Nuclear envelope - plasma membrane - compaction domain in malaria parasites.

A novel compaction domain between the nuclear envelope and the plasma membrane is observed in intraerythrocytic Plasmodium knowlesi and Plasmodium chabaudi. The plasma membrane and the two nuclear membranes stick very closely together thus forming a heptalamina domain. This has an outer diameter of 14.3 +/- 1.1 nm and can be as long as 2/um. Fracture faces in this region appear to be devoid of the typical membrane-intercalating-particles. This compaction domain occurs only in ring-stages and early to mid trophozoites, but never in schizonts.

Monoclonal IgM rheumatoid factor-like anti-globulins enhance the inhibitory effects of Plasmodium falciparum-specific monoclonal antibodies in vitro.

Monoclonal IgM rheumatoid factor-like anti-globulins were produced by in vitro stimulation of naive BALB/c spleen cells with lipopolysaccharide, and by hyperimmunization of mice with merozoites of Plasmodium falciparum, followed by fusion of the spleen cells to mouse myelomas. In vitro, these anti-globulins augmented the inhibitory effects of P. falciparum-specific polyclonal mouse sera and monoclonal IgG1 and IgG2b antibodies by binding to Fc fragments of IgG molecules attached to blood-stage parasites. In some instances, the presence of anti-globulins correlated with an increase in the numbe

Biosynthesis of a putative protective Plasmodium knowlesi merozoite antigen.

A putative protective Plasmodium knowlesi antigen, recognized by two monoclonal antibodies which prevent invasion of erythrocytes by merozoites in vitro, is synthesised only by schizonts with 7 or more nuclei, during the last 1.5-2 h of the 24 h erythrocytic cycle of parasite development. The 66 000 MW antigen, which constitutes a minor parasite protein, is processed at the time of schizont rupture and merozoite release, to give rise to two smaller molecules of 44 000 and 42 000 MW. The 44 000 and 42 000 MW antigens and traces of the 66 000 MW antigen are present on the surface of isolated mer

Antigenic variation and protective immunity in Plasmodium knowlesi malaria.

Rhesus monkeys were immunized with defined strains and variants of Plasmodium knowlesi and their immunity on challenge was correlated with serum levels of schizont agglutinins and specific inhibitory antibody assayed by in vitro parasite culture. The results indicate that the inhibitory antibody provides a consistent index of immune status and probably represents the protective antibody which initiates specific immune reactions in vivo. The relationship between inhibitory and schizont agglutinating antibodies is discussed. Inhibitory antibody is predominantly specific for those variants which

Global gene expression of human malaria parasite liver stages throughout intrahepatocytic development.

Plasmodium falciparum (Pf) is causing the greatest malaria burden, yet the liver stages (LS) of this most important parasite species have remained poorly studied. Here, we used a human liver-chimeric mouse model in combination with a novel fluorescent PfNF54 parasite line (PfNF54cspGFP) to isolate PfLS-infected hepatocytes and generate transcriptomes that cover the major LS developmental phases in human hepatocytes. RNA-seq analysis of early Pf LS trophozoites two days after infection, revealed a central role of translational regulation in the transformation of the extracellular invasive sporo

Clinical presentation and pathology of sarcocystosis in psittaciform birds: 11 cases.

Sarcocystosis in psittaciform birds occurs in several different presentations, making ante-mortem diagnosis challenging without specific laboratory tests. This study followed the course of 11 birds diagnosed with sarcocystosis by serologic analysis and/or post-mortem examinations during a 10-month period in 2006-07. The disease presented in three different clinical forms: an acute pulmonary disease (three birds), muscular disease (five), and neurological disease (three). Early diagnosis of sarcocystosis was possible through the combined used of plasma protein electrophoresis and indirect fluor

SICA-mediated cytoadhesion of Plasmodium knowlesi-infected red blood cells to human umbilical vein endothelial cells.

