Silibinin, or silybin, is a polyphenolic flavonoid and the main active component of silymarin, isolated from the seeds of the milk thistle plant (Silybum marianum). It has been shown to have antioxidant, antineoplastic, hepatoprotective, neuroprotective, anti-inflammatory, antimicrobial, and antidiabetic effects. In this systematic review, a literature search was conducted from inception until January 2024 on major electronic databases (PubMed, Scopus, Web of Science, and Google Scholar) to identify studies assessing the effects of silibinin on diabetes and its associated complications in diff

BACKGROUND AND AIMS: Normal cells become tumorigenic owing to mutations in oncogenes and tumor suppressor genes modulating cell division. Cancer cells break down extracellular matrix to metastasize other tissues. Therefore, the development of natural and synthetic substances that suppress metastatic enzymes such as matrix metalloproteinase (MMP)-2 and MMP-9 is useful to inhibit metastasis. Silibinin is the main ingredient of silymarin extracted from the seeds of milk thistle plants having lung cancer-suppressing effects and liver protection. The purpose of this study was to investigate the inh

Osteoporosis-related bone defects are a major public health concern. Considering poor effects of a singular pharmacological treatment, many have sought combination therapies, including local treatment combined with systemic intervention. Based on recent evidence that selenium and silibinin increase bone formation and bone mineral density, it is hypothesized that systemic administration with silibinin plus local treatment with selenium may have an additive effect on bone regeneration in an OVX rat model with bone defects. To verify this hypothesis, 3-month-old ovariectomized Sprague- Dawley rat

Silibinin (Sil) is used as hepatoprotective drug and is approved for therapeutic use in amanitin poisoning. In our study we compared Sil-bis-succinate (SilBS), a water-soluble drug approved for i.v.-administration, with Sil solved in ethanol (SilEtOH), which is normally used in research. We challenged monocultures or 3D-microtissues consisting of HepG2 cells or primary hepatocytes with α-amanitin and treated with SILBS, SILEtOH, penicillin and combinations thereof. Cell viability and the integrity of the microtissues was monitored. Finally, the expression of the transporters OATP1B1 and B3 was

Diabetic wounds are serious and challenging complications of diabetes mellitus (DM) and are characterized by impaired healing. Its pathogenesis is complex and involves a variety of physiological and pathological changes, including vascular dysfunction, neuropathy, impaired neuropeptide signaling, hyperglycemia, persistent infections, inflammation, oxidative stress, and an impaired immune response. Despite significant advances in the understanding of the pathogenesis of diabetic wounds, current treatment options remain limited and often yield unsatisfactory results. Thus, there is an urgent nee

The escalating threat of antibiotic resistance, particularly from Staphylococcus aureus (S. aureus), has become a critical challenge in both public health and animal husbandry. The extensive use of conventional antibiotics in livestock production accelerates the emergence of resistant strains, heightening risks to food safety and human health. Although plant-derived bioactive compounds are increasingly recognized as promising alternatives to synthetic antimicrobials, the mechanisms underlying their efficacy-and the potential for synergistic action among different plant parts-remain poorly unde

While nickel is required in trace amounts for human health, excessive exposure can be harmful. It can cause significant damage to the liver, kidneys, and other organs, depending on the route of exposure. The aim of this study is to investigate the protective effect of the Silybum marianum L. seeds and leaves on the liver and kidneys as well as blood parameters. The purpose is to examine whether a one-month dietary supplementation with Silybum marianum L. seeds and leaves could prevent and prepare the body to resist nickel-induced damage. Seed and leaf extracts were administered orally to albin

Swertiamarin, a predominant iridoid glycoside from hepatoprotective Swertia herbs, is biotransformed in vivo into nitrogen-containing metabolites (R)-gentiandiol and (S)-gentiandiol. These metabolites may be the real active hepatoprotective agents. A high-fat diet-fed rat model was treated for 12 weeks with swertiamarin, (R)-gentiandiol, (S)-gentiandiol and silybin, and the therapeutic effects on non-alcoholic fatty liver disease (NAFLD) were systematically evaluated through biochemical indices and histopathological observations. Swertiamarin and (R)-gentiandiol reversed high-fat diet-induced

