Immune Reconstitution Syndrome (IRIS) has been associated with a variety of infections in patients with human immunodeficiency virus (HIV). However, we are reporting the first case of IRIS secondary to Rhodococcus equi (R. equi) in a patient with HIV. We report the case of a 48-year-old male found to have HIV infection in the setting of Burkitt's lymphoma. While on anti-retroviral therapy and chemotherapy, he had developed IRIS secondary to R. equi that manifested as a cavitating pneumonia. This report outlines the successful management of the R. equi infection with the use of a combination of

Virulence of Rhocococcus equi ATCC 33701 and its plasmid-cured derivative ATCC 33701P- was compared in BALB/c and C3H/HeJ mice in terms of bacterial growth kinetics and histological changes in the liver, spleen and lungs, and humoral immune responses. Injection with a sublethal dose of 10(6) ATCC 33701 in mice resulted in microabscess formation after rapid multiplication in the liver and spleen by day 4, and then the bacteria were gradually eliminated with the formation of granuloma and the production of specific antibodies against 15- to 17-kDa antigens of the virulent bacteria. By contrast,

Rhodococcus equi is an uncommon opportunistic pathogen increasingly recognized in immunocompromised hosts. It typically presents as cavitary pneumonia with bacteremia, particularly among individuals with advanced HIV infection, but may disseminate to virtually any organ. Because of its diphtheroid morphology, it is frequently dismissed as a contaminant, leading to diagnostic delay. Early recognition, accurate identification using advanced microbiologic tools, and initiation of appropriate combination therapy are essential for optimal outcomes. We describe two cases of R. equi pneumonia with ba

Rhodococcus equi is an important veterinary and zoonotic pathogen that survives inside macrophage phagolysosomes. Nitrate reductase NarGHI is a known virulence factor for this pathogen. This enzyme complex serves as the terminal electron acceptor in anaerobic respiration in other pathogens, such as the phylogenetically related Mycobacterium tuberculosis. A recent study demonstrated that R. equi survives under anaerobic conditions. In this work, we endeavored to uncover the electron transport chain complexes of R. equi. By in silico comparison to known complexes in the Transporter Classificatio

The α,β-desaturation of esters is one of the most challenging transformations in organic synthesis. While transition-metal-catalyzed methods have significantly advanced in the last decades, they often require high catalyst loading and nongreen solvents and rarely enable stereoselective transformations. Here we show that whole cells of Rhodococcus erythropolis enable a dynamic enantioconvergent desaturation of 4,5-disubstituted γ-butyrolactones (Quercus-like lactones), funneling all four initial stereoisomers into a single (R)-configured product with excellent enantioselectivity (up to ee 99%).

Rhodococcus equi is an opportunistic pathogen known to cause pulmonary and extrapulmonary disease among immunocompromised patients. Treatment is frequently challenging due to intrinsic resistance to multiple antibiotic classes. While non-equi Rhodococcus spp. are prevalent, their clinical significance is poorly defined. There is also limited data on antibiotic susceptibility testing (AST) of Rhodococcus infection in humans. We conducted a single-center, retrospective cohort study evaluating clinical characteristics, microbiologic profile, and AST of Rhodococcus infections between June 2012 and

A lipolytic enzyme (Rcut) was discovered from the Rhodococcusstrain (RosL12) isolated from the Antarctic Ross Sea. The corresponding gene composed of 651 bases encoding 216 amino acids. It was found to be a cutinase gene through BLAST search. Rcut has a signal sequence consisting of 29 amino acids. An active Rcut was produced after the intact gene containing the signal sequence was transformed into Escherichia coli Rosetta-gami™ 2 (DE3) pLysS. Rcut was purified through a nickel-nitrilotriacetic acid purification system and a carboxymethyl Sepharose column chromatography. Its specific activity

Rhodococcus erythropolis N9T-4, which is an extremely oligotrophic bacterium, can survive in a completely inorganic medium with no additional carbon source. This bacterium utilizes atmospheric CO2, but does not require any additional energy source such as light and hydrogen gas, required by autotrophic microorganisms. However, its CO2 fixation and energy-acquisition systems in the oligotrophic growth remain unrevealed. We expected N9T-4 to have the transporter(s) that imports essential compound(s) for its oligotrophic growth. Three putative ATP-binding cassette (ABC) transporters were found to

UNLABELLED: The release of SO2 from petroleum products derived from crude oil, which contains sulfur compounds such as dibenzothiophene (DBT), leads to air pollution. The '4S' metabolic pathway catalyzes the sequential conversion of DBT to 2-hydroxybiphenyl via three enzymes encoded by the dsz operon in several bacterial species. DszC (DBT monooxygenase), from Rhodococcus erythropolis D-1 is involved in the first two steps of the '4S' pathway. Here, we determined the first crystal structure of FMN-bound DszC, and found that two distinct conformations occur in the loop region (residues 131-142)

Malakoplakia is a disease especially of the urinary tract with typical plaques most frequently observed in the urinary bladder's mucosa. In the context of immunosuppression malakoplakia can also occur in other organs. Some of these extravesical malakoplakias are associated with an infection by Rhodococcus equi, a rare human pathogen well known from veterinary medicine. Here we present the first case of a pleural malakoplakia without lung involvement caused by a proved Rhodococcus equi infection.

