Biology of bone metastases.

BACKGROUND: Bone metastases cause morbidity and mortality in multiple malignancies. In addition to portending a dire prognosis, bone metastases cause bone pain, fractures, hypercalcemia, spinal cord compression, and other nerve compression syndromes. Improved understanding of the mechanisms that predispose tumor metastases to bone is needed to improve patients' therapeutic options, maintain their quality of life, and improve their survival. METHODS: This review discusses selected preclinical and clinical data regarding bone metastasis development and cytokine/molecular interactions predisposi

Cell Trafficking in Chronic Lymphocytic Leukemia.

Chronic lymphocytic leukemia (CLL) is an indolent lymphoproliferative disorder characterized by both circulating peripheral disease as well as involvement of the lymph nodes and bone marrow. Increasing evidence suggests that the stromal microenvironment provides anti-apoptotic and pro-survival signals to CLL cells, and may contribute significantly to resistance to a wide variety of treatments. Our understanding of the complex interactions involved in CLL cell trafficking continues to grow. Chemokines and corresponding chemokine receptors are key factors for organizing CLL cell trafficking and

Mantle cell lymphoma cells express high levels of CXCR4, CXCR5, and VLA-4 (CD49d): importance for interactions with the stromal microenvironment and specific targeting.

Mantle cell lymphoma (MCL) is characterized by an early, widespread dissemination and residual disease after conventional treatment, but the mechanisms responsible for lymphoma cell motility and drug resistance are largely unknown. There is growing evidence suggesting that chemokine receptors and adhesion molecules are critical for malignant B-cell trafficking and homing to supportive tissue microenvironments, where they receive survival and drug resistance signals. Therefore, we examined chemokine receptor and adhesion molecule expression and function in MCL cells and their importance for mig

Trial Watch: Tumor-targeting monoclonal antibodies in cancer therapy.

In 1997, for the first time in history, a monoclonal antibody (mAb), i.e., the chimeric anti-CD20 molecule rituximab, was approved by the US Food and Drug Administration for use in cancer patients. Since then, the panel of mAbs that are approved by international regulatory agencies for the treatment of hematopoietic and solid malignancies has not stopped to expand, nowadays encompassing a stunning amount of 15 distinct molecules. This therapeutic armamentarium includes mAbs that target tumor-associated antigens, as well as molecules that interfere with tumor-stroma interactions or exert direct

Cd40 expression in uterine tissues: a key regulator of cytokine expression by fibroblasts.

CD40 is a cell surface receptor initially discovered on cells of the hemopoietic lineage. Its primary role on immune cells is to enhance their activation and hence their production of cytokines and immunomodulatory molecules. Recently, CD40 has also been detected on human fibroblasts. An emerging view of the fibroblast is that it is far more than a structural cell, being capable of intimate interaction with cells of the immune system. In fibroblasts from several tissues, the engagement of CD40 with its ligand (CD40L) resulted in the secretion of proinflammatory molecules such as interleukin-6

[The immune microenvironment in malignant lymphoma].

Recent advances in immunotherapy have highlighted the importance of the tumor microenvironment. Lymphomas are a heterogeneous group of malignancies that arise from lymphocytes and typically develop in lymphoid tissues. It has been recognized that the elements of the lymphoma microenvironment are not mere bystanders to a host antitumor inflammatory response but are important components of the tumor that support proliferation, survival, and chemoresistance of lymphoma cells. Lymphoma cells have individual expression patterns of chemokine receptors and adhesion molecules, according to their histo

VISTA: bridging gaps in cancer immunotherapy.

Tumor microenvironment (TME) is complicated by the interaction of different cells of immune system, stromal components, and tumor-associated elements. Immune cells largely influence tumor progression by the means of various activating and inhibitory mechanisms including, immune checkpoint molecules. These molecules have been targeted for treating different types of cancers. For instance, blocking antibodies against CTLA-4, PD-1, or PD-L1 have elicited durable clinical responses and remarkable efficacy. These antibodies have also led to long-term remissions in a subset of patients, especially w

Molecular biology of peritoneal carcinomatosis.

