Probing the solution structure of the E. coli multidrug transporter MdfA using DEER distance measurements with nitroxide and Gd(III) spin labels.

Methodological and technological advances in EPR spectroscopy have enabled novel insight into the structural and dynamic aspects of integral membrane proteins. In addition to an extensive toolkit of EPR methods, multiple spin labels have been developed and utilized, among them Gd(III)-chelates which offer high sensitivity at high magnetic fields. Here, we applied a dual labeling approach, employing nitroxide and Gd(III) spin labels, in conjunction with Q-band and W-band double electron-electron resonance (DEER) measurements to characterize the solution structure of the detergent-solubilized mu

Nitroxide spin labels as EPR reporters of the relaxation and magnetic properties of the heme-copper site in cytochrome bo3, E. coli.

A nitroxide spin label (SL) has been used to probe the electron spin relaxation times and the magnetic states of the oxygen-binding heme-copper dinuclear site in Escherichia coli cytochrome bo(3), a quinol oxidase (QO), in different oxidation states. The spin lattice relaxation times, T(1), of the SL are enhanced by the paramagnetic metal sites in QO and hence show a strong dependence on the oxidation state of the latter. A new, general form of equations and a computer simulation program have been developed for the calculation of relaxation enhancement by an arbitrary fast relaxing spin system

[Study of yeast phosphoglycerate kinase by electron paramagnetic resonance. 1. Modification of protein SH-group by spin labels. Conformational changes of enzyme].

The conditions for modifications of a single SH-group of phosphoglycerate kinase from yeast by maleimid spin label and the mercury-containing spin labels were defined. The time course of immobilization of mercury containing paramagnetic radicals was obtained. Based on changes in EPR spectra we calculated dissociation constants for complexes of phosphoglycerate kinase with 3-phosphoglycerate and sulfate ions. Distinct substances were shown to induce different changes in enzyme conformation. It was concluded that approach of enzyme domains is caused by the binding of 3-phosphoglycerate-a specifi

EPR Techniques to Probe Insertion and Conformation of Spin-Labeled Proteins in Lipid Bilayers.

Electron paramagnetic resonance (EPR) spectroscopy of spin-labeled membrane proteins is a valuable biophysical technique to study structural details and conformational transitions of proteins close to their physiological environment, e.g., in liposomes, membrane bilayers, and nanodiscs. Unlike in nuclear magnetic resonance (NMR) spectroscopy, having only one or a few specific side chains labeled at a time with paramagnetic probes makes the size of the object under investigation irrelevant in terms of technique sensitivity. As a drawback, extensive site-directed mutagenesis is required in order

Probing Electrostatics in a DNA Repair Enzyme with a pH-Sensitive Nitroxyl Spin Label.

Enzymes use electrostatic interactions to recognize their substrates, preorganize active sites, and stabilize reaction transition states. Formamidopyrimidine-DNA glycosylase (Fpg) is a bacterial enzyme that repairs a pro-mutagenic DNA lesion, 8-oxoguanine; its human homologues are important for cancer prevention. General acid-base catalysis in the active site of Fpg requires a finely tuned proton transfer between Pro1 and Glu2 residues. To assess the protonation state of the active site experimentally, we used EPR spectroscopy with a novel imidazolidine nitroxyl spin label responsive in the ph

MOdulation-Guided ENcoding (MOGEN) Scheme for Vessel-Encoded Arterial Spin Labeling.

PURPOSE: Vessel-encoded arterial spin labeling (VEASL) enables simultaneous, non-contrast imaging of multiple vascular territories that is useful for differential diagnosis and treatment monitoring of cerebrovascular diseases. However, the existing encoding methods are signal-to-noise ratio (SNR) inefficient. METHODS: We developed a MOdulation-Guided ENcoding (MOGEN) scheme that directly exploits the inversion spatial modulation profile to obtain SNR-efficient encoding matrix. Simulations, phantom tests, and healthy volunteer scans were performed to demonstrate its feasibility in multiple app

Arterial Spin Labelling Perfusion Changes in Persistent Migrainous Aura Without Infarction.

Migraine is a common disorder characterized by recurrent, usually unilateral, pulsatile headaches often accompanied by nausea, vomiting, photophobia, phonophobia, and fatigue. Visual aura is the most common of all aura symptoms in migraine patients. The aura and headache phases of migraine are caused by intracranial vasoconstriction and extracranial vasodilation, respectively. Migrainous aura without infarction refers to migraine aura symptoms that persist for an extended period of one week or longer without any evidence of infarction on imaging. Arterial spin labelling (ASL) is a non-invasive

Focality in Febrile Seizures: A Retrospective Assessment Using Arterial Spin Labeling MRI.

