Leflunomide monotherapy versus combination therapy with conventional synthetic disease-modifying antirheumatic drugs for rheumatoid arthritis: a retrospective study.

Leflunomide (LEF) is a conventional synthetic disease-modifying antirheumatic drugs (csDMARDs) for the treatment of rheumatoid arthritis. However, there are few reports on the comparison of efficacy between LEF alone and combined with other csDMARDs. Here, the efficacy and safety of LEF monotherapy (88) and combination (361) therapy groups were evaluated. After 3 months, there were no significant differences in 28-joint disease activity score (DAS28), health assessment questionnaire (HAQ), erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP) between the monotherapy and combination

Targeting Alzheimer's disease pathology: influence of nano-based drug delivery systems loaded with a combination of herbal and synthetic drugs.

INTRODUCTION: Alzheimer's Disease (AD) is a progressive neurological disorder marked by cognitive decline and memory loss. Current treatments, including acetylcholinesterase inhibitors (AChEIs) and NMDA receptor antagonists, provide only symptomatic relief due to poor Blood Brain Barrier (BBB) permeability and side effects. The integration of synthetic and natural drug combinations with nanotechnology offers a promising strategy to enhance drug delivery, efficacy, and overall therapeutic outcomes. AREAS COVERED: This review explores the integration of herbal and synthetic drugs in nano-based

[Drug permeation through synthetic lipid membranes. Part 13: The effects of tensides on the permeation of ionized drugs (author's transl)].

The permeation of substances present in the ionized form is most markedly affected by tensides containing ions of opposite sign. These tensides produce a considerable increase in permeation. The effect of micelle formation will become evident only if the transport of the free drug through the membrane is more rapid than the release of the drug from the micelles. If the concentrations of nonionic tensides are less than the critical micelle concentration, a permeation increase occurs which is presumably caused by a reduction of the surface tension of the membrane.

Discordance between self-reported and detected synthetic opioids reveals cychlorphine emergence among people who inject drugs in Estonia.

INTRODUCTION: Since 2002, Estonia has experienced a changing illicit synthetic opioid market, with the emergence of highly potent substances posing a substantial threat to public health. Over the transitional periods, people who inject drugs have faced high risks, often without knowing what substance they are using. This study assesses the agreement between self-reported and detected drug use and examines the presence of synthetic opioids in urine and how long they remain detectable. METHODS: Out of 350 individuals in a cross-sectional bio-behavioural survey among people who inject drugs in K

Risk of Serious Infections in Patients Treated With Biologic or Targeted-synthetic Disease Modifying Antirheumatic Drugs in Qatar.

BACKGROUND: Biologic and targeted-synthetic disease-modifying antirheumatic drugs (b/tsDMARDs), are pivotal in the management of autoimmune-inflammatory disorders, acting by suppressing pathological immune activation. Because of associated immune dysfunction, opportunistic or serious infections (SIs), and latent disease reactivation is frequently reported. This study aimed to investigate the epidemiology, risk factors, and outcomes of SIs in patients treated with b/tsDMARDs in Qatar. METHODS: A retrospective cohort study was conducted at Hamad Medical Corporation, including all the patients t

Synthetic repurposing of drugs against hypertension: a datamining method based on association rules and a novel discrete algorithm.

BACKGROUND: Drug repurposing aims to detect the new therapeutic benefits of the existing drugs and reduce the spent time and cost of the drug development projects. The synthetic repurposing of drugs may prove to be more useful than the single repurposing in terms of reducing toxicity and enhancing efficacy. However, the researchers have not given it serious consideration. To address the issue, a novel datamining method is introduced and applied to repositioning of drugs for hypertension (HT) which is a serious medical condition and needs some improved treatment plans to help treat it. RESULTS

Effects of Conventional Synthetic Disease-Modifying Antirheumatic Drugs on Response to Periodontal Treatment in Patients with Rheumatoid Arthritis.

Rheumatoid arthritis (RA) and periodontitis are common chronic inflammatory diseases and periodontitis is known to be more common and more severe in patients with RA. Based on a paucity of studies about the relationship between common conventional synthetic disease-modifying antirheumatic drugs (csDMARDs) and periodontitis, this prospective study aimed to evaluate the adjunctive effect of csDMARDs on response to nonsurgical periodontal treatment in patients with RA. Thirty-two patients with RA (RA group) and 32 systemically healthy patients (control group) with periodontitis were included in t

EULAR recommendations for the management of rheumatoid arthritis with synthetic and biological disease-modifying antirheumatic drugs: 2013 update.

In this article, the 2010 European League against Rheumatism (EULAR) recommendations for the management of rheumatoid arthritis (RA) with synthetic and biological disease-modifying antirheumatic drugs (sDMARDs and bDMARDs, respectively) have been updated. The 2013 update has been developed by an international task force, which based its decisions mostly on evidence from three systematic literature reviews (one each on sDMARDs, including glucocorticoids, bDMARDs and safety aspects of DMARD therapy); treatment strategies were also covered by the searches. The evidence presented was discussed and