Zoonotic malaria due to Plasmodium knowlesi infection in Southeast Asia is sometimes life-threatening. Post-mortem examination of human knowlesi malaria cases showed sequestration of P. knowlesi-infected red blood cells (iRBCs) in blood vessels, which has been proposed to be linked to disease severity. This sequestration is likely mediated by the cytoadhesion of parasite-iRBCs to vascular endothelial cells; however, the responsible parasite ligands remain undetermined. This study selected P. knowlesi lines with increased iRBC cytoadhesion activity by repeated panning against human umbilical ve

The dynamin-related protein PfDyn2 is essential for both apicoplast and mitochondrial fission in Plasmodium falciparum.

Dynamins, or dynamin-related proteins (DRPs), are large mechano-sensitive GTPases that mediate membrane dynamics or organellar fission/fusion events. Plasmodium falciparum encodes three dynamin-like proteins whose functions are poorly understood. Here, we demonstrate that one of these dynamin-related proteins, PfDyn2, is required to divide both the apicoplast and the mitochondrion, a striking divergence from the biology of related parasites. Using super-resolution and ultrastructure expansion microscopy (U-ExM), we show that PfDyn2 is expressed in dividing schizonts, and that it localizes to b

In vitro and in vivo anti-piroplasm activity of PBT2 against Theileria annulata and Babesia microti.

Theileriosis and babesiosis remain major constraints to livestock production, and increasing drug resistance necessitates alternative therapeutic strategies. Here, we evaluated the metal ionophore PBT2 for anti-piroplasm activity. PBT2 inhibited proliferation of two Theileria annulata schizont infected cell lines with IC50 values of 450.9 nM and 407.8 nM and significantly reduced parasite burden. Colony formation was suppressed in both cell lines, consistent with parasite-associated growth inhibition. Cytotoxicity in BoMAC cells was limited, yielding a selectivity index exceeding 60-fold. Dynamics of detergent-resistant membranes of Plasmodium falciparum during blood stage development.

Membrane microdomains, also known as detergent-resistant membranes (DRMs), are tightly packed assemblies enriched in cholesterol and sphingolipids, where various membrane-associated processes occur. Many intracellular pathogens, including Plasmodium, exploit these raft-like structures to recognize and invade host cells. In particular, Plasmodium depends on host cholesterol to build its own membranes, as it is unable to perform sterol synthesis. Using quantitative proteomics, lipid analysis, and bioinformatics, this study defines the composition and dynamics of Plasmodium falciparum DRMs from k

In Vitro Evaluation of Two Novel Antimalarial Derivatives of SKM13: SKM13-MeO and SKM13-F.

Antimalarial drugs play an important role in the control and treatment of malaria, a deadly disease caused by the protozoan parasite Plasmodium spp. The development of novel antimalarial agents effective against drug-resistant malarial parasites is urgently needed. The novel derivatives, SKM13-MeO and SKM13-F, were designed based on an SKM13 template by replacing the phenyl group with electron-donating (-OMe) or electron-withdrawing groups (-F), respectively, to reverse the electron density. A colorimetric assay was used to quantify cytotoxicity, and in vitro inhibition assays were performed o

Relationship of serum complement levels to events of the malarial paroxysm.

Malarial paroxysms due to Plasmodium vivax were studied for alterations in whole serum complement (C') and certain C' components. The objective was to relate C' values with events of the parasite cycle during schizogony and with the febrile pattern. Substantial decreases in C' were found in 9 of 18 paroxysms studied during relapse. In contrast, only one of 22 paroxysms occuring during the primary attack was associated with a striking depression in C', and this case exhibited certain characteristics of a relapse paroxysm. The mean change in C' levels during paroxysms in relapse (-23%) was signi

Mapping PP1c and Its Inhibitor 2 Interactomes Reveals Conserved and Specific Networks in Asexual and Sexual Stages of Plasmodium.

