Chemogenetic Breakdown of the Dentate Gate Causes Seizures and Spatial Memory Deficits.

The dentate gyrus has often been posited to act as a gate that dampens highly active afferent input into the hippocampus. Effective gating is thought to prevent seizure initiation and propagation in the hippocampus and support learning and memory processes. Pathological changes to DG circuitry that occur in temporal lobe epilepsy (TLE) can increase DG excitability and impair its gating ability which can contribute to seizures and cognitive deficits. There is evidence that TLE pathologies and seizures may independently contribute to learning and memory deficits in TLE through distinct mechanism

Connectivity, Pathology, and ApoE4 Interactions Predict Longitudinal Tau Spatial Progression and Memory.

Tau pathology spread into neocortex indicates a transition from healthy aging to Alzheimer's disease (AD). Connectivity between tau epicenters and later accumulating regions of cortex has been proposed as a mechanism of tau spread, but how this relationship changes with greater AD pathology burden or genotype is not understood. We investigated tau accumulation in two key regions, precuneus and inferior temporal cortex, using resting state functional connectivity (rsFC) and longitudinal PET imaging from a multicohort sample of cognitively unimpaired older adults. We examined how baseline tau PE

Vitamin D3 improves spatial memory and modulates cytokine levels in aged rats.

Vitamin D3 deficiency is associated with an increased risk of dementia. An association between vitamin D3 deficiency and subjective cognitive complaints in geriatric patients has been previously reported. This study aimed to evaluate the effects of two doses of vitamin D3 on spatial memory (using the Radial Maze) and cytokine levels [tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6), and interleukin-10 (IL-10)] on 2-, 6-, 13-, 22-, and 31-month-old male Wistar rats. Animals were supplemented with vitamin D3 at doses of 42 IU/kg and 420 IU/kg for 21 days. A radial ma

Perception and memory have distinct spatial tuning properties in human visual cortex.

Reactivation of earlier perceptual activity is thought to underlie long-term memory recall. Despite evidence for this view, it is unclear whether mnemonic activity exhibits the same tuning properties as feedforward perceptual activity. Here, we leverage population receptive field models to parameterize fMRI activity in human visual cortex during spatial memory retrieval. Though retinotopic organization is present during both perception and memory, large systematic differences in tuning are also evident. Whereas there is a three-fold decline in spatial precision from early to late visual areas

Maternal gastrointestinal nematode infection enhances spatial memory of uninfected juvenile mouse pups.

The developing brain is particularly vulnerable to factors including maternal infection during pregnancy. Establishment of neural networks critical for memory and cognition begins during the perinatal period, when Heligmosomoides bakeri, a gastrointestinal (GI) nematode restricted to the maternal mouse intestine, has been shown to upregulate expression of long-term potentiation genes in the young rodent pup brain. We explored the impact of maternal infection during pregnancy and early lactation on the spatial behavior of uninfected male and female juvenile mice. Pre-weaned pups of H. bakeri in

Distinct patterns of hippocampal activity associated with color and spatial source memory.

The hippocampus is known to be involved in source memory across a wide variety of stimuli and source types. Thus, source memory activity in the hippocampus is thought to be domain-general such that different types of source information are similarly processed in the hippocampus. However, there is some evidence of domain-specificity for spatial and temporal source information. The current fMRI study aimed to determine whether patterns of activity in the hippocampus differed for two types of visual source information: spatial location and background color. Participants completed three runs of a

High-Throughput Task to Study Memory Recall During Spatial Navigation in Rodents.

Spatial navigation is one of the most frequently used behavioral paradigms to study memory formation in rodents. Commonly used tasks to study memory are labor-intensive, preventing the simultaneous testing of multiple animals with the tendency to yield a low number of trials, curtailing the statistical power. Moreover, they are not tailored to be combined with neurophysiology recordings because they are not based on overt stereotyped behavioral responses that can be precisely timed. Here we present a novel task to study long-term memory formation and recall during spatial navigation. The task

Chronic dietary chlorpyrifos causes long-term spatial memory impairment and thigmotaxic behavior.

Little is known about the long-term effects of chronic exposure to low-level organophosphate (OP) pesticides, and the role of neurotransmitter systems, other than the cholinergic system, in mediating OP neurotoxicity. In this study, rats were administered 5mg/kg/day of chlorpyrifos (CPF) for 6 months commencing at 3-months-of-age. The animals were examined 7 months later (at 16-months-of-age) for spatial learning and memory in the Morris water maze (MWM) and locomotor activity. In addition, we assessed the chronic effects of CPF on glutamatergic and gamma-aminobutyric acid (GABAergic) function

Combination of attentional and spatial working memory deficits in Bálint-Holmes syndrome.

This study aims to investigate whether attention and spatiotemporal integration deficits are dissociated in patients with bilateral posterior cortical atrophy (PCA), and whether it is their combination that leads to a severe clinical handicap. We recorded performance and ocular behavior of four PCA patients and four age-matched controls in visual search and counting tasks. We measured the percentage of targets detected and the mean detection time in a "pop-out" search. We also compared counting ability when a set of dots is presented briefly (in healthy individuals, the automatic deployment of

The anterior thalamus provides a subcortical circuit supporting memory and spatial navigation.

The anterior thalamic nuclei (ATN), a central component of Papez' circuit, are generally assumed to be key constituents of the neural circuits responsible for certain categories of learning and memory. Supporting evidence for this contention is that damage to either of two brain regions, the medial temporal lobe and the medial diencephalon, is most consistently associated with anterograde amnesia. Within these respective regions, the hippocampal formation and the ATN (anteromedial, anteroventral, and anterodorsal) are the particular structures of interest. The extensive direct and indirect hip

Spatial memory deficits and thalamic serotonergic metabolite change in thiamine deficient rats.

