, the molecular mechanisms that regulate their development are still elusive. In the present study, we focused on the silkmoth ortholog of fruitless (fru) as a candidate gene that regulates sexual dimorphisms of the brain. fru transcripts were expressed from multiple promoters in various tissu

Chronic Exposure to Oxazepam Pollution Produces Tolerance to Anxiolytic Effects in Zebrafish (Danio rerio).

Environmental concentrations of the anxiolytic drug oxazepam have been found to disrupt antipredator behaviors of wild fish. Most experiments exposed fish for a week, while evidence from mammals suggests that chronic exposure to therapeutic concentrations of benzodiazepines (such as oxazepam) results in the development of tolerance to the anxiolytic effects. If tolerance can also develop in response to the low concentrations found in the aquatic environment, it could mitigate the negative effects of oxazepam pollution. In the current study, we exposed wild-caught zebrafish to oxazepam (∼7 μg L

Histological Properties of Main and Accessory Olfactory Bulbs in the Common Hippopotamus.

The olfactory system of mammals comprises a main olfactory system that detects hundreds of odorants and a vomeronasal system that detects specific chemicals such as pheromones. The main (MOB) and accessory (AOB) olfactory bulbs are the respective primary centers of the main olfactory and vomeronasal systems. Most mammals including artiodactyls possess a large MOB and a comparatively small AOB, whereas most cetaceans lack olfactory bulbs. The common hippopotamus (Hippopotamus amphibius) is semiaquatic and belongs to the order Cetartiodactyla, family Hippopotamidae, which seems to be the closest

The CatSper channel: a polymodal chemosensor in human sperm.

The sperm-specific CatSper channel controls the intracellular Ca(2+) concentration ([Ca(2+)](i)) and, thereby, the swimming behaviour of sperm. In humans, CatSper is directly activated by progesterone and prostaglandins-female factors that stimulate Ca(2+) influx. Other factors including neurotransmitters, chemokines, and odorants also affect sperm function by changing [Ca(2+)](i). Several ligands, notably odorants, have been proposed to control Ca(2+) entry and motility via G protein-coupled receptors (GPCRs) and cAMP-signalling pathways. Here, we show that odorants directly activate CatSper
