Bitter substances from plants used in traditional Chinese medicine exert biased activation of human bitter taste receptors.

The number and variety of bitter compounds originating from plants are vast. Whereas some bitter chemicals are toxic and should not be ingested, other compounds exhibit health beneficial effects, which is manifest in the cross-cultural believe that the bitterness of medicine is correlated with the desired medicinal activity. The bitter taste receptors in the oral cavity serve as sensors for bitter compounds and, as they are expressed in numerous extraoral tissues throughout the body, may also be responsible for some physiological effects exerted by bitter compounds. Chinese herbal medicine use

Berberine inhibits inflammatory bone resorption via bitter taste receptors.

BACKGROUND: Extra-gustatory bitter taste receptors (Tas2rs) have been identified as critical regulators of host inflammation and immune responses. Berberine, a bitter natural alkaloid, has demonstrated effectiveness against bone resorption. However, the potential involvement of Tas2rs-mediated signaling in the anti-resorptive effects of berberine remains to be investigated. PURPOSE: This study aims to elucidate the mechanisms of action of berberine in mitigating inflammatory bone resorption through Tas2rs-mediated pathways. MATERIALS AND METHODS: The expression of bitter taste receptors in m

Pleiotropic Effects of Bitter Taste Receptors on [Ca2+]i Mobilization, Hyperpolarization, and Relaxation of Human Airway Smooth Muscle Cells.

Asthma is characterized by airway inflammation and airflow obstruction from human airway smooth muscle (HASM) constriction due to increased local bronchoconstrictive substances. We have recently found bitter taste receptors (TAS2Rs) on HASM, which increase [Ca2+]i and relax the muscle. We report here that some, but not all, TAS2R agonists decrease [Ca2+]i and relax HASM contracted by G-protein coupled receptors (GPCRs) that stimulate [Ca2+]i. This suggests both a second pathway by which TAS2Rs relax, and, a heterogeneity of the response phenotype. We utilized eight TAS2R agonists and five proc

In Vivo Biofilm Formation, Gram-Negative Infections and TAS2R38 Polymorphisms in CRSw NP Patients.

OBJECTIVES: Among the predisposing factors implicated in the immune response to airway bacterial infections, genetic variations of the bitter taste receptor TAS2R38, which is expressed in the cilia of the human sinonasal epithelial cells, seem to be associated with susceptibility to chronic rhinosinusitis (CRS) and in vitro biofilm formation. Polymorphisms in TAS2R38 generate two common haplotypes: the nonfunctional AVI (Alanine, Valine, Isoleucine) and the functional PAV (Proline, Alanine, Valine) alleles, with the latter protecting against gram-negative sinonasal infections. The aim of this

Taste receptors of the gut: emerging roles in health and disease.

Recent progress in unravelling the nutrient-sensing mechanisms in the taste buds of the tongue has triggered studies on the existence and role of chemosensory cells in the gut. Indeed, the gastrointestinal tract is the key interface between food and the human body and can sense basic tastes in much the same way as the tongue, through the use of similar G-protein-coupled taste receptors. These receptors 'taste' the luminal content and transmit signals that regulate nutrient transporter expression and nutrient uptake, and also the release of gut hormones and neurotransmitters involved in the reg

A biologically informed framework for instrument selection in dietary Mendelian randomization using chemosensory genetics.

BACKGROUND: Mendelian randomization (MR) is increasingly used to strengthen causal inference in nutritional epidemiology. However, dietary MR studies often rely on instruments selected from genome-wide association studies of self-reported intake based solely on statistical significance, increasing vulnerability to pleiotropy and reverse causation and potentially violating key MR assumptions. We aimed to develop and evaluate a biologically informed framework for selecting valid genetic instruments for dietary exposures, leveraging genes encoding taste and olfactory receptors that influence chem

Exploring the Role of TAS2Rs in Obesity: Insights From Human and Rat Models.

OBJECTIVE: Obesity is a chronic disease with various causes and diverse treatments, including dietary restriction and bariatric surgery. Bitter taste receptors (TAS2Rs), found in the oral cavity and gastrointestinal tract (GIT), are implicated in digestive and metabolic regulation, but their role in obesity remains unclear. This study investigates intestinal TAS2R expression concerning obesity and bariatric surgery outcomes. METHODS: TASas2r expression was assessed in intestinal segments of female rats fed a cafeteria diet for 17 weeks. Additionally, jejunal TAS2R expression was measured in w

The human bitter taste receptor T2R38 is broadly tuned for bacterial compounds.

