A species sensitivity distribution-derived generic dose for the phytosanitary irradiation of Pseudococcidae.

Phytosanitary irradiation is an attractive treatment option for fresh fruits and vegetables, as it is effective, leaves no chemical residues, and in most cases results in fewer impacts to commodity quality than other treatment types such as heat-based treatments or methyl bromide fumigation. The dose of irradiation needed to mitigate a specific pest species is determined on the basis of a substantial body of research, reviewed, and approved by a national plant protection organization. Generic doses, used to treat several related species under a single dose, have been developed and approved for

An improved quarantine method for mangoes against the Mexican fruit fly based on high-pressure processing combined with heat.

The Mexican fruit fly Anastrepha ludens Loew (Diptera: Tephritidae) is one of the most important insects infesting mangoes, citrus, and other fruits in Mexico and other Latin-American countries. Quarantine methods approved to destroy this insect decrease the shelf life of commodities. The objective of this study was to determine the effect of high-pressure processing using an initial temperature of 50 degrees C on the survivorship of eggs and larvae of the Mexican fruit fly. Eggs and larvae were pressurized at 25, 50, 75, 100, or 150 MPa for 0, 5, 10, or 20 min. The hatching ability of pressur

A Comparative Transcriptomic Analysis of X-Ray Irradiation Responses Identifies Candidate Biomarker Genes in Bactrocera dorsalis Larvae.

Bactrocera dorsalis (Diptera: Tephritidae) ranks among the most serious quarantine pests threatening global fruit and crop production. Irradiation has been adopted worldwide as an eco-friendly phytosanitary treatment strategy, yet how B. dorsalis responds to irradiation is not fully understood. To investigate the transcriptional response to irradiation, third-instar B. dorsalis larvae were exposed to two X-ray doses: a sub-effective dose of 15 Gy and a phytosanitary dose of 120 Gy. Comparative transcriptomic profiling revealed a clear dose-dependent escalation in the number of differentially e

Resolution of inter and intra-species relationships of the West Indian fruit fly Anastrepha obliqua.

The West Indian fruit fly, Anastrepha obliqua (Diptera: Tephritidae), is an economically important pest that inhabits areas of South and Central America, Mexico and the Caribbean with occasional infestations in the southern United States. We examine intra-specific relationships within A. obliqua as well as interspecific relationships to other Anastrepha species using a multi-locus data set comprising nine loci (seven nuclear, two mitochondrial) with 105 operational taxonomic units. The results based on a concatenated set of nuclear loci strongly support the monophyly of A. obliqua and most of

Endosymbiont-derived microbial volatiles mediate conspecific female attraction in the melon fruit fly Zeugodacus cucurbitae.

Microbial symbionts associated with phytophagous insects are increasingly recognised as key mediators of chemical communication and behaviour. In this study, we investigated the taxonomic diversity and attractancy potential of culturable bacterial symbionts associated with the melon fly, Zeugodacus cucurbitae (Diptera: Tephritidae), with the aim of identifying behaviourally active microbial volatiles with potential application in pest management. Bacteria were isolated from the gut, reproductive organs, and rectal glands of female melon flies from wild and laboratory-reared populations and ide

Genome identification of C-type lectins in Bactrocera dorsalis and functional characterization of BdCTL-S12 with a broad-spectrum pathogen recognition.

BACKGROUND: Bactrocera dorsalis (Diptera: Tephritidae) is a highly adaptable invasive pest that causes significant economic losses. Long-term chemical control strategies have led to high levels of insecticide resistance and severe environmental risks. C-type lectins (CTLs), key pattern recognition receptors in insect innate immunity, are named for their calcium-dependent carbohydrate recognition domains (CRDs) and they play a critical role in detecting and binding to foreign pathogens. This study aims to identify B. dorsalis CTLs (BdCTLs) and investigate their roles in pathogen defense. RESUL

Methoprene treatment and its effect on male reproductive organ size and female remating in a fruit fly.

Methoprene, a juvenile hormone analog, is used to accelerate sexual maturation in males of species of economic importance in support to the sterile insect technique (SIT). In the SIT, mass-reared sterile males are released into the field and need to survive until they reach sexual maturation, find a wild female, mate with her and then induce female sexual refractoriness, so she will not remate with a wild counterpart. The use of methoprene shortens the time between release and copulation. However, in South American fruit flies, Anastrepha fraterculus, the ability of methoprene-treated males to

The Sterile Insect Technique: Success and Perspectives in the Neotropics.

