Deletion of the transmembrane protein Prom1b in zebrafish disrupts outer-segment morphogenesis and causes photoreceptor degeneration.

Mutations in human prominin 1 (PROM1), encoding a transmembrane glycoprotein localized mainly to plasma membrane protrusions, have been reported to cause retinitis pigmentosa, macular degeneration, and cone-rod dystrophy. Although the structural role of PROM1 in outer-segment (OS) morphogenesis has been demonstrated in Prom1-knockout mouse, the mechanisms underlying these complex disease phenotypes remain unclear. Here, we utilized a zebrafish model to further investigate PROM1's role in the retina. The Prom1 orthologs in zebrafish include prom1a and prom1b, and our results showed that prom1b,

Predominant rod photoreceptor degeneration in Leber congenital amaurosis.

PURPOSE: An unusual retinal vascular morphology in an enucleated eye from a patient with Leber congenital amaurosis (LCA) has been associated with a mutation in AIPL1. The AIPL1 protein is expressed in the pineal gland and retinal photoreceptors. In the retina, AIPL1 is expressed in both developing cone and rod photoreceptors, but it is restricted to rod photoreceptors in the adult human retina. Therefore, this study was conducted to determine the photoreceptor phenotype in this LCA patient to determine if photoreceptors were differentially affected. METHODS: Additional genetic screening was

Super-resolution microscopy reveals a Rab6a-dependent trafficking hub for rhodopsin at the mammalian rod photoreceptor Golgi.

Rod photoreceptor stability is critical for retinal health and lifelong vision. The proper intracellular trafficking of the photopigment receptor rhodopsin (Rho) is essential for normal rod homeostasis, as Rho mislocalization precedes rod cell death in inherited retinal disorders such as retinitis pigmentosa. Despite its importance, the molecular mechanisms of Rho trafficking in mammalian rods remain largely undefined. In this study, we combined multiple Rho-labeling strategies with super-resolution microscopy to investigate the subcellular organization of Rho in the Golgi complex of mammalian

Morphological evidence of neurotoxicity in retina after methylmercury exposure.

The visual system is particularly sensitive to methylmercury (MeHg) exposure and, therefore, provides a useful model for investigating the fundamental mechanisms that direct toxic effects. During a period of 70 days, adult of a freshwater fish species Hoplias malabaricus were fed with fish prey previously labeled with two different doses of methylmercury (0.075 and 0.75 μgg(-1)) to determine the mercury distribution and morphological changes in the retina. Mercury deposits were found in the photoreceptor layer, in the inner plexiform layer and in the outer plexiform layer, demonstrating a dose

A light-dependent increase in free Ca2+ concentration in the salamander rod outer segment.

1. The Ca(2+) indicator dye fluo-5F was excited by an argon ion laser to measure changes in free Ca(2+) concentration ([Ca2+]i) in the outer segments of isolated salamander rods rapidly exposed to a 0 Ca(2+), 0 Na(+) solution designed to minimise surface membrane Ca(2+) fluxes. Over 30-60 s of laser illumination, the fluorescence first increased rapidly and then declined at a rate that was much slower than in Ringer solution and consistent with previous physiological evidence that 0 Ca(2+), 0 Na(+) solution greatly retards light-induced changes in [Ca(2+)]i. 2. The initial increase in fluoresc

The distribution of peroxide regulating enzymes in the canine eye.

The retina and rod outer segments (ROS), rich in unsaturated lipids, are highly susceptible to autoxidation via free radical mechanisms. In the canine eye, four enzymes, i.e.; p-phenylenediamine (PPD) peroxidase, glutathione peroxidases, catalase and superoxide dismutase which are involved in the production or degradation of peroxides, were measured. All enzymes were present in the retina, ciliary body and iris, but differed in concentration. Superoxide dismutase showed high activity in the ROS. In the retinal pigment epithelium (RPE) a peroxidase utilizing PPD as cosubstrate and superoxide di

Reduction of phagosomes in the vitiligo (C57BL/6-mivit/mivit) mouse model of retinal degeneration.

