Proneural gene self-stimulation in neural precursors: an essential mechanism for sense organ development that is regulated by Notch signaling.

To learn about the acquisition of neural fate by ectodermal cells, we have analyzed a very early sign of neural commitment in Drosophila, namely the specific accumulation of achaete-scute complex (AS-C) proneural proteins in the cell that becomes a sensory organ mother cell (SMC). We have characterized an AS-C enhancer that directs expression specifically in SMCs. This enhancer promotes Scute protein accumulation in these cells, an event essential for sensory organ development in the absence of other AS-C genes. Interspecific sequence comparisons and site-directed mutagenesis show the presence

Self antigens expressed by solid tumors Do not efficiently stimulate naive or activated T cells: implications for immunotherapy.

Induction and maintenance of cytotoxic T lymphocyte (CTL) activity specific for a primary endogenous tumor was investigated in vivo. The simian virus 40 T antigen (Tag) expressed under the control of the rat insulin promoter (RIP) induced pancreatic beta-cell tumors producing insulin, causing progressive hypoglycemia. As an endogenous tumor antigen, the lymphocytic choriomeningitis virus (LCMV) glycoprotein (GP) was introduced also under the control of the RIP. No significant spontaneous CTL activation against GP was observed. However, LCMV infection induced an antitumor CTL response which eff

Learnt tolerance to sedative effects of chlordiazepoxide on self-stimulation performance, but no tolerance to facilitatory effects after 80 days.

Sedative and facilitatory effects on variable-interval hypothalamic self-stimulation were monitored during chronic treatment with chlordiazepoxide (CDP; 7.5 mg/kg IP), given at 48-h intervals in two groups of rats. Group 1 was injected immediately before each of 40 1-h self-stimulation sessions ("drugged responding"); Group 2 was injected after self-stimulation for the first 20 sessions ("undrugged responding"), and before self-stimulation for a further 20 sessions ("drugged responding"). Significant sedation occurred in both groups in initial sessions of drugged responding, even though Group

Comparison of deficits in electrical self-stimulation after ibotenic acid lesion of the lateral hypothalamus and the medial prefrontal cortex.

The aim of the present study was to compare the self-stimulation deficit produced by a unilateral injection of the neurotoxin, ibotenic acid, in the lateral hypothalamus (LH) to the deficit produced by the same unilateral injection in the medial prefrontal cortex (MPC). Four groups of adult male Sprague-Dawley rats were used: in two control groups, electrodes were bilaterally implanted in the LH (5 rats) or in the MPC (6 rats) and self-stimulation (ICSS) was obtained separately with the right and left electrodes. In the two experimental groups the intrinsic neurons of the LH (8 rats) or of the

Noninvasive tFUS stimulation of the parabrachial nucleus suppresses lever pressing during extinction of cocaine self-administration through aversive circuit engagement.

INTRODUCTION: Drug addiction is a significant public health concern, and persistent drug-seeking behavior remains difficult to control in severe cases. While invasive interventions have been explored to suppress these behaviors, their use is limited by procedural complexity, highlighting the need for effective noninvasive neuromodulatory approaches. Transcranial focused ultrasound (tFUS) provides a noninvasive approach for deep-brain neuromodulation, but its ability to produce persistent behavioral effects by targeting the extralateral parabrachial nucleus (elPBN) remains unknown. METHODS: Mi

The effect of repetitive transcranial magnetic stimulation on food choice-related self-control in patients with severe, enduring anorexia nervosa.

OBJECTIVE: Individuals with anorexia nervosa (AN) pursue low-fat, low-calorie diets even when in a state of emaciation. These maladaptive food choices may involve fronto-limbic circuitry associated with cognitive control, habit, and reward. We assessed whether high-frequency repetitive transcranial magnetic stimulation (rTMS) to the left dorsolateral prefrontal cortex (DLPFC) influenced food-related choice behavior in patients with severe, enduring (SE)-AN. METHOD: Thirty-four females with SE-AN completed a Food Choice Task before and after 20 sessions of real or sham rTMS treatment and at a

Effectiveness of Stimulant Treatment in Preventing Fractures and Head Injuries in Patients With Narcolepsy: A Self-Controlled Case Series Study.

BACKGROUND: Previous cohort studies have investigated whether stimulants reduce injury risk in narcolepsy, but they faced potential biases such as confounding by indication and immortal time bias. We conducted a self-controlled case series study to address these biases and assess the effect of stimulant use on the risk of fracture and head injury in patients with narcolepsy. METHODS: We used a Japanese health insurance claims database to identify patients with narcolepsy who were newly exposed to stimulants and who experienced fracture or head injury. We then applied multivariable conditional

Orally Ingested Self-Powered Stimulators for Targeted Gut-Brain Axis Electrostimulation to Treat Obesity and Metabolic Disorders.