T2R38 has been shown to be a specific bacterial detector implicated in innate immune defense mechanism of human upper airway. Several clinical studies have demonstrated that this receptor is associated with the development of chronic rhinosinusitis (CRS). T2R38 was previously reported to bind to homoserine lactones (HSL), quorum sensing molecules specific of Pseudomonas Aeruginosa and other gram negative species. Nevertheless, these bacteria are not the major pathogens found in CRS. Here we report on the identification of bacterial metabolites acting as new agonists of T2R38 based on a single

Sweet Taste Adaptation to Sugars, Sucralose, and Their Blends: A Human and Rodent Perspective.

Background: Sweet taste adaptation, the decline in perceived sweetness with repeated exposure, may influence dietary behavior and differs across sweeteners. Low-calorie sweeteners (LCSs) such as sucralose strongly activate the T1R2+T1R3 receptor and are generally associated with greater adaptation than sugars, although this effect can be reduced with sweetener blends. Aim: We investigated whether habitual LCS consumption affects sweet taste perception and whether blending sucralose with small amounts of sugars attenuates adaptation using sensory tests in humans and in vivo calcium imaging in a

The expression of bitter taste receptors in mesenteric, cerebral and omental arteries.

AIM: Bitter taste is sensed by the bitter taste receptor (TAS2R), which is mainly expressed in the tongue as well as in extra-oral organs, such as the gastrointestinal tract, respiratory tract, brain, heart and testis. This study aimed to investigate whether TAS2R is expressed in the mesenteric, cerebral and omental arteries. MAIN METHODS: The expression levels of TAS2R mRNA and protein were determined by reverse-transcription polymerase chain reaction and Western blotting, respectively. The location of TAS2R was determined by immunofluorescence imaging. TAS2R agonists were used in a sensitiv

Cigarette Smoke Exposure Attenuates T2R-Mediated Apoptosis in Head and Neck Squamous Cell Carcinoma.

BACKGROUND: Tobacco use is associated with worse outcomes in head and neck squamous cell carcinoma (HNSCC). Bitter taste receptor (T2R) activation induces apoptosis via calcium-dependent signaling, and higher T2R expression correlates with increased survival in HNSCC. However, the effects of cigarette smoke on T2R signaling remain unclear. METHODS: HNSCC cell lines were treated with cigarette smoke condensate (CSC). T2R expression was measured by qPCR, and function evaluated by calcium imaging, viability, and apoptosis assays following stimulation with denatonium benzoate and flufenamic acid.

A targeted genotyping approach enhances identification of variants in taste receptor and appetite/reward genes of potential functional importance for obesity-related porcine traits.

Taste receptors (TASRs) and appetite and reward (AR) mechanisms influence eating behaviour, which in turn affects food intake and risk of obesity. In a previous study, we used next generation sequencing to identify potentially functional mutations in TASR and AR genes and found indications for genetic associations between identified variants and growth and fat deposition in a subgroup of animals (n = 38) from the UNIK resource pig population. This population was created for studying obesity and obesity-related diseases. In the present study we validated results from our previous study by inves

An alternative laboratory designed to address ethical concerns associated with traditional TAS2R38 student genotyping.

The TAS2R38 alleles that code for the PAV/AVI T2R38 proteins have long been viewed as benign taste receptor variants. However, recent studies have demonstrated an expanding and medically relevant role for TAS2R38. The AVI variant of T2R38 is associated with an increased risk of both colorectal cancer and Pseudomonas aeruginosa-associated sinus infection and T2R38 variants have been implicated in off-target drug responses. To address ethical concerns associated with continued student TAS2R38 gene testing, we developed an alternative to the traditional laboratory genotyping exercise. Instead of

The bitter flavor of Banxia Xiexin decoction activates TAS2R38 to ameliorate low-grade inflammation in the duodenum of mice with functional dyspepsia.

ETHNOPHARMACOLOGICAL RELEVANCE: Banxia Xiexin Decoction (BXD) is a traditional herbal formulation with a bitter flavor that has a long-standing history of use in Asia for treating functional dyspepsia (FD). In traditional Chinese medicine, the bitter flavor is believed to play a critical role in the therapeutic activity of BXD. The ethnopharmacological properties of bitter plant extracts are closely associated with their anti-inflammatory effects, which may contribute to their efficacy in FD. However, the specific mechanisms remain unknown. AIM OF THE STUDY: The objective of this study is to

Corpuscular bodies in the palate of the rat. 1. Morphology and distribution.

The morphology and the frequency distribution of corpuscular bodies in the palate was studied in rats using light and electron microscopy. The study extended from the day of birth through 180 days after birth. The corpuscles appeared as round (diameter 35-50 microns) to ovoid (width 40-60 microns, height 60-90 microns) bodies intercalated within the epithelium of the incisal papilla and the soft palate. A subepithelial nerve plexus surrounded the base of a corpuscle. For types of cells were identified within the corpuscle: basal, type I (dark), type II (pale) and type III cells. Some cells bor
