Pramipexole modulates the neural network of reward anticipation.

Pramipexole is widely prescribed to treat Parkinson's disease. It has been found to cause impulse control disorders such as pathological gambling. To examine how pramipexole modulates the network of reward anticipation, we carried out a pharmacological functional magnetic resonance imaging study with a double-blind, within-subject design. During the anticipation of monetary rewards, pramipexole increased the activity of the nucleus accumbens (NAcc), enhanced the interaction between the NAcc and the anterior insula, but weakened the interaction between the NAcc and the prefrontal cortex. These

kappa-Opioid receptor signaling and brain reward function.

The dynorphin-like peptides have profound effects on the state of the brain reward system and human and animal behavior. The dynorphin-like peptides affect locomotor activity, food intake, sexual behavior, anxiety-like behavior, and drug intake. Stimulation of kappa-opioid receptors, the endogenous receptor for the dynorphin-like peptides, inhibits dopamine release in the striatum (nucleus accumbens and caudate putamen) and induces a negative mood state in humans and animals. The administration of drugs of abuse increases the release of dopamine in the striatum and mediates the concomitant rel

An fMRI study of reward-related probability learning.

The human striatum has been implicated in processing reward-related information. More recently, activity in the striatum, particularly the caudate nucleus, has been observed when a contingency between behavior and reward exists, suggesting a role for the caudate in reinforcement-based learning. Using a gambling paradigm, in which affective feedback (reward and punishment) followed simple, random guesses on a trial by trial basis, we sought to investigate the role of the caudate nucleus as reward-related learning progressed. Participants were instructed to make a guess regarding the value of a

Rare Loot Box Rewards Trigger Larger Arousal and Reward Responses, and Greater Urge to Open More Loot Boxes.

Loot boxes are a purchasable video-game feature consisting of randomly determined, in-game virtual items. Due to their chance-based nature, there is much debate as to whether they constitute a form of gambling. We sought to address this issue by examining whether players treat virtual loot box rewards in a way that parallels established reward reactivity for monetary rewards in slots play. Across two sets of experiments, we show that loot boxes containing rarer items are more valuable, arousing, rewarding and urge-inducing to players, similar to the way slots gamblers treat rare large wins in

Disrupting D1-NMDA or D2-NMDA receptor heteromerization prevents cocaine's rewarding effects but preserves natural reward processing.

Addictive drugs increase dopamine in the nucleus accumbens (NAc), where it persistently shapes excitatory glutamate transmission and hijacks natural reward processing. Here, we provide evidence, from mice to humans, that an underlying mechanism relies on drug-evoked heteromerization of glutamate N-methyl-d-aspartate receptors (NMDAR) with dopamine receptor 1 (D1R) or 2 (D2R). Using temporally controlled inhibition of D1R-NMDAR heteromerization, we unraveled their selective implication in early phases of cocaine-mediated synaptic, morphological, and behavioral responses. In contrast, preventing

Contrasting the classic effect of reward on attentional priority maps: behavioral evidence for an attentional gating of spatially-specific reward signals.

The control of selective visuo-spatial attention is known to rely on multiple signals, including reward. While most attention control signals are well acknowledged to interact with each other, the case of reward has led to mixed findings supporting either its interaction with, or its independence from other signals. We addressed this issue using a 4-item search task that entailed spatially biased reward contingencies, with one of the four locations associated with high rewards for correct target discriminations. In different conditions, the target, a distractor or neither of them could be phys

High Risk For High Reward? Latent Profiles Of Impulsivity and Reward Sensitivity in Relation to Alcohol Use Outcomes Among College Students.

Impulsivity and reward sensitivity are risk factors linked to heightened alcohol use and related consequences. However, scant research has explored the influence of impulsivity and reward sensitivity on alcohol-related outcomes. The current study used latent profile analysis to determine (1) whether there were profiles of impulsivity and reward sensitivity and (2) if these profiles were characterized by alcohol use outcomes. Participants were 291 college students who completed a 40-minute online survey that assessed impulsivity, reward sensitivity, alcohol use, and alcohol-related consequences

Attentional disengagement difficulties toward game-related social reward and avoidance toward real-life social reward among individuals with Internet gaming disorder.

