Rare Polyene-polyol Macrolides from Mangrove-derived Streptomyces sp. ZQ4BG.

Bioactive natural products from mangrove-derived actinomycetes are important sources for discovery of drug lead compounds. In this study, an extract prepared from culture of an actinomycete Streptomyces sp. ZQ4BG isolated from mangrove soils was found to have activity in inhibiting proliferation of glioma cells. Large culture of this mangrove actinomycete in Gause's liquid medium resulted in isolation of seven novel polyene-polyol macrolides, named as flavofungins III-IX (3-9), together with known flavofungins I (1) and II (2) and spectinabilin (10). Structures of these isolated compounds were

Engineering Streptomyces coelicolor Carbonyl Reductase for Efficient Atorvastatin Precursor Synthesis.

Streptomyces coelicolor CR1 (ScCR1) has been shown to be a promising biocatalyst for the synthesis of an atorvastatin precursor, ethyl-(S)-4-chloro-3-hydroxybutyrate [(S)-CHBE]. However, limitations of ScCR1 observed for practical application include low activity and poor stability. In this work, protein engineering was employed to improve the catalytic efficiency and stability of ScCR1. First, the crystal structure of ScCR1 complexed with NADH and cosubstrate 2-propanol was solved, and the specific activity of ScCR1 was increased from 38.8 U/mg to 168 U/mg (ScCR1I158V/P168S) by structure-guid

Field efficacy of nonpathogenic Streptomyces species against potato common scab.

AIMS: The primary objective of these experiments was to reduce pathogenicity and virulence of endemic soil pathogenic Streptomyces strains that cause potato common scab (CS) using nonpathogenic Streptomyces strains to suppress CS in a field situation. METHODS AND RESULTS: Nonpathogenic Streptomyces strains that had shown potential for mitigating CS in greenhouse assays were used in Michigan and Pennsylvania fields known to have high CS disease pressure. Five biocontrol (BC) strains and three potato cultivars were used in 2009, and three BC strains and three cultivars were used in 2010 in each

Human cervicofacial mycetoma caused by Streptomyces griseus: First case report.

Streptomyces griseus causes subcutaneous mycetomas in felines and dolphins; however, human mycetoma caused by S. griseus has not previously been reported. Hereby, we report a case of a 50-year-old female presenting with swelling in the left upper cervical region and the left cheek that lasted for 6 months. The fine needle aspiration (FNA) performed on the swelling yielded purulent material; on microscopy, actinomycosis was diagnosed. On culturing, the pus grew S. griseus. To the best of our knowledge, this is the first reported case of human mycetoma caused by S. griseus.

Three pathway-specific regulators control streptolydigin biosynthesis in Streptomyces lydicus.

The streptolydigin biosynthetic gene cluster from Streptomyces lydicus NRRL 2433 contains three putative regulatory genes, slgR1, slgR2 and slgY, encoding proteins belonging to TetR and LuxR transcriptional regulator families and ATP/GTP-binding proteins of DNA and RNA helicase superfamily I, respectively. Inactivation of slgR1 or slgR2 resulted in the abolition of streptolydigin production, suggesting that these proteins are pathway-specific positive regulators. In the case of the slgR1 mutant, low amounts of streptolydigin C were produced instead of streptolydigin. RT-PCR transcription analy

Antimicrobial activity of Streptomyces species against mushroom blotch disease pathogen.

Bacterial blotch caused by Pseudomonas tolaasii is still one of the main diseases in mushroom farms. Effective treatment of the infections caused by P. tolaasii is yet to be established. The isolation and identification of three Streptomyces species that showed antimicrobial activity against P. tolaasii NCPPB 2192(T) were described. Six strains were highly active with an inhibition zone more than 20 mm in diameter. They were assigned to the known Streptomyces cluster groups as S. rochei, S. lydicus and S. antibioticus. The fermentation, preliminary extraction and isolation of bioactive compone

Occurrence of Turnip Scab Caused by Phytotoxin-Producing Streptomyces spp.

Turnips (Brassica rapa L var. rapifera) purchased from a New Brunswick supermarket in the winter of 2003 contained discernable corky scab-like lesions on their surfaces. The turnips were surface-disinfested with a warm-water rinse, and microorganisms from excised lesions were isolated with methodologies developed for the specific isolation of Streptomyces spp. (1). Nine isolates morphologically characteristic of Streptomyces scabies strains (2) were subsequently examined for pathogenicity on potato mini-tubers and production of the scab phytotoxin, thaxtomin A (1). Four of the nine isolates pr

[Studies on regulation of glutamine synthetase activity from Streptomyces lincolnensis].

