Functional oligomerization of the Saccharomyces cerevisiae isoprenylcysteine carboxyl methyltransferase, Ste14p.

The isoprenylcysteine carboxyl methyltransferase (Icmt) from Saccharomyces cerevisiae, also designated Ste14p, is a 26-kDa integral membrane protein that contains six transmembrane spanning segments. This protein is localized to the endoplasmic reticulum membrane where it performs the methylation step of the CAAX post-translational processing pathway. Sequence analysis reveals a putative GXXXG dimerization motif located in transmembrane 1 of Ste14p, but it is not known whether Ste14p forms or functions as a dimer or higher order oligomer. We determined that Ste14p predominantly formed a homodi

[Construction of high sulphite-producing industrial strain of Saccharomyces cerevisiae].

In the process of beer storage and transportation, off-flavor can be produced for oxidation of beer. Sulphite is important for stabilizing the beer flavor because of its antioxidant activity. However, the low level of sulphite synthesized by the brewing yeast is not enough to stabilize beer flavor. Three enzymes involve sulphite biosynthesis in yeast. One of them, APS kinase (encoded by MET14) plays important role in the process of sulphite formation. In order to construct high sulphite-producing brewing yeast strain for beer production, MET14 gene was cloned and overexpressed in industrial st

A cadmium-transporting P1B-type ATPase in yeast Saccharomyces cerevisiae.

Detoxification and homeostatic acquisition of metal ions are vital for all living organisms. We have identified PCA1 in yeast Saccharomyces cerevisiae as an overexpression suppressor of copper toxicity. PCA1 possesses signatures of a P1B-type heavy metal-transporting ATPase that is widely distributed from bacteria to humans. Copper resistance conferred by PCA1 is not dependent on catalytic activity, but it appears that a cysteine-rich region located in the N terminus sequesters copper. Unexpectedly, when compared with two independent natural isolates and an industrial S. cerevisiae strain, the

Anti-Saccharomyces cerevisiae antibodies in twins with inflammatory bowel disease.

BACKGROUND AND AIMS: An increased occurrence of anti-Saccharomyces cerevisiae antibodies (ASCA) is reported in unaffected members of families with Crohn's disease. Whether ASCA is a familial trait due to genetic factors or is caused by exposure to environmental factors is unknown. To assess the genetic influence of ASCA we studied its occurrence in a twin population. PATIENTS AND METHODS: ASCA were analysed in 98 twin pairs with inflammatory bowel disease and were related to clinical phenotype and CARD15/NOD2 genotype. RESULTS: ASCA were more common in Crohn's disease than in ulcerative coli

Protein glycation in Saccharomyces cerevisiae. Argpyrimidine formation and methylglyoxal catabolism.

Methylglyoxal is the most important intracellular glycation agent, formed nonenzymatically from triose phosphates during glycolysis in eukaryotic cells. Methylglyoxal-derived advanced glycation end-products are involved in neurodegenerative disorders (Alzheimer's, Parkinson's and familial amyloidotic polyneurophathy) and in the clinical complications of diabetes. Research models for investigating protein glycation and its relationship to methylglyoxal metabolism are required to understand this process, its implications in cell biochemistry and their role in human diseases. We investigated meth

Saccharomyces cerevisiae and Zygosaccharomyces mellis exhibit different hyperosmotic shock responses.

The effect of hyperosmotic shock on cell volume, vacuole volume, and intracellular pH (pH(i)) of individual cells of Saccharomyces cerevisiae and Zygosaccharomyces mellis was investigated. After transfer from a high water activity (a(w)) medium to low a(w) media, the growth latency periods of Z. mellis were shorter than those of S. cerevisiae. These results demonstrate that Z. mellis manages hyperosmotic shock better than S. cerevisiae. As a response to acute hyperosmotic shock, i.e. the first minute of perfusion with hypertonic buffers, the vacuoles shrank and the pH(i) decreased in both yeas

Las21 participates in extracellular/cell surface phenomena in Saccharomyces cerevisiae.

Las21 (Yj1062W) is a member of the major facilitator super family, possessing multimembrane spanning domains. The LAS21 gene was identified as a responsible gene for a Saccharomyces cerevisiae mutan which shows sensitivity to a local anestheticum, tetracaine. The null las21 mutant (las21 delta) is viable but shows temperature sensitive growth. We found, in addition to this phenotype, that the las21 delta strain shows a number of defects; mating deficiency, calcofluor resistance, and formation of Zymolyase sensitive spores. Temperature sensitive growth of the las21 delta mutant was found to be

Respiratory development in Saccharomyces cerevisiae grown at controlled oxygen tension.

Saccharomyces cerevisiae was grown in batch culture over a wide range of oxygen concentrations, varying from the anaerobic condition to a maximal dissolved oxygen concentration of 3.5 muM. The development of cells was assayed by measuring amounts of the aerobic cytochromes aa(3), b, c, and c(1), the cellular content of unsaturated fatty acids and ergosterol, and the activity of respiratory enzyme complexes. The half-maximal levels of membrane-bound cytochromes aa(3), b, and c(1), were reached in cells grown in O(2) concentrations around 0.1 muM; this was similar to the oxygen concentration req

Can Saccharomyces cerevisiae be used as a therapeutic agent for vulvovaginal candidiasis?

