HTLV-1 Tax: Linking transformation, DNA damage and apoptotic T-cell death.

The human T-cell leukemia virus type I (HTLV-1) is the causative agent of adult T-cell leukemia (ATL), an aggressive CD4-positive T-cell neoplasia. The HTLV-1 proto-oncogene Tax, a potent transcriptional activator of cellular and viral genes, is thought to play a pivotal role in the transforming properties of the virus by deregulating intracellular signaling pathways. During the course of HTLV-1 infection, the dysregulation of cell-cycle checkpoints and the suppression of DNA damage repair is tightly linked to the activity of the viral oncoprotein Tax. Tax activity is associated with productio

Human T Lymphotropic Virus Type 1 protein Tax reduces histone levels.

BACKGROUND: Human T-Lymphotropic Virus Type-1 (HTLV-1) is an oncogenic retrovirus that causes adult T-cell leukemia/lymphoma (ATLL). The virally encoded Tax protein is thought to be necessary and sufficient for T-cell leukemogenesis. Tax promotes inappropriate cellular proliferation, represses multiple DNA repair mechanisms, deregulates cell cycle checkpoints, and induces genomic instability. All of these Tax effects are thought to cooperate in the development of ATLL. RESULTS: In this study, we demonstrate that histone protein levels are reduced in HTLV-1 infected T-cell lines (HuT102, SLB-1

Muscle wasting induced by HTLV-1 tax-1 protein: an in vitro and in vivo study.

Besides tropical spastic paraparesis/human T-cell leukemia virus type-1 (HTLV-1)-associated myelopathy, the human retrovirus HTLV-1 causes inflammatory disorders such as myositis. Although the pathogenesis of HTLV-1-associated myositis is primarily unknown, a direct effect of cytokines or viral proteins in myocytotoxicity is suspected. We have developed an in vitro cell culture model to study the interactions between primary human muscle cells and HTLV-1 chronically infected cells. When HTLV-1-infected cell lines were added to differentiated muscle cultures, cytopathic changes such as fiber sh

Role of Tax protein in human T-cell leukemia virus type-I leukemogenicity.

HTLV-1 is the etiological agent of adult T-cell leukemia (ATL), the neurological syndrome TSP/HAM and certain other clinical disorders. The viral Tax protein is considered to play a central role in the process leading to ATL. Tax modulates the expression of many viral and cellular genes through the CREB/ATF-, SRF- and NF-kappaB-associated pathways. In addition, Tax employs the CBP/p300 and p/CAF co-activators for implementing the full transcriptional activation competence of each of these pathways. Tax also affects the function of various other regulatory proteins by direct protein-protein int

HTLV-1 proviruses encoding non-functional TAX in adult T-cell leukemia.

Adult T-cell leukemia (ATL) is associated with prior infection with human T-cell leukemia virus type 1 (HTLV-1). TAX, the major transactivator of HTLV-1, has been implicated in the immortalization of infected T-cells, but molecular mechanisms of in vivo malignant cell transformation induced by HTLV-1 remain unclear. To investigate the role of TAX in the monoclonal proliferation of ATL cells, we determined the nucleotide sequence of tax DNA clones obtained from 6 ATL patients and analysed the biological function of their products. We found that ATL cells from 2 of these patients possessed tax w

Expression of HTLV-I Env and Tax recombinant peptides in yeast: identification of immunogenic domains.

A peptide library of HTLV-I Env and Tax proteins was constructed in yeast in order to characterize which domains of these proteins are immunogenic in HTLV-I-infected individuals. Five yeast colonies (A to E) were selected using HTLV-I positive plasma, and one yeast colony (F) was selected using rabbit anti-Tax sera. Plasmid DNA present in each positive clone was recovered and sequenced. Overlapping clones A to E covered the C-terminal part of the gp46 exterior glycoprotein (aa 197 to 305) and were all glycosylated. Clone F encoded the C-terminal 25 aa of Tax (aa 329-353). Recombinant peptides

Examining the effectiveness of dissuasive taxes as a policy tool for reducing tobacco and alcohol consumption in Cameroon: A welfare and microsimulation analysis.

Deterrent taxes are a crucial policy tool for reducing the consumption of harmful products like tobacco and alcohol. However, assessing dissuasive taxes impact different income groups is important to ensure that their burden is not disproportionately borne by low-income households. This study examines the effectiveness of deterrent taxes as an economic policy tool for reducing tobacco and alcohol consumption in Cameroon. We analyse the impact on household welfare and distributional effects using microsimulation analysis. The data come from the Cameroon Household Living Conditions Survey and th

Association of cigarette excise taxes and clean indoor air laws with change in smoking behavior in the United States: a Markov modeling analysis.

