Pupillographic assessment of sleepiness in sleep-deprived healthy subjects.

Spontaneous pupillary-behavior in darkness provides information about a subject's level of sleepiness. In the present work, pupil measurements in complete darkness and quiet have been recorded continuously over 11-minute period with infrared video pupillography at 25 Hz. The data have been analyzed to yield three parameters describing pupil behavior; the power of diameter variation at frequencies below 0.8 Hz (slow changes in pupil size), the pupillary unrest index, and the average pupil size. To investigate the changes of these parameters in sleep deprivation, spontaneous pupillary behavior i

Absence epilepsy in rats is reduced by deprivation of REM sleep.

Effects of deprivation of REM sleep on spontaneous occurring spike wave complexes in the WAG/Rij rat strain were studied by means of the pendulum and platform technique. During the deprivation period, which lasted three days, variable results were obtained. While a suppression of the total duration of epileptic activity was found for the pendulum deprived animals, no such evident effect was detected in the platform deprived rats. Probably, instrumental effects are responsible for these differential results. On the other hand, subsequent to deprivation, a prolonged reduction of the total durati

Efficacy of sleep deprivation as an activation procedure in epilepsy patients.

The primary purpose of this article is to determine whether sleep deprivation (SD) of 24 h or longer has been demonstrated to be an effective procedure for eliciting epileptiform and/or seizure patterns in the EEG in epilepsy patients as compared with those of nonepileptics. The relevant literature is reviewed, first chronologically and then critically. Methodological problems are discussed. Although no absolutely definitive study has been done, the preponderance of evidence supports the conclusion that SD is effective in activating the EEGs of epileptics. Recommendations concerning the applic

Circadian rhythm of circulatory functions in depressives and on sleep deprivation.

Continuous examinations of circulatory functions in three 24-hour periods, before, during and after sleep deprivation, were carried out on 26 endogenous depressives and 10 neurotic depressives. The blood pressure recordings scarcely revealed relevant findings. The diurnal rhythm of heart rate in endogenous depressives is less pronounced than in healthy subjects, a phasal shift not being noted. A correlation between diurnal variation of depression symptomatology and the circadian rhythm of hear rate could be presumed on the basis of these findings. The heart rate curve is further flattened, alm

Small platform sleep deprivation selectively increases the average duration of rapid eye movement sleep episodes during sleep rebound.

The single platform-on-water (flower pot) method is extensively used for depriving rapid eye movement sleep (REMS). Detailed comparison of sleep-wake architecture, recorded during the rebound period after spending three days on either a small or large platform, could separate the effects of REMS deficit from other stress factors caused by the procedure. A further aim of the study was to find the most characteristic REMS parameter of the rebound originating from REMS deficit. Rats were kept on a small or large platform for 72 h. Their fronto-parietal electroencephalogram, electromyogram and mot

Sleep deprivation-induced cognitive impairment: Unraveling the role of neuroinflammation.

Sleep deprivation (SD) is a common public health issue linked to cognitive decline and neurodegenerative diseases. SD-induced cognitive impairment is closely linked to neuroinflammation, yet the underlying mechanisms remain incompletely understood. This review highlights neuroinflammation as a key mechanism linking SD to cognitive impairment, focusing on microglial activation via the NF-κB pathway, astrocyte dysfunction, oxidative stress, and gut-brain axis disruption. Chronic SD triggers pro-inflammatory cytokine release, neurotransmitter imbalance, and impaired synaptic plasticity, leading t

Sleep deprivation alters hepatic UGT1A9 and propofol metabolism in mice.

Sleep deprivation (SD) causes circadian misalignment, and circadian clock disruption is associated with metabolic diseases such as obesity, insulin resistance, and diabetes. However, the underlying mechanism for SD-induced circadian clock disruption as well as metabolic enzyme changes is still lacking. Here, we developed SD sensitizes mice with disrupted circadian rhythms to demonstrate the regulation role and mechanism of SD in UDP-glucuronosyltransferases (UGTs) expression and the metabolism of corresponding substrates. We found that UGT Family 1 Member A9 (UGT1A9) expression was significant

Losing Control: Sleep Deprivation Impairs the Suppression of Unwanted Thoughts.

Unwanted memories often enter conscious awareness when individuals confront reminders. People vary widely in their talents at suppressing such memory intrusions; however, the factors that govern suppression ability are poorly understood. We tested the hypothesis that successful memory control requires sleep. Following overnight sleep or total sleep deprivation, participants attempted to suppress intrusions of emotionally negative and neutral scenes when confronted with reminders. The sleep-deprived group experienced significantly more intrusions (unsuccessful suppressions) than the sleep group

Cardioprotective effects of acute sleep deprivation on ischemia/reperfusion injury.

OBJECTIVES: Modulation of sympathetic activity during acute sleep deprivation can produce various effects on body functions. We studied the effects of acute sleep deprivation before ischemia/reperfusion on myocardial injury in isolated rat hearts, and the role of sympathetic nervous system that may mediate these sleep deprivation induced effects. METHODS: The animals were randomized into four groups (n = 11 per group): Ischemia- Reperfusion group (IR), Acute sleep deprivation group (SD), Control group for sleep deprivation (CON-SD) and Sympathectomy + ASD group (SYM-SD). In SD group, sleep de

Relevance of deprivation studies in understanding rapid eye movement sleep.

