Temporo-spatial correlations between scalp and centromedian thalamic EEG activities of stage II slow wave sleep in patients with generalized seizures of the cryptogenic Lennox-Gastaut syndrome.

OBJECTIVES: Temporo-spatial correlations between scalp and centromedian thalamic (CM) normal and abnormal electroencephalographic (EEG) activities of stage II slow wave sleep (SWS II) were investigated in 5 patients with cryptogenic Lennox-Gastaut syndrome (CLGS). METHODS: In each patient, 8h/all-night sleep studies were performed with routine methods; and a total of 1233 normal and 206 abnormal individual activities, spontaneously occurring during 200 epochs of early and late SWS II, were analyzed. Normal activities included scalp-CM K-complexes (KC-CMKC), vertex waves (VW-CMVW), and sleep s

Rationale, design, and methods of electroencephalography-based investigation of the effects of oral desmopressin on improving slow-wave sleep time in nocturnal polyuria patients (the DISTINCT study): protocol for a single-arm, open-label, single-assignment trial.

BACKGROUND: Nocturia is one of the most bothersome lower urinary tract symptoms and often impairs sleep quality in the elderly. Although previous studies on nocturia have indicated that the successful treatment of nocturia improves sleep quality, most used questionnaires and activity devices to analyze sleep/wake patterns. Therefore, there is little information about the treatment effects of desmopressin on objective sleep quality. The aim of the DISTINCT study is to investigate the change in subjective and objective sleep quality using electroencephalography (EEG) and the Pittsburgh Sleep Qua

Does the adenosine deaminase (ADA) gene confer risk of sleepwalking?

Sleepwalking is a common non-rapid eye movement (NREM) parasomnia and a significant cause of sleep-related injuries. While evidence suggest that the occurrence of this condition is partly determined by genetic factors, its pattern of inheritance remains unclear, and few molecular studies have been conducted. One promising candidate is the adenosine deaminase (ADA) gene. Adenosine and the ADA enzyme play an important role in the homeostatic regulation of NREM sleep. In a single sleepwalking family, genome-wide analysis identified a locus on chromosome 20, where ADA lies. In this study, we exami

What single-unit recording studies tell us about the basic mechanisms of sleep and wakefulness.

This paper reviews the presence, localization and characteristics of state-specific neurons in the mouse forebrain, midbrain and hindbrain that are involved in the control of ultradian sleep-wake cycles and shows that all these regions contain basic neural elements capable of generating the sleep-wake cycle. The chronic single-unit recording method in unanaesthetized animals is useful for unravelling the dynamics of sleep-wake switching, in particular because it can analyse events at the level of single neurons, thereby decoding information used by the brain in determining its functional state

Alpha-delta sleep pattern in an acute functional neurological patient with no perception of sleep.

UNLABELLED: Functional neurological disorders are characterized by neurologic symptoms not consistent with a primary neurologic pathology. Although neurological disorders are commonly associated with poor sleep, alpha intrusion of slow-wave sleep is not described in cases of functional neurological disorder. We describe a case demonstrating an alpha-delta sleep pattern in a patient presenting with a functional neurological disorder and no perception of sleep. Although alpha-delta sleep is more commonly associated with fibromyalgia, this pattern may be a potential biomarker for the physiology o

Stimulating the sleeping brain: Current approaches to modulating memory-related sleep physiology.

BACKGROUND: One of the most audacious proposals throughout the history of psychology was the potential ability to learn while we sleep. The idea penetrated culture via sci-fi movies and inspired the invention of devices that claimed to teach foreign languages, facts, and even quit smoking by simply listening to audiocassettes or other devices during sleep. However, the promises from this endeavor didn't stand up to experimental scrutiny, and the dream was shunned from the scientific community. Despite the historic evidence that the sleeping brain cannot learn new complex information (i.e., wor

Nocturnal polysomnographic correlates of daytime sleepiness.

The relations of nighttime polysomnographic sleep variables with daytime sleepiness scores on the Epworth Sleepiness Scale were assessed in a retrospective chart review of 38 patients referred to a sleep clinic. Of the variables assessed, only slow wave sleep was statistically significantly correlated with daytime sleepiness, regardless of whether the analysis was based on absolute minutes of slow wave sleep or percentage of total sleep time spent in slow wave sleep. Stepwise linear regression suggested that other polysomnographic variables did not provide additional predictive value beyond th

Sleep, gender, and depression: an analysis of gender effects on the electroencephalographic sleep of 302 depressed outpatients.

Gender-related differences in electroencephalographic (EEG) sleep were examined in 151 pairs of men and women with major depression, all outpatients, matched for age and severity of depression. Across five decades (age 21-69), depressed men had less slow-wave sleep than did depressed women. Gender differences were small with respect to visually scored measures of slow-wave sleep time and percent, but moderate for gender differences in automated measures of slow-wave density. The time constant of the polygraph preamplifier significantly affected both visually scored and automatically scored slo

Transcranial Electrical Stimulation targeting limbic cortex increases the duration of human deep sleep.