The rates of cigarette smoking in the United States have declined over the past few decades in parallel with increases in cigarette taxes and introduction of more stringent clean indoor air laws. Few longitudinal studies have examined association of taxes and clean indoor air policies with change in smoking nationally. This study examined the association of state and local cigarette taxes and clean indoor laws with change in smoking status of 18,499 adult participants of the longitudinal 2010-2011 Tobacco Use Supplement of the Current Population Survey over a period of 1 year. Every $1 increas

Preparation of soluble and functional recombinant HTLV-1 Tax protein using bacterial chaperones.

HTLV-1 viral protein Tax physically interacts with multiple host proteins to disrupt their normal functions, causing debilitating haematological malignancy and inflammatory disorders. The recombinant Tax protein, like other intrinsically disordered proteins, is insoluble when expressed in Escherichia coli due to the inclusion body formation, hampering structural studies of this important viral protein. We report that the bacterial chaperone Trigger Factor significantly enhances the solubility of HTLV-1 Tax fused to maltose-binding protein (MBP) as a solubility tag. Using a hexahistidine tag in

Splicing-dependent restriction of the HBZ gene by Tax underlies biphasic HTLV-1 infection.

HTLV-1 is an oncovirus that encodes a transactivator Tax and a regulatory gene HBZ. HTLV-1 early or infectious replication depends on Tax; during HTLV-1 late infection, HBZ plays a crucial role in driving the proliferation of infected cells and maintaining viral persistence. The biphasic replication pattern of HTLV-1 dictated by Tax and HBZ represents a result of viral host adaptation, but how HTLV-1 coordinates Tax and HBZ expression to facilitate early and late infection remains elusive. Here we reveal that HBZ RNA splicing exhibits distinct patterns in Tax+ and Tax- HTLV-1 infected cells. W

Intensity Matters: Heterogeneous impact of cigarette tax reform on drinking behaviors by smoking intensity.

This paper investigates the relationship between smoking and drinking behaviors across different smoking intensity groups, using a unique policy shock that increased cigarette taxes in South Korea. Using the Korean Health Panel, we employ a difference-in-differences approach to estimate the causal effect of the cigarette price increase on alcohol consumption among heavy, average, and light smokers. We find that an increase in cigarette prices reduces drinking behavior only among heavy smokers, while cigarette consumption is reduced across all smokers. Subgroup analysis shows that the reduction

Analyzing the impact of export tax rebates and energy conservation on sustainable industrial growth in China.

When China implemented the exports tax rebate policy through administrative framework in 1985, industrial energy consumption increased by more than five times. The purpose of this study is to examine the relationship between industrial energy demand (IED), exports tax rebate, exports, and the value of industrial output in the presence of other variables such as GDP, taking into account the effects of carbon dioxide (CO2) and foreign direct investment (FDI). The present study adopted the Autoregressive-Distributed Lag (ARDL) model and Granger Casuality analysis of the VECM to examine the short

YTHDF1 and YTHDC1 m6A reader proteins regulate HTLV-1 tax and hbz activity.

Human T-cell leukemia virus type 1 (HTLV-1) is a retrovirus responsible for adult T-cell leukemia/lymphoma (ATLL) and HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP), a progressive neurodegenerative disease. Regulation of viral gene expression plays a key role in viral persistence and pathogenesis. However, the molecular mechanisms underlying this fine-tuned regulation remain poorly understood. Little is known regarding RNA chemical modifications of HTLV-1 RNA and how these affect viral biology and disease development. Post-transcriptional chemical modification of RNA is co

A high-resolution photoelectron spectroscopic and computational study of TaX- (X = C, N, O).

We report on the high-resolution photoelectron spectroscopy of diatomic molecules TaX- (X = C, N, O) anions using a cryogenic ion trap combined with the slow electron velocity imaging (cryo-SEVI) method. We determined the electron affinities of TaC, TaN, and TaO to be 2.098(2), 1.576(2), and 1.069(2) eV, respectively. In addition, the electron affinities of TaX molecules were calculated using the CCSD(T) method with the complete basis sets. To interpret the complicated photoelectron spectra, we predicted the excited states of TaX molecules using the MRCI + Q method, accounting for the spin-orb