Obesity is a significant health concern that often leads to metabolic dysfunction and chronic diseases. This study introduces a novel approach to combat obesity using orally ingested self-powered electrostimulators. These electrostimulators consist of piezoelectric BaTiO3 (BTO) particles conjugated with capsaicin (Cap) and aim to activate the vagus nerve. Upon ingestion by diet-induced obese (DIO) mice, the BTO@Cap particles specifically target and bind to Cap-sensitive sensory nerve endings in the gastric mucosa. In response to stomach peristalsis, these particles generate electrical signals.

The effect of transcranial direct current stimulation of the prefrontal cortex on implicit self-esteem is mediated by rumination after criticism.

It has been proposed that a crucial link between cognitive (i.e., self-schemas) and biological vulnerability is prefrontal control. This is because decreased control leads to impaired ability to inhibit ruminative thinking after the activation of negative self-schemas. However, current evidence is mainly correlational. In the current experimental study we tested whether the effect of neurostimulation of the dorsolateral prefrontal cortex (DLPFC) on self-esteem is mediated by momentary ruminative self-referential thinking (MRST) after the induction of negative self-schemas by criticism. We used

Soliton breathing induced by stimulated Raman scattering and self-steepening in octave-spanning Kerr frequency comb generation.

We investigate the impact of stimulated Raman scattering (SRS) and self-steepening (SS) on breather soliton dynamics in octave-spanning Kerr frequency comb generation. SRS and SS can transform chaotic fluctuations in cavity solitons into periodic breathing. Furthermore, with SRS and SS considered, bandwidth of the soliton breathes more than two times stronger. The simultaneous presence of SRS and SS also make the soliton breathe slower and degrades the coherence of the soliton.

[Significance of CRF and dopamine receptors in amygdala for reinforcing effects of opiates and opioids on self-stimulation of lateral hypothalamus in rats].

Bipolar electrodes were implanted in the lateral hypothalamus in a group of 44 Wistar male rats in order to study self-stimulation reaction in the Skinner box. Simultaneously, microcanules were implanted into the central nucleus of the amygdala to inject the drugs (1 microl per injection). The blockade of corticoliberin (CRF) receptors (astressin, 1 microg) or Na+influx currents (xycaine or lidocain 1 microg) by the intrastructural administration of drugs into the amygdala decreased self-stimulation reaction of the lateral hypothalamus in rats by 29-55%. The inhibition of D1 and D2 dopamine re

Electrical self-stimulation in the medial and lateral septum as compared to the lateral hypothalamus: differential intervention of reward and learning processes?

The characteristics of the electrical self-stimulation behavior elicited from the lateral hypothalamus (LH) and from both medial (MS) and lateral (LS) parts of the septal nucleus have been compared in male mice of the BALB/c strain. Using two different experimental situations (the lever-press box and a spatial discrimination test in a Y-maze) the self-stimulation rate-current intensity relations and the performances during both acquisition and reversal of a spatial discrimination were tested successively. In the lever-press box, it was observed that highest self-stimulation rates were obtained

Home-based self-administered transcranial direct current stimulation for women affected by primary dysmenorrhoea in Northeastern Brazil: a protocol study.

BACKGROUND: The prevalence of women with primary dysmenorrhoea is high and negatively impacts physical and mental health. The intense cyclic episodes of pain generate central nervous system dysfunctional processing. In this sense, strategies focused on the central nervous system are important to re-establish normal functioning. Home-based self-administered transcranial direct current stimulation (tDCS) emerges as a strategy to modulate dysfunctional brain areas and improve the symptoms. This protocol aims to evaluate the effects of home-based self-administered tDCS for pain, premenstrual sympt

A self-powered casein hydrogel E-dressing with synergistic photothermal therapy, electrical stimulation, and antibacterial effects for chronic wound management.

Triboelectric nanogenerators (TENGs) have recently demonstrated great application potential for accelerating wound healing in the field of medical research due to their unique electrical stimulation effect. Among the various types of TENGs, solid-liquid TENGs have attracted much attention due to their significant advantages, such as high contact-separation efficiency and a wide range of liquid motion. Therefore, this study innovatively proposed a solid-liquid biphasic TENG electronic dressing constructed from a casein hydrogel enhanced by the metal-organic framework Zeolitic Imidazolate Framew
