A blastoporal organizer in a ctenophore.

In an iconic experiment in 1924, Hilde Mangold and Hans Spemann established that the dorsal blastopore lip of amphibian embryos functions as an organizer and induces a secondary body axis when transplanted into a host embryo1. This discovery demonstrated that specific embryonic regions can regulate embryonic patterning and lead to the establishment of an entire body axis. Subsequent studies have revealed that cnidarians, the sister group to Bilateria, also possess a blastoporal embryonic organizer2,3. However, the evolutionary origin of the organizer remains unclear. Here we report that the bl

A rational nomenclature for naming peptide toxins from spiders and other venomous animals.

Molecular toxinology research was initially driven by an interest in the small subset of animal toxins that are lethal to humans. However, the realization that many venomous creatures possess a complex repertoire of bioactive peptide toxins with potential pharmaceutical and agrochemical applications has led to an explosion in the number of new peptide toxins being discovered and characterized. Unfortunately, this increased awareness of peptide-toxin diversity has not been matched by the development of a generic nomenclature that enables these toxins to be rationally classified, catalogued, and

Some clinical and epidemiological problems of venom poisoning today.

Poisonings caused by bites and stings of venomous animals have not lost their importance from most ancient times until today. Rather, one could say that they in some respects have become even more important due to some moments as: the interesting results of biochemical, pharmacological and immunological researches of their venoms, so as possibilities of their use in treatment and in preparation of drugs; the importation of venomous animals in loads of goods from countries with warm climate into countries where they normally would not be found; the increasing number of fanciers who keep in thei

The toxicogenomic multiverse: convergent recruitment of proteins into animal venoms.

Throughout evolution, numerous proteins have been convergently recruited into the venoms of various animals, including centipedes, cephalopods, cone snails, fish, insects (several independent venom systems), platypus, scorpions, shrews, spiders, toxicoferan reptiles (lizards and snakes), and sea anemones. The protein scaffolds utilized convergently have included AVIT/colipase/prokineticin, CAP, chitinase, cystatin, defensins, hyaluronidase, Kunitz, lectin, lipocalin, natriuretic peptide, peptidase S1, phospholipase A(2), sphingomyelinase D, and SPRY. Many of these same venom protein types have

The pharmacology of voltage-gated sodium channel activators.

Toxins and venom components that target voltage-gated sodium (NaV) channels have evolved numerous times due to the importance of this class of ion channels in the normal physiological function of peripheral and central neurons as well as cardiac and skeletal muscle. NaV channel activators in particular have been isolated from the venom of spiders, wasps, snakes, scorpions, cone snails and sea anemone and are also produced by plants, bacteria and algae. These compounds have provided key insight into the molecular structure, function and pathophysiological roles of NaV channels and are important

Venom toxins in the exploration of molecular, physiological and pathophysiological functions of acid-sensing ion channels.

Acid-sensing ion channels (ASICs) are voltage-independent proton-gated cation channels that are largely expressed in the nervous system as well as in some non-neuronal tissues. In rodents, six different isoforms (ASIC1a, 1b, 2a, 2b, 3 and 4) can associate into homo- or hetero-trimers to form a functional channel. Specific polypeptide toxins targeting ASIC channels have been isolated from the venoms of spider (PcTx1), sea anemone (APETx2) and snakes (MitTx and mambalgins). They exhibit different and sometimes partially overlapping pharmacological profiles and are usually blockers of ASIC channe

Metabolism of Oxybenzone and Dioxybenzone to Phototoxins in Human Skin Cells.

Recent research indicated that exposure of sea anemones in full-spectrum sunlight to the UV filter, oxybenzone, induced increased mortality attributable to the production of phototoxic glucoside metabolites. This study evaluated whether exposure of oxybenzone and dioxybenzone to human skin cell models formed analogous glucuronide conjugates and induced a reduction in cell viability under full-spectrum sunlight. In dose-response evaluations with two human skin cell models, increasing doses of oxybenzone or dioxybenzone (up to 0.1%) or increasing exposure times (up to 24 h) were associated with

Three-dimensional structure of kappa-conotoxin PVIIA, a novel potassium channel-blocking toxin from cone snails.

kappa-Conotoxin PVIIA from the venom of Conus purpurascens is the first cone snail toxin that was described to block potassium channels. We synthesized chemically this toxin and showed that its disulfide bridge pattern is similar to those of omega- and delta-conotoxins. kappa-conotoxin competes with radioactive alpha-dendrotoxin for binding to rat brain synaptosomes, confirming its capacity to bind to potassium channels; however, it behaves as a weak competitor. The three-dimensional structure of kappa-conotoxin PVIIA, as elucidated by NMR spectroscopy and molecular modeling, comprises two lar

The regulatory role of GABAA receptor in Actinia equina nervous system and the possible effect of global ocean acidification.

Global warming and connected acidification of the world ocean attract a substantial amount of research efforts, in particular in a context of their impact on behaviour and metabolism of marine organisms, such as Cnidaria. Nevertheless, mechanisms underlying Cnidarians' neural signalling and behaviour and their (possible) alterations due to the world ocean acidification remain poorly understood. Here we researched for the first time modulation of GABAA receptors (GABAARs) in Actinia equina (Cnidaria: Anthozoa) by pH fluctuations within a range predicted by the world ocean acidification scenario

Treating autoimmune disorders with venom-derived peptides.

The effective treatment of autoimmune diseases remains a challenge. Voltage-gated potassium Kv1.3 channels, which are expressed in lymphocytes, are a new therapeutic target for treating autoimmune disease. Consequently, Kv1.3 channel-inhibiting venom-derived peptides are a prospective resource for new drug discovery and clinical application. Area covered: Preclinical and clinical studies have produced a wealth of information on Kv1.3 channel-inhibiting venom-derived peptides, especially from venomous scorpions and sea anemones. This review highlights the advances in screening and design of the

Enhancing the therapeutic potential of peptide toxins.

