Rhizosphere competitiveness of trichloroethylene-degrading, poplar-colonizing recombinant bacteria.

Indigenous bacteria from poplar tree (Populus canadensis var. eugenei 'Imperial Carolina') and southern California shrub rhizospheres, as well as two tree-colonizing Rhizobium strains (ATCC 10320 and ATCC 35645), were engineered to express constitutively and stably toluene o-monooxygenase (TOM) from Burkholderia cepacia G4 by integrating the tom locus into the chromosome. The poplar and Rhizobium recombinant bacteria degraded trichloroethylene at a rate of 0.8 to 2.1 nmol/min/mg of protein and were competitive against the unengineered hosts in wheat and barley rhizospheres for 1 month (coloniz

Characterization of efficient plant-growth-promoting bacteria isolated from Sulla coronaria resistant to cadmium and to other heavy metals.

The inoculation of plants with plant-growth-promoting rhizobacteria has become a priority in the phytoremediation of heavy-metal-contaminated soils. A total of 82 bacteria were isolated from Sulla coronaria root nodules cultivated on four soil samples differently contaminated by heavy metals. The phenotypic characterization of these isolates demonstrated an increased tolerance to cadmium reaching 4.1mM, and to other metals, including Zn, Cu and Ni. Polymerase Chain Reaction/Restriction Fragment Length Polymorphism (PCR/RFLP) analysis showed a large diversity represented by genera related to Ag

Periplasmic cyclic 1,2-beta-glucan in Brucella spp. is not osmoregulated.

Biosynthesis of periplasmic cyclic 1,2-beta-glucans in Brucella ovis strain REO198 and B. abortus strain 519 was found to be carried out by membrane-bound enzymes that use UDP-glucose (UDP-Glc) as donor substrate. Contrary to what happens in species of the genera Agrobacterium and Rhizobium, the accumulation of the reaction products in Brucella appeared not to be osmotically regulated. Incubation of permeabilized cells with UDP-[14C]Glc led to the formation of soluble neutral cyclic 1,2-beta-glucans and [14C]glucose-labelled glucoproteins. PAGE of pulse-chase experiments carried out with perme

Cytokine inducing activities of rhizobial and mesorhizobial lipopolysaccharides of different lethal toxicity.

The lethality and cytokines-inducing activity of lipopolysaccharides (LPS) obtained from nodulating bacteria, Rhizobium leguminosarum and Mesorhizobium loti, were compared to those of Salmonella typhimurium LPS. The activity of R. leguminosarum LPS was almost comparable to Salmonella endotoxin in terms of lethality, Limulus lysate gelating activity and in vivo tumor necrosis factor-alpha (TNF-alpha), interleukin-1beta (IL-1beta), interleukin-6 (IL-6) and interferon-gamma (IFN-gamma) induction capacity. In contrast to high lethal toxicity of Rhizobium LPS, the lethality of LPS isolated from Mes

Plant-microbe-microbe interactions influence the faba bean nodule colonization by diverse endophytic bacteria.

Legume root nodules harbor rhizobia and other non-nodulating endophytes known as nodule-associated bacteria (NAB) whose role in the legume symbiosis is still unknown. We analysed the genetic diversity of 34 NAB isolates obtained from the root nodules of faba bean grown under various soil conditions in Egypt using 16S rRNA and concatenated sequences of three housekeeping genes. All isolates were identified as members of the family Enterobacteriaceae belonging to the genera Klebsiella, Enterobacter and Raoultella. We identified nine enterobacterial genospecies, most of which have not been previo

Gnotobiotic growth and phosphorus limitation of Arabidopsis thaliana and co-occurring microbes on phosphated iron oxides.

The macronutrient phosphorus is vital for sustaining cellular processes in all life forms. Due to its frequent adsorption on iron minerals, phosphorus bioavailability is low in many soils. While the abiotic adsorption of phosphate on iron minerals has been well studied, the direct effects of this process on bioavailability to plants and microbes has not been thoroughly investigated in a simplified laboratory system. We developed a hydroponic growth system that uses hydrous ferric oxide (HFO) to induce phosphorus limitation and can enable both plant and microbial cultivation as well as gnotobio

Natural genetic variation in rhizobia and alfalfa influence nodule shape and symbiotic performance.

