Quantitative Assessment of Full Size Microplastics in Bottled and Tap Water Samples in Hong Kong.

Human exposure to microplastics (MPs) through drinking water has drawn serious concern recently because of the potential adverse health effects. Although there are reports on the occurrence of MPs in bottled water, little is known about the abundance of a whole spectrum of MPs with sizes ranging from 1 µm to 5 mm due to the restrictions of conventional MPs detection methods. Some studies using micro-Raman spectroscopy can achieve MPs with a size of <10 µm, however, quantitation of all MPs was extremely time consuming and only a small portion (<10%) of MPs would be analyzed. The present study q

Novel analytical methods to interpret large sequencing data from small sample sizes.

BACKGROUND: Targeted therapies have greatly improved cancer patient prognosis. For instance, chronic myeloid leukemia is now well treated with imatinib, a tyrosine kinase inhibitor. Around 80% of the patients reach complete remission. However, despite its great efficiency, some patients are resistant to the drug. This heterogeneity in the response might be associated with pharmacokinetic parameters, varying between individuals because of genetic variants. To assess this issue, next-generation sequencing of large panels of genes can be performed from patient samples. However, the common problem

A general sample size framework for developing or updating a predictive algorithm: with application to clinical prediction models.

BACKGROUND: When developing or updating a predictive algorithm to make predictions (e.g., risk estimates) in individuals, the sample size of the development (training) dataset is an important consideration. Small datasets reduce a model's predictive performance, generalisability and fairness, and may lead to harm. To help identify (minimum) sample sizes required for model development or updating, various sample size calculations exist but their underlying theory is based on standard (unpenalised) regression and focus on minimising overfitting. To address this, we propose a more general approac

Evaluation of sample size for laboratory detection during HAdVs outbreaks in China: a systematic review and model research.

Human adenovirus infection is a common cause of upper and lower respiratory illness and easily breaks out in schools and the army. In an adenovirus epidemic, a large number of samples would be collected for laboratory diagnosis, and it is urgent to optimize the current sampling strategy. We researched the application of laboratory detection in the adenovirus epidemic and optimized the range of laboratory pathogen detection in the adenovirus epidemic by summarizing previous theoretical achievements, research reports, and experts' opinions and by using mathematical model tools. Under certain ass

Sample size considerations using mathematical models: an example with Chlamydia trachomatis infection and its sequelae pelvic inflammatory disease.

BACKGROUND: The success of an intervention to prevent the complications of an infection is influenced by the natural history of the infection. Assumptions about the temporal relationship between infection and the development of sequelae can affect the predicted effect size of an intervention and the sample size calculation. This study investigates how a mathematical model can be used to inform sample size calculations for a randomised controlled trial (RCT) using the example of Chlamydia trachomatis infection and pelvic inflammatory disease (PID). METHODS: We used a compartmental model to imi

Reduction of sample size requirements by bilateral versus unilateral research designs in animal models for cartilage tissue engineering.

Advanced tissue engineering approaches for articular cartilage repair in the knee joint rely on translational animal models. In these investigations, cartilage defects may be established either in one joint (unilateral design) or in both joints of the same animal (bilateral design). We hypothesized that a lower intraindividual variability following the bilateral strategy would reduce the number of required joints. Standardized osteochondral defects were created in the trochlear groove of 18 rabbits. In 12 animals, defects were produced unilaterally (unilateral design; n=12 defects), while defe

Experimental infectious respiratory disease in groups of calves: lobar distribution, variance, and sample-size requirements for vaccine evaluation.

The distribution and variance of respiratory disease produced with aerosols of bovine herpesvirus 1 (BHV-1) and Mannheimia haemolytica in control (183 calves in 44 experiments) and vaccinated calves were studied in experiments conducted at the Animal Diseases Research Institute, Lethbridge, Alberta, from 1975 to 1989. All calves had been born and raised at this institute and exposed similarly for 5 min by means of a face mask to viral and bacterial aerosols produced by a Collison atomizer (particles < 3 microm in diameter). We summarized the macroscopic pathological responses of pneumonia (mai

Cross-technique transfer learning to predict the dose distribution for radiotherapy planning based on a limited sample size.

