Microbial Diversity in Vehicle Windshield Washer Reservoirs: Findings from Legionella Screening.

Legionnaires' disease remains a relevant public health concern, with transmission linked to droplets from diverse aquatic environments, and its burden across Europe, including in Portugal, has been trending up. Vehicle windshield washer reservoirs have been proposed as potential, yet underexplored, habitats for Legionella spp. In this study, we investigated 62 windshield washer fluid samples collected in central Portugal. Cultivation on selective BCYE agar supplemented with GVPC and subsequent molecular identification revealed no evidence of Legionella spp. However, 23 morphologically distinct

Unraveling the allosteric mechanisms of prolyl endopeptidases for celiac disease therapy: Insights from molecular dynamics simulations.

Prolyl endopeptidases (PEP) from Sphingomonas capsulata (sc) and Myxococcus xanthus (mx) selectively degrade gluten peptides in vitro, offering a potential therapeutic strategy for celiac disease. However, the mechanisms governing the interaction of these enzymes with their substrates remain unclear. In this study, conventional molecular dynamics simulations with a microsecond timescale and targeted molecular dynamics simulations were performed to investigate the native states of mxPEP and scPEP enzymes, as well as their allosteric binding with a representative substrate, namely, Z-Ala-Pro-p-n

Effect of goethite-loaded montmorillonite on immobilization of metal(loid)s and the micro-ecological soil response in non-ferrous metal smelting areas.

In this work, the immobilization stabilization and mechanism of heavy metal(loid)s by goethite loaded montmorillonite (GMt) were investigated, and the soil microbial response was explored. The simulated acid rain leaching experiment showed that GMt had a higher acid tolerance and the more stable heavy metal(loid)s fixation ability. The soil incubation demonstrated that GMt significantly decreased the available Cd, Zn, Pb and As concentration. Interestingly, higher immobilization of heavy metals was observed by GMt in highly acid leached and acidic soils. The richness and diversity of bacterial

Characterization of biofilm and corrosion of cast iron pipes in drinking water distribution system with UV/Cl2 disinfection.

The effect of UV/Cl2 disinfection on the biofilm and corrosion of cast iron pipes in drinking water distribution system were studied using annular reactors (ARs). Passivation occurred more rapidly in the AR with UV/Cl2 than in the one with Cl2 alone, decreasing iron release for higher corrosivity of water. Based on functional gene, pyrosequencing assays and principal component analysis, UV disinfection not only reduced the required initial chlorine dose, but also enhanced denitrifying functional bacteria advantage in the biofilm of corrosion scales. The nitrate-reducing bacteria (NRB) Dechloro

Black Tea Quality is Highly Affected during Processing by its Leaf Surface Microbiome.

Microbiomes can greatly affect the quality of fermented food and beverages, including tea. In this study, microbial populations were characterized during black and green tea manufacturing, revealing that tea processing steps can drive both the bacterial and fungal community structure. Tea leaves were found to mostly harbor Proteobacteria, Bacteriodetes, Firmicutes, and Actinobacteria among bacteria and Ascomycetes among fungi. During processing, tea microbial populations changed especially between sterilized and unsterilized samples. The surface sterilization of fresh leaves before processing

Coexistence and survival of pathogenic leptospires by formation of biofilm with Azospirillum.

Pathogenic Leptospira spp. represent one cause of leptospirosis worldwide and have long been regarded as solitary organisms in soil and aquatic environments. However, in the present study, Leptospira interrogans was observed to be associated with environmental biofilms with 21 bacterial isolates belonging to 10 genera. All 21 isolates were examined for their coaggregation and biofilm-forming ability with leptospires in vitro. Among these, Azospirillum brasilense RMRCPB showed maximum interspecies coaggregation with leptospiral strains (>75%, visual score of +4). Other significant coaggregating

Flavonoid-associated microbial defense and Cellvibrio sp. C74 pretreatment collectively enhance straw degradation by Stropharia rugosoannulata.

Stropharia rugosoannulata efficiently utilizes straw in natural habitats, yet how it simultaneously defends against competing environmental microorganisms and degrades lignocellulose remains unclear. Here, transcriptomic, metabolomic, and microbiome analyses were integrated with flavonoid antibacterial assays and bacterial pretreatment experiments to characterize fungus-bacterium interactions during straw degradation. The flavonoid biosynthesis-related genes chi, fns, and fls were significantly upregulated during degradation, and the total flavonoid content reached 101.06 μg/g dry weight at th

Welan Gum, a Bacterial Polysaccharide, Promotes Maize Seedling Growth Through TOR and MAPK Signaling Pathways.

