Orthopedic implants often fail due to bacterial infections and biofilm formation caused by drug-resistant strains, particularly Staphylococcus aureus. Bacteriophages (phages) represent a promising strategy for controlling implant-associated infections. In this study, phages targeting S. aureus were purified from wastewater and chemically grafted onto a chitosan coating applied to a Ti-6Al-4V alloy. The phage-grafted coating was abraded against the spongy portion of a porcine femur bone, disinfected using ethanol and UV exposure, and evaluated in vitro for biofilm prevention and osteoblast mine

BACKGROUND/OBJECTIVES: Methicillin-resistant Staphylococcus aureus (MRSA) continues to pose a significant challenge for infection prevention, particularly because of its ability to persist on surfaces and form resilient biofilms. Although bacteriophages have attracted renewed interest as alternatives or complements to chemical disinfectants, their applied use requires careful assessment of antimicrobial performance, formulation tolerance, and genomic context.
METHODS: Staphylococcus-infecting bacteriophages were isolated from environmental sources and examined against reference Staphylococcus

The rise of antibiotic-resistant bacteria has necessitated the development of alternative therapeutic strategies, such as bacteriophage therapy, where viruses infect bacteria, reducing bacterial burden. However, rapid bacterial resistance to phage treatment remains a critical challenge, potentially leading to failure. Phage steering, which leverages the evolutionary dynamics between phage and bacteria, offers a novel solution by driving bacteria to evolve away from virulence factors or resistance mechanisms. In this study, we examined whether phage steering using bacteriophage Luz19 could func

Staphylococcus saprophyticus is a gram-positive commensal characteristically resistant to novobiocin. Clinically, it is associated with urinary tract and other opportunistic infections. But reports on phages against them are scanty, which underlines the importance of the study. We isolated a Staphylococcus saprophyticus strain from soiled socks. The strain was characterised through biochemical tests and 16S rDNA ribotyping. Furthermore, the antibiogram revealed that the strain is resistant to penicillinase-resistant beta-lactam antibiotics. A double-stranded DNA temperate phage, ØPh_SS01, was

Background:Staphylococcus aureus biofilms contribute negatively to a number of chronic conditions, including chronic rhinosinusitis (CRS). With the inherent tolerance of biofilm-bound bacteria to antibiotics and the global problem of bacterial antibiotic resistance, the need to develop novel therapeutics is paramount. Phage therapy has previously shown promise in treating sinonasal S. aureus biofilms. Methods: This study investigates the long term (20 days) safety of topical sinonasal flushes with bacteriophage suspensions. The bacteriophage cocktail NOV012 against S. aureus selected for this

Bacteriophage (phage) therapy is a promising therapeutic modality for multidrug-resistant bacterial infections, but its application is mainly limited to personalized therapy due to the narrow host range of individual phages. While phage cocktails targeting all possible bacterial receptors could theoretically confer broad coverage, the extensive diversity of bacteria and the complexity of phage-phage interactions render this approach challenging. Here, using screening protocols for identifying "complementarity groups" of phages using non-redundant receptors, we generate effective, broad-range p

Antimicrobial resistance (AMR) has become a major concern in the treatment of bacterial infections, and bacteriophage therapy has emerged as a promising alternative to antibiotics. Bacteriophages are highly specific to their bacterial hosts; hence, isolating phages indigenous to a specific region offers a significant advantage against various pathogen strains. We have isolated a cocktail of bacteriophages against pathogenic E. coli from sewage water at a primary healthcare centre. Characterisation of the isolated phages demonstrated their stability across a broad pH and temperature range, stro

Human milk is the optimal nutrition source for infants and contains a complex mix of bioactive compounds and microorganisms. When unavailable, pasteurized donor milk may be provided, particularly to preterm infants. Holder pasteurization (HP) is typically implemented in human milk banks to prevent pathogen transmission. Given the impact of heat on milk bioactives, ultraviolet-C irradiation (UV-C) is an alternative being explored and has demonstrated effective bactericidal activity. In addition to bacteria, milk contains viruses, including primarily bacteriophages (phages) and which likely infl

One hundred and fifty hospital Staphylococcus aureus strains isolated on Polish hospitals during an outbreaks were used to evaluate usefulness of S. aureus phenotyping methods. According to the expression of resistance to methicillin (a marker of resistance to all beta-lactam antibiotics) all strains were classified as homogeneously of heterogeneously resistant to methicillin (methicillin resistant S. aureus--MRSA) and as susceptible to this antibiotic (methicillin-susceptible S. aureus--MSSA). All strains were analysed according to the resistance patterns to fifteen antistaphylococcal drugs,

The SaPIs are 15- to 17-kb mobile pathogenicity islands in staphylococci. They usually carry two or more superantigens and are responsible for most superantigen-related human diseases, especially staphylococcal toxic shock syndrome. SaPIs are extremely common in Staphylococcus aureus, with all but one of the sequenced genomes containing one or more. The SaPIs have a highly conserved overall genome organization, parallel to that of typical temperate phages. Each occupies a specific chromosomal site from which it is induced to excise and replicate by one or more specific staphylococcal phages. F