Glutamine synthetase in crude extracts from Streptomyces lincolnensis growing under different nitrogen sources were studied. The results showed that NH4+ in high concentration repressed the biosynthesis of the enzyme. To determine whether Streptomyces lincolnensis has undergone covalent modification, a comparison of the glutamine synthetase isolated from cells grown on different nitrogen sources was made. No significant difference was observed in specific activity, pH optima, divalent cation response, and ultraviolet absorption spectra. Glutamine synthetase activity was not influenced by ammon

Relationship between alkaline phosphatase and neomycin formation in Streptomyces fradiae.

Studies on phosphatase activity of Streptomyces fradiae 3535 grown in three different media indicate that neomycin formation varies directly with enzyme activity, sodium nitrate-maltose-mineral salts medium giving the highest yields of alkaline phosphatase and neomycin. S. fradiae contains more than one alkaline phosphatase and the phosphatase responsible for hydrolysis of neomycin phosphate appears to be substrate specific. The same enzyme apparently hydrolyses both the N-P and P-O-P bonds of neomycin pyrophosphate. The enzyme is stimulated by Ca(2+), is inactive at a pH below 7 and is inhibi

Heterologous production of kasugamycin, an aminoglycoside antibiotic from Streptomyces kasugaensis, in Streptomyces lividans and Rhodococcus erythropolis L-88 by constitutive expression of the biosynthetic gene cluster.

Kasugamycin (KSM), an aminoglycoside antibiotic isolated from Streptomyces kasugaensis cultures, has been used against rice blast disease for more than 50 years. We cloned the KSM biosynthetic gene (KBG) cluster from S. kasugaensis MB273-C4 and constructed three KBG cassettes (i.e., cassettes I-III) to enable heterologous production of KSM in many actinomycetes by constitutive expression of KBGs. Cassette I comprised all putative transcriptional units in the cluster, but it was placed under the control of the P neo promoter from Tn5. It was not maintained stably in Streptomyces lividans and di

Antibiotic production reduces the cost of resource exploitation in Streptomyces coelicolor.

Soil is a competitive environment containing a variety of resources used for bacterial growth. Complex polysaccharides, like chitin or starch, require the secretion of enzymes that degrade these resources into smaller units before they can be consumed. However, exoenzymes and the products they create are public goods, meaning they can be used by competitors who consume and benefit from these public goods even if they do not produce enzymes themselves. Here, we test the hypothesis that antibiotics produced by Streptomyces are used to privatize public goods by restricting access to resource expl

Dual Silencing of Aminoglycoside Biosynthesis for the Discovery of Novel Antimicrobials in Streptomyces.

Streptomyces species are prolific sources of antimicrobials, yet their vast biosynthetic capacity remains underexploited due to the rediscovery of common antibiotics (e.g., ~1% for streptomycin and ~10% for streptothricin). To address this challenge, we developed a dual-layer biosynthetic blockade system, designated CI-ARS3, which integrates two orthogonal silencing modules: a competitive transcriptional silencer that represses transcription of the native biosynthetic gene cluster (BGC), and an antisense RNA-based silencer that facilitates the degradation of BGC-derived mRNAs and/or translatio

The involvement of multiple ABC transporters in daunorubicin efflux in Streptomyces coeruleorubidus.

Streptomyces genus produces a large number of antibiotics, which are always synthesized by specific biosynthetic gene clusters (BGCs). To resist autotoxicity, transporters encoded by genes located within BGC occasionally pump antibiotic along with transporter encoded by gene located outside BGC. Daunorubicin is an anthracycline antibiotic biosynthesized by Streptomyces species, playing a crucial role in the treatment of leukaemia. In existing studies, only one two-component ATP-binding cassette (ABC) transporter, encoded by drrA1-drrB1 (abbreviated as drrAB1) and located within the daunorubici

Mining and engineering exporters for titer improvement of macrolide biopesticides in Streptomyces.

Exporter engineering is a promising strategy to construct high-yield Streptomyces for natural product pharmaceuticals in industrial biotechnology. However, available exporters are scarce, due to the limited knowledge of bacterial transporters. Here, we built a workflow for exporter mining and devised a tunable plug-and-play exporter (TuPPE) module to improve the production of macrolide biopesticides in Streptomyces. Combining genome analyses and experimental confirmations, we found three ATP-binding cassette transporters that contribute to milbemycin production in Streptomyces bingchenggensis.