Individuals with Internet Gaming Disorder (IGD) exhibit behaviors of immersion in game social interactions and detachment from real-life social interactions. Social Motivation Theory posits that social rewards incentivize social activities, while impairments in social reward processing may lead to such behaviors. Moreover, attentional bias plays a crucial role in reward processing. However, no studies have examined attentional bias toward social rewards in individuals with IGD. Therefore, this study utilized a dot-probe paradigm to compare attentional bias toward game-related social rewards (G

Prior Cocaine Exposure Increases Firing to Immediate Reward While Attenuating Cue and Context Signals Related to Reward Value in the Insula.

The insula contributes to behavioral control and is disrupted by substance abuse, yet we know little about the neural signals underlying these functions or how they are disrupted after chronic drug self-administration. Here, male and female rats self-administered either cocaine (experimental group) or sucrose (control) for 12 consecutive days. After a 1 month withdrawal period, we recorded from insula while rats performed a previously learned reward-guided decision-making task. Cocaine-exposed rats were more sensitive to value manipulations and were faster to respond. These behavioral changes

The protective effect of operant social reward on cocaine self-administration, choice, and relapse is dependent on delay and effort for the social reward.

Social reinforcement-based treatments are effective for many, but not all, people with addictions to drugs. We recently developed an operant rat model that mimics features of one such treatment, the community-reinforcement approach. In this model, rats uniformly choose social interaction over methamphetamine or heroin. Abstinence induced by social preference protects against the incubation of drug-seeking that would emerge during forced abstinence. Here, we determined whether these findings generalize to cocaine and whether delaying or increasing effort for social interaction could reveal poss

Size and probability of rewards modulate the feedback error-related negativity associated with wins but not losses in a monetarily rewarded gambling task.

Feedback error-related negativity (fERN) has been referred to as a negative deflection in the event related potential (ERP), which distinguishes between wins and losses in terms of expected and unexpected outcomes. Some studies refer to the "expected outcome" as the probability to win vs. to lose, and others as expected size of rewards. We still do not know much about whether these alternative interpretations of "expected outcome" affect the fERN in a different manner, nor do we know the effect of their interaction in an expected value fashion. We set a gambling task with four game categories;

The claustrum as a central hub for reward and threat processing.

The claustrum (CLA) complex is composed of the CLA and the dorsal endopiriform nucleus. The CLA is a small, evolutionarily conserved brain structure situated between the striatum and insular cortex. Although the CLA is present in nearly all mammals, its precise anatomical delineation, particularly in rodents, remains poorly defined due to unclear boundaries with adjacent cortical regions. Interest in the CLA has augmented following the influential hypothesis proposed by Crick and Koch regarding its potential involvement in consciousness. Over the past two decades, accumulating evidence has imp

Blunted anticipation, but not consummation, of food rewards in depression.

Anhedonia is a core symptom of major depressive disorder (MDD) and includes anticipatory and consummatory reward processes. Yet some measures still equate anhedonia with reduced consummatory pleasure and have rarely been validated against behavioral tasks. In a cross-sectional study, 52 patients with MDD and 51 healthy control participants (HCPs) completed metabolic assessments and repeated food reward ratings, measuring wanting and liking from anticipation to consumption. Patients with MDD showed blunted anticipatory wanting instead of consummatory liking. Once food rewards were available, wa

Perceived reward certainty in the assessment of delay discounting.

Reward delays are often associated with reduced probability of reward, although standard assessments of delay discounting do not specify degree of reward certainty. Thus, the extent to which estimates of delay discounting are influenced by uncontrolled variance in perceived reward certainty remains unclear. Here we examine 370 participants who were randomly assigned to complete a delay discounting task when reward certainty was either unspecified (n=184) or specified as 100% (n = 186) in the task trials and task instructions. We examined potential group differences in (a) perceived reward cert

A short burst of reward curbs the addictiveness of ketamine.

An analysis of ketamine and cocaine use in mice reveals that the drugs trigger release of the neurotransmitter dopamine through different mechanisms, and indicates that the risk of addiction to ketamine is low.

FGF2 is an endogenous regulator of alcohol reward and consumption.

Alcohol use disorder (AUD) is a chronic, relapsing disorder, characterized by escalating alcohol drinking and loss of control, with very limited available treatments. We recently reported that the expression of fibroblast growth factor 2 (Fgf2) is increased in the striatum of rodents following long-term excessive alcohol drinking and that the systemic or intra-striatal administration of recombinant FGF2 increases alcohol consumption. Here, we set out to determine whether the endogenous FGF2 plays a role in alcohol drinking and reward, by testing the behavioural phenotype of Fgf2 knockout mice.