Peritoneal carcinomatosis can be thought of as a sequence of events that together form a peritoneal metastatic cascade. Presently our understanding of the molecular mediators that orchestrate this cascade is ill-understood. Initial tumour-mesothelial interaction appears to involve several adhesion molecules, including CD44, the Selectins, and various leukocyte associated antigens. The exact molecules involved are probably determined by the nature of the metastatic tumour cell. Invasion of the mesothelial monolayer appears to occur by tumour-induced mesothelial apoptosis, at least in part via t

Localization of a corneal basement membrane glycoconjugate in bovine eye.

Lectin histochemistry was used to analyze the ocular surface basement membrane in order to identify novel, tissue-specific glycoconjugates. Soybean agglutinin (SBA), a lectin marker for N-acetylgalactosamine residues, recognized a 130 kDa glycoconjugate in corneal but not conjunctival basement membrane. Following corneal epithelial cell wounding in vitro, the glycoconjugate was not expressed at the epithelial cell-stromal interface until 72-96 h in culture, much later than the expression of other basement membrane molecules. In epithelial cells maintained on various extracellular matrix substr

Ovine conceptuses express phospholipase inhibitory genes on days 14-15 of pregnancy, interacting with IFNT pathways.

IN BRIEF: Ovine conceptuses highly express phospholipase inhibitory genes just before the conceptus attachment period. Phospholipase inhibitors could synergistically work with the interferon pathway on the endometrium. ABSTRACT: In mammals, various molecules are involved in the biochemical interaction between the conceptus and endometrium for pregnancy recognition and establishment. In ruminants, interferon tau (IFNT) is the pregnancy recognition factor; however, IFNT alone does not explain corpus luteum maintenance. Although data on factors expressed during implantation have been accumulated

Monomeric hapten dissociates FcεRI-evoked STIM1-Orai1 interaction and immediately terminates Ca2+ influx in rat basophilic leukemia cells.

Crosslinking of the high affinity IgE receptor (FcεRI) by multivalent antigen increases intracellular Ca2+ concentration ([Ca2+]i) through two sequential steps: (i) Ca2+ release from the endoplasmic reticulum (ER) and (ii) store-operated Ca2+ entry (SOCE) mediated by stromal interaction molecule 1 (STIM1)-Orai1 interaction. This process is essential for basophil and mast cell activation. We previously showed that FcεRI disaggregation by monomeric hapten rapidly and persistently inhibited [Ca2+]i increase although upstream signal disruption was transient in rat basophilic leukemia cells. STIM1 functionally couples to transient receptor potential ankyrin 1 contributing to nociception.

STIM1 is a calcium sensor that can sense calcium level changes in the endoplasmic reticulum (ER) and respond to extracellular stimuli. We have reported that STIM1 is expressed in nociceptors. However, its functional significance remains unclear. Here, we show that STIM1 plays an important role in sensing cold, chemical, and noxious mechanical stimuli in both male and female mice. We found that activation of transient receptor potential ankyrin 1 (TRPA1) triggers ER Ca 2+ release, STIM1 translocation, and store-operated Ca 2+ entry (SOCE). Immunostaining and western blot results reveal that TRP

Voluntary Wheel Running Mitigates Disease in an Orai1 Gain-of-Function Mouse Model of Tubular Aggregate Myopathy.

Tubular aggregate myopathy (TAM) is an inherited skeletal muscle disease associated with progressive muscle weakness, cramps, and myalgia. Tubular aggregates (TAs) are regular arrays of highly ordered and densely packed straight-tubules observed in muscle biopsies; the extensive presence of TAs represent a key histopathological hallmark of this disease in TAM patients. TAM is caused by gain-of-function mutations in proteins that coordinate store-operated Ca2+ entry (SOCE): STIM1 Ca2+ sensor proteins in the sarcoplasmic reticulum (SR) and Ca2+-permeable ORAI1 channels in the surface membrane. The spectrum of neuromuscular diseases with tubular aggregates.

Tubular aggregates are granular inclusions of unclear function postulated to be massive proliferations from lateral sacs of the sarcoplasmic reticulum. However, immunohistochemical studies suggest a more complex origin. They usually accumulate in type 2 fibers and do not react to the mitochondrial oxidative enzymes. Tubular aggregates are present in diverse acquired and inherited clinical conditions. Myopathies in which the predominant pathological hallmark is tubular aggregates usually manifest in one of three major phenotypes: 1) slowly progressive, proximal predominant weakness; 2) myalgia

STIM1 ablation impairs exercise-induced physiological cardiac hypertrophy and dysregulates autophagy in mouse hearts.