The sterile insect technique (SIT), an environmentally friendly means of control, is currently used against plant, animal, and human pests under the area-wide integrated pest management. It consists in the mass production, sterilization, and release of insects in an affected area where sterile males mate with wild females leading to no reproduction. Here, we review SIT in the Neotropics and focus on particular recent successful cases of eradication of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann), as well as effective programs used against the Mexican fruit fly Anastrepha ludens

Potential for high hydrostatic pressure processing to control quarantine insects in fruit.

Tests were conducted to determine the potential for high hydrostatic pressure (HPP) to control codling moth, Cydia pomonella (L.), and western cherry fruit fly, Rhagoletis indifferens Curran. Apples (Malus spp.) with codling moth larvae or eggs were treated at 24 and 72 h, respectively, after infestation at a series of pressures between 14,000 and 26,000 pounds per inch2 (psi). Survivorship was determined the next day for larvae and after 10 d for eggs. Codling moth eggs were more tolerant of HPP treatment than larvae. Mortality of larvae was 97% at 22,000 psi, whereas mortality of eggs at thi

Nanomaterial-encapsulated dsRNA of ecdysone-induced early gene E75, a potential RNAi-based SIT strategy for pest control against Bactrocera dorsalis.

Bactrocera dorsalis is a notorious pest widely distributed across most Asian countries. With the rapid development of pesticide resistance, new pest control methods are urgently needed. RNAi-based sterile insect technique (SIT) is a species-specific pest management strategy for B. dorsalis control. It is of great significance to identify more target genes from B. dorsalis, and improve the RNAi efficiency. In this study, microinjection-based RNAi results showed that six 20E response genes were necessary for male fertility of B. dorsalis, of which E75 was identified as the key target according t

Sampling technique biases in the analysis of fruit fly volatiles: a case study of Queensland fruit fly.

Diverse methods have been used to sample insect semiochemicals. Sampling methods can differ in efficiency and affinity and this can introduce significant biases when interpreting biological patterns. We compare common methods used to sample tephritid fruit fly rectal gland volatiles ('pheromones'), focusing on Queensland fruit fly, Bactrocera tryoni. Solvents of different polarity, n-hexane, dichloromethane and ethanol, were compared using intact and crushed glands. Polydimethylsiloxane, polydimethylsiloxane/divinylbenzene and polyacrylate were compared as adsorbents for solid phase microextra

miR-318 regulates juvenile hormone biosynthesis to control ovarian development in Bactrocera dorsalis.

Reproduction is a fundamental process in the insect life cycle, with the ovary serving as the central organ governing oogenesis and fertility. Hormonal homeostasis, particularly juvenile hormone (JH) regulation, is critical for proper ovarian development. Bactrocera dorsalis (Hendel) is a globally invasive agricultural insect pest characterized by high reproductive capacity. MicroRNAs (miRNAs) are key post-transcriptional regulators of diverse biological processes, yet their roles in insect ovarian development remain incompletely understood. Here, we identify miR-318 as an ovary-enriched miRNA

Multiwavelength Optical Rotation Detection: An Effective Approach for the Recognition of Analytical and Semi-Preparative HPLC Enantioseparation of the Chiral Pheromone Olean and Its Stereochemical Characterization.

Olean is the chiral spiroacetal sex pheromone of the female olive fruit fly Bactrocera oleae. Laboratory tests have demonstrated that the (R)-(-)-olean enantiomer is active on males, whereas females respond to (S)-(+)-olean. Here we present the first HPLC enantioseparation of olean using polysaccharide derivatives as chiral stationary phases and a polarimetric detector equipped with a micro-flow cell capable of detecting optical rotation at six different wavelengths. Unlike blind UV detection, the polarimetric detector allows the chiral spiroacetal, which lacks a chromophore, to be detected as

CYP4G100 contributes to desiccation resistance by mediating cuticular hydrocarbon synthesis in Bactrocera dorsalis.

