Morphological and functional diversity of spatially resolved vestibular ganglion neuron cell types.

Vestibular ganglion neurons (VGNs) are the primary sensory neurons of the vestibular system, a relatively understudied sensory modality that is essential for maintaining visual stability and postural balance. Compared with other sensory systems, our understanding of the molecular heterogeneity of VGNs and its contribution to the diverse functions of the vestibular system remains limited. Using single-cell and spatial transcriptomics, we defined five transcriptionally distinct VGN cell types with discrete spatial distributions across the ganglion. These VGN cell types demonstrate organ-specific

Capsaicin-sensitive intestinal mucosal afferent mechanism and body fat distribution.

This report summarizes clinical and experimental data in support of the hypothesis that capsaicin-sensitive intestinal mucosal afferent mechanism plays a role in regulating body fat distribution. Epidemiological data have revealed that the consumption of foods containing capsaicin is associated with a lower prevalence of obesity. Rural Thai people consume diets containing 0.014% capsaicin. Rodents fed a diet containing 0.014% capsaicin showed no change in caloric intake but a significant 24% and 29% reduction in the visceral (peri-renal) fat weight. Increase in intestinal blood flow facilitate

How to differentiate induced pluripotent stem cells into sensory neurons for disease modelling: a functional assessment.

BACKGROUND: Human induced pluripotent stem cell (iPSC)-derived peripheral sensory neurons present a valuable tool to model human diseases and are a source for applications in drug discovery and regenerative medicine. Clinically, peripheral sensory neuropathies can result in maladies ranging from a complete loss of pain to severe painful neuropathic disorders. Sensory neurons are located in the dorsal root ganglion and are comprised of functionally diverse neuronal types. Low efficiency, reproducibility concerns, variations arising due to genetic factors and time needed to generate functionally

In Vitro Visualization of Cell-to-Cell Interactions Between Natural Killer Cells and Sensory Neurons.

Cell-to-cell interactions between the immune and nervous systems are increasingly recognized for their importance in health and disease. Assessment of cellular neuro-immune interactions can be aided by co-culture of two (or more) cells in an in vitro model system that preserves the morphology of neuronal cells. Here we describe methods to investigate the cytotoxic effector functions of natural killer cells on sensory neurons isolated from syngeneic embryonic and adult mice. We present methods for the morphological analysis of axon fragmentation (pruning) and dynamic cell function via live conf

IPSC-Derived Sensory Neurons Directing Fate Commitment of Human BMSC-Derived Schwann Cells: Applications in Traumatic Neural Injuries.

The in vitro derivation of Schwann cells from human bone marrow stromal cells (hBMSCs) opens avenues for autologous transplantation to achieve remyelination therapy for post-traumatic neural regeneration. Towards this end, we exploited human induced pluripotent stem-cell-derived sensory neurons to direct Schwann-cell-like cells derived from among the hBMSC-neurosphere cells into lineage-committed Schwann cells (hBMSC-dSCs). These cells were seeded into synthetic conduits for bridging critical gaps in a rat model of sciatic nerve injury. With improvement in gait by 12-week post-bridging, evoked

Conserved molecular signatures of hygrosensory neurons in two dipteran species.

Small poikilothermic animals like insects rely on environmental sensing for survival. The ability to detect humidity through specialized sensory neurons is particularly critical, allowing them to maintain water balance across diverse environments. While recent studies have identified key receptors associated with humidity sensing, our understanding of the underlying molecular architecture of these sensory systems remains incomplete. Here, we conducted a comparative analysis of single-nucleus transcriptomes of humidity receptor neurons (HRNs) between the vinegar fly Drosophila melanogaster and

Targeting TRP channels for chronic cough: from bench to bedside.

Cough is currently the most common reason for patients to visit a primary care physician in the UK, yet it remains an unmet medical need. Current therapies have limited efficacy or have potentially dangerous side effects. Under normal circumstances, cough is a protective reflex to clear the lungs of harmful particles; however, in disease, cough can become excessive, dramatically impacting patients' lives. In many cases, this condition is linked to inflammatory diseases such as asthma and chronic obstructive pulmonary disease (COPD), but can also be refractory to treatment and idiopathic in nat

Effect of the resection of the sciatic nerve on the Th1/Th2 balance in the synovia of the ankle joint of adjuvant arthritic rats.

Inflamed synovia of the ankle joint after 2-4 weeks of adjuvant injection receives dense sensory innervation. To study the role of sensory nerves on the local inflammation, the relative expression of T helper 1 and 2 lymphocyte (Th1 and Th2) markers was investigated on both axotomized adjuvant arthritic (AA) rats, whose sciatic nerves were resected before adjuvant injection, and on sham-operated ones. Immunohistochemical expressions of CXC chemokine receptor 3 (CXCR3) and CC chemokine receptor 4 (CCR4) were examined and compared with those of Th1 cytokine (interferon-gamma, IFN-gamma), Th2 cyt

Interleukin-10 signaling in somatosensory neurons controls CCL2 release and inflammatory response.

Appropriate regulation of the inflammatory response is essential for survival. Interleukin-10 (IL-10), a well-known anti-inflammatory cytokine, plays a major role in controlling inflammation. In addition to immune cells, we previously demonstrated that the IL-10 receptor (IL-10R1) is expressed in dorsal root ganglion sensory neurons. There is emerging evidence that these sensory neurons contribute to immunoregulation, and we hypothesized that IL-10 signaling in dorsal root ganglion (DRG) neurons facilitates the regulation of the inflammatory response. We showed that mice that lack IL-10R1 spec

Entangled cellular and molecular relationships at the sensory neuron-cancer interface.

