Topical application of silk fibroin-based hydrogel in preventing hypertrophic scars.

Current medications for the treatment of hypertrophic scars suffer from bottlenecks of limited therapeutic efficacy and a slow recovery rate. Silk fibroin (SF) has gained attention for its ability to promote wound healing in burns and cutaneous wounds, but its therapeutic effects against hypertrophic scar have not been thoroughly investigated. We prepared SF-based hydrogels (SFHs) with various SF concentrations (1.5 %, 3 %, and 6 %) and characterized their physicochemical properties. Cell experiments showed that these SFHs had favorable biocompatibility in vitro. Further animal experiments in

Enzymatically Cross-Linked Silk Fibroin-Based Hierarchical Scaffolds for Osteochondral Regeneration.

Osteochondral (OC) regeneration faces several limitations in orthopedic surgery, owing to the complexity of the OC tissue that simultaneously entails the restoration of articular cartilage and subchondral bone diseases. In this study, novel biofunctional hierarchical scaffolds composed of a horseradish peroxidase (HRP)-cross-linked silk fibroin (SF) cartilage-like layer (HRP-SF layer) fully integrated into a HRP-SF/ZnSr-doped β-tricalcium phosphate (β-TCP) subchondral bone-like layer (HRP-SF/dTCP layer) were proposed as a promising strategy for OC tissue regeneration. For comparative purposes,

Carbonic anhydrase generates a pH gradient in Bombyx mori silk glands.

Silk is a protein of interest to both biological and industrial sciences. The silkworm, Bombyx mori, forms this protein into strong threads starting from soluble silk proteins using a number of biochemical and physical cues to allow the transition from liquid to fibrous silk. A pH gradient has been measured along the gland, but the methodology employed was not able to precisely determine the pH at specific regions of interest in the silk gland. Furthermore, the physiological mechanisms responsible for the generation of this pH gradient are unknown. In this study, concentric ion selective micro

Highly flexible and lightweight organic solar cells on biocompatible silk fibroin.

Organic electronics have gained widespread attention due to their flexibility, lightness, and low-cost potential. It is attractive due to the possibility of large-scale roll-to-roll processing. However, organic electronics require additional development before they can be made commercially available and fully integrated into everyday life. To achieve feasibility for commercial use, these devices must be biocompatible and flexible while maintaining high performance. In this study, biocompatible silk fibroin (SF) was integrated with a mesh of silver nanowires (AgNWs) to build up flexible organic

Fabrication and intermolecular interactions of silk fibroin/hydroxybutyl chitosan blended nanofibers.

The native extracellular matrix (ECM) is composed of a cross-linked porous network of multifibril collagens and glycosaminoglycans. Nanofibrous scaffolds of silk fibroin (SF) and hydroxybutyl chitosan (HBC) blends were fabricated using 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and trifluoroacetic acid (TFA) as solvents to biomimic the native ECM via electrospinning. Scanning electronic microscope (SEM) showed that relatively uniform nanofibers could be obtained when 12% SF was blended with 6% HBC at the weight ratio of 50:50. Meanwhile, the average nanofibrous diameter increased when the conten

Targeted nanotherapeutic strategy for melanoma using silk-fibroin based artemisinin nanoparticles.

Skin cancer is one of the most common cancers in the world, and malignant melanoma is the most aggressive and life-threatening form of skin cancer. It often quickly spreads to other parts of the body and may not always respond well to standard treatments. This calls for the urgent development of more effective and targeted treatment strategies. Protein-based nanoparticles have gained considerable attention in oncology owing to their improved bioavailability, tumour-targeted delivery potential, and reduced systemic toxicity. Silk fibroin, a versatile natural biopolymer, has emerged as a promisi

Antigen-Containing Silk Nanoparticles: A New Potential as Oral Vaccine Carriers.

PURPOSE: Oral vaccines are safer, easier to administer, and more cost-effective than injectable vaccines are. They induce systemic and mucosal immunity, thereby providing broad protection. However, developing an orally administered vaccine that can traverse the stomach and safely reach the intestinal tract to induce antigen-specific immune responses is challenging. Recently, many effective nanoparticle (NP) drug delivery systems have been developed. Various polymeric materials, including synthetic biodegradable and natural polymers, have been used as drug delivery matrices. Silk fibroin, a nat

Bioengineering toughened, stretchable bacterial cellulose-based films inspired by spider silk.