Cardiac stromal interaction molecule 1 (STIM1), a key mediator of store-operated Ca2+ entry (SOCE), is a known determinant of cardiomyocyte pathological growth in hypertrophic cardiomyopathy. We examined the role of STIM1 and SOCE in response to exercise-dependent physiological hypertrophy. Wild-type (WT) mice subjected to exercise training (WT-Ex) showed a significant increase in exercise capacity and heart weight compared with sedentary (WT-Sed) mice. Moreover, myocytes from WT-Ex hearts displayed an increase in length, but not width, compared with WT-Sed myocytes. Conversely, exercised card

Silencing of the Ca2+ Channel ORAI1 Improves the Multi-Systemic Phenotype of Tubular Aggregate Myopathy (TAM) and Stormorken Syndrome (STRMK) in Mice.

Tubular aggregate myopathy (TAM) and Stormorken syndrome (STRMK) form a clinical continuum associating progressive muscle weakness with additional multi-systemic anomalies of the bones, skin, spleen, and platelets. TAM/STRMK arises from excessive extracellular Ca2+ entry due to gain-of-function mutations in the Ca2+ sensor STIM1 or the Ca2+ channel ORAI1. Currently, no treatment is available. Here we assessed the therapeutic potential of ORAI1 downregulation to anticipate and reverse disease development in a faithful mouse model carrying the most common TAM/STRMK mutation and recapitulating th

Stromal Vascular Fraction and its Role in the Management of Alopecia: A Review.

Adipose cells organized in small clusters under the reticular dermis closely interact with hair follicular cells and regulate the hair cycle. Intradermal adipocyte progenitor cells are activated toward the end of the telogen phase to proliferate and differentiate into mature adipocytes. These cells, surrounding the hair follicles, secrete signaling molecules that control the progression of the hair cycle. Diseases associated with defects in adipocyte homeostasis, such as lipodystrophy and focal dermal hypoplasia, lead to alopecia. In this review, we discuss the potential influence of stromal v

IGF-1 increases survival of CD4+ lineage in a 2D model of thymocyte/thymic stromal cell co-culture.

Insulin-like growth factor-1 (IGF-1), in addition to its classic effects on cell proliferation and organism growth, has pleiotropic actions on the immune system, particularly on the thymus. Thus, the objective of this study was to evaluate the influence of IGF-1 on molecules involved in the survival of thymocytes in vitro using a co-culture system with thymic stromal cells obtained from C57BL/6 mice. The obtained thymic stroma has contained thymic epithelial cells, macrophages, dendritic cells, fibroblasts, and preserved the expression of the major histocompatibility complex (MHC) molecules. F

Inhibition of focal adhesion kinase overcomes resistance of mantle cell lymphoma to ibrutinib in the bone marrow microenvironment.

Mantle cell lymphoma and other lymphoma subtypes often spread to the bone marrow, and stromal interactions mediated by focal adhesion kinase frequently enhance survival and drug resistance of the lymphoma cells. To study the role of focal adhesion kinase in mantle cell lymphoma, immunohistochemistry of primary cases and functional analysis of mantle cell lymphoma cell lines and primary mantle cell lymphoma cells co-cultured with bone marrow stromal cells (BMSC) using small molecule inhibitors and RNAi-based focal adhesion kinase silencing was performed. We showed that focal adhesion kinase is

Hypercapnia enhances airway smooth muscle contractility via STIM1-dependent Ca2+ signaling.

Hypercapnia, elevated carbon dioxide (CO2), is common in advanced chronic obstructive pulmonary disease (COPD) and predicts poor clinical outcomes. Traditionally considered a consequence of disease severity, hypercapnia may drive disease progression by promoting airway dysfunction. Here, we show that hypercapnia acts as an active stressor, driving airway smooth muscle (ASM) constriction through a stromal interaction molecule 1-dependent (STIM1-dependent) pathway. Hypercapnia rapidly activates ERK, triggering sarcoplasmic reticulum calcium (Ca2+) release via phosphorylation of the inositol 1,4,