BACKGROUND: Accurate dose prediction is challenged by the lack of available training samples and the rapid evolution of radiotherapy techniques. PURPOSE: A cross-technique transfer learning strategy was developed to predict the dose distribution for radiotherapy planning using limited training samples. METHODS: Data were collected from 154 patients with nasopharyngeal carcinoma: 60 treated with intensity-modulated radiotherapy (IMRT) and 94 treated with volumetric modulated arc therapy (VMAT). The Res-U Net was selected as the base deep learning network. Cross-technique models were pretraine

Sample Size Determination for Pilot Studies Based on Event Thresholds: A Framework for Evaluating Feasibility of Research Processes.

BACKGROUND: Pilot studies are essential for assessing feasibility and operational processes before large-scale research. This study presents a Poisson-based framework for determining the minimum pilot sample size based on discrete event thresholds. METHODS: The number of events represents occurrences such as errors, deviations, or missing data that indicate whether a process is functioning as intended. Setting a maximum tolerable threshold for these events provides a clear decision rule for proceeding or refining study procedures. A Poisson-based model was applied for sample size calculation.

A diffusion model for mosquito control trials with spillover: application to calculations of power, sample size, and bias.

BACKGROUND: Mosquito movement is ubiquitous and complicates the design of cluster randomized trials (CRTs) of vector control interventions because it can cause spillover between trial arms. There has been no analytical approach to quantify the impact of this spillover on trial power, required sample sizes, or bias. This precludes formal allowance for spillover in formulae for study power, and a precautionary principle is generally used in trial design, leading to CRTs with very large clusters and extensive buffer zones between arms. METHODS: The diffusion equation was solved to give a mathema

Impact of correlation structure on sample size requirements of statistical methods for multiple binary outcomes: A simulation study.

BackgroundIn randomized clinical trials, multiple-testing procedures, composite endpoints, and prioritized outcome approaches are increasingly used to analyze multiple binary outcomes. Previous studies have shown that correlations between outcomes influence their sample size requirements. Although sample size is an important factor affecting the choice of statistical methods, the power and required sample sizes of methods for analyzing multiple binary outcomes have yet to be compared under the influence of outcome correlations.MethodsWe conducted simulations to evaluate the power of co-primary

A systematic review of sample size estimation accuracy on power in malaria cluster randomised trials measuring epidemiological outcomes.

INTRODUCTION: Cluster randomised trials (CRTs) are the gold standard for measuring the community-wide impacts of malaria control tools. CRTs rely on well-defined sample size estimations to detect statistically significant effects of trialled interventions, however these are often predicted poorly by triallists. Here, we review the accuracy of predicted parameters used in sample size calculations for malaria CRTs with epidemiological outcomes. METHODS: We searched for published malaria CRTs using four online databases in March 2022. Eligible trials included those with malaria-specific epidemio

Effect of Sample Preprocessing and Size-Based Extraction Methods on the Physical and Molecular Profiles of Extracellular Vesicles.

Extracellular vesicles (EVs) are nanometric lipid vesicles that shuttle cargo between cells. Their analysis could shed light on health and disease conditions, but EVs must first be preserved, extracted, and often preconcentrated. Here we first compare plasma preservation agents, and second, using both plasma and cell supernatant, four EV extraction methods, including (i) ultracentrifugation (UC), (ii) size-exclusion chromatography (SEC), (iii) centrifugal filtration (LoDF), and (iv) accousto-sorting (AcS). We benchmarked them by characterizing the integrity, size distribution, concentration, p

The Causal Effects of Cholelithiasis on Acute Pancreatitis and Pancreatic Cancer: A Large Sample Size Mendelian Randomization Analysis.

BACKGROUND: The aim of two-sample Mendelian randomization (MR) with a large sample size was to explore the causal cholelithiasis impact on acute pancreatitis and pancreatic cancer. METHODS: We performed the two-sample MR analysis with two models. Publicly available summary- level information for cholelithiasis was acquired from the Genome-Wide Summary Association Studies (GWAS) of FinnGen Biobank. The inverse variance weighted (IVW) method was the main method to obtain the MR estimates. Other methods were also used as supplementary methods, including MR-Egger, maximum likelihood, MR-Robust Ad

Estimating the minimum sample size for interventional and observational studies using the lung clearance index as an endpoint✰.

BACKGROUND: With the increasing availability of highly effective modulators for people living with cystic fibrosis (CF), there is a need to re-design research studies to reflect the changing epidemiology of the CF population. The lung clearance index (LCI), a sensitive physiological measure of lung function, may be ideally suited as an endpoint in the era of CF modulator therapies. In this study we describe study design considerations for implementing LCI into interventional and observational research. METHODS: Simulations were used to estimate the required sample size to detect a range of tr