Peripheral sensory neurons, once regarded merely as a passive route for nociceptive signals, are now acknowledged as active participants in solid tumor progression. This review explores how sensory neurons influence and are influenced by the tumor microenvironment (TME) through both chemical and electrical signaling, underscoring their pivotal role in the emerging field of cancer neuroscience. We summarize recent findings indicating that cancer-neuron interactions vary among different organs and experimental models, highlighting the ways in which various tumors recruit and reprogram sensory ne

In-Vivo Calcium Imaging of Sensory Neurons in the Rat Trigeminal Ganglion.

Genetically encoded calcium indicators (GECIs) enable imaging techniques to monitor changes in intracellular calcium in targeted cell populations. Their large signal-to-noise ratio makes GECIs a powerful tool for detecting stimulus-evoked activity in sensory neurons. GECIs facilitate population-level analysis of stimulus encoding with the number of neurons that can be studied simultaneously. This population encoding is most appropriately done in vivo. Dorsal root ganglia (DRG), which house the soma of sensory neurons innervating somatic and visceral structures below the neck, are used most ext

Unique nasal septal island in dromedary camels may play a role in pain perception: microscopic studies.

The septal organs are islands or patches of sensory epithelium, located in the ventral parts of the nasal septum and innervated by the olfactory nerve. The septal island in dromedaries (Camelus dromedarius) was unusually located in the rostro-dorsal part of the nasal septum, where the ethmoidal branch of the trigeminal nerve provides innervation to the island mucosa. Therefore, the objectives of this study were to reveal the microscopic and ultrastructure of this island and to explain the probable functions. Twelve septal islands from 12 healthy male camels were used. Unlike the olfactory epit

A quantitative analysis of changes in the olfactory epithelium following bulbectomy in hamster.

Continuous replacement of sensory neurons is a normal process in the olfactory system of adult vertebrates. The capacity for replacement following experimentally induced degeneration of sensory neurons makes this system ideal for the study of the dynamics of neural populations. A quantitative analysis was made to determine the time course and degree of cell replacement in the olfactory epithelium following unilateral bulbectomy in adult hamsters. Histological measurements of number of cells and epithelial thickness were made for up to 194 days postoperatively. Results for each experimental ani

Upside-Down Preference in the Forskolin-Induced In Vitro Differentiation of 50B11 Sensory Neurons: A Morphological Investigation by Label-Free Non-Linear Microscopy.

In this study, we revealed a peculiar morphological feature of 50B11 nociceptive sensory neurons in in vitro culture related to the forskolin-induced differentiation of these cells growing upside-down on cover glass supports. Multi-photon non-linear microscopy was applied to monitor increased neurite arborization and elongation. Under live and unstained conditions, second harmonic generation (SHG) microscopy could monitor microtubule organization inside the cells while also correlating with the detection of cellular multi-photon autofluorescence, probably derived from mitochondria metabolites.

Peripheral sensory neurons promote angiogenesis in neurovascular models derived from hESCs.

In the adult tissues, blood vessels traverse the body with neurons side by side; and share common signaling molecules. Developmental studies on animal models have shown that peripheral sensory neurons (PSNs) secrete angiogenic factors and endothelial cells (ECs) secrete neurotrophic factors which contribute to their coexistence, thereby forming the peripheral neurovascular (PNV) unit. Despite the large number of studies showing that innervation and vascularization complement each other, the interaction between human PSNs and ECs is still largely unknown. To study this interaction and to evalua

An analysis of Na+ currents in rat olfactory receptor neurons.

Na+ currents were observed in acutely-dissociated adult rat olfactory receptor neurons using the whole-cell recording techniques. The threshold for current activation was near -70 mV and currents were fully activated by -10 mV (midpoint: -45 mV). Steady-state inactivation was complete at potentials more positive than -70 mV and half complete at -110 mV (+/- less than 1, n = 8). Complete recovery from inactivation required one second at -100 mV (n = 7). The addition of 10 microM tetrodotoxin or 1 mM Zn2+ to the external solution was required to completely block the current. The current differs

Spared but Not Silent: Ion Channel Plasticity in Uninjured Sensory Neurons in Neuropathic Pain.

Chronic pain is maintained by persistent, maladaptive molecular processes, many of which converge on ion channels. In sensory neurons, these processes include transcriptional reprogramming, altered trafficking, and remodeling of the channel complexes that set membrane excitability. Accumulating evidence indicates that the major ion channel changes driving chronification of neuropathic pain are not restricted to injured sensory neurons but also arise within the adjacent population of spared neurons. Here we review the ion channel alterations that sustain chronic pain states, the mechanisms by w

DiI Dye-Filling as a Simple and Inexpensive Tool to Visualize Ciliated Sensory Neurons in C. elegans.

C. elegans have long been used as a simple and accessible model to study neuronal structure and the many functions of the nervous system. Of the 302 neurons within the adult hermaphrodite nervous system, 60 are classified as ciliated sensory neurons. These neurons are central to a number of C. elegans behaviors, including but not limited to chemo-, mechano-, and osmosensing, male mating, and dauer formation. For several decades now, members of the C. elegans community have used the red fluorescent lipophilic dye DiI to visualize a subset of the ciliated sensory neurons that are directly expose