BACKGROUND: The purpose of the study was to evaluate the in vitro antibacterial effect of experimental eye drops with bacteriophages in elimination of Staphylococcus spp. isolated from dogs with bacterial conjunctivitis.. The bacterial material was collected from dogs with independent clinical signs of bacterial conjunctivitis. Staphylococcus spp. were identified by phenotypic and genotypic methods (MALDI-TOF MS mass spectrometry). Antibiotic resistance was determined by the disc-diffusion method. Phage activity (Plaque forming units, PFU) was determined on double-layer agar plates. Phages wit

OBJECTIVES: Staphylococcus aureus and Pseudomonas aeruginosa co-infections in patients with cystic fibrosis (CF) are associated with disease severity. Their treatment is complicated by biofilm formation in the sticky mucus obstructing the airways. We investigated the activity of phages-antibiotics combinations using a dual species biofilm (P. aeruginosa/S. aureus) formed in artificial sputum medium.
METHODS: Biofilmswere incubated with broad-spectrum antibiotics (meropenem, ceftazidime, ciprofloxacin, tobramycin) combined with a cocktail of two (bacterio)phages (PSP3 and ISP) proven active vi

The evolution of the virulence and antibiotic resistance of staphylococci, important opportunistic pathogens, is strongly determined by their mobilome, which can spread by phage virions or small-headed particles resulting from the hijacking of helper phage machinery by phage satellites named phage-inducible chromosomal islands (PICIs). Despite known mechanisms of the formation of transducing particles, it has not yet been possible to analyze their DNA content at the single-virion level. Using the Staphylococcus epidermidis model and long-read nanopore sequencing, we determined the sequence str

PURPOSE: Methicillin-resistant Staphylococcus aureus (MRSA) continues to compromise the efficacy of conventional antibiotics, highlighting the urgent need for alternative antimicrobial strategies. This study aimed to isolate, characterize, and evaluate the therapeutic potential of two strictly lytic bacteriophages, Staphylococcus phages EAS1 and EAS2, against MRSA, including their safety and efficacy in an in vivo myositis model.
METHODS: EAS1 and EAS2 were isolated from wastewater and characterized using transmission electron microscopy, whole-genome sequencing, and in vitro functional assay

Staphylococcus aureus is a very successful opportunistic pathogen capable of causing a variety of diseases ranging from mild skin infections to life-threatening sepsis, meningitis and pneumonia. Its ability to display numerous virulence mechanisms matches its skill to display resistance to several antibiotics, including β-lactams, underscoring the fact that new anti-S. aureus drugs are urgently required. In this scenario, the utilization of lytic bacteriophages that kill bacteria in a genus -or even species- specific way, has become an attractive field of study. In this report, we describe the

BACKGROUND: Chronic Staphylococcus aureus prosthetic joint infection (PJI) remains difficult to manage when surgical revision and long-term antibiotics are not feasible. Bacteriophage therapy is emerging as a potential adjunct, though experience in orthopedic infections, particularly in the United Kingdom, is limited.
CASE SUMMARY: We report the first UK case of intra-articular bacteriophage therapy administered alongside debridement, antibiotics, and implant retention (DAIR) for chronic methicillin-sensitive Staphylococcus aureus knee PJI. An 81-year-old man with multiple comorbidities devel

Holins are phage-encoded hydrophobic membrane proteins that spontaneously and non-specifically accumulate and form lesions in the cytoplasmic membrane. The ORF72 gene (also designated HolGH15) derived from the genome of the Staphylococcus aureus phage GH15 was predicted to encode a membrane protein. An analysis indicated that the protein encoded by HolGH15 potentially consisted of two hydrophobic transmembrane helices. This protein exhibited the structural characteristics of class II holins and belonged to the phage_holin_1 superfamily. Expression of HolGH15 in Escherichia coli BL21 cells resu

Engineered luciferase reporter bacteriophages provide specific, sensitive, rapid and low-cost detection of target bacteria and address growing diagnostic needs in multiple industries. Detection of methicillin-resistant Staphylococcus aureus (MRSA) nasal colonization and antibiotic susceptibility play a critical supportive role in preventing hospital-acquired infections and facilitating antibiotic stewardship. We describe the development and evaluation of a novel phage-based MRSA diagnostic screen for nasal swab specimens. The screen utilizes two luciferase reporter phages capable of recognizin

A total of 190 Staphylococcus epidermidis isolates from healthy carriers and clinical material were biotyped according to the Baird-Parker scheme, phage typed by a provisional set of 18 typing phages and tested for antimicrobial susceptibility. The majority of the strains from both groups were found to belong to biotype 1. A considerable number of strains were lysed at 100 x RTD by at least one phage. The strains from healthy carriers were distributed in 24 phage types and about half of them were clustered among three major phage types. Nine of the 30 clinical isolates were of the phage type 1

Enterotoxin production by strains of Staphylococcus aureus isolated from clinical specimens of human and animal origin and from healthy human carriers was investigated. All nine patients admitted to hospital with symptoms of toxic shock syndrome (TSS) yielded enterotoxin-producing strains of S. aureus. Eight of these produced staphylococcal enterotoxin F (SEF). A significantly smaller proportion of strains (42% of 50 strains tested) isolated from other clinical specimens of hospitalized patients produced SEF. Production of SEF by strains isolated from clinical specimens of animal origin (48 st