PURPOSE: The vitiligo (C57BL/6-mivit/mivit) mouse has a slowly progressing retinal degeneration, in which photoreceptor cell nuclei are gradually lost and the retinal pigment epithelium (RPE) is unevenly pigmented. The purpose of the present study was to assess the phagocytic ability of the RPE in the vitiligo mouse by determining whether and when a phagocytic burst occurs in affected mice and whether the number of phagosomes varies between control and affected animals. METHODS: Eyes of control and vitiligo mice 4 to 20 weeks of age were embedded in Spurr. Thin sections were cut and examined

Slowly progressive changes of the retina and retinal pigment epithelium in Briard dogs with hereditary retinal dystrophy. A morphological study.

Seven eyes from 2 generations of Briard dogs (5 weeks--7 years old) with congenital night blindness and (in the second generation) impairment of day vision to varying degrees, were examined by light and electron microscopy. Specimens from 4 locations were studied: the central area, the midperiphery of the tapetal area, the upper periphery and the lower periphery. Disorientation of rod outer segment disc membranes was seen in the 5-week-old dog. Large electron-lucent inclusions were found in the RPE at 3.5 months of age. These inclusions occurred most frequently in the central and midperipheral

Modulation of retinal transducin and phosphodiesterase activities by synthetic peptides of the phosphodiesterase gamma-subunit.

Synthetic peptides corresponding to various regions of the light-activated guanosine 3',5'-cyclic monophosphate phosphodiesterase (PDE) gamma-subunit (PDE gamma) from bovine retinal rod outer segments were synthesized and tested for their ability to inhibit PDE activity, and GTPase activity of transducin. One of these peptides, corresponding to PDE gamma residues 31-45, inhibited PDE activity and GTPase activity in a dose-dependent manner. The GTPase activity was inhibited by PDE gamma-3 non-competitively. This region of the PDE gamma subunit may be involved in the direct interaction of transd

Retinal degeneration in cats fed casein. IV. The early receptor potential.

Electroretinographic studies of casein-fed cats with retinal taurine deficiency revealed that the early receptor potential (ERP) was initially normal in amplitude at a time when the a-wave and b-wave of the electroretinogram were substantially reduced or even nondetectable. The preserved ERP's in these taurine-deficient cats could be correlated with the histologic finding that their outer segments were relatively intact over 90% of the retinal area subtended by the test flash. The sequence of electroretinographic changes in these taurine-deficient cats was also consistent with previous biochem

Physiological Postnatal Neurogenesis, Development, and Maturation in the Regenerable Retina.

The retina enlarges postnatally by ∼440% in axolotls and ∼150% in humans, predicting a reduction in neuron densities by 81.5% and 60%, respectively, if neurogenesis is absent. The concurrent retinal remodeling is poorly characterized. We studied retinas of 2-15 cm axolotls. Results showed that hatchling (∼2 cm) retinas, particularly the peripheral (Peri) had 1-2 more soma rows in the ganglion cell (GCL) layer and outer (ONL) and inner nuclear (INL) layers. Cell density reduced in GCL (GCs, -49%) in juveniles (4-12 cm) but increased in INL (74%) and ONL (55.5%) in young juveniles (4-6 cm), demo

Immunocytochemical demonstration of visual pigments in the degenerate retinal and pineal photoreceptors of the blind cave salamander (Proteus anguinus).

Visual pigments in the regressed eye and pineal of the depigmented neotenic urodele, the blind cave salamander (Proteus anguinus anguinus), were studied by immunocytochemistry with anti-opsin antibodies. The study included light- and electron-microscopic investigations of both the eye and the pineal organ. A comparison was made with the black pigmented subspecies Proteus anguinus parkelj (black proteus), which has a normal eye structure. In the retina of the black proteus, we found principal rods, red-sensitive cones and a third photoreceptor type, which might represent a blue- or UV-sensitive

The retinal phenotype of Usher syndrome: pathophysiological insights from animal models.