Malaria parasites require multiple phosphorylation and dephosphorylation steps to drive signaling pathways for proper differentiation and transformation. Several protein phosphatases, including protein phosphatase 1 (PP1), one of the main dephosphorylation enzymes, have been shown to be indispensable for the Plasmodium life cycle. The catalytic subunit of PP1 (PP1c) participates in cellular processes via dynamic interactions with a vast number of binding partners that contribute to its diversity of action. In this study, we used Plasmodium berghei transgenic parasite strains stably expressing

Detection and stage classification of Plasmodium falciparum from images of Giemsa stained thin blood films using random forest classifiers.

BACKGROUND: The conventional method for the diagnosis of malaria parasites is the microscopic examination of stained blood films, which is time consuming and requires expertise. We introduce computer-based image segmentation and life stage classification with a random forest classifier. Segmentation and stage classification are performed on a large dataset of malaria parasites with ground truth labels provided by experts. METHODS: We made use of Giemsa stained images obtained from the blood of 16 patients infected with Plasmodium falciparum. Experts labeled the parasite types from each of the

Cytauxzoonosis in domestic cats: report of two cases in Oklahoma, with a review and discussion of the disease.

In 2 cases of cytauxzoonosis in domestic cats in Oklahoma, the clinical features, gross lesions, and microscopic findings were of similar nature and degree to those previously reported by others, both in the naturally occurring and experimentally induced disease. Intraerythrocytic forms of Cytauxzoon felis organisms were demonstrated in stained blood smears from one cat. Blood from the other cat was not examined. The diagnosis was established by the finding of schizonts in endothelial-associated macrophages within vascular channels in tissues from both cats.

Respiratory cryptosporidiosis in a peacock chick.

Respiratory cryptosporidiosis was diagnosed in a 2-week-old peacock chick, whose symptoms were a gurgling respiration, coughing and sneezing, and a serous oculonasal discharge. Light microscopy detected cryptosporidial-like organisms attached to the surface of conjunctival, nasal-sinus, and tracheal-epithelial cells, and electron microscopy demonstrated stages typical of cryptosporidial trophozoites, schizonts, and oocysts. About 30 other in-contact peacock chicks also developed the same clinical symptoms, and all died despite treatment with oxytetracycline HCl at 200 ppm in the drinking water

In vitro maturation of the exoerythrocytic stage of Plasmodium knowlesi observed under phase contrast microscopy.

Exoerythrocytic stages of Plasmodium knowlesi were obtained in primary culture of rhesus monkey hepatocytes. The development of a single parasite was followed with phase contrast microscopy until release of merozoites in slightly less than 5 days. This direct observation may offer opportunities to determine visually factors that may influence specific steps of schizont development.

Time course of in vitro maturation of intra-erythrocytic malaria parasite: a comparison between Plasmodium falciparum and Plasmodium knowlesi.

The schizont maturation assay for in vitro drug sensitivity tests has been a standard method employed in the global baseline assessment and monitoring of drug response in Plasmodium falciparum. This test is limited in its application to synchronous plasmodial infections because it evaluates the effect of drug on the maturation of parasite especially from ring to schizont stage and therefore synchronized P. falciparum cultures are required. On the other hand, P. knowlesi, a simian malaria parasite has a unique 24-h periodicity and maintains high natural synchronicity in monkeys. The present rep

Single-cell RNA sequencing reveals a signature of sexual commitment in malaria parasites.

Pathogens have to balance transmission with persistence. For Plasmodium falciparum, the most widespread and virulent malaria parasite, persistence within its human host requires continuous asexual replication within red blood cells, while its mosquito-borne transmission depends on intra-erythrocytic differentiation into non-replicating sexual stages called gametocytes. Commitment to either fate is determined during the preceding cell cycle that begins with invasion by a single, asexually committed merozoite and ends, 48 hours later, with a schizont releasing newly formed merozoites, all commit