Hydrogen polysulfides (H2Sn), a key reactive sulfur species, play pivotal roles in cellular signaling, antioxidative stress, and cell death regulation. Revealing the dynamic changes of H2Sn levels in vivo is crucial for elucidating its physiological functions. However, real-time detection of H2Sn in vivo faces significant challenges due to its inherent instability. In this study, we screened a near-infrared fluorogenic (NIRF) probe DCICl-H2Sn for the detection of H2Sn in cells and mice models. This probe employed a dicyanoisophorone scaffold functionalized with ortho-NO2 substituted phenyl sul

Xiaochaihutang (XCHT) is a decoction of seven botanical extracts used for liver diseases traditionally in East Asia. However, few studies have investigated its anti-hepatic fibrosis effects and the mechanisms of action. Oxidative stress is one of the key factors responsible for occurrence and development of hepatic fibrosis and nuclear factor erythroid 2-related factor 2 (Nrf2) serves as a major regulator of the cellular defense system against oxidative stress. The aim of the present study was to investigate the effect of XCHT on liver fibrosis and focuse on the Nrf2 pathway in the protection

Silymarin (SM), an extract from the Silybum marianum (milk thistle) plant containing various flavonolignans (with silybin being the major one), has received a tremendous amount of attention over the last decade as a herbal remedy for liver treatment. In many cases, the antioxidant properties of SM are considered to be responsible for its protective actions. Possible antioxidant mechanisms of SM are evaluated in this review. (1) Direct scavenging free radicals and chelating free Fe and Cu are mainly effective in the gut. (2) Preventing free radical formation by inhibiting specific ROS-producing

Anastatica hierochuntica (Brassicaceae) is a desert plant used globally to treat various diseases. To advance research on A. hierochuntica, we conducted studies on its morphology, anatomy, physicochemical parameters, and antioxidant properties. Morphological analysis revealed that the whole plant was light yellow with an irregular umbrella shape; the stem, roots, leaves, and fruits each have distinct structural features. Light microscopy and scanning electron microscope (SEM) identified numerous vessels, including reticulate and spiral vessels, along with non-glandular hairs, phloem fibers, ep

Abrin is a toxin from the seeds of Abrus precatorius. Abrin is considerably more toxic than ricin and a potent bio-warfare agent. The mechanism of abrin induced hepatotoxicity remains unclear. Silibinin has antioxidant, anti-inflammatory and hepatoprotective activities. But, its therapeutic potential in abrin toxicity is unknown. In view of these facts, the purpose of this study was to delineate the mechanisms and ameliorative role of silibinin against abrin induced hepatotoxicity. Parameters related to liver functions, oxidative stress, inflammation, Fas pathway and histopathology were evalua

The molecular mechanisms by which different proteins assemble into highly ordered fibrillar deposits and cause disease remain topics of debate. Human amylin (also known as islet amyloid polypeptide/hIAPP) is found in vivo as amyloid deposits in the pancreatic islets of sufferers of type II diabetes mellitus, and its self-aggregation is thought to be a pathogenic factor in disease and to contribute to the failure of islet transplants. Here, electrospray ionization-ion mobility spectrometry-mass spectrometry (ESI-IMS-MS) has been used to monitor oligomer formation from IAPP. The detection, ident

Sleep deprivation (SD) impairs cognitive function through oxidative stress, neuroinflammation, and neuroendocrine dysregulation, whereas recovery sleep may not fully reverse these alterations. Silibinin possesses antioxidant, anti-inflammatory, and neuroprotective properties; however, its potential to facilitate recovery following prolonged SD remains unclear. This study investigated the effects of silibinin on cognitive dysfunction and hippocampal alterations induced by 5 days of SD, with or without a subsequent 24-h recovery period, in rats. Forty-eight adult male rats were allocated to cont

Alcohol liver disease (ALD) is a chronic liver disorder resulting from long-term heavy alcohol consumption. The pathogenesis of ALD is multifactorial, and existing therapeutic agents primarily target specific aspects of the disease while presenting significant side effects, including drug-induced liver injury and hepatobiliary disease. Silibinin (SLB) has attracted widespread attention for its hepatoprotective effects and favorable safety profile. However, inherent limitations associated with SLB, such as poor solubility and bioavailability, have significantly limited its clinical application.
