Staphylococcus aureus produces an antiphagocytic polysaccharide capsule to evade neutrophil-mediated killing. Many vaccines against encapsulated bacterial pathogens require generation of functional anti-capsular antibodies to mediate protection against infection and disease. Here it is shown that the generation of such antibody responses to S. aureus in vivo and in vitro requires the presence of O-acetyl modifications on the capsular polysaccharides. O-acetylation of S. aureus capsular polysaccharide therefore should be monitored carefully during vaccine development and production. This findin

Staphylococcus aureus (S. aureus) as common Gram-positive pathogenic bacteria, causes local and systemic infections, including sepsis and bacteremia. In particular, the high prevalence of drug-resistant S. aureus further complicates the post-infection treatment. Highly effective S. aureus vaccines are urgently desired. Herein, a novel S. aureus vaccine (MnO2@FS) is developed via biomineralizing manganese dioxide (MnO2) on formaldehyde-fixed S. aureus (FS). In such vaccine, with FS to induce bacteria-specific immune responses, MnO2 via releasing Mn2+ can activate the cyclic GMP-AMP synthase-sti

Staphylococcus lugdunensis has emerged as a significant human pathogen, responsible for a range of infections from skin and soft tissue infections to endocarditis and bacteremia. Notably, abscess formation is a common manifestation, reflecting its potential shift from a benign skin commensal to a serious pathogen, akin to infective endocarditis. With the rising prevalence of antibiotic resistance, there is a pressing need for novel therapeutic strategies. This study addresses this need by exploring the development of an effective S. lugdunensis vaccine. Multiepitope vaccines, which incorporate

Skin problems are common in returning travellers, despite the fact that vaccination, insect repellent techniques, adequate clothing and footwear are all ways to prevent skin disease. When travelling to unusual areas, the traveller can still expect to pick up diseases that are common across the world, such as lice, scabies, and staphylococcal or streptococcal infections. What can differ are the different arthropods and the diseases they can spread. Given the plethora of dermatoses in returning travellers, a methodological approach is necessary. We attempted such an approach by creating a practi

This study was conducted to analyse the clinical characteristics of measles in infants and to compare them with those in older children. Clinical and sociodemographic details of all hospitalised children with suspected measles were recorded and confirmed cases were analysed further. Twenty-four infants and 34 children aged >1 year were included. Most infants were boys and 15 (62.5%) of them were aged <9 months. Early onset of rash (within three days of fever in 62.5% of cases) occurred in infants. The complications of measles in infants included bronchopneumonia (n = 17, 71%), acute diarrhoea

Colonisation of the human skin by Staphylococcus aureus is a precursor for a variety of infections ranging from boils to sepsis and pneumonia. The rapid emergence of methicillin-resistant S. aureus following the clinical introduction of this antimicrobial drug and reports of resistance to all currently used anti-staphylococcal drugs has added to its formidable reputation. S. aureus survival on the skin and in vivo virulence is underpinned by a remarkable environmental adaptability, made possible by highly orchestrated regulation of gene expression and a capacity to undertake genome remodelling

Immunotherapy can be a sensible alternative because invasive Staphylococcus aureus infection mortality, morbidity, and cost are still alarmingly high despite the development of multiple new medications to treat methicillin-resistant S. aureus infections. Herein, killed whole-cell Staphylococcus aureus was formulated in Montanide ISA266 and Alum adjuvants, and the potency and efficacy of the vaccine were studied. After the preparation of two kinds of whole-cell vaccine (bacterin and lysate), 20 µg of each vaccine candidate was formulated in Montanide ISA266 and Alum adjuvants, then subcutaneous

INTRODUCTION: Chagas disease is a chronic parasitic infection endemic to Latin America that affects more than 7 million people and is increasingly spreading worldwide due to human migration. Trypanosoma cruzi is the etiological agent of this disease, for which no effective vaccine has yet been approved for human use. We previously developed a heterologous chimeric immunogen, CruSEG, composed of the N-terminal domain of the major cysteine protease cruzipain (Nt-Cz) fused to a genetically detoxified mutant of the staphylococcal superantigen G (SEGN24A). This modified superantigen preserves innat

Staphylococcus pseudintermedius is a major opportunistic bacterial pathogen and the leading cause of pyoderma in dogs. In canines it is also often associated with infections of the urinary system and wounds and occasionally infects people. Widespread antimicrobial resistance has made the development of alternative treatments a high priority. The development of a staphylococcal vaccine, however, has proven challenging. Identification of virulence factors that inhibit phagocytosis and avoid innate immunity may play a significant role in preventing or treating infection with S. pseudintermedius.