Welan gum, a high-molecular-weight exopolysaccharide from Sphingomonas sp., exhibits exceptional rheological stability and robust hydrogel properties, emerging as a highly promising agricultural biomaterial. In this study, we show that the application of welan gum promotes maize seedling growth by enhancing nitrogen use efficiency via the coordinated transcriptional reprogramming of nitrogen regulators and transporters. Mechanistically, welan gum activates the TOR pathway and the MAPK cascade, leading to the phosphorylation of ZmS6K and the terminal MAPK ZmMPK3-1. Notably, TOR inhibition reduc

Skin microbiota of fallow deer: Composition, diversity, and influencing factors.

Understanding the microbiota of wild animals can contribute to the monitoring of population health or habitat changes. The skin microbiota of fallow deer remains poorly characterized and represents a gap in current veterinary microbiology research. The aim of this study was to analyse skin microbiota of fifteen clinically healthy fallow deer using next-generation sequencing. Genomic DNA was extracted from superficial skin swabs collected from three anatomical sites per individual (n = 45 samples) and sequenced targeting the V3-V4 region of the bacterial 16S rRNA gene using Illumina sequencing.

Seminal Plasma Microbiome Composition and Its Association with Sperm Morphology in Breeding Boars.

Semen quality is a key determinant of reproductive performance in breeding boars, and emerging evidence suggests the seminal microbiome may influence sperm function. However, the composition of the seminal plasma microbiome and its relationship to sperm quality remain poorly characterized. This study aimed to investigate the microbial composition of boar seminal plasma and its association with sperm quality. Semen ejaculates collected from Duroc boars were analyzed and classified as Passed (≥70% normal morphology) or Failed (<70% normal morphology). Seminal plasma was isolated by centrifugatio

Chemical fumigants control apple replant disease: Microbial community structure-mediated inhibition of Fusarium and degradation of phenolic acids.

Fusarium and phenolic acids in apple replant soil have deleterious effects on soil, which affects the growth of young replanted apple trees. Here, we studied the effects of different chemical fumigants (metham sodium, dazomet, calcium cyanamide, 1,3-dichloropropene, and methyl bromide) on Fusarium and phenolic acids in soil. The chemical fumigants disturbed the apple replant soil microbial community to different degrees in the order from highest to the lowest as methyl bromide > 1,3-dichloropropene > dazomet > metham sodium > calcium cyanamide. Compared with the control, the total numbers of O

Dissipation of Sulfonamides in Soil Emphasizing Taxonomy and Function of Microbiomes by Metagenomic Analysis.

Sulfonamides (SAs) are widespread in soils, and their dissipation behavior is important for their fate, risk assessment, and pollution control. In this work, we investigated the dissipation behavior of different SAs in a soil under aerobic condition, focusing on revealing the relationship between overall dissipation (without sterilization and in dark) and individual abiotic (sorption, hydrolysis)/biotic (with sterilization and in dark) factors and taxonomy/function of microbiomes. The results showed that dissipation of all SAs in the soil followed the pseudo-first-order kinetic model with diss

Characterization of intestinal microbiota in alcoholic patients with and without alcoholic hepatitis or chronic alcoholic pancreatitis.

Excessive alcohol consumption leads to severe alcoholic hepatitis (sAH) or chronic alcoholic pancreatitis (CAP) only in a subset of patients. We aimed to characterize the intestinal microbiota profiles of alcoholic patients according to the presence and nature of the complications observed: sAH or CAP. Eighty two alcoholic patients were included according to their complications: CAP (N = 24), sAH (N = 13) or no complications (alcoholic controls, AC, N = 45). We analyzed the intestinal microbiota by high-throughput sequencing. Bacterial diversity was lower in patients with CAP, who had a global

Stochastic processes govern bacterial communities from the blood of pikas and from their arthropod vectors.

Vector-borne microbes influence pathogen transmission and blood microbiomes, thereby affecting the emergence of infectious diseases. Thus, understanding the relationship between host and vector microbiomes is of importance. In this study, we investigated the bacterial community composition, diversity and assembly of the flea (Rhadinopsylla dahurica vicina), torsalo (Hypoderma curzonial), and the blood and gut of their shared pika host, Ochotona curzoniae. Bartonella, Sphingomonas and Bradyrhizobium were enriched in blood, while Wolbachia and Fusobacterium were more abundant in fleas and torsal