Rapid eye movement sleep (REMS) is a unique phenomenon essential for maintaining normal physiological processes and is expressed at least in species higher in the evolution. The basic scaffold of the neuronal network responsible for REMS regulation is present in the brainstem, which may be directly or indirectly influenced by most other physiological processes. It is regulated by the neurons in the brainstem. Various manipulations including chemical, elec-trophysiological, lesion, stimulation, behavioral, ontogenic and deprivation studies have been designed to understand REMS genesis, maintena

Caffeine/sleep-deprivation interaction in mice produces complex memory effects.

BACKGROUND: Sleep deprivation negatively impacts memory, causing deficits in memory processes. Of interest is any agent that can offset such deficits. Mice were given varying doses of caffeine for 14 days and then deprived of sleep for 6 hours by the 'gentle handling' method. Memory was assessed using the Novel Object Recognition Test and Y maze alternation. PURPOSE: The study was designed to ascertain the impact of varying doses of caffeine combined with total sleep-deprivation on spatial and non spatial memory in mice. METHODS: Adult Swiss Webster mice of both sexes were assigned to six gr

Sleep deprivation impairs inhibitory control during wakefulness in adult sleepwalkers.

Sleepwalkers often complain of excessive daytime somnolence. Although excessive daytime somnolence has been associated with cognitive impairment in several sleep disorders, very few data exist concerning sleepwalking. This study aimed to investigate daytime cognitive functioning in adults diagnosed with idiopathic sleepwalking. Fifteen sleepwalkers and 15 matched controls were administered the Continuous Performance Test and Stroop Colour-Word Test in the morning after an overnight polysomnographic assessment. Participants were tested a week later on the same neuropsychological battery, but af

Does central fatigue explain reduced cycling after complete sleep deprivation?

PURPOSE: Sleep deprivation (SD) is characterized by reduced cognitive capabilities and endurance exercise performance and increased perceived exertion (RPE) during exercise. The combined effects of SD and exercise-induced changes in neuromuscular function and cognition are unknown. This study aimed to determine whether central fatigue is greater with SD, and if so, whether this corresponds to diminished cognitive and physical responses. METHODS: Twelve active males performed two 2-d conditions (SD and control (CO)). On day 1, subjects performed baseline cognitive and neuromuscular testing. Af

Partial sleep deprivation reduces phase advances to light in humans.

Partial sleep deprivation is increasingly common in modern society. This study examined for the first time if partial sleep deprivation alters circadian phase shifts to bright light in humans. Thirteen young healthy subjects participated in a repeated-measures counterbalanced design with 2 conditions. Each condition had baseline sleep, a dim-light circadian phase assessment, a 3-day phase-advancing protocol with morning bright light, then another phase assessment. In one condition (no sleep deprivation), subjects had an 8-h sleep opportunity per night during the advancing protocol. In the othe

Decreased arousals among healthy infants after short-term sleep deprivation.

OBJECTIVE: Sleep deprivation is a risk factor for sudden infant death syndrome (SIDS). Recent changes in normal life routines were more common among SIDS victims, compared with control infants. Sleep deprivation can result from handling conditions or from sleep fragmentation attributable to respiratory or digestive conditions, fever, or airway obstructions during sleep. Compared with matched control infants, future SIDS victims exhibited fewer complete arousals by the end of the night, when most SIDS cases occur. Arousal from sleep could be an important defense against potentially dangerous si

Paradoxical sleep deprivation in female rats alters drug-induced behaviors.

Paradoxical sleep deprivation (PSD) induces changes in behaviors induced by dopaminergic and cholinergic agonists, including increased aggressive behavior and stereotypy, decreased number of yawns, and shedding of bloody tears in male rats. In female rats, however, very little is known about the relationship between PSD and the effect of these drugs. The present study sought to examine this issue. As in males, PSD in females resulted in increased apomorphine-induced stereotypy, decreased pilocarpine-induced chromodacryorrhea, and hyperthermia. Unlike males, however, no apomorphine-induced aggr

Sex-specific acceleration of Alzheimer's pathogenesis by chronic sleep-deprivation.

INTRODUCTION: Sleep impairments likely contribute to Alzheimer's pathology, though specific contributions to disease progression are incompletely understood. We propose that autophagic impairment is associated with chronic sleep disruption and examine how sleep loss and stress influence disease development, including having impacts on proteostasis, cognition, and neural circuitry. METHODS: We sleep-disrupted 6-month-old APPNL-G-F mice for 2 weeks and behaviorally assessed sleep recovery, nesting, stress, and cognition. Subsequently, we analyzed markers of Alzheimer's pathology, stress, neuroi

Chronic sleep deprivation impairs retinal circadian transcriptome and visual function.

Sleep loss is common in modern society and is increasingly associated with eye diseases. However, the precise effects of sleep loss on retinal structure and function, particularly on the retinal circadian system, remain largely unexplored. This study investigates these effects using a chronic sleep deprivation (CSD) model in mice. Our investigation reveals that CSD significantly alters the retinal circadian transcriptome, leading to remarkable changes in the temporal patterns of enriched pathways. This perturbation extends to metabolic and immune-related transcriptomes, coupled with an accumul

Psychophysical changes after total sleep deprivation and experimental muscle pain.

Sleep disturbances exacerbate chronic pain, increase psychological load, and increase inflammation. Delayed onset muscle soreness (DOMS) mimics aspects of chronic pain, predominantly affecting peripheral pain mechanisms, while experimental sleep provocations have been shown to impact central pain mechanisms. This study aimed to combine a DOMS model with total sleep deprivation (TSD) to create a novel model affecting both peripheral and central pain mechanisms. A total of 30 healthy participants attended two sessions (baseline and follow-up) separated by 24 h of TSD and a home rating after 48 h