INTRODUCTION: Vulvovaginal candidiasis (VVC) is a highly prevalent fungal vaginal infection with a substantial impact on women's quality of life. Increasing evidence indicates that VVC pathogenesis is driven not only by high fungal burden but also by dysregulated host-immune responses, highlighting the need for therapeutic strategies that extend beyond conventional antifungal treatment. AREAS COVERED: In this review, recent advances elucidating the antifungal and immunomodulatory properties of Saccharomyces cerevisiae in the context of (recurrent) VVC are critically examined. Evidence from in

Encapsulation of Saccharomyces boulardii into sugar pellets coated with an enteric polymer.

Many current probiotic products do not optimally release enough viable cells at the site of action due to the loss of viability during manufacturing and passage through the stomach. This work aimed to formulate a modified release system that protects S. boulardii during the manufacturing process and under simulated acidic gastric conditions. S. boulardii was layered to sugar pellets, coated at 5% and 10% with the enteric polymer Eudragit® S12.5, and encapsulated in hard gelatin capsules. During manufacturing, the main determinant of viability was the incorporation of yeast onto the sugar pelle

Biocontrol potential of Saccharomyces as a sustainable approach targeting Spodoptera frugiperda.

Spodoptera frugiperda, known as the fall armyworm, is a highly destructive pest that greatly impacts worldwide agriculture, particularly maize and other key crops. Its rapid expansion and resistance to standard insecticides pose a significant threat to food security. As a result, effective and long-term pest management solutions are required. In this context, microbial biocontrol agents, such as Saccharomyces cerevisiae, offer an environmentally friendly alternative to chemical pesticides. This study investigates S. cerevisiae's efficiency as a biocontrol agent against S. frugiperda. The S. ce

Kinetic Evaluation of the Production of Mead from a Non-Saccharomyces Strain.

There is a growing market for craft beverages with unique flavors. This study aimed to obtain a palate-pleasing mead derived from Pichia kudriavzevii 4A as a monoculture. Different culture media were evaluated to compare the fermentation kinetics and final products. The crucial factors in the medium were ~200 mg L-1 of yeast assimilable nitrogen and a pH of 3.5-5.0. A panel of judges favored the mead derived from Pichia kudriavzevii 4A (fermented in a medium with honey initially at 23 °Bx) over a commercial sample produced from Saccharomyces cerevisiae, considering its appearance, fruity and f

Pib2 is a cysteine sensor involved in TORC1 activation in Saccharomyces cerevisiae.

Target of rapamycin complex 1 (TORC1) is a master regulator that monitors the availability of various amino acids to promote cell growth in Saccharomyces cerevisiae. It is activated via two distinct upstream pathways: the Gtr pathway, which corresponds to mammalian Rag, and the Pib2 pathway. This study shows that Ser3 was phosphorylated exclusively in a Pib2-dependent manner. Using Ser3 as an indicator of TORC1 activity, together with the established TORC1 substrate Sch9, we investigated which pathways were employed by individual amino acids. Different amino acids exhibited different dependenc

Fermentation of Persimmon Leaves Extract by Lactiplantibacillus plantarum and Saccharomyces cerevisiae.

Persimmon leaves usually as agricultural and forestry waste were fermented by Lactiplantibacillus plantarum and Saccharomyces cerevisiae. Growth and metabolic performances of L. plantarum and S. cerevisiae, as well as the effect of fermentation on the antioxidant abilities of the extract was investigated, including the content of flavonoids, 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical clearance rates. Growth of L. plantarum was limited, even though the acid production was sustainable, while S. cerevisiae was more suitable to inhabit in the persimmon leaves extract. A symbiotic re

Purification of Bioactive Peptide with Antimicrobial Properties Produced by Saccharomyces cerevisiae.

A variety of organisms produce bioactive peptides that express inhibition activity against other organisms. Saccharomyces cerevisiae is considered the best example of a unicellular organism that is useful for studying peptide production. In this study, an antibacterial peptide was produced and isolated from Saccharomyces cerevisiae (Baker's yeast) by an ultrafiltration process (two membranes with cut-offs of 2 and 10 kDa) and purified using the ÄKTA Pure 25 system. Antibacterial peptide activity was characterized and examined against four bacterial strains including Gram-positive and Gram-nega

Attenuated apoptotic BAX expression as a xenobiotic reporter in Saccharomyces cerevisiae.

Fungal infections are a major challenge to medicine and agriculture. Repeated and prophylactic use of antifungals can lead to pathogen cross-resistance to different classes of drugs. The early development of multidrug resistance in pathogenic fungi includes drug tolerance mediated by drug-dependent activation of drug efflux. In Saccharomyces cerevisiae and the fungal pathogen Candida glabrata, xenobiotic sensing motifs in transcription factors upregulate expression of several ATP-binding cassette (ABC) drug efflux pumps. We have therefore considered how drug candidates that trigger or prevent