The Usher syndrome (USH) is the most prevalent cause of inherited deaf-blindness. Three clinical subtypes, USH1-3, have been defined, and ten USH genes identified. The hearing impairment due to USH gene defects has been shown to result from improper organisation of the hair bundle, the sound receptive structure of sensory hair cells. In contrast, the cellular basis of the visual defect is less well understood as this phenotype is absent in almost all the USH mouse models that faithfully mimic the human hearing impairment. Structural and molecular interspecies discrepancies regarding photorecep

Temporal Transformation of the Rod Single-Photon Response in the Retinal Circuitry.

PURPOSE: The temporal sensitivity of rod vision is higher than expected based on slow single-photon responses produced by phototransduction in rod outer segments. We sought to establish the retinal basis for the temporal speeding of rod responses and their dependence on phototransduction. METHODS: We made patch-clamp recordings of photocurrent and photovoltage from rods and rod bipolar cells (RBCs), and loose-patch and voltage-clamp recordings from ON α retinal ganglion cells, in three mouse strains: wild-type (WT), rhodopsin hemizygotes (Rh+/-), and GCAPs-/-. RESULTS: Single-photon response

Usher syndrome type 1-associated gene, pcdh15b, is required for photoreceptor structural integrity in zebrafish.

Blindness associated with Usher syndrome type 1 (USH1) is typically characterized as rod photoreceptor degeneration, followed by secondary loss of cones. The mechanisms leading to blindness are unknown because most genetic mouse models only recapitulate auditory defects. We generated zebrafish mutants for one of the USH1 genes, protocadherin-15b (pcdh15b), a putative cell adhesion molecule. Zebrafish Pcdh15 is expressed exclusively in photoreceptors within calyceal processes (CPs), at the base of the outer segment (OS) and within the synapse. In our mutants, rod and cone photoreceptor integrit

Accessory Inner Segment-Like Structures Develop in Human Retinal Organoids.

PURPOSE: A novel photoreceptor structure, called the accessory inner segment (aIS), was recently identified in human rod photoreceptors. It is described as a microtubule-based, mitochondria-rich extension of the conventional inner segment attached externally to the outer segment. Human retinal organoids recapitulate many developmental and mature cellular features of the human retina. Thus, we sought to determine if retinal organoids exhibit an aIS-like structure, to support future in vitro studies of its role in photoreceptor structure and disease. METHODS: Mature retinal organoids were deriv

PIP2 binding at allosteric site blocks activation in human rod CNG channels.

Phosphatidylinositol-4,5-bisphosphate (PIP2) is a signaling lipid that regulates multiple ion channels. In rod cyclic nucleotide-gated (CNG) channels, PIP2 was reported to inhibit activity, controlling light sensitivity and dynamic range. Reports of low PIP2 levels in rod outer segment (ROS) membranes questioned the physiological relevance of such inhibition, and the underlying mechanism remained unclear. Here we define the mechanism of PIP2 inhibition of human CNGA1, the principal rod CNG channel subunit. Flux assays and single-channel electrophysiology of purified, liposome-reconstituted CNG

Maintenance of opsin density in photoreceptor outer segments of retinoid-deprived rats.

Dietary deficiency in the retinoid precursors of the visual pigment chromophore 11-cis-retinal eventually results in selective degeneration of the photoreceptor cells of the vertebrate retina. Early effects of retinoid deficiency are depletion of rhodopsin from the retina and vesiculation of the photoreceptor outer segment disc membranes. Experiments were conducted to determine whether these early changes were accompanied by an alteration of the opsin content of the disc membranes. After being fed a retinoid-deficient diet containing retinoic acid for 26 weeks, the rhodopsin content of rat ret

Exogenous PDE5 Expression Rescues Photoreceptors in RD1 Mice.

BACKGROUND: Catalytic hydrolysis of cyclic guanosine monophosphate (cGMP) by phosphodiesterase 6 (PDE6) is critical in phototransduction signalling in photoreceptors. Mutations in the genes encoding any of the three PDE6 subunits are associated with retinitis pigmentosa, the most common form of inherited retinal diseases. The RD1 mouse carries a naturally occurring nonsense mutation in the Pde6b gene. The RD1 mouse retina rapidly degenerates and fails to form rod photoreceptor outer segments due to the elevated cGMP level and subsequent excessive Ca2+ influx. In this study, we aim to test whet