Many viruses often have closely related yet antigenically distinct serotypes. An ideal vaccine against viral infections should induce a multivalent and protective immune response against all serotypes. Inspired by bacterial membrane vesicles (MVs) that carry different protein components, we constructed an agr locus deletion mutant of the Staphylococcus aureus strain (RN4220-Δagr) to reduce potential toxicity. Nanoscale vesicles derived from this strain (ΔagrMVs) carry at least four major components that can deliver heterologous antigens. These components were each fused with a triple FLAG tag,

The globally prominent pathogen Streptococcus pyogenes secretes potent immunomodulatory proteins known as superantigens (SAgs), which engage lateral surfaces of major histocompatibility class II molecules and T-cell receptor (TCR) β-chain variable domains (Vβs). These interactions result in the activation of numerous Vβ-specific T cells, which is the defining activity of a SAg. Although streptococcal SAgs are known virulence factors in scarlet fever and toxic shock syndrome, mechanisms by how SAgs contribute to the life cycle of S. pyogenes remain poorly understood. Herein, we demonstrate that

Ciclosporin (Atopica; Novartis Animal Health) has been licensed for canine atopic dermatitis (AD) since 2002. Adverse events (AEs) have been reported in 55 per cent of 759 dogs in 15 clinical trials, but are rare in pharmacovigilance data (71.81 AEs/million capsules sold). Gastrointestinal reactions were most common, but were mild and rarely required intervention. Other AEs were rare (≤1 per cent in clinical trials; <10/million capsules sold). Hirsutism, gingival hyperplasia and hyperplastic dermatitis were rarely significant and resolved on dose reduction. Ciclosporin decreases staphylococcal

Staphylococcus aureus (S. aureus) remains a critical global public health threat, and the development of effective non-antibiotic prophylactic strategies is an urgent priority. To date, no licensed vaccine exists for the prevention of invasive S. aureus infections in humans. In the present study, a comprehensive multi-method pipeline was employed to design a bivalent self-amplifying mRNA (saRNA) vaccine cocktail targeting the ClfA, Hly, and SraP virulence antigens of S. aureus. An integrated framework combining immunoinformatics, structural bioinformatics, molecular simulations, and repository

Staphylococcus aureus is a common pathogen found in the community and in hospitals. Most notably, methicillin-resistant S. aureus is resistant to many antibiotics, which is a growing public health concern. The emergence of drug-resistant strains has prompted the search for alternative treatments, such as immunotherapeutic approaches. To date, most clinical trials of vaccines or of passive immunization against S. aureus have ended in failure. In this study, we investigated two ESAT-6-like proteins secreted by S. aureus, S. aureus EsxA (SaEsxA) and SaEsxB, as possible targets for a vaccine. Mice

Secretion of extracellular vesicles (EVs), a process common to eukaryotes, archae, and bacteria, represents a secretory pathway that allows cell-free intercellular communication. Microbial EVs package diverse proteins and influence the host-pathogen interaction, but the mechanisms underlying EV production in Gram-positive bacteria are poorly understood. Here we show that EVs purified from community-associated methicillin-resistant Staphylococcus aureus package cytosolic, surface, and secreted proteins, including cytolysins. Staphylococcal alpha-type phenol-soluble modulins promote EV biogenesi

Lack of known mechanisms of protection against Staphylococcus aureus in humans represents an important risk factor for skin infections and bacteremia in patients, intern hindering the development of efficacious vaccines. However, development of effective humoral response may be dampened by converging immune-evasion mechanisms of S. aureus. To develop a promising vaccine against S. aureus, it is pre-requisite to clear understanding of cutaneous, innate and adaptive immune response. The S. aureus dampening the humoral response, T cell help, blocking complement factors, and killing immune players

Mastitis caused by Staphylococcus aureus remains a significant challenge to control due to increasing antimicrobial resistance and the limited efficacy of existing vaccines. Staphylococcal enterotoxin C (SEC) is a critical virulence factor and a highly immunogenic determinant in S. aureus strains frequently isolated from bovine mastitis cases. In this study, we evaluated the immunogenicity and protective efficacy of recombinant SEC (rSEC) encapsulated within poly(lactide-co-glycolide) microparticles (PLGA-rSEC) using a murine mastitis model. Immunization with PLGA-rSEC induced significantly hi