Bacterial cellulose (BC) has emerged as a promising sustainable biomaterial with potential across multiple fields. However, its intrinsic stiffness and limited toughness restrict its use in applications such as textiles or wearable devices. Spider silk is one of nature's toughest materials, combining high strength and extensibility. The recombinant mini-spidroin (A3I)3-A14, a minimized synthetic variant of a spider silk protein, can be spun under mild conditions into ductile and tough fibers. Here, we present a bioinspired strategy to toughen BC with (A3I)3-A14 by tuning the incorporation rout

Silk Fibroin Gels Fabricated from Ajisawa Reagent with Mechanochromic Sensing Capability.

Silk fibroin (SF) derived from Bombyx mori cocoons was employed to fabricate gels using Ajisawa reagent (AJ) as the extraction medium. The resulting regenerated SF solution was subsequently directly dialyzed against an ethanol-water mixture, inducing a gradual and controllable solution-gel transition. Obtained SF gels exhibit a pronounced increase in compressive strength aligned with rising initial fibroin concentration, along with excellent water stability. FESEM, FTIR, and synchrotron X-ray diffraction analyses reveal a homogeneous network structure, likely governed by uniformly distributed

Isolation, characterization, antimicrobial and cytotoxic properties of Cydalima perspectalis silk proteins.

Natural silk produced by silkworms and some arthropod spiders stands out as a traditional protein polymer. Fibroin and sericin, which are silk proteins obtained from cocoons, have been used in the medical field for centuries. This study is the first to evaluate the physicochemical properties and biological activities of these proteins, which were isolated from the boxwood moth (Cydalima perspectalis), as a biotechnological material. This followed this separation of the fibroin and sericin proteins obtained from the cocoons using a high-temperature and high-pressure method. The morphological, t

Radiopaque Silk Sericin Nanoparticles for Computed Tomography Imaging of Solid Tumors.

Computed tomography (CT) is a non-invasive technology which is commonly used for cancer diagnosis owing to its high efficiency and widespread availability. But the inability to differentiate soft tissues with similar densities is the major limitation of CT. In the present study, silk sericin nanoparticles (SNP) prepared using the ethanol desolvation method were functionalized using diatrizoic acid (DTA). The prepared DTA-functionalized SNP (D-SNP) were spherical in shape with a mean hydrodynamic diameter of 52.6 nm. D-SNP were non-hemolytic and non-cytotoxic when treated with human erythrocyte

Novel Silk Fibroin Based Bilayer Scaffolds for Bioabsorbable Internal Biliary Stenting.

Biliary duct reconstruction is one of the most challenging parts of liver transplantation and accounts for 40%-60% of complications. While current stent-based devices on the market show promising results in reducing complications, they are manufactured from permanent synthetic materials and require a second reintervention for their removal. This exposes the patients to other potential complications and increases healthcare costs. This study develops a fabrication technique to produce a bioabsorbable biliary stent based on silk fibroin. The process used a dip-coating procedure for silk fibroin

Streamlining Skin Regeneration: A Ready-To-Use Silk Bilayer Wound Dressing.

Silk proteins have been highlighted in the past decade for tissue engineering (TE) and skin regeneration due to their biocompatibility, biodegradability, and exceptional mechanical properties. While silk fibroin (SF) has high structural and mechanical stability with high potential as an external protective layer, traditionally discarded sericin (SS) has shown great potential as a natural-based hydrogel, promoting cell-cell interactions, making it an ideal material for direct wound contact. In this context, the present study proposes a new wound dressing approach by developing an SS/SF bilayer

High-performance peptide biosensor based on unified structure of lotus silk.

The sensitive materials of current gas sensors are fabricated on planar substrates, significantly limiting the quantity of sensitive material available on the sensor and the complete exposure of the sensitive material to the target gas. In this work, we harnessed the finest, resilient, naturally degradable, and low-cost lotus silk derived from plant fibers, to fabricate a high-performance bio-sensor for toxic and harmful gas detection, employing peptides with full surface connectivity. The proposed approach to fabricate gas sensors eliminated the need for substrates and electrodes. To ascertai

Silk Fibroin Formed Bioadhesive Ophthalmic Gel for Dry Eye Syndrome Treatment.

PURPOSE: Dry eye syndrome (DES), arising from various etiologic factors, leads to tear film instability and ocular surface damage. Given its anti-inflammatory effects, cyclosporine A (CsA) has been widely used as a short-term treatment option for DES. However, poor bioavailability and solubility of CsA in aqueous phase make the development of a cyclosporine A-based eye drop for ocular topical application a huge challenge. METHODS: In this study, a novel strategy for preparing cyclosporine A-loaded silk fibroin nanoemulsion gel (CsA NBGs) was proposed to address these barriers. Additionally, t