In many host-microbe symbioses, hosts develop specialized organs that house microbial symbionts. In legume-rhizobium symbiosis, these organs are called root nodules. While many studies have focused on host traits like plant growth, nodule number, and their structural changes, nodule morphology may influence rhizobial benefits (e.g., bacterial population size). Here, we investigated how variation in alfalfa nodule morphologies influences the fitness outcome of both partners. We used 117 strains and 3 alfalfa varieties to test how genetic variation influenced 1) nodule branch number, area, and r

Differential bacterial endophytome in Foc-resistant banana cultivar displays enhanced antagonistic activity against Fusarium oxysporum f.sp. cubense (Foc).

Diverse endophytes with multiple functions exist in different banana cultivars. However, the diversity of cultivable bacterial endophytome that contributes to antifungal activity against Fusarium oxysporum f.sp. cubense (Foc) in resistant and susceptible banana cultivars is mostly unknown. In the present study, we isolated bacterial endophytes from resistant Yengambi KM5 (AAA) and susceptible banana cultivar Ney Poovan (AB) to determine the diversity of cultivable bacterial endophytes. Our study revealed the presence of 56 cultivable bacterial endophytes and 6 nectar-associated bacteria in YKM

Hydrogen Sulfide Promotes Nodulation and Nitrogen Fixation in Soybean-Rhizobia Symbiotic System.

The rhizobium-legume symbiotic system is crucial for nitrogen cycle balance in agriculture. Hydrogen sulfide (H2S), a gaseous signaling molecule, may regulate various physiological processes in plants. However, whether H2S has regulatory effect in this symbiotic system remains unknown. Herein, we investigated the possible role of H2S in the symbiosis between soybean (Glycine max) and rhizobium (Sinorhizobium fredii). Our results demonstrated that an exogenous H2S donor (sodium hydrosulfide [NaHS]) treatment promoted soybean growth, nodulation, and nitrogenase (Nase) activity. Western blotting

Intracellular lifestyle of Brucella spp. Common genes with other animal pathogens, plant pathogens, and endosymbionts.

Brucella spp. are intracellular pathogens that belong, like Agrobacterium, Rhizobium and Rickettsia, to the alpha-2-subgroup of proteobacteria. The genome organization of most Brucella spp. is characterized by the presence of two chromosomes. The intracellular lifestyle of Brucella, as well as the possible genes involved in pathogenesis and host cell signaling, are discussed, including the presence of genes with high similarity to those from other animal pathogens, plant pathogens and endosymbionts.

Successional dynamics of microbial communities and remediation of petroleum-contaminated soils during pilot-scale bioaugmentation.

Petroleum-contaminated soils pose serious global ecological and health risks. This pilot-scale study (120 days, 25 m2) aimed to evaluate the field efficacy of a compound microbial agent, ECT (composed of Ralstonia sp. CP, Rhizobium sp. BX, and Acinetobacter sp. FL), for bioaugmented remediation of heavily petroleum-polluted soil (initial TPH: 45,178 mg kg-1). ECT achieved an excellent TPH removal rate of 91.61%, with exogenous strains successfully colonizing the soil and maintaining high relative abundances (Ralstonia 42.1%, Rhizobium 10.0%, Acinetobacter 12.3% at day 120). Post-Release Changes of the Goat's Rue Rhizobia (Neorhizobium galegae bv. orientalis) Genomes in the Field: Prospects for Breeding the Symbiotically Effective Strains.

Root nodule bacteria (rhizobia) fixing N2 in cooperation with legumes are broadly used for improving production of these plants inoculated with strains selected for high symbiotic activity. These bacteria are characterized by enormous evolutionary plasticity based on genomic rearrangements and horizontal gene transfer. However, this native plasticity is still poorly used for symbiotic improvement of the inoculant strains. In the presented paper, we analyzed genomic structure and symbiotic efficacy in ten clones isolated from nodules formed by the highly effective strain Gr30-13 of goat's rue r

H2 S works synergistically with rhizobia to modify photosynthetic carbon assimilation and metabolism in nitrogen-deficient soybeans.

