Numerical modelling of blood rheology and platelet activation through a stenosed left coronary artery bifurcation.

Coronary bifurcations are prone to atherosclerotic plaque growth, experiencing regions of reduced wall shear stress (WSS) and increased platelet adhesion. This study compares effects across different rheological approaches on hemodynamics, combined with a shear stress exposure history model of platelets within a stenosed porcine bifurcation. Simulations used both single/multiphase blood models to determine which approach best predicts phenomena associated with atherosclerosis and atherothrombosis. A novel Lagrangian platelet tracking model was used to evaluate residence time and shear history

Promotion of whole blood rheology after vitamin C supplementation: focus on red blood cells 1.

Hemorheological properties represent significant contributors in the pathogenesis of cardiovascular diseases. As plasma vitamin C is inversely associated with blood viscosity in humans, we aimed to characterize the effect of vitamin C supplementation on hemorheology with an emphasis on erythrocyte functions. Twenty young healthy volunteers were asked to take vitamin C (1000 mg per day) for 3 weeks. We observed beneficial effect of intervention on multiple hemorheological parameters: whole blood viscosity in the range of medium to high shear rates, Casson yield stress, complex viscosity, and st

Reduced Mucin-7 (Muc7) Sialylation and Altered Saliva Rheology in Sjögren's Syndrome Associated Oral Dryness.

Sjögren's syndrome is a chronic autoimmune disorder characterized by lymphocytic infiltration and hypofunction of salivary and lacrimal glands. This loss of salivary function leads to oral dryness, impaired swallowing and speech, and increased infection and is associated with other autoimmune diseases and an increased risk of certain cancers. Despite the implications of this prevalent disease, diagnosis currently takes years, partly due to the diversity in patient presentation. Saliva is a complicated biological fluid with major constituents, including heavily glycosylated mucins MUC5B and MUC

Shear rheology of mixed protein adsorption layers vs their structure studied by surface force measurements.

The hydrophobins are proteins that form the most rigid adsorption layers at liquid interfaces in comparison with all other investigated proteins. The mixing of hydrophobin HFBII with other conventional proteins is expected to reduce the surface shear elasticity and viscosity, E(sh) and η(sh), proportional to the fraction of the conventional protein. However, the experiments show that the effect of mixing can be rather different depending on the nature of the additive. If the additive is a globular protein, like β-lactoglobulin and ovalbumin, the surface rigidity is preserved, and even enhanced

Blood rheology abnormalities and vascular cell adhesion mechanisms in sickle cell trait carriers during exercise.

Sickle cell trait (SCT) is usually considered a benign disorder compared with sickle cell anemia (SS hemoglobinopathy). However, several authors have reported cases of exercise-related sudden death in this population. Among the mechanisms that could be involved in these fatal complications, vaso-occlusive processes, such as those occurring in SS hemoglobinopathy, may play a role. In sickle cell anemia, these vaso-occlusive processes involve inflammatory and adhesion molecules such as the cell adhesion molecules (CAM family), which play a role in the firm adhesion of reticulocytes and leukocyte

Altering the interfacial rheology of Pseudomonas aeruginosa and Staphylococcus aureus with N-acetyl cysteine and cysteamine.

INTRODUCTION: Chronic lung infection due to bacterial biofilms is one of the leading causes of mortality in cystic fibrosis (CF) patients. Among many species colonizing the lung airways, Pseudomonas aeruginosa and Staphylococcus aureus are two virulent pathogens involved in mechanically robust biofilms that are difficult to eradicate using airway clearance techniques like lung lavage. To remove such biological materials, glycoside hydrolase-based compounds are commonly employed for targeting and breaking down the biofilm matrix, and subsequently increasing cell susceptibility to antibiotics.

Hyaluronic Acid in the Intestinal Tract: Influence of Structure, Rheology, and Mucoadhesion on the Intestinal Uptake in Rats.

Oral hyaluronic acid (HA) is a ubiquitous biopolymer that has gained attention as a treatment for local or systemic diseases. Here, we prepared and characterized structures of free HA (f-HA) with a high (>105 Da), intermediate (≤105 Da), and low (≤104 Da) average molar mass (MM); nanoparticles crosslinked with adipic dihydrazide (n-HA); and mixed formulations (mixed-HA) containing f-HA and n-HA. MM distribution determined the structure, hydrodynamic diameter, and zeta potential of the f-HAs. Crosslinking changed the physicochemical properties in n-HA. In vitro tack adhesion assays, using mucin

The effect of HPMC and MC as pore formers on the rheology of the implant microenvironment and the drug release in vitro.

Porous implants or implantable scaffolds used for tissue regeneration can encourage tissue growth inside the implant and provide extended drug release. Water-soluble polymers incorporated into a biodegradable or inert implant matrix may leach out upon contact with biological fluids and thereby gradually increasing the porosity of the implant and simultaneously release drug from the implant matrix. Different molecular weight grades of methylcellulose (MC) and hydroxypropyl methylcellulose (HPMC) were mixed with polylactide and extruded into model implants containing nitrofurantoin as a model dr

Polysaccharides As Viscosupplementation Agents: Structural Molecular Characteristics but Not Rheology Appear Crucial to the Therapeutic Response.

INTRODUCTION: Most clinical studies and basic research document viscosupplementation (VS) in terms of effectiveness and safety, but only a few highlight its molecular mechanisms of action. Besides, there is generally focus on hyaluronic acid (HA) as being the most relevant polysaccharide to reach the clinical endpoints, attributing its effect mainly to its unique viscoelastic properties, related to a high-molecular weight and gel formulation. Usually, studies do not approach the possible biological pathways where HA may interfere, and there is a lack of reports on other biocompatible polysacch

Effects of CaCO3 treatment on the morphology, crystallinity, rheology and hydrolysis of gelatinized maize starch dispersions.