BACKGROUND: Researchers have proposed that impaired sleep may be a causal link in the progression from Mild Cognitive Impairment (MCI) to Alzheimer's Disease (AD). Several recent findings suggest that enhancing deep sleep (N3) may improve neurological health in persons with MCI, and buffer the risk for AD. Specifically, Transcranial Electrical Stimulation (TES) of frontal brain areas, the inferred source of the Slow Oscillations (SOs) of N3 sleep, can extend N3 sleep duration and improve declarative memory for recently learned information. Recent work in our laboratory using dense array Electr

Why do we sleepwalk? A noradrenergic hypothesis of NREM sleep parasomnias.

Disorders of arousal (DOA), including sleepwalking, sleep terrors, and confusional arousals, are characterized by complex behaviors emerging from non-rapid eye movement (NREM) sleep. Although these conditions are defined by abnormal arousal, their neurobiological basis remains poorly understood, and the contribution of neuromodulatory systems, particularly the locus coeruleus-noradrenaline (LC-NA) system, has received limited attention. Here, we present a hypothesis-driven conceptual framework integrating existing evidence from neurophysiology, neuroimaging, autonomic regulation, and genetics.

Worse sleep architecture but not self-reported insomnia and sleepiness is associated with higher cortisol levels in menopausal women.

OBJECTIVE: Worsening of sleep quality during menopause is well recognized. However, the underlying hormonal regulation is insufficiently described. In this study, we evaluated associations between sleep and cortisol levels. STUDY DESIGN: Seventeen perimenopausal and 18 postmenopausal women were enrolled in a three-night sleep study. Diurnal blood sampling was performed during the third night and the following day. MAIN OUTCOME MEASURES: Self-reported insomnia and sleepiness were evaluated with the Basic Nordic Sleep Questionnaire and sleep architecture with all-night polysomnography. Diurnal

Temporal distribution of sleep onset REM periods and N3 sleep in the MSLT and night polysomnogram of narcolepsy type 1 and other hypersomnias.

INTRODUCTION: The presence of ≥2 sleep onset REM periods (SOREMP) in the Multiple Sleep Latency Test (MSLT) and the previous night polysomnogram (PSG) is part of the diagnostic criteria of narcolepsy, with every SOREMP having the same diagnostic value, despite evidence suggesting that time of SOREMP appearance and their preceding sleep stage might be relevant. We studied the temporal distribution of SOREMPs and associated sleep stages in the MSLT of patients with narcolepsy type 1 (NT1) and other hypersomnias (OH). METHODS: We reviewed consecutive five-nap MSLTs and their preceding PSG from 8

Evidence for histaminergic arousal mechanisms in the hypothalamus of cat.

Polygraphic 23-hr recordings were carried out in 25 adult cats in order to examine the effects of both systemic and local injections of various histaminergic and antihistaminergic drugs on sleep-waking cycles. alpha-Fluoromethylhistidine (alpha-FMH), a specific inhibitor of histidine decarboxylase, when injected intraperitoneally at a dose of 20 mg/kg, induced a significant increase in deep slow wave sleep (S2) and a decrease in wakefulness (W), without modifying light slow wave sleep (S1) and paradoxical sleep (PS). Recent Evidence of Non-Rapid Eye Movement Sleep Oscillation Abnormalities in Psychiatric Disorders.

PURPOSE OF REVIEW: We review recent studies published from 2019 to 2024 examining slow waves and sleep spindles abnormalities across neurodevelopmental, mood, trauma-related, and psychotic disorders using polysomnography and Electroencephalogram (EEG). RECENT FINDINGS: Individuals with attention-deficit/hyperactivity disorder (ADHD) showed higher slow-spindle activity, while findings on slow-wave activity were mixed. Individuals with autism spectrum disorder (ASD) showed inconsistent results with some evidence of lower spindle chirp and slow-wave amplitude. Individuals with depression display

Design of Experiments and Sobol' sensitivity analysis of a hippocampus computational model.

The hippocampus is a brain area involved in many memory processes. This structure can also be affected in neurological diseases such as mesial temporal lobe epilepsy. A better understanding of its electrophysiological activity could benefit both the neuroscientific and clinical communities. We proposed, in a previous paper, a detailed bio-realistic conductance-based mathematical model of more than thirty thousand neurons to reproduce the main oscillatory features of the healthy hippocampus during slow-wave sleep and wakefulness, from slow to very fast frequencies. One big challenge of this mod

Towards threshold invariance in defining hippocampal ripples.

Objective.Hippocampal ripples are transient neuronal features observed in high-frequency oscillatory bands of local field potentials (LFPs), and they occur primarily during periods of behavioral immobility and slow-wave sleep. Ripples have been defined based on mathematically engineered features, such as magnitudes, durations, and cycles per event. However, the 'ripple' could vary from laboratory to laboratory because their definition is subject to human bias, including the arbitrary choice of parameters and thresholds. In addition, LFPs are often influenced by myoelectric noise arising from a

Spontaneous slow oscillation-slow spindle features predict induced overnight memory retention.

STUDY OBJECTIVES: Synchronization of neural activity within local networks and between brain regions is a major contributor to rhythmic field potentials such as the EEG. On the other hand, dynamic changes in microstructure and activity are reflected in the EEG, for instance slow oscillation (SO) slope can reflect synaptic strength. SO-spindle coupling is a measure for neural communication. It was previously associated with memory consolidation, but also shown to reveal strong interindividual differences. In studies, weak electric current stimulation has modulated brain rhythms and memory reten