An organic solvent-tolerant bacterium, Rhodococcus opacus B-4, was metabolically engineered to remove sulfur from dibenzothiophene (DBT), a component of crude oil. The resulting recombinant strain ROD2-8 constitutively expressed the Rhodococcus erythropolis IGTS8 genes dszA, dszB, and dszC, encoding dibenzothiophene sulfone monooxygenase, 2-(2'-hydroxyphenyl) benzenesulfinate desulfinase, and dibenzothiophene monooxygenase, respectively, of the 4S pathway to avoid transcriptional inhibition by the sulfate end-product. Unlike the wild-type strain, ROD2-8 grew in mineral salts medium containing

The siderophore produced by Rhodococcus rhodochrous strain OFS, rhodobactin, was isolated from iron-deficient cultures and purified by a combination of XAD-7 absorptive/partition resin column and semi-preparative HPLC. The siderophore structure was characterized using 1D and 2D (1)H, (13)C and (15)N NMR techniques (DQFCOSY, TOCSY, NOESY, HSQC and LR-HSQC) and was confirmed using ESI-MS and MS/MS experiments. The structural characterization revealed that the siderophore, rhodobactin, is a mixed ligand hexadentate siderophore with two catecholate and one hydroxamate moieties for iron chelation.

This work reports on the immobilization and performance of a hydrocarbon-degrading microorganism on polyurethane foam (PUF) in the bioremediation of petroleum hydrocarbons. The ability of four different microorganisms to immobilize on PUF and to degrade various petroleum products (Arabian light crude (ALC), Al-Shaheen crude (ASC), diesel and oil slops) was assessed by measuring the n-alkane fraction remaining in the petroleum products over time. A Rhodococcus sp. (designated as F92) had the highest number of immobilized viable cells (10(9) cells per cm3 PUF) and a maximum attachment efficiency

Rhodococcus ruber MSA14, isolated from oil-polluted coastal sediments in Baja California, Mexico, has previously been shown to degrade pyrene, highlighting its potential for polycyclic aromatic hydrocarbons (PAH) bioremediation. In the present study, MSA14 was subjected to adaptive laboratory evolution using progressively increasing concentrations of pyrene as the sole carbon and energy source, resulting in the isolation of a high-pyrene-adapted strain, R. ruber AD0911. Compared with the parental strain, AD0911 displayed improved growth and pyrene tolerance, modified colony morphology, enhance

Background/Objectives: Microbial biotransformation of monoterpenoids is a promising approach for obtaining bioactive compounds. Rhodococcus species are attractive biocatalysts due to their metabolic versatility and ability to transform hydrophobic substrates. In this study, we investigated the catalytic potential of Rhodococcus qingshengii IEGM 267 toward carveol isomers and explored genomic features that may underlie this activity. Methods: The strain was cultivated in mineral medium supplemented with (-)-trans-carveol. Biotransformation products were analyzed by TLC and GC-MS. The draft geno

INTRODUCTION: We describe cases of contamination of Mycobacterium ulcerans infections (Buruli ulcer) with Rhodococcus erythropolis, a bacterium of environmental origin that is rarely associated with human infection.
CASE PRESENTATION: The infectious pathogen of Buruli ulcer, Mycobacterium ulcerans, was detected and cultured in vitro from two lesion swabs taken from clinically described Buruli ulcer-like patients (4 and 34 years old). Infection by M. ulcerans was confirmed using the WHO-recommended IS2404/IPC-qPCR multiplex analysis of DNA extracts from patient samples, which were subsequently

We present the case of a 47-year-old man living with HIV who had experienced fever, night sweats, cough and occasional haemoptysis for the previous two months. Computed tomography of the chest revealed a lesion measuring 57 mm in diameter with cavitation in the left lower superior segment. His CD4+ T cell count was 220 cells/mm³ and his HIV RNA level was 406,756 copies/ml. Acid-resistant coccobacilli were isolated from the transthoracic biopsy of the lesion and were initially identified as Mycobacterium lentiflavum. However, 16S rDNA sequence analysis identified it as Rhodococcus equi. A histo

Rhodococcus sp. strain HA01, isolated through its ability to utilize dibenzofuran (DBF) as the sole carbon and energy source, was also capable, albeit with low activity, of transforming dibenzo-p-dioxin (DD). This strain could also transform 3-chlorodibenzofuran (3CDBF), mainly by angular oxygenation at the ether bond-carrying carbon (the angular position) and an adjacent carbon atom, to 4-chlorosalicylate as the end product. Similarly, 2-chlorodibenzofuran (2CDBF) was transformed to 5-chlorosalicylate. However, lateral oxygenation at the 3,4-positions was also observed and yielded the novel p

We report a heart transplant recipient with Rhodococcus equi and Cryptococcus neoformans infection. Despite an initially relapsing course and the acquisition of resistance in vivo, the patient responded to 5 months of linezolid and 6 months of fluconazole. This is the 1st R. equi infection to be cured with linezolid in a heart transplant recipient.

Infections of Rhodococcus equi, a well-known pathogen in animals which causes cavitated pneumonia similar to that caused by mycobacteria, were studied in three HIV-infected patients. This microorganism was isolated in the bronchoalveolar washings of two patients and in the sputum of the third. In two patients, Rh. equi represented the first clinical opportunistic manifestation of HIV disease. One patient died of concomitant Pneumocystis infection. The eradication of the microorganism occurred in two out of three patients. It was found that no isolates were resistant to erythromycin, claritromy