PURPOSE: Defining focality of febrile seizures (FS) in clinical practice remains controversial. We investigated focality issues in FS with a postictal arterial spin labeling (ASL) sequence. METHODS: We retrospectively reviewed 77 children (median: 19.0 months, range: 15.0-33.0 months) who consecutively visited our emergency room for FS and underwent brain magnetic resonance imaging (MRI), including the ASL sequence, within 24 hours of seizure onset. ASL data were visually analyzed to assess perfusion changes. Factors related to the perfusion changes were investigated. RESULTS: The mean time

Arterial spin labeling perfusion in acute Wernicke encephalopathy: a case series discussion.

Wernicke's encephalopathy (WE) is a life-threatening neurologic disorder resulting from thiamine (vitamin B1) deficiency that can be secondary to chronic alcohol abuse, gastrointestinal surgery, systemic infectious and non-infectious diseases, and chemotherapy. WE is classically characterized on MRI by reduced diffusion and T2 prolongation along the mammillothalamic tracts, periaqueductal gray and tectal plate. We present two patients with acute WE who had baseline arterial spin labeling (ASL) perfusion at the time of presentation, demonstrating increase in cerebral blood flow (CBF) within the

The value of a shorter-delay arterial spin labeling protocol for detecting cerebrovascular impairment.

BACKGROUND: The aim of this study was to determine the relationship between blood oxygen level dependent (BOLD) cerebrovascular reactivity (CVR) and cerebral blood flow (CBF) obtained from arterial spin labeling (ASL) using different post labeling delays (PLD). METHODS: Forty-two patients with steno-occlusive diseases and impaired CVR were divided into two groups, one scanned with a 1.5-second (1.5-s) and the other with a 2.5-second (2.5-s) PLD ASL protocol. For all patients, a region of interest (ROI) was drawn around the CVR impairment. This affected ROI was then left-right flipped across t

Arterial spin labeling imaging correlates with the angiographic and clinical vascularity of vestibular schwannomas.

PURPOSE: Hypervascular vestibular schwannomas (HVSs) are a type of the vestibular schwannomas (VSs) that are extremely difficult to remove. We examined whether HVSs can be predicted by using arterial spin labeling (ASL) imaging. METHODS: A total of 103 patients with VSs underwent ASL imaging and digital subtraction angiography (DSA) before surgery. Regional cerebral blood flow (CBF) of gray matter and regional tumor blood flow (TBF) were calculated from ASL imaging, and we defined the ratio of TBF to CBF as the relative TBF (rTBF = TBF/CBF). Angiographic vascularity was evaluated by DSA, and

Combining site-directed spin labeling in vivo and in-cell EPR distance determination.

Structural studies on proteins directly in their native environment are required for a comprehensive understanding of their function. Electron paramagnetic resonance (EPR) spectroscopy and in particular double electron-electron resonance (DEER) distance determination are suited to investigate spin-labeled proteins directly in the cell. The combination of intracellular bioorthogonal labeling with in-cell DEER measurements does not require additional purification or delivery steps of spin-labeled protein to the cells. In this study, we express eGFP in E. coli and use copper-catalyzed azide-alkyn

Contribution of arterial spin-labelling MRI in a case with immune reconstitution inflammatory syndrome.

Central nervous system immune reconstitution inflammatory syndrome (CNS-IRIS), which occurs most often in HIV-infected patients, is an exacerbation of inflammatory reactions related to opportunistic infections as well as primary CNS malignancies both of which mostly occur in HIV-infected patients. However, differential diagnoses are challenging both clinically and radiologically. We describe a patient with CNS-IRIS due to toxoplasmosis whose 11C-methionine uptake suggested lymphoma but whose arterial spin-labelling MRI led to the correct diagnosis.

Mild Thyrotoxicosis Leads to Brain Perfusion Changes: An Arterial Spin Labelling Study.

Hypo- and hyperthyroidism have effects on brain structure and function, as well as cognitive processes, including memory. However, little is known about the influence of thyroid hormones on brain perfusion and the relationship of such perfusion changes with cognition. The present study aimed to demonstrate the effect of short-term experimental hyperthyroidism on brain perfusion in healthy volunteers and to assess whether perfusion changes, if present, are related to cognitive performance. It is known that an interaction exists between brain perfusion and cerebral oxygen consumption rate and it