Peptide toxins are potent and often exquisitely selective probes of the structure and function of ion channels and receptors, and as such are of significant interest to the pharmaceutical and biotech industries as both therapeutic leads and pharmacological tools. Their progression as clinical candidates, however, faces many of the challenges that are common to peptide drugs generally. Areas covered: The attributes of peptide toxins as therapeutic leads are outlined, as well as some of the limiting factors that have hampered the clinical development of many promising candidates. Strategies to o

Diversification of neurotoxins by C-tail 'wiggling': a scorpion recipe for survival.

The structure of bioactive surfaces of proteins is a subject of intensive research, yet the mechanisms by which such surfaces have evolved are largely unknown. Polypeptide toxins produced by venomous animals such as sea anemones, cone snails, scorpions, and snakes show multiple routes for active site diversification, each maintaining a typical conserved scaffold. Comparative analysis of an array of genetically related scorpion polypeptide toxins that modulate sodium channels in neuronal membranes suggests a unique route of toxic site diversification. This premise is based on recent identificat

[Effective Anticoagulation Therapy Prior to Surgical Excision of an Aortic Valve Papillary Fibroelastoma Diagnosed after a Transient Cerebral Ischemic Attack;Report of a Case].

Cardiac papillary fibroelastoma is reported to be the 2nd most common cardiac tumor following myxoma. Owing to the risk of embolism, early surgical excision is the treatment of choice. We report a case of effective anticoagulation therapy prior to surgical excision of an aortic valve papillary fibroelastoma. A 78-year-old man was admitted to our hospital because of transient cerebral ischemic attack. The symptom was relieved in a short period. Echocardiography revealed a tumor at the aortic valve. Cardiac computed tomography revealed a sea-anemone-like appearance of the tumor. Cardiac papillar

Biology of FGFRL1, the fifth fibroblast growth factor receptor.

FGFRL1 (fibroblast growth factor receptor like 1) is the most recently discovered member of the FGFR family. It contains three extracellular Ig-like domains similar to the classical FGFRs, but it lacks the protein tyrosine kinase domain and instead contains a short intracellular tail with a peculiar histidine-rich motif. The gene for FGFRL1 is found in all metazoans from sea anemone to mammals. FGFRL1 binds to FGF ligands and heparin with high affinity. It exerts a negative effect on cell proliferation, but a positive effect on cell differentiation. Mice with a targeted deletion of the Fgfrl1

Rapid development of a papillary fibroelastoma in the aortic valve: report of a case.

We report a case of a fast-growing cardiac papillary fibroelastoma (CPF) arising in the aortic valve. A 71-year-old woman was referred to a cardiologist for investigation of palpitations. Cardiac catheterization, coronary angiography, and echocardiography showed no stenosis of the coronary arteries, nor any other cardiovascular abnormalities. However, 12 months later she was referred to our hospital for further investigations when she began experiencing chest discomfort on exertion. This time, echocardiography showed an abnormal mass arising from the aortic valve, floating between the left sin

Toxins interacting with ether-à-go-go-related gene voltage-dependent potassium channels.

The critical role that ether-à-go-go-related gene (erg) K(+) channels play in mating in Caenorhabditis elegans, neuronal seizures in Drosophila and cardiac action potential repolarization in humans has been well documented. Three erg genes (erg1, erg2 and erg3) have been identified and characterized. A structurally diverse number of compounds block these channels, but do not display specificity among the different channel isoforms. In this review we describe the blocking properties of several peptides, purified from scorpion, sea anemone and spider venoms, which are selective for certain membe

Molecular determinants of two neurotoxins that regulate sodium current inactivation in rat hippocampal neurons.

We studied functional and structural differences between the two neurotoxins, wasp toxin pompilidotoxin (PMTX) and sea anemone toxin (ATXII). Although PMTX and ATXII inhibited inactivation of sodium currents both toxins had distinct actions on the lobster axon and on the rat hippocampal cells. To determine structural basis of the difference we compared arrangement of polar and non-polar amino acids of the two toxins and found that similar sequence of PMTX exist in a discrete position of three-dimensional structure of ATXII. The sequence may be responsible for the binding site in the neuronal N

Binding of an alpha scorpion toxin to insect sodium channels is not dependent on membrane potential.

The insect-specific Lqh alpha IT toxin resembles alpha scorpion toxins affecting mammals by its amino acid sequence and effects on sodium conductance. The present study reveals that Lqh alpha IT does not bind to rat brain membranes and possesses in locust neuronal membranes a single class of high affinity (Kd = 1.06 +/- 0.15 nM) and low capacity (Bmax = 0.7 +/- 0.19 pmol/mg protein) binding sites. The latter are: (1) distinct from binding sites of other sodium channel neurotoxins; (2) inhibited by sea anemone toxin II; (3) cooperatively interacting with veratridine; (4) not dependent on membra

RAG1 core and V(D)J recombination signal sequences were derived from Transib transposons.

The V(D)J recombination reaction in jawed vertebrates is catalyzed by the RAG1 and RAG2 proteins, which are believed to have emerged approximately 500 million years ago from transposon-encoded proteins. Yet no transposase sequence similar to RAG1 or RAG2 has been found. Here we show that the approximately 600-amino acid "core" region of RAG1 required for its catalytic activity is significantly similar to the transposase encoded by DNA transposons that belong to the Transib superfamily. This superfamily was discovered recently based on computational analysis of the fruit fly and African malaria