The purposes of the present study were to verify the effects of a severe thiamine deficiency episode on spatial cognitive aspects and thalamic serotonergic parameters. The animals were submitted to a severe thiamine deficiency treatment that was interrupted after the onset of the last neurological signs. The results obtained confirm previous findings about TD deficiency effects on cognitive function and, further show that this vitamin increases the thalamic serotonine metabolite, 5-hidroxyindolacetic acid (5-HIAA), level. In addition, the present data shed light on the importance of this metab

Abstinent adolescent marijuana users show altered fMRI response during spatial working memory.

Marijuana is the most widely used illicit substance among teenagers, yet little is known about the possible neural influence of heavy marijuana use during adolescence. We previously demonstrated an altered functional magnetic resonance imaging (fMRI) activity related to spatial working memory (SWM) among adolescents who were heavy users of after an average of 8 days of abstinence, but the persisting neural effects remain unclear. To characterize the potentially persisting neurocognitive effects of heavy marijuana use in adolescence, we examined fMRI response during SWM among abstinent marijuan

Remote spatial memory in an amnesic person with extensive bilateral hippocampal lesions.

The hippocampus may have a time-limited role in memory, being needed only until information is permanently stored elsewhere, or this region may permanently represent long-term allocentric spatial information or cognitive maps in memory. To test these ideas, we investigated remote spatial memory in K.C., a patient with bilateral hippocampal lesions and amnesia for autobiographical events. In his spatial knowledge, general aspects were preserved, but details were lost, a pattern that resembled his memory loss in other domains. K.C. performed normally on allocentric spatial tests of his neighborh

Spatial working memory is preserved in rats treated with anabolic-androgenic steroids.

The effects of anabolic-androgenic steroid (AAS) compounds on spatial working memory were evaluated in male rats. Thirty days of administration of a high dose of three individual AAS compounds (17 alpha-methyltestosterone, methandrostenolone, or testosterone cypionate) had no effects on spatial memory or motivation as tested on a delayed non-match-to-sample radial arm maze task. Administration of these AAS compounds at doses within the human abuse range does not impair spatial working memory in rats.

Antisense oligonucleotides targeting basal forebrain ATXN2 enhances spatial memory and ameliorates sleep deprivation-induced fear memory impairment in mice.

INTRODUCTION: Regulation of brain-derived neurotrophic factor (BDNF) in the basal forebrain ameliorates sleep deprivation-induced fear memory impairments in rodents. Antisense oligonucleotides (ASOs) targeting ATXN2 was a potential therapy for spinocerebellar ataxia, whose pathogenic mechanism associates with reduced BDNF expression. We tested the hypothesis that ASO7 targeting ATXN2 could affect BDNF levels in mouse basal forebrain and ameliorate sleep deprivation-induced fear memory impairments. METHODS: Adult male C57BL/6 mice were used to evaluate the effects of ASO7 targeting ATXN2 micro

Hippocampal lesions impair spatial memory performance, but not song--a developmental study of independent memory systems in the zebra finch.

Songbirds demonstrate song- and spatial-learning, forms of memory that appear distinct in formal characteristics and fitting the descriptions and criteria of procedural and episodic-like memory function, respectively. As in other vertebrates, the neural pathways underlying these forms of memory may also be dissociable, and include the corresponding song circuit and hippocampus (HP). Whether (or not) these two memory systems interact is unknown. Interestingly, the HP distinguishes itself as a site of immediate early gene expression in response to song and as a site of estrogen synthesis, a ster

Visual-spatial learning and memory in schizotypal personality disorder: continued evidence for the importance of working memory in the schizophrenia spectrum.

Verbal episodic memory deficits, a well-established feature of the schizophrenia spectrum, have also been found in individuals with schizotypal personality disorder (SPD), although visual-spatial episodic memory has proven harder to examine. To address this, we administered the Visual Object Learning Test (VOLT), a measure of visual-spatial learning and memory, as well as the California Verbal Learning Test (CVLT) and a verbal working memory test, to 50 individuals with SPD, 19 with other personality disorders (OPD), and 17 healthy volunteers. Compared to both other groups, individuals with SP

Spatial generalization of peripherally encoded memories emerges before selection for report.

Visual working memory has been shown to transition from location-specific to spatially generalized representations. It remains unclear what the time course of this generalization is and to which information processing stages and signals it is linked. Here we used EEG to investigate spatial generalisation of peripherally encoded stimuli before and after an item was selected for report. Thirty healthy participants memorized two orientations presented serially at fifteen degrees eccentricity to the left and right of central fixation. One of the two items was subsequently cued as task-relevant. Du

Mental programming of spatial sequences in working memory in the macaque frontal cortex.

How the brain mentally sorts a series of items in a specific order within working memory (WM) remains largely unknown. We investigated mental sorting using high-throughput electrophysiological recordings in the frontal cortex of macaque monkeys, who memorized and sorted spatial sequences in forward or backward orders according to visual cues. We discovered that items at each ordinal rank in WM were encoded in separate rank-WM subspaces and then, depending on cues, were maintained or reordered between the subspaces, accompanied by two extra temporary subspaces in two operation steps. The Influence of Music Reading on Spatial Working Memory and Self-Assessment Accuracy.

BACKGROUND/OBJECTIVES: Previous research has suggested that Western musicians, who generally demonstrate proficiency in reading musical scores, exhibit superior performance in visuospatial working memory tasks compared to non-musicians. Evidence indicates brain activation in regions such as the left inferior parietal lobe and the right posterior fusiform gyrus during music reading, which are associated with visuospatial processing. This study aimed to explore how musical training influences spatial working memory and to examine the relationship between self-assessment accuracy and cognitive pe