The oriental fruit fly Bactrocera dorsalis (Hendel) is expanding its distribution to higher latitudes. Our goal in this study was to understand how B. dorsalis adapts to higher latitude environments that are more arid than tropical regions. Cuticular hydrocarbons (CHCs) on the surface of the epicuticle in insects act as a hydrophobic barrier against water loss. The essential decarbonylation reaction in CHC synthesis is catalysed by CYP4G, a cytochrome P450 subfamily protein. Hence, in B. dorsalis it is necessary to clarify the function of the CYP4G gene and its role in desiccation resistance.

Behavioral evidence for fruit odor discrimination and sympatric host races of Rhagoletis pomonella flies in the Western United States.

The recent shift of Rhagoletis pomonella (Diptera: Tephritidae) from its native host downy hawthorn, Crataegus mollis, to introduced domesticated apple, Malus domestica, in the eastern United States is a model for sympatric host race formation. However, the fly is also present in the western United States, where it may have been introduced via infested apples within the last 60 years. In addition to apple, R. pomonella also infests two hawthorns in the West, one the native black hawthorn, C. douglasii, and the other the introduced English ornamental hawthorn, C. monogyna. Here, we test for beh

The relative importance of resources and natural enemies in determining herbivore abundance: thistles, tephritids and parasitoids.

1. The relative importance of host-plant resources and natural enemies in influencing the abundance of insect herbivores was investigated in potted plant and natural population experiments, using tephritid (Diptera: Tephritidae) flies, their host plant, creeping thistle Cirsium arvense, and their Hymenoptera parasitoids. 2. Experimental manipulation of host-plant quality (i.e. levels of host-plant nutrients) and resource availability (i.e. the number of buds) increased tephritid abundance. There was no evidence that the seed-feeding tephritid fly Xyphosia miliaria preferentially oviposited on

Effects of DNA 6mA methylation on female reproduction in Bactrocera dorsalis.

BACKGROUND: DNA 6mA methylation is a critical epigenetic modification that significantly regulates a diverse range of biological processes. Nevertheless, the molecular mechanisms underlying its regulation of the reproductive development of insects remain largely unknown. Here, we demonstrate the molecular mechanism by which DNA 6mA methylation regulates female reproduction in Bactrocera dorsalis. RESULTS: RNA interference-mediated knockdown of BdJumu, which binds to 6mA-labeled DNA, significantly decreases the level of DNA 6mA methylation in the ovary of B. dorsalis. Moreover, compared with f

Identification and functional verification of Y-chromosome-specific gene typo-gyf in Bactrocera dorsalis.

Genes on the Y chromosome play important roles in male sex determination and development. The identification of Y-chromosome-specific genes not only provides a theoretical basis for the study of male reproductive development, but also offers genetic control targets for agricultural pests. However, Y-chromosome genes are rarely characterized due to their high repeatability and high heterochromatinization, especially in the oriental fruit fly. In this study, 1 011 Y-chromosome-specific candidate sequences were screened from 2 to 4 h Bactrocera dorsalis embryo datasets with the chromosome quotien

Arylalkylamine N-acetyltransferase 1 gene (AANAT1) regulates cuticle pigmentation and ovary development of the adult oriental fruit fly, Bactrocera dorsalis.

The arylalkylamine N-acetyltransferase (AANAT) enzymes catalyze the acetyl-CoA-dependent acetylation of an amine or arylalkylamine, which is involved in important biological processes of insects. Here, we carried out the molecular and biochemical identification of an arylalkylamine N-acetyltransferase (AANAT) from the oriental fruit fly, Bactrocera dorsalis. Using a bacterial expression system, we expressed and purified the encoded recombinant BdorAANAT1-V3 protein. The purified recombinant protein acts on a wide range of substrates, including dopamine, tyramine, octopamine, serotonin, methoxytry

Chlorination Is Ineffective at Eliminating Insects from Wastewater: A Case Study Using Ceratitis capitata.

Eradicating insects from small bodies of water without resorting to environmentally harmful methods is a challenge worldwide. Among the practices used to treat the water waste of arthropod containment facilities, chlorination is still used, despite the known impacts of chlorine byproducts on the environment and limited scientific literature documenting its potency against arthropods. Here, we evaluated the efficacy of chlorination as a means to kill insects in water by immersing eggs of the Mediterranean fruit fly Ceratitis capitata (Diptera: Tephritidae) in 10 solutions containing different c
