Metagenomic characterization of gut microbiota in rheumatoid arthritis-associated interstitial lung disease: taxonomic shifts and clinical correlations.

BACKGROUND: Rheumatoid arthritis-associated interstitial lung disease (RA-ILD) is a severe extra-articular manifestation with limited diagnostic biomarkers. While gut microbiota dysbiosis contributes to rheumatoid arthritis (RA) pathogenesis, its specific role in RA-ILD remains poorly characterized. METHODS: We performed shotgun metagenomic sequencing on fecal samples from 41 participants: 10 RA-ILD patients, 20 RA patients without ILD (RA-non-ILD), and 11 healthy controls (HCs). We assessed alpha and beta diversity, differential abundance (Wilcoxon rank-sum tests with FDR correction), Spearm

Effects of α-amylase and coated α-amylase supplementation on growth performance, nutrient digestion, and rumen fermentation in Holstein bulls.

This study evaluated the impacts of α-amylase (AM) and coated α-amylase (CAM) on bull performance, nutrient digestibility, and ruminal fermentation. This study randomized 60 Holstein bulls of 365 ± 11.5 days of age and 457.5 ± 9.35 kg body weight into three groups: without AM addition, adding AM 0.6 g/kg dry matter (DM), and adding CAM 0.6 g AM/kg DM, separately. This whole experimental period was 80 days, including a 20-day adaptation period and a 60-day data and sample acquisition period. In comparison with the unsupplemented control, dry matter intake (DMI) was unaltered; however, average d

A Large Genetic Causal Analysis of the Gut Microbiota and Urological Cancers: A Bidirectional Mendelian Randomization Study.

BACKGROUND: Several observational studies and clinical trials have shown that the gut microbiota is associated with urological cancers. However, the causal relationship between gut microbiota and urological cancers remains to be elucidated due to many confounding factors. METHODS: In this study, we used two thresholds to identify gut microbiota GWAS from the MiBioGen consortium and obtained data for five urological cancers from the UK biobank and Finngen consortium, respectively. We then performed a two-sample Mendelian randomization (MR) analysis with Wald ratio or inverse variance weighted

Prevalence of vitamin D deficiency and rickets in children with cholestasis in Iran.

BACKGROUND: Ruminant gastrointestinal microbiota are spatially heterogeneous, yet whether the rumen microbiota contributes to bile acid-related gut-liver associations beyond its local fermentative role remains unclear. Here, paired rumen, ileum, and colon microbiota were integrated with liver transcriptomic data, blood biochemical and tail fat-related phenotypes to determine which gastrointestinal sector microbiota play prioritized host interactions in a sheep model. RESULTS: The rumen, ileum, and colon showed marked taxonomic and predicted functional divergence, indicating strong spatial res

Undernutrition disrupts jejunal and ileal microbiota and epithelial tissue homeostasis in a pregnant sheep model.

Nutrition consistently affects microbe-host interactions in the gastrointestinal tract. This study aimed to unravel how undernutrition reshapes the microbial composition and the homeostasis of epithelium in the jejunum and ileum. Sixteen late-gestation Hu-sheep were randomly assigned to the control group (n = 8, 100% ad libitum feeding levels) or the undernutrition group (n = 8, which received 30% ad libitum feeding levels). After 15-d treatment, all ewes were slaughtered, and jejunal and ileal digesta and epithelium samples were collected for 16S rRNA gene sequencing and transcriptome sequenc

Longitudinal body mass index trajectories at preschool age: children with rapid growth have differential composition of the gut microbiota in the first year of life.

BACKGROUND/OBJECTIVE: The steep rise in childhood obesity has emerged as a worldwide public health problem. The first 4 years of life are a critical window where long-term developmental patterns of body mass index (BMI) are established and a critical period for microbiota maturation. Understanding how the early-life microbiota relate to preschool growth may be useful for identifying preventive interventions for childhood obesity. We aim to investigate whether longitudinal shifts within the bacterial community between 3 months and 1 year of life are associated with preschool BMI z-score traject

Role of gut microbiota dysbiosis in the pathogenesis of non-alcoholic fatty liver disease among obese individuals.

Non-alcoholic fatty liver disease (NAFLD) is highly prevalent in obese individuals, and gut microbiota dysbiosis is increasingly recognized as a major contributor to its pathogenesis. In this cross-sectional study of 80 obese participants (40 NAFLD and 40 controls), 16S rRNA sequencing revealed significantly reduced microbial diversity in NAFLD patients (Shannon index 2.9 vs. 3.6; p=0.001), with increased Firmicutes, Enterobacteriaceae, and Ruminococcus, and decreased Bacteroidetes. These patients also showed elevated liver enzymes (ALT, AST) and inflammatory markers (TNF-α, IL-6). A strong co

The gut microbiome can be used to predict the gastrointestinal response and efficacy of lung cancer patients undergoing chemotherapy.

BACKGROUND: Lung cancer has the highest incidence and mortality rate of any cancer worldwide. Platinum-based combination chemotherapy is still the standard treatment for advanced lung cancer. However, the clinical efficacy of this treatment can be affected by its adverse reactions, especially gastrointestinal mucositis. The adverse reactions often lead to delayed and reduced medication. The role played by gut microbiome in the treatment of cancer is becoming clearer, and evidence suggests that regulation of the gut microbiome may affect the response to multiple types of cancer treatment.