Using calcium salts instead of lime allows for an ecological nixtamalization of maize grains, where the negative contamination impact of the traditional lime nixtamalization is reduced. This work assessed the effects of calcium carbonate (0.0-2.0%w/w CaCO3) on the morphology, crystallinity, rheology and hydrolysis of gelatinized maize starch dispersions (GMSD). Microscopy analysis showed that CaCO3 changed the morphology of insoluble remnants (ghosts) and decreased the degree of syneresis. Analysis of particle size distribution showed a slight shift to smaller sizes as the CaCO3 was increased.

Solvent effects on modulus of poly(propylene oxide)-based organogels as measured by cavitation rheology.

A series of novel organogels were synthesized from poly(propylene oxide) (PPO) functionalized with main chain urea moieties which provided rapid gelation and high moduli in a variety of solvents. Three different molecular weight PPOs were used in this study: 430, 2000, and 4000 g mol(-1), each with α,ω-amino-end groups. Four urea groups were introduced into the main chain by reaction with hexamethylene diisocyanate followed by subsequent reaction with a monofunctional alkyl or aromatic amine. This PPO/urea gelator was found to form gels in carbon tetrachloride, chloroform, dichloromethane, tol

Effects of hydration and dehydration on blood rheology in sickle cell trait carriers during exercise.

This study compared the hemorheological responses of a group of sickle cell trait (SCT) carriers with those of a control (Cont) group in response to 40 min of submaximal exercise (exercise intensity, 55% aerobic peak power) performed in two conditions: one with water offered ad libitum, i.e., the hydration (Hyd) condition, and one without water, i.e., the dehydration (Dehyd) condition. Blood and plasma viscosities, as well as red blood cell rigidity, were determined at rest, at the end of exercise, and at 2 h recovery with a cone plate viscometer at high shear rate and 37 degrees C. [Rheology of human cervical mucus--with special reference to measurement of viscoelasticity using various rheometers].

Cervical mucus is egg white-like mucus that increases in the female cervical canal and external os of the uterus at ovulatory phase. Spermatozoa climb up through this mucus to the uterine tube, where they meet an oocyte causing fertilization. If this cervical mucus has quality or quantity problems, spermatozoa are unable to ascend. Then, so-called infertility due to cervical hostility would occur(5% in general). One day before ovulation, because there is the highest amount of cervical mucus, the greatest degree of Spinnbarkeit and the lowest degree of viscoelasticity, spermatozoa are able to p

Hydroxypropyl-phenyltriazole-functionalized chitosan as an adaptive amphiphilic emulsifier: Interfacial stabilization, rheology, and antioxidant activity.

Chitosan (CS) is an attractive bio-based macromolecule for emulsion stabilization, but its strong hydrophilicity, pH-dependent solubility, and limited oil-water interfacial affinity restrict its performance as a robust polymeric emulsifier. Herein, hydroxypropyl-phenyltriazole-functionalized chitosan (CS-HP-PhTrz) was synthesized via CuAAC click chemistry as an amphiphilic CS derivative with improved interfacial adaptability and antioxidant functionality. Successful functionalization was supported by FTIR, UV-Vis, 1H NMR, XRD, TGA, SEM-EDX, and XPS analyses, with an estimated degree of substit

Tuning the Gel Network Structure and Rheology of Acid-Induced Casein Gels via Thiol Blocking.

This study systematically investigates how thiol-disulfide interactions influence the structure and mechanical properties of casein gels. Acid gels were prepared from suspensions of micellar casein (MC) powder that were heat-treated at 70 °C. Thiol groups were variably blocked with N-ethylmaleimide (NEM). The gels were characterized using stress-strain measurements, rheological analyses, and confocal microscopy. The stress-strain curves exhibited a biphasic behavior, with an initial linear elastic phase followed by a linear plastic region and a nonlinear failure zone. Compared to control sampl

Fallopian tube rheology regulates epithelial cell differentiation and function to enhance cilia formation and coordination.

The rheological properties of the extracellular fluid in the female reproductive tract vary spatiotemporally, however, the effect on the behaviour of epithelial cells that line the tract is unexplored. Here, we reveal that epithelial cells respond to the elevated viscosity of culture media by modulating their development and functionality to enhance cilia formation and coordination. Specifically, ciliation increases by 4-fold and cilia beating frequency decreases by 30% when cells are cultured at 100 mPa·s. Further, cilia manifest a coordinated beating pattern that can facilitate the formation

Experimental characterization of blood rheology and development of a computational rheological Index during progressive hemorrhagic shock.

Background: Hemorrhagic shock is associated with profound disturbances of microcirculatory perfusion in which alterations of blood rheology may contribute to tissue hypoxia and organ dysfunction. Historically, the concept of hemorrhagic shock has evolved from an initial focus on systemic hemodynamic failure toward recognition of the critical role of microcirculatory dysfunction and impaired tissue oxygen delivery. More recent experimental and clinical research has further emphasized the contribution of blood rheological abnormalities to the progression of hemorrhagic shock and organ dysfunctio

Rheology of CHO Cell Suspensions and Its Effects on High-Density Cultivation Process and Bioreactor Design.

With the rapidly growing demand for monoclonal antibodies (mAbs) worldwide, optimizing the high-density and ultra-high-density cultivation processes of Chinese hamster ovary (CHO) cells has become crucial for enhancing production efficiency. Shear stress and mass transfer have always been the vital operating parameters for the bioreactor in creating a suitable microenvironment for cell growth and antibody production. However, researchers have not actively focused on the rheology of CHO cell suspensions and its impact on these parameters in bioreactors. The factors influencing the rheology of s