Hydrogen sulfide (H2 S) performs a crucial role in plant development and abiotic stress responses by interacting with other signalling molecules. However, the synergistic involvement of H2 S and rhizobia in photosynthetic carbon (C) metabolism in soybean (Glycine max) under nitrogen (N) deficiency has been largely overlooked. Therefore, we scrutinised how H2 S drives photosynthetic C fixation, utilisation, and accumulation in soybean-rhizobia symbiotic systems. When soybeans encountered N deficiency, organ growth, grain output, and nodule N-fixation performance were considerably improved owing

Diverse plant growth-promoting bacteria as microsymbionts in nodules of Leucaena leucocephala.

This study characterized 18 endophytic bacterial isolates in association with the root nodules of Leucaena leucocephala through phenotypic and genotypic analyses. The endophytes were associated with the plants and exhibited diverse plant growth-promoting (PGP) traits. Phosphate solubilization was observed in 39% of isolates at high levels and 33.3% at moderate levels. Siderophore production was prevalent, with 38.9% displaying high and 33.3% moderate production, aiding iron uptake. Indole-3-acetic acid (IAA) production varied (32.15 to 86.28 µg/ml) among the isolates. Notably, 94.4% of isolate

The 75-kilodalton antigen of Bartonella bacilliformis is a structural homolog of the cell division protein FtsZ.

A genomic library of Bartonella bacilliformis was constructed and screened with human anti-Bartonella serum from a patient with the chronic, verruga peruana phase of bartonellosis. An immunoreactive clone isolated from this library was found to code for a 591-amino-acid protein with a high degree of sequence similarity to the FtsZ family of proteins. The degree of amino acid identity between the B. bacilliformis protein (FtsZ[Bb]) and the other FtsZ proteins is especially pronounced over the N-terminal 321 amino acids (N-terminal domain) of the sequence, with values ranging from 45% identity f

Modulation of Symbiotic Compatibility by Rhizobial Zinc Starvation Machinery.

Pathogenic bacteria need high-affinity zinc uptake systems to counteract the nutritional immunity exerted by infected hosts. However, our understanding of zinc homeostasis in mutualistic systems such as the rhizobium-legume symbiosis is limited. Here, we show that the conserved high-affinity zinc transporter ZnuABC and accessory transporter proteins (Zip1, Zip2, and c06450) made cumulative contributions to nodulation of the broad-host-range strain Sinorhizobium fredii CCBAU45436. Zur acted as a zinc-dependent repressor for the znuC-znuB-zur operon, znuA, and c06450 by binding to the associated

Optimization of trichloroethylene degradation using soluble methane monooxygenase of Methylosinus trichosporium OB3b expressed in recombinant bacteria.

By complementing cell-free extracts of Pseudomonas putida F1/pSMMO20 with purified soluble methane monooxygenase (sMMO) components of Methylosinus trichosporium OB3b, the low cloned-gene sMMO activity in the recombinant strain was found to be due to incomplete activity of the hydroxylase component. To address this incomplete activity, additional sMMO-expressing strains were formed by transferring mmo-containing pSMMO20 and pSMMO50 into various bacterial species including pseudomonads and alpha-2 subdivision strains such as methanotrophs, methylotrophs, Agrobacterium tumefaciens A114, and Rhizo

Managing Pneumonia Due to Rare Non-Fermenting Gram-Negative Bacteria: Epidemiology, Risk Factors and Therapeutic Strategies.

Pneumonia remains a leading cause of morbidity and mortality worldwide, with bacterial pathogens contributing significantly to its burden. While Pseudomonas aeruginosa and Acinetobacter baumannii complex are well-recognized non-fermenting Gram-negative bacteria (NFGNB) causing severe pneumonia, particularly in healthcare settings, an expanding array of other, rarer NFGNB species is increasingly implicated. These species include, but are not limited to, Achromobacter spp., Ochrobactrum spp., Burkholderia spp., Aeromonas spp., Roseomonas spp., Elizabethkingia spp., Chryseobacterium spp. Alcalige
